Indica Rice Grown in Korea (우리나라에 재배된 Indica벼)

  • A few Korean native rice cultivars, 'Sharei' and red rice which are regarded as weedy rice and a rice sample enshrined in the Buddha's Image were examined for the grain shape, seed coat color, amylose content, alkali digestibility and esterase response pattern. Among the Korean native varieties which are collected during 1920's and preserved up to now, there are some varieties belong to the typical Indicas. Some lines of 'Sharei' and some collections of red rice from farmer's field showed the evidence of Indica rice. A rice sample which was enshrined in the Buddha's image in 1302 looked like a mixtures of Indica and Japonica. Reviewing the articles from the old books describing characteristics and some reports on the Indica rice in Korea, it was deduced that the Indica rice was grown in Korea in some extent before 1300.

Inhibitory effect of natural extract mixtures on microbial growth and lipid oxidation of sausages during storage

  • Large amounts of additives are used during meat product processing to maintain product quality and shelf life. With the growing interest in healthy foods, natural plant-based additives are being used as alternatives to synthetic additives. In this study, six types of natural extracts with excellent antibacterial activity were selected, and their antibacterial and antioxidant activities against four types of pathogens were evaluated in various combinations. In addition, the pH, color, amount of thiobarbituric acid reactive substances (TBARS), and growth of pathogenic microorganisms were analyzed during the storage of sausages treated with various combinations of these extracts. The natural extract mixtures exhibited different antibacterial activities, depending on the combination. Compared to grapefruit seed extract, a mixture of natural extracts extracted with ethanol (M4) reduced the Escherichia coli content by more than 99.9% after 8 days of storage and slowed the growth of Listeria monocytogenes and Salmonella spp. by more than 80% after 14 days. Compared to untreated (NC) and grapefruit extract (PC)-treated sausages, sausages treated with the natural extract mixtures showed a significant decrease in CIE L* and an increase in CIE a* and CIE b* (p < 0.05). The pH value was significantly lower in sausages containing natural extract mixtures than in the NC and PC sausages (p < 0.05). The natural plant extract mixtures significantly prevented lipid oxidation (p < 0.05). In summary, different types of natural extract mixtures have a synergistic effect when used together, suggesting that natural preservatives can generally inhibit the growth of microorganisms and oxidation of processed meat.

Isolation and Characterization of Lipoxygenase-producing Bacteria for Industrial Applications (산업적 응용을 위한 Lipoxygenase 생산 세균의 분리 및 특성)

  • Lipoxygenase is an enzyme, mainly produced by plants, capable of converting unsaturated fatty acids to fatty acids. It has vast application potential in the food, pharmaceutical and agricultural industries. The aim of this study was to isolate novel lipoxygenase-producing bacteria from the environment and to investigate the lipoxygenase enzymatic properties for industrial production. The strain, NC1, isolated from cultivation soils, was identified as Bacillus subtilis based on the phenotypic characteristics and 16S rRNA gene sequencing. This strain formed a pink color around the colony when cultured on indamine dye formation plates. The production of lipoxygenase by B. subtilis NC1 was influenced by the composition of the medium and linoleic acid concentrations. The optimum temperature and pH for lipoxygenase activity was determined to be 40 ℃ and pH 6, respectively. The enzyme showed relatively high stability at temperatures ranging from 20-50 ℃ and acid-neutral regions. In addition, the lipoxygenase produced by B. subtilis NC1 was able to degrade commercially available oils including sunflower seed oil and Perilla oil. In this study, a useful indigenous bacterium was isolated, and the fundamental physicochemical data of bacterial lipoxygenase giving it industrial potential are presented.

Characterization of Purple-discolored, Uppermost Leaves of Soybean; QTL Mapping, HyperspectraI Imaging, and TEM Observation

  • Purple-discoloration of the uppermost leaves has been observed in some soybean cultivars in recent years. The purpose of this study was to characterize the novel phenotypic changes between the uppermost and middle leaves via multiple approaches. First, quantitative trait loci mapping was conducted to detect loci associated with the novel phenotype using 85 recombinant inbred lines (RILs) of the 'Daepung' × PI 96983 population. 180K SNP data, a major quantitative trait locus (QTL) was identified at around 60 cM of chromosome 6, which accounts for 56% of total phenotypic variance. The genomic interval is about ~700kb, and a list of annotated genes includes the T-gene which is known to control pubescence and seed coat color and is presumed to encode flavonoid 35-hydroxylase (F3'H). Based on Hyperspectral imaging, the reflectance at 528-554 nm wavelength band was extremely reduced in the uppermost leaves compared to the middle (green leaves), which is presumed die to the accumulation of anthocyanins. In addition, purple-discolored leaf tissues were observed and compared to normal leaves using a transmission electronic microscope (TEM). Base on observations of the cell organelles, the purple-discolored uppermost leaves had many pigments formed in the epidermal cells unlike the normal middle leaves, and the cell wall thickness was twice as thick in the discolored leaves. The thickness of the thylakoid layer in the chloroplast the number of starch grains, the size of starch all decreased in the discolored leaves, while the number of plastoglobule and mitochondria increased.

