흰가루병 저항성 동양종 호박 품종 육성 (Breeding for Powdery Mildew Resistant Varieties in Cucurbita moschata)

  • To breed powdery mildew resistant varieties, 104 accessions of Cucurbita spp. germplasm were evaluated. Powdery mildew resistance and horticultural characteristics of the accessions were investigated in the field conditions. C. martinezii was selected for high resistance to powdery mildew in the field. The growth of powdery mildew pathogen and response of leaf surface tissue to the pathogen were observed after artificial inoculation to resistant C martinezii and susceptible C. moschata cv. 'Wonye No.402'. Mycelium growth was very clearly different between resistant C. martinezii and susceptible C moschata cv. 'Wonye No. 402' at 5 days after inoculation at the adult stage. Interspecific hybridization was attempted to transfer powdery mildew resistance gene(s) from C. martinezii to C. moschata. A single dominant gene action was postulated as deter mining powdery mildew resistance at the seedling stage. But for the effective selection of powdery mildew resistant individuals, it was established that adult plant resistance should be confirmed in the field conditions after the seedling test. Through the seedling and field tests, C. moschata lines resistant to powdery mildew and superior to horticultural characteristics, were selected after 2 backcrossing and 3 selfing generations. The fruit characteristics of newly developed lines were similar to those of the recurrent parent, 'Jaechenjarae'.

알팔파 꽃 추출물의 Autotoxicity와 돌피에 대한 Allelopathy (Autotoxicity of alfalfa flower extract and its allelopathy to Echinochloa crus-galli)

  • 본 실험은 알파파 꽃 추출물로부터 Allelopathy와 Autotoxicity에 관련되는 활성물질을 분리, 동정하기 위하여 실시하였다. 알팔파 꽃 200g을 80% MeOH로 추출하여 $CHCl_3$층을 silica gel thin layer chromatography (TLC), microcrystalline cellulose thin layer chromatography (MCTLC)와 droplet countercurrent chromatography(DCCC)의 분획을 이용하여 생물검정을 행하고 DCCC의 생물검정에서 가장 억제력을 보였던 분획 5와 6은 HPLC로 활성물질을 동정하였다. HPLC분석 결과 분획 5에서는 ferulic acid, caffeic acid, vanillic acid, rutin, narringin 이, 분획 6에서는 ferulic acid, caffeic acid, vanillic acid, rutin, coumarin이 동정되었다. 이들 동정된 물질을 이용한 알팔파와 돌피에 대한 발아실험에서도 모두 알팔파와 돌피의 발아와 생육에 억제적으로 작용하였으며, 이중 coumarin의 처리가 가장 억제적이었다. 따라서 이들 물질이 알팔파와 돌피의 autotoxicity와 allelopathy에 관련하는 것으로 생각되었다.

플러그 트레이를 이용한 황금줄사철(Euonymus fortunei 'Emerald and Gold') 삽목 시 셀 크기 및 육묘기가 생장에 미치는 영향 (Effect of Cell Size and Seedling Raising Stage on Growth of Cuttings form Euonymus fortunei 'Emerald and Gold' Seedlings in a Plug Tray)

  • This study aimed to find a suitable size and a seedling raising stage for growing cuttings of Euonymus fortunei 'Emerald and Gold' using plug trays. The experimental method, involved cutting two nodes from a solitary branch of E. fortunei 'Emerald and Gold', and the use of 32 (143 mL/cell), 50 (70 mL/cell), 105 (18 mL/cell), 200 (13 mL/cell) plug trays. The cuttings were transplanted to trays after they were filled with a universal horticultural medium. To compare the growths, plant heights, the numbers of leaves, longest root lengths, thickness/radius ratios, dry weights, and fresh weights were measured from July to October, and statistical analyses were performed using both the two-way repeated-measures analysis of variance (ANOVA) and Tukey's post-test. The results confirmed that the size of the plug tray and the seedling raising stage had a significant effect on the growth of E. fortunei 'Emerald and Gold.' In addition, the overall growth was high and the change in growth was relatively rapid in districts 50 and 105. Therefore, it can be considered appropriate to use 50 and 105 trays when growing cuttings of E. fortunei 'Emerald and Gold' on plug trays.

