Effect of covering materials on seedling quality of rice (못자리 피복재가 벼의 묘소질에 미치는 영향)

  • This study was carried out to investigate the availability of covering material used for raising seedlings in the rice nursery bed. The pre-germinated seeds of a rice cultivar 'Chilbo' were sown into seedling tray ($60{\times}30{\times}2.5cm$) on 20th April and 3rd May 2010. Polypropylene spunbonded fabric (PSF) and lagging were used for covering materials in rice nursery bed. The growth characteristics of seedlings were evaluated at 8, 15 and 25 days after sowing (DAS) in the nursery beds covered with PSF and lagging, respectively. The plant height of 8-day old seedlings grown in lagging nursery bed was longer by 2cm than those in PSF nursery bed when the seeds were sown on 20th April, but seedling quality grown in lagging nursery bed was poor compared with that in PSF nursery bed. The growth characteristics of 15-day old seedlings were similar to that of 8-day old seedlings. The plant height of 8, 15 and 25-day old seedlings grown in lagging nursery was longer than those in PSF nursery when the seeds were sown on 3rd May. The seedling quality grown in PSF nursery was better than that in lagging nursery. The seedlings grown in PSF nursery have more leaves and better seedling quality compared with that of lagging nursery. The seedling quality was remarkably decreased with the increase of nursery period in lagging bed but seedlings quality were recovered at 5 days after hardening treatment by removing the lagging.

Salt Tolerance of Vigna angularis during Germination and Early Seedling Growth

  • The present study was undertaken in investigate the response to salinity and effect of plant growth regulators and proline under salinity stress on the germination and seedling growth of Vigna angularis. The protective effect of external Ca2+ on root elongation under saline conditions was also investigated. The seed germination of Vigna angularis decreased with an increase in salinity. The growth regulators GA3 was more effective than kinetin. At a higher salinity, low concentrations of kinetin and high concentrations of GA3 were more effective. The external application of proline and betaine improved germination under saline conditions. At a low salinity proline and betaine alleviated the salinity-induced inhibition of germination, yet at higher NaCl concentrations, proline and betaine were both ineffective. Exposure to salinity during germination was accompanied by an increase in the proline content, thereby suggesting that one compatible solute in the germinating seed would seem to be proline. The inhibition of germination by high NaCl concentrations was relatively more severe in scarified seeds than in intact seeds, indicating that the seed coat acts as a partial barrier to an Na2+ ameliorated the adverse effect of salinity stress.

  • Experiments were carried out to asses the impotance of environmental conditions upon the growth rate, length compositions seedling and spore liberation of a red alga Glacilaric verrucosa. The alga grown in water of low salinity mixed with fresh water shows better growth rate, lager length composition and excellent seedling, whereas the one vegetated in an area of tidal exposure shows a somewhat retarded growth rate and poor seedlings. The rate of growth Increases from August to October and begins to diminish from December continuing to diminish through February. Therefore the spore settling time extends from May to lune. Although the sun-drying method is the most eacient way of inducing spore liberation, further investigations are necessary in oiler to use this method for the application to artificial spore settling.

Phytotoxic Effect of Lettuce (Lactuca sativa L.) Leaf Extract on Seedling Growth of Crops and Weeds

  • Lettuce (Lactuca sativa L.) is known to contain water-soluble substances that are biologically active. Aqueous or methanol extracts and residues from leaves of lettuce plants were assayed to determine their allelopathic effects, and the causative allelochemicals from fractions were quantified by means of HPLC analysis and bioassayed. Extracts from oven-dried leaf samples were more phytotoxic than those from freeze-dried samples. Leaf extracts of 40 g L$^{-1}$ were completely inhibitory on root growth of alfalfa (Medicago sativa L.), while root growths of barley (Hordeum vulgare L.) and soybean (Glycine max L.) were less sensitive. Early seedling growth of both alfalfa and barnyard grass (Echinochloa crus-galli) was significantly reduced by methanol leaf extracts. The major allelopathic substances analyzed by HPLC were coumarin, trans-cinnamic acid, o-coumaric acid, p-coumaric acid and chlorogenic acid. Of them p-coumaric acid was found as the greatest amount (8.9 mg 100 g$^{-1}$ ) in the EtOAc fraction; only coumarin was found in all the fractions. Hexane and EtOAc fractions of L. sativa reduced alfalfa root growth more than did BuOH and water fractions. These results suggest that lettuce had potent herbicidal activity, and that its activity differed depending on type and amount of causative compounds by fraction.

