Germination Characteristics of PEG Priming Seed in Barley (Hordeum vulgare L.) (PEG로 프라이밍 된 보리(Hordeum vulgare L.)종자의 발아특성)

  • These experiments were conducted to evaluate the variability of seed germination, and seedling growth of PEG priming barley. The average germination percentage(AGP) of PEG priming seed was higher than control, but those were low with extend the treatment period. The AGP of washing seed after PEG priming was higher than unwashed seed, and that of redried seed after priming was lower than control. The germination time of priming seed was short compare to control seed, and that was prolonged with extend the priming period, and that of washing seed after priming was shortening, and that of redried seed after priming was prolonging. The emergence percentage(EP) of priming seed was higher than control, and the emergence time was shortest in 50% field moisture capacity soil. The seedling and root length of priming seed was shorter than control.

Lilium longiflorum 'Charm' as a F1 Hybrid for Pot Plant (종자번식 일대잡종 분화용 나팔나리(Lilium longiflorum) 'Charm' 육성)

  • Lilium longiflorum 'Charm' as a $F_1$ hybrid cultivar was released by crossing inbred line '$L_2$-14' and '$L_2$-21' which were obtained from 5 self crosses originated from 'Nellie White', 'Ace' and 'Hinomoto'. The growth and flowering characteristics were evaluated in a greenhouse maintained at a minimum of $13^{\circ}C$ at night during winter in 2006 and 2007. The flower of 'Charm' is white color and horizontal-facing. The flower number of a plant and its diameter is 7.4 and 16.5 cm with 24.5 ornamental(flowering) days. The plant height is 60.3 cm with 70.3 number of leaves. The stem diameter and internode length is 1.2 cm and 1.1 cm, respectively, meaning the plant is compact and sturdy. And the number of seed per a capsule is 251.1. The results of these evaluation, therefore, suggest that seedling Lilium longiflorum 'Charm' can be used as a pot plant due to its short stems, many number of flowers per plant, long ornamental period, strong growth habit with many leaves and thick stem diameter.

Studies on Pathogenicity of Wheat Scab Fungus(Gibberella zeae) to Various Crop Seedlings

  • 1. Purpose of this experiment is to investigate the influence of wheat scab fungus(Gibberella zeae) remaining in soil or residues of infected plants to the growth of the seedlings of 14 different crops. 2. Susceptibility of these crops to the infection by G. zeae was investigated by planting these crops both to the artificially inoculated soils and naturally infected soils where both serious and light wheat scab epidemic have occurred previously. 3. In artifical inoculation tests, bailey, wheat, rye, soybean, rice, buckwheat, corn, cotton and greenbean has shown susceptible reaction, while indianbean, sesame, sorghum, chinese cabbage and radish has shown resistant reaction. 4. In artificial inoculation tests, there was significant difference in the germination ratio of the susceptible crops between the plants planted in inoculated soils and uninoculated soils with the exception of rice, com and cotton. Preemergence seedling blight was confined only to barley and corn, whereas postemergence seedling blight was confined to rye, wheat, rice buckwheat, barley and corn. la most of the susceptible crops secondary roots were almost rotted and the primary roots were either partially rotted or discolorated in inoculated soil. There was significant difference in the stem length of the plants grown in inoculated soils and uninoculated soils in susceptible crops. 5. No infection by G. zeae was observed when wheat, barley and rye plants were sown to the soils where both serious and light wheat scab epidemic occurred naturally.

Suggestion of Spring Seedling Amounts and Drone Spreader Type for Italian Ryegrass using Drones

  • The production area of Italian ryegrass feed is gradually increasing and labor-saving technologies are being developed. If sowing and fertilization are carried out agricultural drones, working hours and labor are reduced. The purpose of this study is to suggest an appropriate seedling amount for feed production by drone spreading of Italian ryegrass in spring. In addition, we would like to review the productivity of the drone seeding machine that is being developed by Rural Development Administration(RDA) of Korea. Italian ryegrass was sown by a drone in February at the NICS located in Gyehwa-hwa, Jeollabuk-do, South Korea. In Experiment 1, 50kg/ha, 60, 70, and 80 seeding rates were sown with a horizontal spreader drone. In Experiment 2, uniform spreaders type drone and horizontal spreader type were sown with the same seeding amount and compared. The drone was sown using the AF-52 aircraft. The higher the seeding amount, the higher the emergence rate. As the seeding amount increased, the plant length increased, but the number of tillers per individual decreased. The dry matter weight of the feed was the highest at 1,326kg/10a at the seeding rate of 70kg/ha, and decreased by 20.5% at the seeding rate of 80kg/ha. The coverage ratio was the highest at 96 at the seeding rate of 70kg/ha, which was the most advantageous for spring sowing. In the comparative experiment according to the spreader type, the uniform spreader had a high emergence rate per unit area. When the uniform spreader was used, the dry matter weight of the feed was 17% higher than that of the horizontal one, and the coverage was about 5% higher.

