A New Medium for the Selective Isolation of Soil Actinomycetes (토양중 방선균의 선택적 분리를 위한 배지)

  • For the more effective isolation of soil actinomycetes, we have developed HHV (Hair hydrolysate-vitamin) agar medium, containing hair as the sole source of carbon and nitrogen. The HHV agar medium was superior to other media such as colloidal chintin agar, glycerol-arginine agar and starch-casein-nitrate agar, and HV (humic acid-vitamin) agar. The maximum effect of this medium has been shown in hair dry weight 0.4 g/l medium. Of each soil sample, the greatestest number of actinomycetes was isolated from the potato annual planted soil among the tested samp- les. The genus of actinomycetes isolated from the potato annual planted soil sample was identified such 5 group as Stretomyces, Micromonospora, Microbispora, Nocardia and Saccharopolyspora.

Determination of best enrichment media for growth of Salmonella injured from cold temperature during process and storage (저온저장으로 인해 손상된 살모넬라를 배양하기 위한 최적의 배지 선정에 관한 연구)

  • This purpose of this study was to determine the best enrichment medium for rejuvenating and recovering Salmonella placed in cold temperature prior to the employment of the gold biosensor combined with a light microscopic imaging system. A mixture of nalidixic-resistant Salmonella Typhimurium and Enteritidis were inoculated onto chicken (1,000 CFU/chicken). After cold injury at $4^{\circ}C$ for 24 hr, Salmonella on chicken was enriched for 6 hr with six non-selective media including buffered peptone water broth, lactose broth, brain heart infusion broth (BHI), universal pre-enrichment broth, nutrient broth, and tryptic soy broth, and five selective media including brilliant green broth (BG), rappaport-vassiliadis R10 broth, selenite cystine broth, selenite broth, and tetrathionate brilliant green broth (TBG) for the comparison of Salmonella growth. Various concentrations of Salmonella (10, 50, 100, 500, and 1,000 CFU/chicken) were then enriched for 6 hr in both BHI and BG media to select the best media. BHI was selected as the most effective non-selective enrichment medium, while BG was selected as the most effective selective enrichment medium. Finally, BHI medium was selected as the most efficient enrichment medium for Salmonella growth injured from cold temperature during processing or storage.

Comparative Evaluation of Selective Media for Isolation of Bifidobacterium Species in Human Fecal Sample (인체 분변에서 Bifidobacterium species의 선택적 분리를 위한 배지 비교)

  • This study aimed to evaluate the appropriateness of MRS-C (0.05% L-cystein; pH 5) and BHI-CM (0.05% L-cystein, 0.5% mucin) agars for the selective isolation of bifidobacteria in fecal samples compared to blood-liver-NPNL (BL-NPNL) agar. Over 200 isolated colonies were characterized morphologically and biochemically. Genomic DNA was extracted from pure cultures of the isolated strains, followed by PCR amplification of the 16S rRNA gene. Bifidobacterium longum and B. animalis were selectively isolated from MRS-C agar and Lactobacillus acidophilus and Enterococcus avium were also isolated. B. longum, B. faecale, and B. animalis were isolated from feces on BHI-CM agar; however, different Bacteroides strains (including Bac. fragilis, Bac. kiribbi, Bac. ovatus, Bac. koreensis, and Bac. salyersiae) were also detected. BL-NPNL agar successfully isolated B. longum and Bacillus, while other Bifidobacterium and Bacteroides species could not grow owing to the presence of antibiotics in the medium. The use of antibiotics in a medium can enhance the selectivity; however, antibiotics may inhibit the growth of certain bacteria in a sample. Hence, adjusting pH or adding non-antibiotic nutrients to the medium is more advantageous, than relying on antibiotics.

Resistance of Fusarium fujikuroi Isolates to Hydrogen Peroxide and Its Application for Fungal Isolation

  • The ascomycete fungus Fusarium fujikuroi causes bakanae disease in rice and this disease has been reemerging in Korea. Other fungal species including F. graminearum and Magnaporthe oryzae are often associated with F. fujikuroi, hampering pure isolation of F. fujikuroi from rice. In this study, we modified a selective medium for F. fujikuroi as supplementing both pentachloronitrobenzene and hydrogen peroxide into minimal medium. This medium efficiently suppressed the vegetative growth of F. graminearum and M. oryzae, but did not significantly reduce F. fujikuroi growth, providing an efficient tool for isolating F. fujikuroi.

  • Selective culture of actinomycetes from soil microbes and their antibiotic producing characters by agar-disk method were examined. Some of the organisms which produced antibiotics on agar disk did not produce antibiotics in liquid culture. Further examination indicated that production of antibiotic was dependent on the composition of medium. Many streptomycestes produced antibacterial substances in tryptic soy broth but others produced antifungal antibiotics in V-8 broth. Production of antibacterial substances by Streptomyces sp. was also dependent on the medium composition.

