For the comparision of the differences in dry matter weight and the chemical composition of soybean seeds in relation to utilization styles and storage durations, the seed coats and cotyledons including embryo axis separated from the seeds of 3 recommended cultivars stored for 3 years and of 5 recommended cultivars and 2 local varieties with black seed coat and green cotyledons were observed in dry weight, germination percentage, protein, oil, soluble sugar, oilgosaccharide, and starch. In the seeds stored for 3 years, dry weight and germination rate were decreased distinctively. This decrease of seed dry matter resulted from the decrease of the cotyledon weight, and the ratio of seed coat to whole seed was increased. For the change of chemical compositions by the decrement of seed dry weight during the storage, protein, oil, and carbohydrate contents of the seed coat were decreased, and protein content of the cotyledon was markedly decreased in the seeds stored for 3 years. The ratio of seed coat to whole seed was higher in small seed than large seed cultivars. In the local varieties, protein and oil contents were 43.59% and 18.25% in the cotyledons, respectively, which were lower than those of recommended cultivars. But starch and sugar contents were 5.70% and 11.58%, respectively, which were higher than those of recommended cultivars. Sucrose, raffinose and stachyose contents were also higher in the cotyledon of the local varieties.
Buffer soluble storage proteins of ginseng seed have been localized by electron microscopy using post-embedding immunocytochemical gold labelling technique. Major components of the storage proteins were revealed to be storage protein-1($SP_{1}$, MW 160,000) and storage protein-2($SP_{2}$, MW 70,000). Both of the storage proteins are glycoproteins. Anti-$SP_{1}$ and anti-$SP_{2}$ from rabbit, against $SP_1$ and $SP_2$, respectively, reacted on sections of ginseng endosperm tissue embedded in Spurr's epoxy resin. The rabbit antibodies were visualized indirectly by reaction with protein A labelled with colloidal gold. Both storage proteins were found to be accumulated together in the same protein bodies, but their relative contents are not equal.
Seed morphology, physiology and environmental conditions have influence on germination of any tropical seeds and their appropriate handling, processing and handling enhances seeds emergent. This study therefore investigated effects of storage durations under different temperatures on germination of Bombax costatum seeds from different provenances. Fresh 25 seeds of B. costatum in four replicates were collected from four different provenances (Aponmu, Oluwa, Ibadan and Oyo). They were surface sterilized, thoroughly washed in distilled water and stored at four different temperature regimes: 28℃, 21℃, 5℃ and -17℃. Samples were taken every 2 months for germination test for 18 months. Germination assessment was carried out daily and recorded. The experimental design was 4×4×10 factorial in 4 replicates. Data were subjected to percentages and analysis of variance (ANOVA). Results showed that there were significant differences among storage temperatures, storage durations and provenances on germination percentage. Freshly collected seeds from Aponmu and Oyo had the highest mean germination of 100% followed by seeds from Ibadan (89.3%). Seeds from Oyo provenance stored 5℃ had germination of 94.0%, 88.70% and 78.7% at 2nd, 4th and 6th month respectively. Seeds from Ibadan and Oyo stored at 28℃, 21℃ recorded 0.0% starting from 8 months of storage. Germination of B. costatum seeds from the four provenances decreased with increase in storage duration at different temperatures.

Understanding the mechanism(s) to overcome or prevent seed ageing deterioration during storage is of fundamental interest to seed physiologists. Vitamin E (tocols) is known as a key metabolite to efficiently scavenge lipid peroxy radicals which cause membrane breakdown resulting in seed ageing. However, in rice research this hypothesis has been tested for very few lines only without considering intraspecific variation in genomic structure. Here, we present a correlation study between tocols and seed longevity using a diverse rice panel. Seeds of 20 rice accessions held in the International Rice Genebank at the International Rice Research Institute, representing aus, indica, temperate japonica and tropical japonica subpopulations, were used for tocols analysis (quantification of ${\alpha}$-, ${\beta}$-, ${\gamma}$-, ${\delta}$-tocopherol/tocotrienol by ultra performance liquid chromatography) and storage experiments at $45^{\circ}C$ and 10.9% seed moisture content (sample taken for germination testing every 3 days up to 60 days). To examine interactions between DNA sequences and phenotype, the 700k high-density single-nucleotide polymorphism marker data-set was utilized. Both seed longevity (time for viability to fall to 50%; $p_{50}$) and tocols content varied across subpopulations due to heterogeneity in the genetic architecture. Among eight types of tocol homologues, ${\alpha}$-tocopherol and ${\gamma}$-tocotrienol were significantly correlated with $p_{50}$ (negatively and positively, respectively). While temperate japonica varieties were most abundant in ${\alpha}$-tocopherol, indica varieties recorded 1.3 to 1.7-fold higher ${\gamma}$-tocotrienol than those of other subpopulations. It was highlighted that specific ratio of tocol homologues rather than total tocols content plays an important role in the seed longevity mechanism.
