Selective Medium for Isolation and Enumeration of Eubacterium sp.from the Feces of the Korean People (한국인의 분변으로부터 Eubacterium을 분리하기 위한 선택 배지 조사)

  • Eubacterium is one of the predominant bacteria in the human large intestine. currently ES (Eubacterium Selective) medium developed by T. Mitsuoka is commonly used as a selective medium. neomycin sulfate which is one of the selective agents of ES medium inhibited about 50% of the growth of Eubacterium isolated, whereas malidixic acid inhibited only 5% while inhibiting other intestinal bacteria. NES medium which replaced neomycin with nalidixic acid in the ES medium was designed and shown to be better for the isolation and enumeration of Eubacterium sp. than ES medium.

Construction of Probability Identification Matrix and Selective Medium for Acidophilic Actinomycetes Using Numerical Classification Data

  • A probability identification matrix of acidophilic Streptomyces was constructed. The phenetic data of the strains were derived from numerical classification described by Seong et al. The minimum number of diagnostic characters was determined using computer programs for calculation of different separation indices. The resulting matrix consisted of 25 clusters versus 53 characters. Theoretical evaluation of this matrix was achieved by estimating the chuster overlap and the identification scores for the Hypothetical Median Organisms (HMO) and for the representatives of each cluster. Cluster overlap was found to be relatively small. Identification scores for the HMO and the randomly selected representatives of each cluster were satisfactory. The matrix was assessed practically by applying the matrix to the identification of unknown isolates. Of the unknown isolates, 71.9% were clearly identified to one of eight clusters. The numerical classification data was also used to design a selective isolation medium for antibiotic-producing organisms. Four chemical substances including 2 antibiotics were determined by the DLACHAR program as diagnostic for the isolation of target organisms which have antimicrobial activity against Micrococcus luteus. It was possible to detect the increased rate of selective isolation on the synthesized medium. Theresults show that the numerical phenetic data can be applied to a variety of purposes, such as construction of identification matrix and selective isolation medium for acidophilic antinomycetes.

Study on Selective Media for Isolation of Entomopathogenic Fungi

  • To determine the optimal isolation conditions of the entomopathogenic fungi from soil, we compared their growth characteristics with non-entomopathogenic fungi on agar media containing various concentrations of cooper (II) chloride ($CuCl_2$) or dodine. The result showed that dodine medium is more selective, and the optimal concentration of dodine is determined with $50{\mu}g$/ml. We could isolate several putative entomopathogenic fungi from soil using this, and identify them using ITS analysis. As a result, 64% fungi were identified as typical entomopathogenic fungi. This selective medium may be useful to the rapid and simple isolation of entomopathogenic fungi from soil.

An Improved Selective Isolation of Rare Actinomycetes from Forest Soil

  • Various pretreatment procedures and selective media were applied to assess the optimal conditions for the isolation of rare actinomycetes from soil. Pretreatment of wet-heating for 15 min at 70$^{\circ}C$ and phenol treatment of soil suspension were the most effective methods for the isolation of these microorganisms. Hair hydrolysate vitamin agar (HHVA) was the most suitable medium for the recovery of rare actinomycetes. Thirty-five rare actinomycete strains were chosen using selective isolation approaches, then morphological and chemical properties of the isolates were determined. The isolates belonged to one of the following genus, Micromonospora, Microbispora, Actinoplanes and Streptosporangium.

  • Three kinds of pretreatment methods were used for selectivel isolation of soil actinomy cetes. One hundred and six strains were isolated on Bennet's agar and 114 strains were on humic acid-vitamin agar from 5 domestic soil samples. All of these isolates were identified to the genus level based on morphological and physiological characheristics. Among three methods, dry heat was most effective to isolate many different strains including rare actinomycetes. On genus, Bennet's agar was effective for selective isolation of Nocardia but bumic acid-vitamin agar for another rare actinomycetes.

  • This study was conducted to evaluate the selective media listed in currently available Food Code in Korea. The 29 different types of media of five different types of foodborne bacteria including Salmonella spp., Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, Staphylococcus aureus were tested in the broth and food. The recovery test for five different types of foodborne bacteria was performed in the artificially inoculated into chicken, rice, pork and mackerel. There was no significant differences in isolation capabilities among twenty nine different types of isolation selective media for five different types of foodborne bacteria in broth condition, while there was significantly a little differences in isolation capabilities among those on foods (P<0.05). The higher number of foodborne pathogens were isolated from conventional selective media approved in Food Code than newly developed selective media such as chromogenic media. This results suggest that there was differences of selectivities among currently available isolation selective media in many countries and further studies are needed to be approved by Korean Food and Drug Administration.

  • The effect of SiH2Cl2 -HCl gas on the growth rate of epitaxial layer is studied. The temperature, pressure and gas mixing ratio of SiH2Cl2 and HCl are varied to study the growth rate dependence and selective Si epitaxy. The P-n junction diode is fabricated on the epitaxial layer and electrical characteristics are examined. Also, using selective Si epitaxy, a possibility of thin dielectric isolation process, that gives an independent isolation width on the mask dimension, is examined.

  • Phage utilizing medium and BL agar supplemented with antibiotic Tc(tetracycline) were developed as selective media for the isolation and counting of bifidobacteria in dairy products. The former was based on the host specificity of phage. When bifidobacteria and Lactobacillus casei HY 2782 were mixed together in dairy product, L. casei HY 2782 was laysed by J1 phage which has host specificity to L. casei HY 2782 whereas bifidobacteria grew well on the selective medium added wIth J1 phage. The latter was found to inhibit the growth of S. salivarius subsp. thermophilus, L. acidophilus, and L. casei, but commercial bifidobacteria grew well in Tc-containing BL agar.

  • Selective media for isolation and enumeration of lactic acid bacteria from kimchi were compared using 36 strains of lactic acid bacteria from kimchi and dairy products. Among the selective media, KF Streptococcus agar showed the best result for the selection of Enterococcus group and Pediococus group, and M-17 agar with pH indicator for Lactococcus group, the m-LBS agar for Lactobacillus group, and PES-3 agar for Leuconostoc group.

  • This paper presents the design of reconfigurable frequency selective surfaces for X-band bandpass operation with improved isolation. The proposed reconfigurable FSS is composed of a four-legged loaded element, a inductive stub and a bias grid. The PIN diode is located between the four-legged loaded element and the stub, which can control the frequency response of reconfigurable FSS by ON/OFF state. By adjusting the length of the stub, the desired bandpass frequency and the improved isolation between ON and OFF state can be obtained. For validation of simulated results, we have carried out transmission characteristic measurements using rectangular waveguide of WR-90. The measured results are in good agreements with the simulated results.
