Stable production of fermented kanjang containing 1.8% (v/v) ethanol was obtained within four days using traditional kanjang containing 4% added glucose in packed-bed bioreactor systems filled with immobilized Zygosaccharomyces rouxii and Candida versatilis on porous alumina ceramic bead carrier at $28{\pm}0.5^{\circ}C$ and aeration rate of 0.05 vvm. Specific rates of alcohol production for Z. rouxii and C. versatilis were 0.0033 and 0.0031/day, respectively, and those of glucose consumption were both -0.0087/day in the batch type of alcoholic fermentation. In semi-continuous alcoholic fermentation at a dilution rate of 0.25/day, specific rates of alcohol production for Z. rouxii and C. versatilis were 0.0045 and 0.0029/day, and those of glucose consumption were -0.01 and -0.008/day, respectively, using identical bioreactor system. Similar specific rates of alcohol production were observed both in the batch or semi-continuous process and in the continuous one at the dilution rate of 0.25/day. Sensory characteristics of all alcoholic-fermented kanjang by Z. rouxii, C. versatilis, and a mixture of both yeasts (2:1, w/w) were shown to be significantly superior to those of home-made kanjang as revealed through organoleptic evaluation tests (p<0.05).

The objective of this study was to investigate feasibility of semi-dry anaerobic digestion using dairy cattle manure and to evaluate solidified fuel value of semi-dry anaerobic digestate. To evaluate semi-dry anaerobic digestion using dairy cattle manure, 950 mL bottle type anaerobic reactor was set in the constant temperature room maintained at $35^{\circ}C$. To produce anaerobic digestate for making solidified fuel, acrylic cylindrical anaerobic digester(1,000 mm width ${\times}$ 450 mm height) was set in the constant room temperature to carry out batch test of semi-dry anaerobic digestion using same dairy cattle manure. Moisture content of dairy cattle manure and inoculum solution for anaerobic digestion were 80.64% and 96.83%, respectively. The dairy cattle manure and the inoculum solution was mixed by 1:1 ratio(v/v) for anaerobic digestion. Water content and VS/TS(Volatile Solids/Total Solids) of mixture of substrate and inoculum were 89.74% and 83.35%, respectively. In case of non-inoculated anaerobic digester, the biogas was not produced. By the semi-dry anaerobic digestion, the calorific value of the digestate was reduced by 20% compare to fresh dairy cattle manure. In other hand, ash content increased from 15% to 18.4%. The contents of Cr, Pb, Cd and S of pellet produced from anaerobically digested dairy cattle manure were not against the standard regulation for livestock manure solidified fuel. Therefore, it can be used as fuel that anaerobic digestate produced after semi-dry anaerobic digestion using dairy cattle manure.
Recombinant human epidermal growth factor (rhEGF) was produced by E. coli BL2l (DE3) harboring a plasmid pYHB101. The production of rhEGF was 44.5 mg/L when the E. coli BL2l (pYHB101) was cultured at 27$^{\circ}C$ for 48 hr in the modified MBL medium containing 10 $\mu\textrm{g}$/L glucose with 10 $\mu\textrm{m}$ IPTG/lactose induction at 2 hr after inoculation. It was shown that lactose is able to induce the rhEGF expression of E. coli BL2l (pYHB101) with the same efficiency as IPTG. In the batch culture system, when induced with 10 $\mu\textrm{m}$ lactose, E. coli BL2l (pYHB101) produced maximum 45 mg/L of the rhEGF at 28 hr culture in the modified MBL medium containing 10 g/L glucose. In the semi-fed batch culture system, the volumetric yield was 160 mg/L when the culture was added with 0.5% (w/v) lactose and 0.25% (w/v) yeast extract in the late logarithmic phase and 94.3% of rhEGF was secreted as soluble form. However, when the culture was added with them in the early logarithmic phase, the volumetric yield was 120 mg/L and 20.9% of rhEGF was found in cytoplasmic insoluble aggregates. It was found that the addition time of lactose was important for production of soluble rhEGF from E. coli BL21 (pYHB101).

Thermophilic aerobic bacteria were applied in the degradation of industrial waste activated sludge (WAS) on a laboratory scale expreiment. The performance of digestion was estimated by measuring the reduction of total suspended solids (TSS), dissolved organic carbon (DOC), and total organic carbon (TOC). Among three strains of Bacillus stearothermophilus and three strains of Thermus species, B. stearothemophilus ATCC 31197 showed the best overall efficiency level for the degradation of industrial WAS, which was collected from a wastewater treatment plant in an oil refinery factory. Industrial WAS coul be successfully detraded in a batch digestion with ATCC 31197. The stability of the digestion process with ATCC 31197 was successfully verified by semi-continuous (fill-and-draw) digestion experiment. From the results of this study, it was shown that the thermophilic aerobic digestion process with ATCC 31197 could efficiently be applied to the degradation of industrial WAS.
