A Pumilio Activity Sensor Reveals Bag-of-Marbles Inhibition of Pum Activity in the Drosophila Ovary

  • Pumilio (Pum) is an RNA-binding protein and translational repressor important to diverse biological processes. In the Drosophila ovary, Pum is expressed in female germline stem cells (GSCs), wherein it acts as an intrinsic stem cell maintenance factor via repressing target mRNAs that are as yet mostly unknown. Pum recognizes the Pum binding sequence (PBS) in the mRNA 3'UTR through its C-terminus Puf domain. Translational repression is mediated through its N-terminal domain, but the molecular mechanism remains largely unknown. We previously showed that Bag-of-marbles, a critical differentiation-promoting factor of female GSCs, physically interacts with the N-terminus of Pum. We further showed that this interaction is critical to Bam inhibition of Pum repressive action in cultured cells, but the physiological relevance was not addressed. Here we design an in vivo GFP translational reporter bearing the PBS in its 3'UTR and show that GFP expression is reduced in cells wherein Pum is known to be active. Furthermore, we demonstrate in pum mutant ovary that this GFP repression requires Pum, and also that the sensor faithfully monitors Pum activity. Finally, we show that forced expression of Bam inhibits Pum-mediated repression, validating that Bam inhibits Pum activity in vivo.

Design of a Displacement and Velocity Measurement System Based on Environmental Characteristic Analysis of Laser Sensors for Automatic Mooring Devices (레이저 센서의 환경적 특성 분석에 기반한 선박 자동계류장치용 변위 및 속도 측정시스템 설계)

  • To prevent accidents near the quay caused by a ship, ports are generally designed and constructed through navigation and berthing safety assessment. However, unpredictable accidents such as ship collisions with the quay or personal accidents caused by ropes still occur sometimes during the ship berthing or mooring process. Automatic mooring systems, which are equipped with an attachment mechanism composed of robotic manipulators and vacuum pads, are designed for rapid and safe mooring of ships. This paper deals with a displacement and velocity measurement system for the automatic mooring device, which is essential for the position and speed control of the vacuum pads. To design a suitable system for an automatic mooring device, we first analyze the sensor's performance and outdoor environmental characteristics. Based on the analysis results, we describe the configuration and design methods of a displacement and velocity measurement system for application in outdoor environments. Additionally, several algorithms for detecting the sensor's state and estimating a ship's velocity are developed. The proposed method is verified through some experiments for displacement and speed measurement targeted at a moving object with constant speed.

Cutting force estimation using spindle and feeddrive motor currents in milling processes (밀링공정에서 이송모터와 주축모터의 전류신호를 이용한 절삭력 추정)

  • Advanced sensor design and filtering technology have been studied to obtain information for condition monitoring and diagnostics inmachining processes. To develope and economic monitoring system in end milling processes, indirect and reliable type of cutting force estimators were required. In this paper, an estimation method of cutting forces during end milling processes was studied through the measurement of current signals obtained from spindle and feeddrive motors. Cutting force and torque models were derived from the cutting geometry in down milling processes. Relationships between motor currents and cutting forces were also developed in the form of AC and DC components from the developed force models. The validity of the cutting force estimator was confirmed by the experiments under various cutting conditions.

  • Pneumatic servo valve is an electro-mechanical device which change electric signals to a proper pneumatic signals, that is, flowrate and pressure. In this study, a pneumatic servo valve was designed and each simulation was conducted on any variation in the flowrate depending upon the magnetic force of the linear force motor and the displacement of the spool. And permanent magnet was used as a material for the plunger of the servo valve. Thereby, a low electrical power consumption type coil was desinged. And a modeling for the coil design was conducted by using the magnetic circuit. also, the feasibility of the modeling was verified by using a commercial magnetic field analysis program. The designed and fabrication of the spool and sleeve, position sensor, servo controller and the dynamic characteristic verified by the experiment.

  • COMS Satellite is automatically able to recover from any defined failure thanks to a full redundancy. This study assesses the effects of gyro failure on the COMS mission and analyzes the mechanism of Gyro Failure Detection, Isolation and Recovery about failure detection means, isolation and recovery actions and their consequences. At last, it checks the FDIR behavior from an injected failure on COMS simulator.

  • In this study, automatic assembly and evaluation system fer phone camera module is conceptually designed. The designed core(Auto focus & UV curing, Image Test) equipments adopts a clustering mechanism and compactible structure using index table for minimum tact time. Using a ball screw actuator and absolute encoder in each axis, we can verifies the repeatability and position accuracy of system within ${\pm}3{\mu}m$. In result of simulation test, the proposed system is expected up to 30% in productivity than manual operation.

  • Sensitivity analysis of the cylindrical zigzag cams under acceleration and centrifugal forces is performed. A Lagrangian method is used to determine the mechanism constant of zigzag track, And the equation of motion for cylindrical zigzag cam under rectangular pulse is derived by the governing equations of a single spring mass system. The ratio of the drive force tn resisting force is derived by angular acceleration, centrifugal force and setback force on the operation of the munition. The theoretical sensitivity curves for 3 models are analyzed. And experiments for 3 models are conducted to check safe and functional zone. Zigzag cam types can be satisfied all major design requirements for switch system of munition.

  • Energy savings and environmental protection policies have been the general trend in the engine design. The friction power loss associated with the cam mechanism has become important. But it is difficult to measure the torque of rotating valvetrain in real engine environment because most of conventional torque meters are axial type. The objective of this paper is to develop new equipment which can be installed in a rotating camshaft. It uses strain gages to measure the elastic deformation of torque sensor which replaces the cam sprocket. It includes telemetry to transmit torque data via Bluetooth and induction power system to provide adequate power to rotating torque meter. The developed torque meter has good linearity and thermostability. It was installed in a real engine, and successfully measured the valvetrain torque.

  • A new small humanoid robot system is developed in this paper. The humanoid robot has total 20 DOFs : 6 DOFs in each legs, 3 DOFs in each arms, and 2 DOFs in head, 34cms in height, and 2kgs in weight. The robot has the following characteristics: (1) PDA as host controller (2) network-based joint controller (3) wireless camera attached in robot's head (4) mechanism design by CATIA and high speed laser prototyping (5) graphic MMI(Man-Machine Interface) utilizing the CATIA data. By using ADXL inclination sensor, we implement the rope swing with the robot leg motion as well as walking.

  • The security of wireless sensor networks is a challenging research area where the resources constraints are a bottleneck for any successful security design. Due to their computational feasibility, symmetric key algorithms that require key pre-distribution are more desirable for use in these networks. In the pre-distribution scheme, keys or keying materials are assigned to each node prior deployment to guarantee a secure communication within the entire network. Though several works are introduced on this issue, yet the connectivity and resiliency are imperfectly handled. In this paper, we revisit the grid based key pre-distribution scheme aiming to improve the connectivity, introduce a higher resiliency level, simplify the logic of key establishment and maintain same level of used of resources usage. The core of our modification relies on introducing the novel plat-based polynomial assignment and key establishment mechanism. To demonstrate the advantageous properties of our scheme over the revisited one, details of consumed resources, resulting connectivity, security and comparisons with relevant works are introduced.