보리의 수발아정도 및 탈곡방법이 종자활력 관련 형질에 미치는 영향 (Effects of Sprouting Degree and Threshing Methods on Germination and Seedling Vigor in Barley)

  • 과맥에서 종자활력의 저하요인을 구명하고자 과맥 2계통(피, 과 isogenic line포함) 및 피맥 1품종의 종자의 수발아를 유도하여 손과 기계탈곡(700 rpm ) 한 종자를 이용하였다. 1. 수발아된 종자를 손 탈곡하여 배부분의 손상이 없어서 발아세, 발아율, TTC치은 수발아 정도에 따른 차이가 적었으나, 출아력은 수발아가 클수록 감소되었다. 2. 수발아된 종자는 기계탈곡시 배의 부분적인 손상이 관찰되었으며, 손 탈곡에 비해서 종자활력에 관계된 형질들이 고도로 유의적인 감소를 가져왔고 효소활력은 크게 증가되었다. 3 손 탈곡구에서 수발아 정도는 효소활력 ( r =0.9 31**, 0.951**) 및 발아력 ( r=0.46*)과 정상관이 있었다.

새만금 간척지 밭 토양의 관개 방식별 관개용수량과 토양 용적수분함량 변화 분석 연구 (Analysis of Irrigation Amounts and Soil Volumetric Water Contents by Irrigation Method in Saemangeum Reclaimed Tideland)

  • This study was conducted to analyze changes of irrigation water and soil volumetric water content by irrigation method of field soil in Saemangeum reclaimed tideland. The main test irrigation methods was surface drip irrigation, sprinkler irrigation, and sub drip irrigation. In addition, the correlation between irrigation amounts and crop yield by irrigation method was investigated. For soil volumetric water contents increases by 25%, surface drip irrigation took 1.5 hour, sprinkler irrigation took 2.0 hours, and sub drip irrigation took 3.0 hours. As a result of analyzing the irrigation amounts according to the yield, the surface drip irrigation was 2.66 mm/day in the seedling stages, 3.31 mm/day in the vegetative growth stages, and 5.09 mm/day in the flowering stages. Sprinkler irrigation was 2.90 mm/day in the seedling stages, 3.87 mm/day in the vegetative growth stages, and 7.11 mm/day in the flowering stages. Sub drip irrigation was 2.42 mm/day in the seedling stages, 3.09 mm/day in the vegetative growth stages, and 4.87 mm/day in the flowering stages. It was analyzed that there was a statistically significant difference in irrigation amounts by fresh weight and irrigation method (F=4.002, p=0.022), and irrigation amounts by dry weight and irrigation method (F=3.499 p=0.034). Surface drip irrigation was judged to be more appropriate than sprinkler irrigation or sub drip irrigation for field crops in Saemangeum reclaimed land.

팥 포장출현력 예측을 위한 종자세 검사 (Application of Seed Vigor Test for Predicting Field Emergence in Azuki Bean (Vigna angularis Wight))

  • 경실로 인해 다른 두과작물에 비해 포장 입묘가 불량한 팥의 포장출현율의 효과적인 예측 방법을 개발하기 위해 인위로 노화 정도를 조절한 종자에 대해 실험실에서 몇 가지 종자세 검사를 실시하여 포장출현력과 비교하였다. 얻어진 종자세 지표들에 대해 포장출현력과의 상관분석을 통해 예측을 위한 효율적인 지표들을 도출하였다. 팥에서 종자세가 높을 경우에는 표준발아검사에서의 종자활력률이 포장출현력 예측에 효과적이며, 약간 노화된 종자의 경우에는 저온발아검사(CGT)에서의 비정상묘율과 배축의 길이는 포장출현력과 높은 상관관계를 보였으며, 전기전도도와 CSVT에서의 활력률과 테트라졸리움 검사에서 종자세와 예측발아율은 포장출현력과 5% 수준에서 상관관계가 인정되었다. 종자세가 낮은 종자에서는 CSVT에서 불발아종자율이 포장출현력과 5% 수준에서 상관을 보였다. 포장출현력 예측에 효과적인 몇 가지 종자세지수를 이용하여 stepwise multiple regression 분석을 실시한 결과 테트라졸리움(TZ) 검사에서의 예측발아율은 결정계수($R^2$)가 0.820으로 포장출현력 예측에 가장 효과적인 검사방법임을 알 수 있었다. 표준발아검사(SGT)에서의 종자활력률, 저온발아검사(CGT)에서의 정상묘율과 건물중을 회귀방정식에 추가함에 따라 86.9% 까지 포장출현력 예측효율을 증가시킬 수 있었다.

