Bahavior of Some Herbicides Applied to Oil-bearing Crops (유지작물(油脂作物)에 시용(施用)한 몇가지 제초제(除草劑)의 행방(行方))

  • Three herbicides including nitrofen, alachlor and butachlor f?ere applied to farm soils of oil-bearing crops rape, soybean and paddy rice and their residual levels in the soil and seeds were determined by ECD-attached gas chromatograph without hydrolysis. Applied herbicides were decreased abruptly 2 weeks after application and slowly thereafter, reaching below 10% level while the extent of disappearance varied depending on the herbicides, crops and soil conditions. The herbicides were not detected at all in the seeds of tested crops within the detection limit of the analytical methods employed.

PDAT1 genome editing reduces hydroxy fatty acid production in transgenic Arabidopsis

  • The fatty acids content of castor (Ricinus communis L.) seed oil is 80-90% ricinoleic acid, which is a hydroxy fatty acid (HFA). The structures and functional groups of HFAs are different from those of common fatty acids and are useful for various industrial applications. However, castor seeds contain the toxin ricin and an allergenic protein, which limit their cultivation. Accordingly, many researchers are conducting studies to enhance the production of HFAs in Arabidopsis thaliana, a model plant for oil crops. Oleate 12-hydroxylase from castor (RcFAH12), which synthesizes HFA (18:1-OH), was transformed into an Arabidopsis fae1 mutant, resulting in the CL37 line producing a maximum of 17% HFA content. In addition, castor phospholipid:diacylglycerol acyltransferase 1-2 (RcPDAT1-2), which catalyzes the production of triacylglycerol by transferring HFA from phosphatidylcholine to diacylglycerol, was transformed into the CL37 line to develop a P327 line that produces 25% HFA. In this study, we investigated changes in HFA content when endogenous Arabidopsis PDAT1 (AtPDAT1) of the P327 line was edited using the CRISPR/Cas9 technique. The successful mutation resulted in three independent lines with different mutation patterns, which were transmitted until the T4 generation. Fatty acid analysis of the seeds showed that HFA content decreased in all three mutant lines. These findings indicate that AtPDAT1 as well as RcPDAT1-2 in the P327 line are involved in transferring and increasing HFAs to triacylglycerol.

Distribution of Fatty Acids in Newly Developed Tissues of Soybean Seedlings

  • The objective of this study was to determine the fatty acid composition of newly developed tissues of germinated soybean seeds. Five soybean accessions with varied fatty acid composition were allowed to germinate in sand under greenhouse conditions. Seedlings were picked up after 4, 6, 8, 10 and 12 days of germination and freeze dried. The fatty acid composition of the newly developed tissues was analyzed by gas chromatography. Significant variation in fatty acid composition was observed between accessions, days of germination, and variety ${\times}$ day of germination in whole and the cotyledons. In the case of newly developed five tissues, significant variation in fatty acid composition were observed between days of germination except oleic acid for root, hypocotyl and epicotyl stem and except stearic acid for hypocotyl and unifoliate leaves while all the parameters were significantly different for accession. Significant interactions of accession and days of germination were observed for palmitic, linoleic and linolenic acid in all tissues; only for oleic acid in hypocotyl, epicotyl and unifoliate leaves; and only for stearic acid in root, hypocotyl, epicotyl and unifoliate leaves. During germination, the fatty acid composition of newly developed tissues changed dramatically but whole seedlings and cotyledons changed slightly. These tissues contained five major fatty acids as found in original seeds, but compositions were totally different from that of the seed: higher in palmitic, stearic and linolenic acid and lower in oleic and linoleic acid. New tissues conserved their fatty acid compositions regardless of genotypic variation in the original seeds.

