The filter elution technique was used to assay $^{60}Co$$\gamma$ ray-induced DNA strand breaks(SB) in EL4 mouse leukemia cell and mouse spleen lymphocyte. The lymphocytes were stimulated with lipopolysaccharide (LPS, $20{\mu}g/ml$) to label $[^3H]$ thymidine. EL4 cells and lymphocytes in suspension were exposed at $0^{\circ}C$ to 0Gy, 1Gy, 5Gy, 10Gy or l5Gy for DNA single strand breaks(SSB) assay and 0Gy, 25Gy, 50Gy, 75Gy or 100Gy for DNA double strand breaks(DSB) assay of $^{60}Co$ radiation and elution procedure was performed at pH12.1 and 9.6. The number of DNA strand breaks increased with increasing doses of r rays. The strand scission factor(SSF) was estimated in each experiment (eluted volume 21ml). The slope of SSB EL4 cells was $0.01301{\pm}0.00096Gy^{-1}$ (n=5), the slope of SSB for lymphocytes was $0.01097{\pm}0.00091Gy^{-1}$ (n=5) and the slope of DSB for lymphocytes was $0.001707{\pm}0.0000573Gy^{-1}$ (n=5). Thus EL4 cells were more sensitive to induction of DNA SSB by ionizing radiation than lymphocytes (p<0.005). The ratio of slope of dose-response relationship (SSF versus dose) of lymphocytes DNA SSB as compared with the slope of DNA DSB was 6.4.

In recent years, monitoring of spent nuclear fuel inside dry cask storage has become an important area of national security. Muon tomography is a useful method for monitoring spent nuclear fuel because it uses high energy muons that penetrate deep into the target material and provides a 3-D structure of the inner materials. We designed a muon tomography system consisting of four 2-D position sensitive detector and characterized and optimized the system parameters. Each detector, measuring 200 × 200 cm2, consists of a plastic scintillator, wavelength shifting (WLS) fibers and, SiPMs. The reconstructed image is obtained by extracting the intersection of the incoming and outgoing muon tracks using a Point-of-Closest-Approach (PoCA) algorithm. The Geant4 simulation was used to evaluate the performance of the muon tomography system and to optimize the design parameters including the pixel size of the muon detector, the field of view (FOV), and the distance between detectors. Based on the optimized design parameters, the spent fuel assemblies were modeled and the line profile was analyzed to conduct a feasibility study. Line profile analysis confirmed that muon tomography system can monitor nuclear spent fuel in dry storage container.

The objective of this investigation was to evaluate of dose-incidence relationships on the prenatal effects of gamma-radiation. Pregnant ICR mice were exposed on day 11.5 after conception, coincident with the most sensitive stage for the induction of major congenital malformations, with 0.5-4.0 Gy of gamma-radiations. The animals were sacrificed on day 18 of gestation and the fetuses were examined for mortality, growth retardation, change in head size and any other morphological abnormalities. With increasing radiation dose, incidence of small head, growth retarded fetus, cleft palate, dilatation of cerebral ventricle and abnormalities of the extremities in live fetuses rose. The threshold doses of radiation that induced cleft palate and dilatation of cerebral ventricle, and abnormal extremities were between 1.0 and 2.0 Gy, and between 0.5 and 1.0 Gy, respectively.
A multi-wire proportional counter with large sensitive area was designed and constructed considering diameter of anode wire. its material and space. A preamplifier connecting detector to main amplifier or counter was also designed and constructed for measurement output pulse from multi-wire proportional counter. To test the performance of this equipment, terminal output pulse from the preamplifier was measured and compared with noise For these tests $^{239}Pu(360 Bq)\;and\; ^{90}Sr/^{90}Y(250 Bq)$ were used as radiation sources. The noise ingredient contributing to the maximum amplitude(180mV from $^{239}Pu$ and 200 mV from $^{90}Sr/^{90}Y$) was found to be very small(8 mV) Piled up pulse occurring at the output pulse of charge-sensitive differential circuit was measured as an independent pulse since this affected the amplification in the clipping circuit and amplification circuit. This information can be used to improve the loss of measurement due to piled up pulse.

The effects of carbon-ion irradiation on cancer cell telomere function have not been comprehensively studied. In our previous report cancer cells with telomere dysfunction were more sensitive to carbon-ion irradiation, but the underlying mechanisms remained unclear. Here we found that telomerase activity was suppressed by carbon-ion irradiation via hTERT down-regulation. Inhibition of telomere activity by MST-312 further increased cancer cell radiosensitivity to carbon-ion radiation. hTERT suppression caused by either carbon-ion irradiation or MST-312 impaired mitochondrial function, as indicated by decreased membrane potential, mtDNA copy number, mitochondrial mass, total ATP levels and elevated reactive oxygen species (ROS). PGC-$1{\alpha}$ expression was repressed after carbion-ion irradiation, and hTERT inhibition by MST-312 could further exacerbate this effect. Lowering the mitochondrial ROS level by MitoTEMPO could partially counteract the induction of cellular senescence induced by carbon-ion radiation and MST-312 incubation. Taken together, the current data suggest that telomere-mitochondrion links play a role in the induction of senescence in MCF-7 cells after carbon-ion irradiation.
Radiofrequency (RF) radiation at the frequency of mobile phones has been not reported to induce cellular responses in in vitro and in vivo models. We exposed HEI-OC1, conditionally-immortalized mouse auditory cells, to RF radiation to characterize cellular responses to 1763 MHz RF radiation. While we could not detect any differences upon RF exposure, whole-genome expression profiling might provide the most sensitive method to find the molecular responses to RF radiation. HEI-OC1 cells were exposed to 1763 MHz RF radiation at an average specific absorption rate (SAR) of 20 W/kg for 24 hr and harvested after 5 hr of recovery (R5), alongside sham-exposed samples (S5). From the whole-genome profiles of mouse neurons, we selected 9 differentially-expressed genes between the S5 and R5 groups using information gain-based recursive feature elimination procedure. Based on support vector machine (SVM), we designed a prediction model using the 9 genes to discriminate the two groups. Our prediction model could predict the target class without any error. From these results, we developed a prediction model using biomarkers to determine the RF radiation exposure in mouse auditory cells with perfect accuracy, which may need validation in in vivo RF-exposure models.