복합종자세지수를 이용한 보리 포장출아률과 수량예측 (The Use of Multiple Seed Vigor Indices to Predict Field Emergence and Grain Yield of Naked and Malting Barley)

  • 보리종실의 샐리적 특성을 포장출아력과 비교함으로써 포장유묘출아율과 수량의 예측에 적합한 검정방법을 구명하고자 맥주보리 2품종과 쌀보리 2품종을 인위노화처리시켜 고온발아검정(warm germination test), 저온발아검정(cold germination test), Tetrazolium 유모세검정(TZ vigor test), 전기전도도검정(electroconductivity test)의 결과를 포장출아율 및 수량과 비교 검토 하였다. 고온발아검정과 저온발아검정이 포장출아율 예측에 효과적인 검정법으로 나타났으며, 이들의 관계는 맥주보리에서는 Y(포장출아율)=-2.962+0.229X$_1$ (% warm germination) -0.001X$_2$ (vigor, WGT) +0.354X$_3$ (vigor, CT) -0.558X$_4$ (% cold germination)으로 나타났다. 다중화귀방정식의 결정계수는 맥주보리의 경우 고온발아검정에서의 발아율만으로 포장출아율을 64%예측할 수 있었다. 고온발아유묘세, 저온발아유묘세, 저온발아율은 각각 4, 7, 9%씩 결정계수를 높혔다. 네가지 검정치의 누가적 예측효율은 맥주보리에서 84%, 쌀보리 54%였다. 수량에서는 포장출아율에서 보다 맥주보리, 쌀보리 공히 예측효과가 낮게 나타났다.

  • History , achievement and future of direct-wet-rice-seeding technique in Korea and problems encountered in on-farming research were introduced. Some farmers are very eager to lower production cost and save labor. Their effort results in several direct-wet-seeding methods and seeding machines. The design problems and improvement points related to the direct -wet-rice-seeding machines were drawn out of field test.

Alantolactone이 식물생장에 미치는 영향과 L-Cysteine과의 상호작용 (Inhibitory Effect of Alantolactone on the Growth of Plant and Interaction with L-Cysteine)

  • Inhibitory effect of alantolactone and isoalantolactone was shwon in Avena straight growth test and in the formation of adventitious root in Phaseolus seedling. However, di-, and tetrahydroalantolactones given no effect on the elongation and the rooting. Inhibitory effect of alantolactone could partly be removed by cysteine, cystine, and reduced glutthione. The plant materials were made less sensitive to alantolactone by the pretreatment of cysteine, but cysteine supplied after the treatment of alantolactone brought about no effect on the action of alantolactone. A new spot was shown on TLC plate from the mixture of alantolactone and cysteine, indicating that alantolactone can be inactivated by cysteine, not cystine, without any biological processes.

Salinity Tolerance of Progenies between Korean Cultivars and IRRI's New Plant Type Lines in Rice

  • To select new germplasm for salinity tolerance from new plant type (NPT) breeding lines, the sixty F$_4$ lines selected from the crosses between Korean cultivars and IRRI's NPT lines were evaluated for salinity tolerance at the seedling stage with salinized culture solution (EC=12 dS/m) in the controlled conditions. Two NTP lines derived from a cross between 'Ilmibyeo' and 'IR66152-AC5-1', 'HR15258-7-1' and 'HR15258-27-1', were found to have good tolerance. The salinity tolerance of the lines was compared to their parents and the sensitive ('IR29') and tolerant ('Pokkali') checks in three salinity levels, no salinity (control) and an EC of 12 and 16 dS/m. Visual salinity score, shoot Na+ and Na-K ratio in two NPT lines was significantly low compared with the parents and IR29. Indicating that salinity tolerance of the lines might be derived from a transgressive segregation. The relative water content of the lines was higher than Pokkali, and the dry weight of shoot and root was proportionally decreased to salinity score and salinizing concentration. The visual salinity scores were significantly correlated with shoot Na concentration, Na-K ratio, relative water content, and reduction of dry weight (P<0.01). Their tolerance was attributed to root and shoot characteristics that led to high shoot water content, thus diluting the toxic effect of salts.