Pesticide residues in chili pepper seeds and their transfer into the seed oil (고추씨 중 농약 잔류와 고추씨 기름으로 농약의 이행)

  • A pesticide mix solution containing difenoconazole, lambda-cyhalothrin, and lufenuron was applied 3 times on field grown chili pepper at a fivefold overdose dilution concentration of the spray solution at a pre-harvest interval of 7 day. Difenoconazole, lambda-cyhalothrin, and lufenuron were detected at 4.43, 0.334, and 1.56 mg/kg, respectively, in raw chili pepper. Washing with water reduced the residue levels to 91.4, 94.3, and 85.3%, respectively. In dried chili pepper, the residues of difenoconazole, lambda-cyhalothrin, and lufenuron were 22.2 mg/kg (processing factor, Pf =5.01), 1.65 mg/kg (Pf =4.94), and 6.54 mg/kg (Pf =4.19). In the seeds, difenoconazole and lambda-cyhalothrin were not detected, and lufenuron was detected at 0.0075 mg/kg (n=1) and <0.005 mg/kg (n=2). Thus the pesticide residues in the seeds was negligible. In the seed oil, difenoconazole and lufenuron residues were 0.0263 and 0.0295 mg/kg, respectively (concentration factors=5.26 and 4.72). These concentration factors supported the theoretical concentration factor of 6.8, assuming that all of compound present in the seed are transferred into the oil.

The Effects of Phosphate to the Growth and Oil Contents in Sesame(Sesamum indicum L.) (참깨의 단경, 분지형에 있어서 인산함량이 함유율과 지상부 생육에 미치는 영향)

  • Effects of phosphorous fertilizer on the growth habit and yield of monoculm and branch types of two leading sesame varieties was investigated. The photosynthetic rate of at each growing stage was high at the flowering stage. The photosynthetic rate of monoculm sesame variety, Pungnyeon was high prior to flowering stage, while the Kwangsan variety, branch type was high after the flowering stage. The level of phosphate were most effective in increasing photosynthesis when standard level of fertilizer was applied in both varieties. Kwangsan variety showed higher photosynthetic rate per unit area, higher NAR, and CGR and higher LAR and RGR at the maximum flowering stage than the Pungnyoeon variety. Those characters attained maximum level when standard levels of phosphorous fertilizer were applied. The protein and oil content of seeds were higher in Kwangsan variety compared with Pungnyeon. However, the carbohydrate of seeds was high for Pungnyeon variety. The highest protein content was attained when standard level of fertilizer were applied. The oil content appeared to increase as the level of applied fertilizer increased. The results suggest that the vegetative growth and seed yields of sesame may be enough with present level of fertilizers. However, higher amount of phosphorous fertilizer may be required to increase the oil content of sesame seeds.

Effect of extraction method on sesame oil quality

  • Sesame has been consumed for centuries as flavoring ingredient in eastern Asian countries, especially Korea. Sesame seeds have been used as health food for traditional medicine to prevent disease in Asian countries for several thousand years. Sesame seed has higher oil content (around 50%) than most of the known oilseeds. Sesame oil is rich in monounsaturated and polyunsaturated fatty acids. Extraction of sesame has developed significantly over the years. The mechanical method was an early means of separation which was physical pressure to squeeze the oil out. Nowadays, solvent extraction becomes the commonly used commercial technique to recover oil from oilseeds. In this study, we investigated extraction efficiency and quality of oil affected by cultivars and extraction methods of sesame seed. Different variables were investigated; roasting temperature ($170{\sim}220^{\circ}C$), extraction methods (solvent and physical pressure), forced ventilation system and cultivars. The Contents of B(a)P in sesame oil after roasting at $170{\sim}220^{\circ}C$ were 0.30~2.53 ppm. When we introduced forced ventilation system during roasting, B(a)P Contents were decreased up to 36%. The Oil extraction efficiency on sesame seed was statistically depending on the cultivars and extraction methods. The oil extraction yields of solvent and physical pressure extraction were 56.3% and 44.6%, respectively. Many of sesame cultivars and genetic resources are linolenic acid content of less than 0.5%. The results supported that we have developed a safe and high quality sesame oil processing methods for small and medium-sized companies.