Effects of CLA-vegetable Oils and CLA-lard Additives on Quality Characteristics of Emulsion-type Sausage (유화형 Sausage의 품질특성에 식물성유와 동물성유 CLA 첨가가 미치는 영향)

  • Emulsion-type sausages were manufactured to investigate the effects of CLA-vegetable oils and CLA-lard on quality characteristics of emulsion-type sausage. Each treatments replaced pork back fat with CLA-corn oil (CLA-CO), CLA-safflower seed oil (CLA-SSO) and CLA-lard (CLA-LD) were stored during 1, 7, 14, 21 and 28 days at 4$^{\circ}C$. The changes in physico-chemical properties, texture, thiobarbituric acid reactive substances (TBARS), fatty acid composition and CLA content of each treatments were measured during 1, 7, 14, 21 and 28 days at 4$^{\circ}C$. During storage, pH of control showed higher pH value than that of CLA-vegetable and CLA-lard treatments. The pH values of all treatments was significantly low at storage 14 days (p<0.05). The color L* and a*-value of all the treatments was not significantly changed with the storage periods. Color b*-value of CLA-treatments was higher than that of control during the storage period. Fatty acids composition was changed by addition of CLA-vegetable oils and CLA-lard. All kinds of fatty acids composition were decreased with CLA replacements than that of the control. Whereas linolenic acid content extremely increased by replacement of CLA-vegetable. CLA content of CLA-treatments was extremely increased than that of the control. The cohesiveness, springness, gumminess and brittleness of CLA-LD treatment was significantly higher (P<0.05) than that of the other treatments. The TBARS value of all treatments were significantly increased (P<0.05) as the storage period was passed. TBARS of sausage products containing CLA-CO was the lowest. Summing up the a forementioned results, emulsion-type sausage manufacturing with CLA-vegetable oil and CLA-lard was not affected in physico-chemical properties and texture characteristics. Also, it may be assumed that the high quality sausage can be manufactured with the extent of storage period and CLA accumulation.

The Quality Characteristics of Fermented and Soaked Jujube Wine (발효 및 침출 대추주의 품질 특성)

  • The chemical composition characteristics of jujube wine using different preparation methods including extraction in -soju (25%) and fermentation were investigated. The jujube wine was prepared by soaking in soju using- whole fruit(WJ-S1) or seed-removed fruit(WJ-S2). Fermentations of the jujube wine were started by using whole fruit(WJ-F1), seed-removed fruit(WJ-F2) or extracts of whole fruit(WJ-F3) after adding 24% sucrose. The pH of all samples decreased from a range of 4.92~5.42 at the start time to 3.66~4.97 after 100 days. The treatment WJ-F3 showed the lowest pH among all treatments. Total acid content was 0.67~1.01% at the initial stage and then changed to 0.51~0.88% after 100 days. Total sugar and reducing sugar contents were 1.20~13.8% and 0.50~4.45% at initial stage and then changed to 1.53~4.52% and 1.75~3.82% after 100 days respectively. These sugars decreased during the preparation and fermentation of the jujube wine. The amounts of free sugars including fructose, glucose, and sucrose ranged from 1.53~4.52% and treatment WJ-F3 showed the highest amount(1.95~13.64%) compared the other treatment. Glucose level were high in treatment WJ-S1 and WJ-S2, and fructose was increased in treatments WJ-F1, WJ-F2, and WJ-F3 after 60 days. Amino nitrogen content was 0.02~0.11% after 20 days and 0.07~0.14% after 40 days respectively. Solid content was 2.68~7.76% at the initial fermentation stage and changed to 4.81~9.73% after 100 days. Hunter color(L values) were 88.45~96.74 at the initial stage and then changed to 92.62~100.45 after 100 days. Preference tests a significant difference between the two types (extraction and fermentation) of jujube wine. And treatment WJ-S2 received the highest preference scores among the all treatments.

Morphological Variation of Foxtail Millet (Setaria italica (L.) P. Beauv.) Germplasm Collected in Korea, China and Pakistan (우리나라와 중국, 파키스탄에서 수집한 조 계통들에 대한 형태적 변이)

  • To understand the morphological differentiation of the 26 accessions of Foxtail Millet collected in Korea (15 accessions), China (7 accessions) and Pakistan (4 accessions), we analyzed 9 morphological characteristics such as plant height, panicle length, leaf number, tiller number, heading time, seed weight and panicle color etc. Most accessions of foxtail millet collected in Korea showed late heading time, tall plant height and long conical panicles. While foxtail millet accessions of Pakistan showed early heading time, short plant height and short conical panicles. In case of Chinese accessions, some accessions of them showed similar characteristics with Korean accessions, and the other showed similar characteristics to Pakistan accessions. In ANOVA analysis, most of quantitative characteristics such as plant height, leaf number, internode number and heading time showed significant differences among foxtail millet accessions collected from Korea, China and Pakistan. Principal component analyses clearly discriminate foxtail millet accessions of Korea from those of China and Pakistan. In PCA analysis, most of quantitative characters such as panicle length, leaf number and internode number greatly contributed in positive direction, whereas several quantitative characters such as tiller number, seed weight and panicle color contributed in negative direction on the first axis. Thus, these morphological characteristics could be used to classify the foxtail millet accessions collected in Korea, China and Pakistan. The present results could expand our understanding of the morphological variation in foxtail millet accessions from Korea, China and Pakistan, and also could be useful for foxtail millet germplasm preservation.