Effects of different day length and wind conditions to the seedling growth performance of Phragmites australis

  • Background: To understand shade and wind effects on seedling traits of common reed (Phragmites australis), we conducted a mesocosm experiment manipulating day length (10 h daytime a day as open canopy conditions or 6 h daytime a day as partially closed canopy conditions) and wind speed (0 m/s as windless conditions or 4 m/s as windy conditions). Results: Most values of functional traits of leaf blades, culms, and biomass production of P. australis were higher under long day length. In particular, we found sole positive effects of long day length in several functional traits such as internode and leaf blade lengths and the values of above-ground dry weight (DW), rhizome DW, and total DW. Wind-induced effects on functional traits were different depending on functional traits. Wind contributed to relatively low values of chlorophyll contents, angles between leaf blades, mean culm height, and maximum culm height. In contrast, wind contributed to relatively high values of culm density and below-ground DW. Conclusions: Although wind appeared to inhibit the vertical growth of P. australis through physiological and morphological changes in leaf blades, it seemed that P. australis might compensate the inhibited vertical growth with increased horizontal growth such as more numerous culms, indicating a highly adaptive characteristic of P. australis in terms of phenotypic plasticity under windy environments.

Development of Raising Seedling Technology for Veronica pyrethrina Nakai Using Plug Trays

  • Background and objective: This study was carried out to develop an effective technique for raising seedlings of Veronica pyrethrina Nakai, a native plant species in the Korean Peninsula, in plug trays. Methods: To investigate the optimum plug cell size and sowing media, we sowed seed in to plug trays with 34, 21, and 10 mL cells and filled with a commercial horticultural substrate and mixtures of peatmoss and perlite in 1:1, 3:1, and 4:1 ratios. Fertilization levels were set at 0, 500, 1000 and 2000 mg·L-1. Results: Plug cell size did not significantly influence the seedling growth of V. pyrethrina. By substrate type, the growth rate was highest in the horticultural substrate, followed by 4:1, 3:1, and 1:1. Growth by fertilization level was higher in all fertilized treatment groups than in the control group, and there was no difference among 500, 1000, and 2000 mg·L-1. Conclusion: The results of this study proved that it is most suitable for raising seedlings of V. pyrethrina to sow the seeds in a 10 mL cell plug tray filled with horticultural substrates, and apply fertilizers with less than 500 mg·L-1 concentration.

Analysis of Antioxidant Enzyme Activity During Seedling Growth of Leymus chinensis Trin Under Salt and Dehydration Stresses (고염과 건조 스트레스 처리 조건 동안 양초 유식물체의 항산화효소 활성 분석)

  • To understand the adaptability of Leymus chinensis forage grass to environmental stresses, we analyzed the $H_2O_2$ scavenging activity based on several antioxidant enzymes and total phenolics content, including peroxidase (POD), ascorbate peroxidase (APX), and catalase (CAT), in shoots and roots subjected to salt and dehydration stresses during seedling growth. After NaCl or PEG treatment, plants showed reduced seedling growth under over 200 mM NaCl or 30% PEG treatment condition in shoots and roots compared with the control condition. In addition, plants showed high enzymatic activity of CAT in the shoots, whereas they exhibited high activity levels of APX and POD in the roots in both the NaCl and PEG treatment conditions. These results seem to indicate that Leymus chinensis seedlings responding to salt and dehydration stresses during initial growth is associated with enhanced activity of $H_2O_2$ scavenging antioxidant enzymes in the shoots or roots. The plants also showed high levels of total phenolics under NaCl treatment, with a high concentration in both the shoots and roots. Our results showed that the induced activity patterns of APX in the roots and CAT in the shoots indicate that low $H_2O_2$ levels were mainly maintained through tissue-specific redox homeostasis involving enzymes such as APX and CAT during salt and dehydration stresses. This study highlights the importance of antioxidant enzymes in the establishment of Leymus chinensis seedlings under high salinity conditions, such as typical desertification.