GWAS of Salt Tolerance and Drought Tolerance in Korean Wheat Core Collection

  • Abiotic stress is a major problem in global agriculture as it negatively affects crop growth, yield, and quality. Wheat (Triticum aestivum) is the world's second-highest-producing food resource, so the importance of mitigating damage caused by abiotic stress has been emerging. In this study, we performed GWAS to search for SNPs associated with salt tolerance and drought tolerance. NaCl (200 mM) treatment was performed at the seedling stage using 613 wheat varieties in Korean wheat core collection. Root length, root surface area, root average diameter, and root volume were measured. Drought stress was applied at the seedling stage, and the above phenotypes were measured. GW AS was performed for each phenotype data using the MLM, MLMM, and FarmCPU models. The best salt-tolerant wheat varieties were 'MK2402', 'Gyeongnam Geochang-1985-3698', and 'Milyang 13', showing superior root growth. The significant SNP AX-94704125 (BA00756838) were identified in all models. The genes closely located to the significant SNP were searched within ± 250 kb of the corresponding SNP. A total of 11 genes were identified within the region. NB-ARC involved in the defense response, FKSI involved in cell wall biosynthesis, and putative BP Ml involved in abiotic stress responses were discovered in the 11 genes. The best drought-tolerant wheat varieties were 'PI 534284', 'Moro of Sind', and 'CM92354-33M-0Y-0M-6Y-0B-0BGD', showing superior root growth. This study discovered SNPs associated with salt tolerance in Korean wheat core collection through GWAS. GWAS of drought tolerance is now proceeding, and the GWAS results will be represented on a poster. The SNPs identified by GWAS can be useful for studying molecular mechanisms of salt tolerance and drought tolerance in wheat.

Comparative Transcriptome Analysis of Sucrose Biosynthesis-Associated Gene Expression Using RNA-Seq at Various Growth Periods in Sugar Beet (Beta vulgaris L.)

  • Sugar beet (Beta vulgaris L.) is one of the most important sugar crops and provides up to 30% of the world's sugar production. In this study, we mainly performed RNA-sequencing to obtain identify putative genes involved in biosynthesis pathway of sucrose in sugar beet and comparative transcriptomic analyses in the four developmental stages (50, 90, 160 and 330 days after seedling). As a result of the sugar content analysis, it was increased significantly from 50 to 160 days after seedling (DAS), and then decreased at 330 DAS. On the other hand, the taproot weight, length, and width were increased during all the growth periods. Out of 21,451 genes with expressed value, 21,402 (99.77%) genes had functional descriptions. Among the three comparisons, S1 (50 DAS) vs. S2 (90 DAS), S1 vs. S3 (160 DAS), and S1 vs. S4 (330 DAS), expression profiling of the transcripts was identified 4,991 with differentially expressed genes (DEGs). By comparing the top 20 enriched gene ontology (GO) terms as three comparisons, the top GO terms were commonly confirmed with external encapsulating structure, cell wall, and extracellular regions. In addition, the 38 enriched candidate genes related to sucrose biosynthetic pathway were screened from the entire DEG pool, and the candidate genes might be providing a basic data for further sugar metabolism studies in development of sugar beet taproot.

Comparison of Growth and Survival Rate in Shortnecked Clams, Ruditapes philippinarum from Different Seedling Production Areas (산지별 바지락, Ruditapes philippinarum의 성장과 생존율 비교)

  • Growth and survival rate in shortnecked clam, Ruditapes philippinarum were studied from different seedling production areas, Hadong, Ulsan and Kochang to Kwangyang Bay, Korea, and three stocking densities (1,000, 1,500 and 3,000 individuals/$m^2$) of clam from Ulsan seedling production area were examined. During the experimental period, water temperature and specific gravity were ranged from 4.5 to $26.0^{\circ}C$ and from 1.0216 to 1.0248, respectively. Shell length was grown from $2.30{\pm}0.05\;to\;3,29{\pm}0.02$ cm in Hadong, $1.32{\pm}0.02\;to\;3.15{\pm}2.02$ cm in Ulsan, $1.24{\pm}0.03\;to\;3.18{\pm}0.02$ cm in Kochang. Kochang's seed was displayed fast growth rate than others. In different rearing densities, the lower density group was showed a rapid growth rate. Shell length from the 1,000 individuals per $m^2$ group grew longer than that from the 2,000 individuals group. Total weight from the 1,000 individuals group was higher than that from the 1,5()0 individual and the 2,000 individual groups. Survival rate by seedling production areas, Hadong, Ulsan and Kochang showed $69.4\%,\;63.8\%\;and\;41.7\%$, respectively. Survival rate by rearing density, 1,000, 1,500 and 2,000 individual groups showed $70.8\%,\;67.8\%\;and\;59.0\%$, respectively.