Seed viability is affected by storage conditions and rapid loss of viability in storage is the major cause for low germination. This study was carried out to examine the effect of packaging materials and storage temperature on seed germination rate over 10 years in two species (Capsicum annuum L. and Brassica rapa L. ssp. pekinensis) and determine effective storage conditions for maintaining seed viability. Seeds were packaged in aluminum poly pouches under vacuum, polyethylene bottles, and paper bags containing silica gel and stored under one of two controlled conditions ($15^{\circ}C$, RH 40% or $5^{\circ}C$, RH 30%) or at ambient condition. Seed germination was recorded at 6-month intervals for 10 years. The seeds of both species showed no decline in viability until 6.5 years at 15 or $5^{\circ}C$, irrespective of packaging materials. However, under ambient conditions, the seeds of chili pepper and Chinese cabbage in paper bags lost viability after 4 and 5 years, respectively. By contrast, seeds of both species in vacuum-aluminum poly pouches exhibited a 99% germination rate after 6 years under ambient conditions. Pepper seeds in the vacuum-aluminum poly pouches maintained a 93% germination rate after 10 years in ambient conditions. These results indicated that a special seed storage facility for maintaining viability of chili pepper and Chinese cabbage seed might not be essential and seed testing would not be necessary for 10 years, if chili pepper and Chinese cabbage seeds were packed in ambient/vacuum-aluminum poly pouches or $5^{\circ}C$/vacuum-aluminum poly pouches.
This study investigated the effect of the powder produced by ball-milling the outer layer of annatto (Bixa orellana L.) seeds on the physicochemical properties as well as the antioxidant and antimicrobial activities of pork patties over 14 d of refrigerated storage (4±1℃). Five pork patty treatments were produced containing three different concentrations of annatto seeds, 0.1, 0.25 and 0.5% (ANT0.1, ANT0.25, ANT0.5), 0.1% ascorbic acid (AA0.1), and a control (CTL). Based on the results, annatto seed powder appeared to show antioxidant activity. The Hunter color values of pork patties were affected by the addition of annatto seed powder, which increased the redness and yellowness values, but decreased the lightness of the patties (p<0.05). To evaluate the antioxidative effects of annatto on pork patties, thiobarbituric acid reactive substances (TBARS) and peroxide values (POV) were analyzed over 14 d of refrigerated storage. Treatments containing annatto seed showed lower TBARS and POV than control (CTL) samples (p<0.05). The volatile basic nitrogen (VBN) of the pork patties containing annatto seeds were lower than that of CTL at the end of storage (p<0.05). Although no differences in total bacterial counts were observed between control and treated patties, those containing annatto seeds had lower microbial counts for Enterobacteriacease than CTL or AA 0.1%. Therefore, annatto seed powder might be a good source of natural antioxidants for the production of meat products.

