A Case of Clostridium difficile Pseudomembranous Colitis (Clostridium difficile 감염에 의한 위막성 대장염 1례)

  • Pseudomembranous colitis, thought to be uncommon in children, is a bacterial, toxin-mediated inflammatory process resulting in acute or chronic diarrhea and is characterized by colonic pseudomembranes. It is mediated by toxins produced by Clostridium difficile and is increasingly recognized in pediatric population. Diagnosis is based on positive culture of C. difficile in selective media and positive test of C. difficile toxin. Oral metronidazole or vancomycin are the main treatment options but avoidance of further antibiotics should also be encouraged where possible. We have experienced a case of pseudomembranous colitis in a 4-year-old female presented with septic shock and colitis. This case was diagnosed with positive test of C. difficile toxin B and confirmed by isolation of the organism on cultire in selective media. Symptoms have been ameliorated by discontinuation of antibiotics and administration of metronidazole and oral vancomycin, and ICU care.

  • This paper proposes a novel switching strategy of 1Mvar STATCON using cascade multilevel H-bridge inverter(HBI) for FACTS application. To control the reactive power instantaneously, the d-q dynamic system model is described and analyzed. A single pulse pattern based on the SHEM(Selective Harmonic Elimination Method) technique is determined from the look-up table to reduce the line current harmonics and a rotating fundamental frequency switching scheme is presented to adjust the DC voltage of each inverter capacitor at the same value. So the voltage unbalance problem between separately DC bus voltage is improved by using the proposed switching scheme. As a result, the presented inverter configuration not only reduces the system complexity by eliminating the isolation at the AC input side transformer but also improves the dynamic response to the step change of reactive power.

  • In this study we attempted to develop a novel genomic method to selectively isolate target functional microbial genomes from environmental samples. For this purpose, stable isotope probing (SIP) was applied in selectively isolating organic pollutant-assimilating populations. When soil microbes were fed with $^{13}C-labeled $ biphenyl, biphenyl-utilizing cells were incorporated with the heavy carbon isotope. The heavy DNA portion was successfully separated by CsCl equilibrium density gradient. And the diversity in the heavy DNA was sufficiently reduced, being suitable for the current DNA microarray techniques to detect biphenyl-utilizing populations in the soil. In addition, we proposed a new way to get more genetic information by combining this SIP method with selective metagenomic approach. The increased selective power of these new DNA isolation methods will be expected to provide a good quality of new genetic information, which, in turn, will result in development of a variety of biomarkers that may be used in assessing ecotoxicology issues including the impacts of organic hazards, and antibiotic-resistant pathogens on human and ecological systems.

  • A novel lipase gene, Lip-1420, was isolated from a metagenomic library constructed from reed marsh from Mt. Jumbong in Korea, comprising 112,500 members of recombinant plasmids. The DNA sequence of Lip-1420-subclone (5,513 bp) was found to contain at least 11 ORFs according to the GenBank database. The ORF-3 gene was inserted into the pET21a plasmid containing the C-terminal 6-His tag and transformed into E. coli BL21(DE3) to express the recombinant lipase protein. Lip-1420 was purified using a fast protein liquid chromatography system. The gene was registered in GenBank (MH628529). The values of Km and Vmax were determined as 0.268 mM and 1.821 units, respectively, at 40℃ and pH 8.0, using p-nitrophenyl palmitate as the substrate. This lipase belongs to family IV taxonomically because it has conserved HGGG and GDSAG motifs in the constitutive amino acid sequence. According to the predicted structural model, the binding sites are represented by residues H78, G81, D150, S151, A152, V181, and D236. Finally, Lip-1420 showed triolein selectivity for methanolysis between triolein (18:1) and tristearin (18:0) substrates. Further study of the selective mechanism and structure-function relationship of this new lipase could be useful for more practical applications.

Isolation and structural elucidation of the herbicidal active compounds from Ligularia stenocephala M.

  • Screening was conducted using 200 kinds of plant extracts to explore herbicide-activated components of plant origin. We separated and purified active substances and elucidated chemical structures using Ligularia stenocephala M., which has strong activity and has not yet been studied. When the solvent fractions of the leaves of Ligularia stenocephala M. were tested for their herbicidal activity, ethyl acetate and chloroform layer showed an inhibition rate of 95.2% and 94.1%, respectively. In particular, the chloroform layer exerted more than 50% herbicidal activity at 10 ppm. From the chloroform layer with the highest herbicidal activity, we isolated three herbicidal active compounds using stepwise chromatography, specifically silica gel or octadecyl silica (ODS) column chromatography, Sep-pak cartridges, and high performance liquid chromatography (HPLC). Based on the analysis of the active compounds using electron ionization mass spectroscopy (EI-MS), 1H-NMR, and 13C-NMR, we identified the active compounds as euparin, 5,6-dimethoxy-2-isopropenylbenzofuran, and liguhodgsonal. When the herbicidal activity of the identified compounds was tested, euparin showed selective herbicidal activity for lettuce at 10-3 M, and both liguhogsonal and 5,6-dimethoxy-2-isoprophenylbenzofuran exerted selective activity for rice and Echinochloa crus-galli.