Effect of finite substrate plane on the radiation characteristics of microstrip patch antennas is investigated. The variation of the characteristics of the radiation pattern due to the change of the length of a substrate is much larger than that due to the change of the width of a substrate. As the thickness of a substrate increases, the characteristics of the radiation pattern reveal more sensitive for the change of the size of a substrate.
In this study, to develop highly sensitive radiation sensors, $CaSO_4$ : Tm phosphors and its disc-type TLD elements embedded PTFE(polytetrafluoroethylene) are fabricated. The highest sensitivity of $CaSO_4$ : Tm phosphors is obtained when phosphors have been doped with 0.5mol % Tm and sintered in atmosphere at $600^{\circ}C$ for two hours. Fabricated disc-type elements are made from a homogeneous mixture of phosphors and PTFE powder. They are first cold-pressed and then polymerized at $370^{\circ}C$ in air for one hour. The dose dependence of the prepared $CaSO_4$ : Tm TLD radiation sensors is linear within the range of $100{\mu}Gy{\sim}10Gy$ for X-rays and ${\gamma}-rays$. The response of $CaSO_4$ : Tm to 30keV X-rays is ten times higher than that of 1.25MeV $^{60}Co\;{\gamma}-rays$. The fading rate of the main peak is about 2% per a month. The spectral peaks of TL emission spectrum are at about 350nm and 475nm. The $CaSO_4$ : Tm TLD radiation sensors prepared in this work may be used as radiation dosimeter for personal and environmental monitoring because of their high sensitivity and little fading.
Background: In order to measure neutron energy spectra, the conventional Bonner Sphere Spectrometers (BSS) are widely used. In this spectrometer, several measurements with different size Bonner spheres are required. Operators should, therefore, place these spheres in several times to a measurement point where radiation dose might be relatively high. In order to reduce this effort, novel neutron energy spectrometer using an onion-like single Bonner sphere was proposed in our group. This Bonner sphere has multiple sensitive spherical shell layers in the single sphere. In this spectrometer, a band-shaped thermal neutron detection medium, which consists of a LiF-ZnS mixed powder scintillator sheet and a wavelength-shifting (WLS) fiber readout, was looped to each sphere at equal angular intervals. Amount of LiF neutron converter is reduced near polar region, where the band-shaped detectors are concentrated, in order to uniform the directional sensitivity. The LiF-ZnS mixed powder has an advantage of extremely high light yield. However, since it is opaque, scintillation photons cannot be collect uniformly. This type of detector shows no characteristic shape in the pulse height spectrum. Subsequently, it is difficult to set the pulse height discrimination level. This issue causes sensitivity fluctuation due to gain instability of photodetectors and/or electric modules. Materials and Methods: In order to solve this problem, we propose to replace the LiF-ZnS mixed powder into a flexible and Transparent RUbber SheeT type $LiCaAlF_6$ (TRUST LiCAF) scintillator. TRUST LiCAF scintillator can show a peak shape corresponding to neutron absorption events in the pulse height spectrum. Results and Discussion: We fabricated the prototype detector with five sensitive layers using TRUST LiCAF scintillator and conducted basic experiments to evaluate the directional uniformity of the sensitivity. Conclusion: The fabricated detector shows excellent directional uniformity of the neutron sensitivity.
Poly-ADP-ribosylation (PAR) is a post-translational modification of mainly chromosomal proteins. It is known to be strongly involved in several molecular events, including nucleosome-remodelling and carcinogenesis. In this investigation, it was attempted to evaluate PAR level as a reliable biomarker for early detection of cancer in blood lymphocyte histones. PAR of isolated histone proteins was monitored in normal and dimethylnitrosamine (DMN)-exposed mice tissues using a novel ELISA-based immuno-probe assay developed in our laboratory. An inverse relationship was found between the level of PAR and period of DMN exposure in various histone proteins of blood lymphocytes and spleen cells. With the increase in the DMN exposure period, there was reduction in the PAR level of individual histones in both cases. It was also observed that the decrease in the level of PAR of histones resulted in progressive relaxation of genomic DNA, perhaps triggering activation of genes that are involved in initiation of transformation. The observed effect of carcinogen on the PAR of blood lymphocyte histones provided us with a handy tool for monitoring biochemical or physiological status of individuals exposed to carcinogens without obtaining biopsies of cancerous tissues, which involves several medical and ethical issues. Obtaining blood from any patient and separating blood lymphocytes are routine medical practices involving virtually no medical intervention, post-procedure medical care or trauma to a patient. Moreover, the immuno-probe assay is very simple, sensitive, reliable and cost-effective. Therefore, combined with the ease of preparation of blood lymphocytes and the simplicity of the technique, immuno-probe assay of PAR has the potential to be applied for mass screening of cancer. It appears to be a promising step in the ultimate goal of making cancer detection simple, sensitive and reliable in the near future.