Leaf Characteristics and Growth Performance in Progenies of Hovenia dulcis var. koreana Nakai (헛개나무 선발개체 차대의 생장 및 엽특성)

  • This study was conducted by estimate of leaf characteristics and growth performance among selected 20 families progenies of Hovenia dulcis var. koreana Nakai. Mean seedling height and root-collar diameter in whole progenies showed 61.6cm, 6.98mm, respectively. Mean seedling height of KW 2-5 family was 79.3cm and it was higher than that of other families. JN 2-2 family had highest value, 9.16mm, in mean of root-collar diameter. Growth performances of KW 2-5 and JN 2-2 families showed 57% (in seedling height), 53% (in root-collar diameter) superior to those of lowest marked families. The selection level based on growth performances, which are seedling height over 75.0cm and root-collar diameter over 8.0mm, were applied on whole families, and two families selected (KW 2-5, JN 2-2). The selection effects from selected families was evaluated as 125% compared to the mean of whole families. The coefficient of variation (C.V) appeared wide range, $9.4{\sim}42.4%$, in estimated leaf characteristics. Particularly, C.V of petiole length (PL) and petiole diameter (PD) showed 42.4% and 28.7%, respectively. Their were higher value than other leaf characteristics. Analysis of variance for all leaf characteristics were significantly different among families. Also, there had positive correlation between most leaf characteristics except the correlation between leaf length (LL) and leaf morphological index (LMI, LW/LL). Particularly, the correlation between leaf length and leaf width (LW) showed the highest correlation.

Effect of Growth Medium and Extract Sterilization on a Seedling Bioassay of Alfalfa Autotoxicity (생장배지 및 추출액의 멸균이 알팔파 Autotoxicity 의 유묘 생물검정에 미치는 영향)

  • Autotoxic substance(s) from alfalfa(Medicago sativa L.) plants reduces germination and growth of adjacent new alfalfa after alfalfa. The autotoxic chemical(s) in alfalfa are clearly unknown. Our objective was to improve the sensitivity of an alfalfa seedling bioassay for evaluating autotoxic leaf extracts. We determined critical extract concentrations that inhibit seed germination and seedling growth, compared two different culture media, and evaluated the effects of extract sterilization on the sensitivity of the assay, by using streptomycin and autoclaving method. An agar medium in petri plate gave better responses of germination and seedling growth to the extracts than using filter paper in the plate. On agar medium, the concentration of extract required to reach 50% inhibition of root length was 2.7 g $kg^{-1}$, and of germination and hypocotyl length were 3.8 and 9.9 g $kg^{-1}$, respectively. Leaf extracts with 100 ppm streptomycin stimulated germination significantly compared to Leaf extract alone but reduced root length of control by 43%. Root length was more sensitive to the autotoxin(s) than was germination or hypocotyl length. These results suggest that agar medium mixed with extract and sterilization by autoclaving could be improved the consistency and precision of bioassay, and that root length was the best parameter of autotoxic effect of alfalfa leaf extract.

Effects of Solvent-extracts Extracted from Coptis chinensis on Seed Germination and Seedling Growth (종자발아 및 유묘생장에 미치는 황련(Coptis chinensis) 추출물의 영향)

  • For searching the natural herbicide-components, the seed germination and seedling growth of receptor plant species (Brassica campestris, Sesamum indicum, Perilla frutescens and Echinochloa crus-galli) were investigated in four solvent-extracts extracted from Coptis chinensis Franch. The seed germination of receptor plant species was largely inhibited in 2,000 ppm of ethyl acetate compared to the control, and it was inhibited in order of P. frutescens, B. campestris, E. crux-galli, and S. indicum. In seedling growth, the shoot and root elongations of receptor plant species were inhibited in order of S. indicum, P. frutescens, B. campostris, and E. crus-galli. Root elongation was remarkably reduced in order of $H_2O$, butyl alcohol, and hexane, ethyl acetate extracts. Of four receptor plant species, seed germination and seedling growth of B. campestris and S. indicum showed the species-specific reaction to the solvent-extracts extracted from C. chinemis. $H_2O$ extract had a natural herbicide potential to the seed germination or root elongation in B. campestris and S. indicum. The result can be provided a basic data f3r the development of natural herbicide.