The emission of carbon dioxide from the burning of fossil fuels has been identified as a major contributor to green house emissions and subsequent global warming and climate changes. For these reasons, it is necessary to separate and recover $CO_2$ gas. A new process based on gas hydrate crystallization is proposed for the $CO_2$ separation/recovery of the gas mixture. In this study, gas hydrate from $CO_2/H_2$ gas mixtures was formed in a semi-batch stirred vessel at a constant pressure and temperature. This mixture is of interest to $CO_2$ separation and recovery in Integrated Coal Gasification (IGCC) plants. The impact of tetrahydrofuran (THF) on hydrate formation from the $CO_2/H_2$ was observed. The addition of THF not only reduced the equilibrium formation conditions significantly but also helped ease the formation of hydrates. This study illustrates the concept and provides the basic operations of the separation/recovery of $CO_2$ (pre-combustion capture) from a fuel gas ($CO_2/H_2$) mixture.
Generally pancreas consist of lipid, water and protein, digestion enzyme complex (pro-tease, lipase, amylase). The sample used in this work was frozen dry and treated by a semi-batch flow type. In order to develop a supercritical fluid extraction process to rem-ove lipid from the pancreas, experiments were conducted at various operating conditions(pressure range $1500{\sim}2800psi$, temperature range $25{\sim}40^{cdot}C$, particle size$(0.25{\sim}1.0mm$, flow rate $20{\sim}80m{\ell}/min)$. Also cholesterol in the pancreas was removed. The highest extraction efficiency was 2500psi, $35^{\cdot}C$, 0.25mm of pancreas size. The enzyme activity of the pancreas produced from this work showed high value compared with imported pancreas.
The purpose of this study is to develop a continuous process for sorbitol production using dried Zymomonas mobilis immobilized in K-carrageenan. The methods of glularaldehyde cross-linking of enzymes in CTAB (celyltrimetylammoniumbromide) treated cells immobilized in K-carrageenan showed stability for the production of sorbitol for 30 days of operation. K-carrageenan beads entrapping permeabilized cells were dried to Improve bead rigidity and storage stability. A semi-batch process with dry beads was carried out and only a small loss of enzyme activity (less than 8%) was observed during 72h. The value of Vmax for the dry K-carrageenan beads was found to be one half or that for free cells. It was shown that the productivities of the continuous process with wet K-carrageenan beads and dry beads at a dilution rate 0.1h-1 were 3.4g/L-h and 2.88h/L-h, respectively.
Culture conditions for biological hydrogen production were investigated in wastewater of Takju manufacturing factory. Rhodobacter spaeroides KCTC1425, photosynthesis bacteria, and Enterobacter cloacae YJ-1, anaerobic bacteria were used. The hydrogen production were $195.3m{\ell}{\cdot}H_2/{\ell}$ broth for Rhodobacter spaeroides KCTC1425 and $271.8m{\ell}{\cdot}H_2/{\ell}$ broth for Enterobacter cloacae YJ-1 during 36 h. The hydrogen production increased with light intensity, and were highest over 12000Lux. In mixed culture of Rhodobacter spaeroides KCTC1425 and Enterobacter cloacae Y J-1, the optimum mixing ratio of hydrogen production was 20 and 80. Adding volume of yeast extract for maximum hydrogen production was 15 $g/{\ell}$, but there was no effect over that. $Na_2MoO_4$ was most effective among the inorganic salts, and the optimum volume was 0.4 $g/{\ell}$. In semi-continuous culture, total hydrogen production was $13086m{\ell}{\cdot}H_2/{\ell}$ broth for 144 h with operating period of 24 h.
The production of vinegar containing 16.0-18.0% of acetic acid was examined in two stage fermentation consisting of semi-continuous and fed-batch type. The optimum conditions were obtained when the fermentation was carried out at agitation of 600 rpm, aeration of 0.1 vvm and temperature of $30^{\circ}C$. The initial and residual ethanol concentration in 1st stage were $50.0g/{\ell}$ and $5.0g/{\ell}$, respectively, and the ethanol concentration in 2nd stage was maintained from 5.0 to $10.0g/{\ell}$. The maximum productivity was 3.3 gll-hr and the acidity was 17.6% after the two days of acetic acid fermentation.