Selective Media Containing Antibiotics for Counting Bifidobacteria (항생제를 이용한 Bifidobacteria의 선택배지)

  • Selective agar media were constructed for the counting of bifidobacteria in dairy produ- cts containing bifidobacteria, lactobacilli and streptococci. This media containing antibiotics inhibited the growth of lactobacilli and streptococci at less than 10$^{5}$ cfu/ml, but had no influence on the recovery of bifidobacteria. In order to inhibit the growth of 10$^{5}$ cfu/ml of lactobacilli and streptoco- cci, the addition of 1.0~2.0 $\mu$g/ml of tetracycline in BL agar medium was needed. When 25 $\mu$g/ml of neomycin and paromomycin were mixed with 1.0 $\mu$g/ml of tetracycline in BL agar medium, it was able to inhibit the growth of 10$^{6}$ cfu/ml of lactobacilli and streptococci but had a little negative effect on the recovery of colonies of bifidobacteria. The results revealed that the BL agar medium containing 1.0 $\mu$g/ml of tetracycline was suitable to count the cell number of bifidobacteria selecti- vely in the presence of a 1 to 10$^{5}$-fold excess of L. casei, L. acidophilus and S. salivarius subsp.thermophilus.

  • HZSM-5, synthesized in fluoride medium, showed high selectivity for the formation of 1,4-diethylbenzene (1,4-DEB) in the vapour phase ethylation of ethylbenzene (EB) with ethanol and hence becomes a convenient eco-friendly substitute for hazardous mineral acid catalysts. De-ethylation of EB to benzene was also minimized over this catalyst. As the medium pore size and presence of weak and medium acid sites might be the cause of such benefits, fluoride mediated synthesis of ZSM-5 is proven to be advantageous for para-selective alkylation of alkyl aromatics.

Selection of Glyphosate-Resistant Clones form MNNG-treated Mesophyll Protoplasts of Haploid Tobacco Plants (반수체 담배의 엽육 원형질체로부터 MNNG 처리에 의한 Glyphosate 저항성 클론의 선별)

  • Selection of glyphosate-resistant clones from MNNG-treated mesophyll protoplasts of haploid tobacco and their differentiation were studied. The protoplasts were treated with 0.1 to 100 $\mu\textrm{g}$/mL N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) for 30 min when they expanded to oval shapes. After the treatment, the protoplasts in 4-16 cell stages were transferred to the selective medium containing 1 mM glyphosate for the selection of the glyphosate-resistant colonies. The efficiency of the cell division of the protoplasts in the selective medium decreased as the MNNG concentrations in creased. Optimal MNNG concentration for induction of the glyphosate-resistant clones was 10$\mu\textrm{g}$/mL and mutation frequency was 2.66$\times$10-6. The stability of the glypohsate-resistance of the clones was examined by prolonged subculture in the medium with 1 mM glyphosate, and the resistant clones were survived more than 10 months. Among them one clone has been proliferating and greening and the others were proliferating without greening or greening with slower proliferating.

  • Some physical and physiological properties of di-D-fructofuranose dianhydrideIII (DFAIII), as a new sweetener, were investigated via in vitro experiments. The disaccharide was prepared by decomposing inulin with inulin fructotransferase (depolymerizing) from Arthrobacter sp. A-6. DFAIII had more excellent heat and acid stability than sucrose. This was one of the most desirable properties especially for the oligomer types of sweetener. DFAIII showed the least pH drop in the Streptococcus mutans culture, compared with the other saccharides examined. This indicates that the sugar will be fairly effective for preventing dental caries. The saccharide also had a selective Bifidus growth-promoting effect in PYF medium. Whereas, E. coli did not show growth promotion in the DFAIII-containing medium. In the co-culture of Bifidus longum and E. coli in the BL medium, Bifidus longum had a selective growth while the growth of E. coli appeared rather to be inhibited.

Effect of the paper acidity on the cellulolytic activity of fungi (종이의 산성화가 미생물의 분해능에 미치는 영향)

  • The effect of pH on degradation of paper by some fungi, which able to degrade cellulose, was investigated. Trichoderma koningii, Aspergillus nigerand Penicillium nigulosum were cultured at $28^{\circ}C$ for 16 days in the selective medium (PH3, PH4, PH5, PH6, PH7, PH8, PH9, PH10, PHC) containing paper as substrate. Each paper was pretreated with each pH buffer (pH 3∼pH 10, D.W.)prior to addition to the selective medium. Enzyme activities in the each culture medium were measured spectroph to metrically using C.M.C., Avicel, PNPG as the substrates for endoglucanase, exoglucanase and $\beta$-glucosidase, respectively. In all experimental fungi, the enzyme activities of PH3 and PH9 medium were usually much higher than those of other experimental groups. However in the PH6medium, enzyme activity was lower than other groups. To analyze the concentration and pattern of protein in the each culture medium, the medium was concentrated by lyophilization. The protein concentration of PH3 and PH9 medium were relatively high (T.koningii; 6.31mg, 6,19mg, A.niger; 1.62mg, 1.96mg, P.nigulosum;2.50mg, 2.73mg, respectively), but that of PH6 was relatively low. The protein pattern of each medium was analyzed by using SDS-PAGE and VDS Image Master Analysis Program. The concentrations of bands in the each lane were usually high at lane2 (PH3) and lane8 (PH9) and low at lane5 (PH6). Therefore, the incresed cellulolytic activity of fungus against acidified paper could be result of structural change and deterioration of paper caused by being acidified.