Effects of Filtration or Centrifugation on the Oxidative Stabilities of Sesame Oil (여과 및 원심 분리가 참기름의 산화 안정성에 미치는 영향)

  • Effects of filtration and centrifugation on the oxidative stabilities of sesame oils during storage at $70^{\circ}C$ were studied by combination of determining peroxide values and conjugated dienoic acid values of oils and measuring the hexanal formation using headspace gas chromatography. Crude sesame oil from roasted seeds contained more free fatty acids, conjugated dienes, and metals (Fe, Cu, Mg and Zn); on the other hand, higher contents of moisture and ${\gamma}-tocopherol$ were found in the filtered or centrifuged oil. Only filtered oil contained more peroxides than the crude oil in spite of the color advantage of the highest L and b values among three oils. All the oils showed the tendency of increasing in total color difference during storage at $70^{\circ}C$, fatty acid compositions were relatively constant except for decreasing tendency of linoleic acid in filtered oil. No significant difference at 1% in the oxidative stabilities was observed between centrifuged oil and crude oil with higher susceptibility to the oxidation in the filtered oil. Centrifuged sesame oil was the best in the aspect of both oxidative stability and the oil color.

Changes of Volatile Flavor Compounds in Sesame Oils during Industrial Process (가공공정에 따른 참기름 휘발성 향기성분의 변화)

  • Changes of volatile flavor compounds in sesame oil during industrial process (roasting temp. $225{\pm}2^{\circ}C$, roasting time 15 min) were investigated. Total volatile flavor compounds of 1 st expressed oil from roasted sesame seeds were 536.3 ppm, and those of 2nd expressed oil from sesame seed cake, residue of 1st expression, were 266.8 ppm. Those of 1st filtered oil, fixed oil and 2nd filtered oil were 472.2 ppm, 472.4 ppm and 443.0 ppm, respectively. Volatile flavor compounds were gradually decreased during processing. Top notes $(peak{\;}No.1{\sim}26)$ playing an important role in the aromatic character of sesame oil, of 2nd expressed oil were markedly reduced (70.67% of initial content). Especially pyrazine compounds showed the largest reduction in 2nd expressed oil. Total volatile flavor compounds of fixed oil, filtered oil were reduced slightly.

Effect for Oxidation Stability of Refined Perilla Oil Use in Extract of Black Rice (Oryza sativa L.) (정제들깨기름의 산화안정성에 미치는 흑미 추출물의 영향)

  • Proximate compositions of black rice and antioxidative effects of its methanol extract on refined perilla oil were investigated. Black rice seeds were composed of water 12.4%, crude protein 7.9%, crude fat 2.2%, crude fiber 1.0%, ash 1.4% and N-free extract 75.1%. The hexane and 80% methanol fractions in yields were higher than 80% ethanol and chloroform. In AOM(active oxygen method) test, antioxidative effects of black rice added to the refined perilla oil was increased with dose-dependent fashion in concentration of the black rice extracts. Among several extracts tested, 80% aqueous methanol extract showed the highest antioxidative activity, followed by hexane, chloroform, in that order. The peroxide value and TBA value for their antioxidation stability were also lower than that of control. This result was similar to that of AOM test.

Effect of Essential Oil from San-Jo-In (Zizyphus jujuba Mill. seeds) on Human Electroencephalographic Activity (산조인(Zizyphus jujuba Mill.) 에센셜오일 흡입이 인간의 뇌파에 미치는 영향)

  • Essential oils have been used to treat psychophysiological disorders, but their effects on human electroencephalographic (EEG) activity have not been thoroughly investigated. We evaluated the effects of essential oil of Zizyphus jujuba seeds, a Korean folk medicine known as San-Jo-In, on human EEG activity. For this purpose, essential oil was extracted from San-Jo-In by the supercritical carbon dioxide extraction method. The effect of its inhalation on EEG activity was evaluated by measuring the EEG power spectrum (25 indices) in 20 healthy participants. The results of the EEG power spectrum indicated that the values of the theta wave decreased significantly (p<0.05) in the left (from 17.277 to $13.854{\mu}V$) and right parietal (from 15.324 to $13.020{\mu}V$) regions compared to the other regions. During the inhalation of San-Jo-In oil, the EEG spectrum values of fast alpha, relative gamma, and spectral edge frequency increased 50% compared to those before inhalation. The values of the fast alpha wave increased significantly (p<0.05) in the left prefrontal (from 0.063 to $0.085{\mu}V$), right prefrontal (from 0.064 to $0.085{\mu}V$), and left frontal (from 0.073 to $0.100{\mu}V$) regions following inhalation of the San-Jo-In essential oil. The changes in the EEG activities following inhalation of San-Jo-In suggest that the oil can improve psychological well-being by increasing attention and relaxation.