Design of a Frequency Selective Surface Using DSRRs (DSRR을 이용한 주파수 선택적 표면 설계)

  • We propose a frequency selective surface(FSS) using double split ring resonators(DSRRs) for isolation enhancement between CDMA and RFID. The structure consists of an outer SRR and an inner SRR, and the gaps are formed in the same direction. By properly adjusting the gap and line width, the resonant frequency and skirt characteristics can be adjusted without varying the unit cell size. The proposed structure has a different field distribution from that of an ordinary SRR for magneto-dielectric materials. One layer consists of $9{\times}9$ unit cells and the other layer was separated by 50 mm. To validate the simulation results, we fabricated the patch antenna and the FSSs, and the measured results show a good agreement with the simulated ones. The electrical size of the unit cell is $0.110\;{\lambda}{\times}0.110\;{\lambda}{\times}0.002\;{\lambda}$, and the size of the two layer FSS is $1.058\;{\lambda}{\times}1.058\;{\lambda}{\times}0.153\;{\lambda}$. The two layer FSS maintain gain in CDMA frequency and has 6.9 dB reduced gain in RFID frequency.

Isolation and identification of Agrobacterium spp. using carrot disc and transformation of Nicotiana gluca by selected strains (당근 절편을 이용한 Agrobacterium spp. 분리, 동정과 선발균주에 의한 연초의 형질전환)

  • These studies were carred out to obtain the transformant from tobacco cells by Agrobacterium spp. from crown gall and soil at the natural field in Korea, and identified their virulence. Kodo's and Clark's selective media were used for isolation of Agrobacterium spp. In these media, total of 99 strains were characterized based on the morphological characteristics of colonies. Among them 34 strains were able to induce on carrot discs. And hypervirulent strains C23-1 and K29-1 were identified as Agrobacterium tumefaciens biotype 1 and biotype 2, respectively. These strains formed fast growing, larger gall as compared to those induced by other strains on the carrot discs. Transformed tobacco callus was initiated on the phytohormone free MS medium with 250$\mu\textrm{g}$/ml carbenicillin after co-cultivation of tobacco stem explants and Agrobacteria. On the phytohormone free media, shoot was rarely formed from transformed callus. However, these shoot were teratoma shoots which were not grown as normal shoot, and teratoma shoot from transformant by C23-1 was smaller than that of K29-1.

Isolation and characterization of Clostridium Perfringens on bovine and porcine carcass (소와 돼지의 도체에서 Clostridium perfringens의 분리와 특성)

  • A total of 1,848 samples was taken from bovine and porcine from January 2003 to November 2005. They were examined for the presence of Clostridiuml perfringens. The properties of the isolates were characterized for Gram staining, biochemical features and enterotoxin production. Forty-one strains (2.2%) of C perfringens were isolated from the 1,848 bovine and porcine carcass using selective media. 30 (3.2%) C perfringens were isolated from the 925 of bovine cacasses, and 11(1.2%) were isolated from the 923 of porcine cacasses. In TSC agar, all isolates showed lecithinase activity. The isolation rate was higher in spring and summer than in autumn and winter. Among 41 isolates, only 1 isolate detected the cpe gene in PCR. The PCR amplified band was observed 233 bp.

Isolation of Vibrio vulnificus from Shellfish (어패류에서의 Vibrio vulnificus 분리)

  • Vibrio vulnificus is an organism capable of causing septicemia and wound infection in compromised patients. The source of infection is known to be raw oysters and others. The prevalence of the organism in Korean sea water and shellfish is not known. The authors surveyed shellfish and other specimens obtained mainly from a market in Seoul and from ones in Inchon. Five cultures of V. vulnificus were isolated from oyster and clam samples. Two isolates had typical characteristics of the strains isolated from patients, i.e., definite hemolysis and typical biochemical reactions. However, other 2 isolates were sucrose positive, although the identity were confirmed by Center for Disease Control. We do not know wether such strains are pathogenic or not. For the isolation of V. vulnificus from environmental samples, TCBS agar and VV agar were not very selective or differential. We isolated our strains with the use of OF-lactose agar and SPS agar. However OF-lactose agar did not support good growth of V. vulnificus, while SPS agar did not suppress other vibrios.

A Semiselective Medium for the Isolation of Xanthomonas campestris pv. oryzae from Rice Seed (벼 종자에서 Xanthomonas campestris pv. oryzae의 분리를 위한 선택배지)

  • A semiselective agar medium (XCO) was developed for the isolation of bacterial blight pathogen, Xanthomonas campestris pv. oryzae, from rice seed. The medium contained yeast extract 1.0 g, peptone 2.0 g, sucrose 5.0 g, sodium glutamate 1.0 g, FeSO4.7H2O 0.05 g, Fe.EDTA 1 mg, cephalexin 20 mg, Evan blue (0.1%) 1.5 ml, bromcresol purple (0.1%) 2.5ml, cycloheximide 100 mg and agar 15.0 g per liter. Colonies of X. c. pv. oryzae were 4~5 mm in diameter, smooth, round, blue (darker center) and convex after 6 days incubation at 28$^{\circ}C$. The recovery of 6 isolates of X. c. pv. oryzae on the XOC medium ranged from 81% to 120% (mean 98.2%) in comparison to Wakimoto's medium. The number of saprophytic bacteria from 10 rice seed lots on XCO medium was reduced to 70.4% of that on Wakimoto's medium. The recovery of X. c. pv. oryzae added to rice seed on XOC medium ranged from 67% to 87% (mean 75.6%) of that on Wakimoto's medium.