Growth and Histological Characteristics of Barley (Hordium vulgare L.) Seedling to NaCl Stress (NaCl Stress에 따른 보리 유묘의 생육특성 및 세포학적 반응)

  • This study was conducted to determine the morphological responses of barley seedlings to NaCl stress and to investigate histological changes of cells with transmission electron microscope(TEM) after NaCl stress. Plant height and root length of 10-day old barley seedlings with NaCl stress were reduced and inhibition level was found to be more severe in the plant height than in the root length. The leaf length, leaf width and leaf area were shorter as well with NaCl stress than without NaCl stress. However, there was no difference in the number of roots between NaCl treatments. The weight of dry matter decreased at higher NaCl concentrations, especially at 100mM NaCl. The water content of shoots tend to decrease at higher NaCl concentrations, but there was no difference in the water content of roots, The reduced sugar content was greatly increase than starch. Cellulose content was higher in NaCl stressed-plant than control, and tended to decreased at higher NaCl concentrations. Lignin content also decreased NaCl stressed-plant but there was no tendency at NaCl stress concentrations. Electric conductivity of cell sap with seedlings was high with NaCl stressed-plant. Amount of cell sap gradually increased with time in the roots than in the shoots, The grana of chloroplasts was changed by 150mM NaCl concentration. The christe of mitochondria in root meristematic sells ruined in structure and cell wall of leaf and root was also ruined by NaCl stress.

Analysis of Leaf Morphological Variation of 11 Natural Populations of Acer pictum subsp. mono (Maxim.) Ohashi (고로쇠나무 11개 자연집단의 잎의 형질 변이 분석)

  • Characteristics of leaf morphology was examined in 11 natural populations of Acer pictum subsp. mono (Maxim.) Ohashi in Korea. Nested Anova showed that there were statistically significant differences among populations as well as among individuals within populations in all 14 quantitative characters. In all the leaf characters, variance components among individuals within populations were higher than those among populations. Some morphological characters such as the ration of Blade length to Petiole length (0.90), Uower lobe width (0.85), Blade length (0.83), and Middle lobe width (0.83) showed the relative high differences in the values of differentiation (Dij) among populations. Of the ten populations, Gwangyang (1.36) and Cheongyang (1.30) were highly differentiated based on the total mean Dij of fourteen morphological characters, while Ulju (0.30) and Jeongseon (0.44) were scarcely differentiated.

Effects of seed sources and shade on vigor of Brant's oak seedling

  • The use of local seed provenance is often recommended in forest restoration. Early vigor is a combination of the performance of seed germination and emergence after planting. The ability of young Brant's oak plants to grow and develop after emergence and its dependence on local habitat conditions was investigated in this study. The effects of seed source and shade on early growing seedlings of Brant's oak (Quercus brantii L.) were determined in field measurements. Seeds of Quercus brantii L. were collected from 4 forest areas (seed sources) in southern Zagros (Provinces of Kohkilouyeh-Bouyer Ahmad and Fars) at altitudes of 850, 1,100, 1,500, 2,100 m a.s.l., and planted in a nursery constructed in southwestern Iran. According to a split-plot design consisting of four blocks, each containing two main treatment plots (no shading, partial shading), each main plot was sub-divided into four sub-plots (for elevations of 850, 1,100, 1,500 and 2,100 m). Results showed that shade treatments had significant effects on emergence percentage and rate, shoot length, shoot dry weight (SDW), root dry weight (RDW), leaf area (LA), and chlorophyll content. Ecological factors also had an effect on seed performance. Altitude of seed source had a very significant effect on root length, LA, SDW, and RDW. The seeds collected from 850 m a.s.l. elevation showed the highest performance, especially in leaf area, root length, shoot dry weight, and root dry weight. Our results showed that the altitude of 850 m a.s.l. was the best for collecting Brant's oak seeds.