Adaptability of the high first pod height, shattering-resistant soybean cultivar 'Saegeum' to mechanized harvesting (고착협 내탈립 기계수확 적응 장류·두부용 콩 품종 '새금')

  • The soybean cultivar, 'Saegeum', has been developed for preparing soy-paste and tofu. The soybean cultivars 'Daepung' and 'SS98207-3SSD-168' were crossed in 2003 to obtain 'Saegeum'. Single seed descent method was used to advance the generation from F3 to F5, and the plant lines with promising traits were selected from F6 to F7 by pedigree method. The preliminary yield (PYT) and advanced yield trials (AYT) were conducted from 2009 to 2010, and the regional yield trial (RYT) was conducted in 12 regions between 2011 and 2013. The morphological characteristics of 'Saegeum' were as follows: determinate plant type, white flower, tawny pubescence color, and brown pod color. Flowering and maturity dates were August 2, XXXX and October 17, XXXX, respectively. Plant height, first pod height, number of nodes, number of branches, and number of pods were 79 cm, 18 cm, 16, 2.3, and 44, respectively. The seed characteristics of 'Saegeum' were as follows: yellow spherical shape, yellow hilum, and the 100-seed weight was 25.4 g. 'Saegeum' was resistant to bacterial pustule and SMV in the field test, and its lodging resistance was mildly strong, whereas its shattering resistance was excellent. The ability of this cultivar to be processed into tofu, soybean malt, and other fermented products was comparable with that of 'Daewonkong'. The yield of 'Saegeum' in the adaptable regions was 3.02 ton ha-1. Thus, 'Saegeum' is adaptable to mechanized harvesting because of its high first pod height, as well as lodging and shattering resistance. (Registration number: 5929)

Selection and Horticultural Characteristics Evalution of High ${\alpha}-Glucosidase$ Inhibitor in Pepper (고추의 ${\alpha}-glucosidase$ 저해제 고 활성 계통 선발 및 특성 평가)

  • This study was carried out to investigate the distribution of ${\alpha}-glucosidase$ inhibitor (AGI) activity and to evaluate horticultural characteristics of pepper (Capsicum spp.). AGI activities of pepper fruits and leaves were different from 1.0 to 20.5 times and 1.0 to 5.9 times, respectively. Weight, length and width of evaluated pepper fruit were distributed from 0.5 to 56.0 g, 0.8 to 15.4 cm and 0.5 to 6.3 cm per fruit respectively. Stem colors before transplanting varied from green to violet. Length and width of leaf were distributed from 3.1 to 5.0 cm and 2.1 to 3.0 cm. Immature fruit color was almost green and mature fruit color was almost red. In horticultural characteristics of selected pepper lines with high AGI activity, the fruit position was downward position. The immature fruit color was green in all lines except one and the mature fruit color was red in all lines. Fruit weight and fruit length of selected pepper lines with high AGI activity were distributed from 5.9 to 41.1 g and 5.9 to 17.0 cm and leaf width and leaf length were distributed from 5.8 to 29.7 cm and 3.9 to 8.7 cm, respectively. The AGI activities of pepper is widely variable between leaf and fruit. According to this result, it suggested the possibility of developing a new pepper line with high AGI activity.

Effect of LED as Light Quality on the Germination, Growth and Physiological Activities of Broccoli Sprouts (LED 광질이 브로콜리 새싹의 발아, 생장 및 생리활성에 미치는 영향)

  • This study was carried out to investigate into the effect of light-emitting diode (LED) for the light quality as a light source on the broccoli seed germination and the physiological activity of vegetable sprouts. We have also germinated seeds of the broccoli and applied LED as a light quality such as blue, green, red, white, yellow and red + blue color lights to their sprouts for 14 hours and kept dark for 10 hours at the temperature of $25^{\circ}C$ (day)/$18^{\circ}C$ (night). Broccoli sprouts were extracted by methanol and their physiological activities were examined. All broccoli seeds were germinated at 3 days after seeding regardless of the light color. Total sprout fresh weight were mostly became highest by 0.389g (10 plants) at 8 days after seeding when their sprouts were grown under blue color light. Total phenol compound contents in broccoli sprouts were extremely increased by $83.0\;mg{\cdot}L^{-1}$ under the white light, and total flavonoid contents were most much more by $72.6\;mg{\cdot}L^{-1}$ under the blue light. DPPH radical scavenging activity at $2,000\;mg{\cdot}L^{-1}$ were most highest by 93.5% in broccoli sprouts grown under the white light. Nitrite radical scavenging activity at the concentration of $500\;mg{\cdot}L^{-1}$ in sprout extracts were the most increased by 66.9% under the yellow light, and tyrosinase inhibition activity at $2,000\;mg{\cdot}L^{-1}$ in sprout extracts were by 14.5% under red light.