Green stem disorder (GSD) of soybean (Glycine max (L.) Merr.) is characterized by delayed senescence of stems with normal pod ripening and seed maturation (Hobbs, 2006). GSD complicates harvesting of soybeans by significantly increasing the difficulty in cutting the affected plants. There is also the potential for moisture in the stems to be scattered on the seed, reducing the grade and storability of the seed. Not only the cause of GSD is yet unknown, but also GSD cannot be evaluated until maturity, therefore the method to evaluate GSD in early growth stage with high sensitivity is necessary. In previous studies, it has been reported that vegetative storage protein (VSP) accumulates and the syndrome of GSD appears in soybean after depod treatment (Fischer, 1999). Soybean VSP is a storage protein which is abundant in young sink leaves and degraded during seed fill (Wittenbach, 1982). Hence, we have established a system to quantify VSP of high sensitivity by using standard protein made by genetically transformed E. coli and specific antibody against VSP, and studied the relationship between VSP and GSD, by depod experiment and drought/excess wet experiments. The result of depod experiment with the cultivar 'Yukihomare' was the same with the previous studies, VSP accumulated much more than control and the syndrome of GSD appeared in soybean in depod treatment. Drought and excess wet had different impact on GSD. Excess wet caused GSD of the cultivar 'Tachinagaha (GSD susceptible)', while drought caused a little syndrome of GSD in the cultivar 'Touhoku 129 (GSD resistant)'. The accumulation of VSP differed between the two cultivars over time. In conclusion, the accumulation of VSP came along with the emergence of GSD. Different cultivars showed different response to drought and excess wet. In the future, it is expected that the dynamics of VSP will be elucidated in detail, leading to the development of early diagnosis technology for green stem disorder and the elucidation of mechanism of soybean GSD.
This study was conducted to determine the effects of seed storage methods ($-20^{\circ}C$, stratification, $2^{\circ}C$ dry, $2^{\circ}C$ wet and room temperature) and $GA_3$ application (control, $dH_2O$, 10, 100, 1000 ppm) on seed germination and seedling growth of S. lyratum. As a result seed germination rate of S. lyratum was the highest at 91% when seeds were stored at room temperature and then soaked for 24 hours in $GA_3$ 10 ppm. And seedlings of S. lyratum showed the best quality when seed were stored at $2^{\circ}C$ dry and then soaked for 24 hours in $GA_3$ 1000 ppm, with the growth characteristics of plant height (47 mm), number of leaves (8), leaf width (12 mm), leaf length (19 mm), fresh weight (aerial/root part; 471/476 mg), dry weight (aerial/root; 106/41 mg) and seedling quality indices (106). Therefore, S. lyratum seed were stored at $2^{\circ}C$ dry, and then soaked for 24 hours in $GA_3$ 1000 ppm, seed germination rate was more than 80% and production of superior quality container seedlings.
This study was conducted to determine the yield, free radical scavengering effect and total phenol contents of various solvent fractions on the crude and defatted grape seed extract during storage. The optimal condition for the extraction yield, free radical scavengering effect and total phenol contents was $90\%$ ethanol for 6 hour at $70^{\circ}C$. The extraction yield for crude and defatted grape seed at optimal condition was $8.9\%\;and\;9.16\%$, respectively. Also, the strongest free radical scavengering effect with $41.52\;{\mu}g/mL$ was observed in $95\%$ ethanol of defatted grape seed extracted for 6 hour at $70^{\circ}C$. Similar result was observed in total phenol contents of defatted grape seed. The ethyl acetate fraction obtained from ethanol extract of defatted grape seed showed the strongest RC50($12.35\;{\mu}g/mL$) compared to other organic fractions. Free radical scavengering effect of crude and defatted grape seed extracts treated with alkali condition(pH 10) was reduced compared to that of acidic condition(pH 2) during storage far 1 month at $50^{\circ}C$. Overall, more stronger free radical scavengering effect and higher total phenol contents in defatted grape seed extracts was observed than that of crude grape seed.
Seed storage proteins (SSPs) are important for seed germination and early seedling growth. We studied the accumulation and degradation profiles of four major Arabidopsis 12S SSPs using a 2-DE scheme combined with mass spectrometric methods. On the 2-DE map of 23 dpa (days post anthesis) siliques, 48 protein spots were identified as putative full-length or partial $\alpha$, $\delta$ subunits. Only 9 of them were found in 12 dpa siliques with none in younger than 8 dpa siliques, indicating that the accumulation of 12S SSPs started after the completion of cell elongation processes both in siliques and in developing seeds. The length and strength of transcription activity for each gene determined the final contents of respective SSP. At the beginning of imbibition, 68 SSP spots were identified while only 2 spots were found at the end of the 4 d germination period, with $\alpha$, subunits degraded more rapidly than the $\alpha$ subunits. The CRC $\delta$ subunit was found to degrade from its C-terminus with conserved sequence motifs. Our data provide an important basis for understanding the nutritional value of developing plant seeds and may serve as a useful platform for other species.
