Changing Wheat Quality with the Modification of Storage Protein Structure

  • The visco-elastic properties of gluten are major determinants of the processing properties of doughs. These visco-elastic properties are strongly influenced by the ratio of monomeric and polymeric proteins and the size distribution of the polymeric proteins, which make up the gluten fraction of the dough. Recent studies have revealed that other features, such as the number of the cysteine residues of the HMW-GS, also play an important role in determining the functional characteristics. To modify the processing properties at molecular level, the relationship between the structure of molecules and dough properties has to be understood. In order to explore the relationships between individual proteins and dough properties, we have developed procedures for incorporating bacterially expressed proteins into doughs, and measuring their functional properties in small-scale equipment. A major problem in investigating the structure/function relationships of individual seed storage proteins is to obtain sufficient amounts of pure polypeptides from the complex families of proteins expressed in the endosperm. Therefore, we have established a simplified model system in which we produce specific protein genes through bacterial expression and test their functional properties in smallscale apparatus after incorporation into base flour. An S poor protein gene has been chosen as a template gene. This template gene has been modified using standard recombinant DNA techniques in order to test the effects of varying the number and position of cysteine residues, and the size of the protein. Doughs have been mixed in small scale apparatus and characterized with respect to their polymeric composition and their functional properties, including dough mixing, extensibility and small scale bating. We conclude that dough characteristics can be manipulated in a predictable manner by altering the cysteine residues and the size of high molecular weight glutenins.

Seed-born Burkholderia glumae Infects Rice Seedling and Maintains Bacterial Population during Vegetative and Reproductive Growth Stage

  • Rice world production is affected due to the growing impact of diseases such as bacterial panicle blight, produced by Burkholderia glumae. The pathogen-induced symptoms include seedling rot, grain rot and leafsheath browning in rice plants. It is currently recognized the entrance of this pathogen to the plant, from infected seeds and from environmental sources of the microorganism. However, it is still not fully elucidated the dynamics and permanence of the pathogen in the plant, from its entry until the development of disease symptoms in seedlings or panicles. In this work it was evaluated the infection of B. glumae rice plants, starting from inoculated seeds and substrates, and its subsequent monitoring after infection. Various organs of the plant during the vegetative stage and until the beginning of the reproductive stage, were evaluated. In both inoculation models, the bacteria was maintained in the plant as an endophyte between $1{\times}10^1$ and $1{\times}10^5cfu$ of B. $glumae.g^{-1}$ of plant throughout the vegetative stage. An increase of bacterial population towards initiation of the panicle was observed, and in the maturity of the grain, an endophyte population was identified in the flag leaf at $1{\times}10^6cfu$ of B. $glumae.g^{-1}$ fresh weight of rice plant, conducting towards the symptoms of bacterial panicle blight. The results found, suggest that B. glumae in rice plants developed from infected seeds or from the substrate, can colonize seedlings, establishing and maintaining a bacterial population over time, using rice plants as habitat to survive endophyticly until formation of bacterial panicle blight symptoms.

  • This study was conducted to investigate the relationships between yearly variations of climatic elements and yearly variations of productivity in sesame. In addition, correlation coefficients among yield and yield components were estimated. The data of yield and yield components were investigated for 10 years from 1992 to 2001. The meteorological data gathered at the Yeosu Weather Station for the same period were used to find out the relationships between climatic elements and productivity. Yearly variation of the amount of precipitation in July and September were large with coefficients of variation(c.v.) of 64.59, 92.47%, respectively, but the variation of the average temperature in June and August were relative small. Yield and plant height greatly with c. v. of 26.24, 23.41 %, respectively, 1, 000 grain weights show more or less c.v. of 3.83% and length capsule setting show still less variation. Correlation coefficients between maximun temperature in period of cultivation(from June to September) and yield are positively significant at the level of 5.1 %, respectively. Correlation coefficients amount the plant height, length capsule setting, number of capsules per plant, weight of 1, 000 grains and seed yield were positively significant at the level of 1 %, respectively. Simple linear regression equations by the least square method are estimated for number of capsules per plant(Y$_1$) and the maximun temperature in August(X) as $Y_1$=10.1255+0.1725X, and for yield(Y$_2$) and the maximun temperature in August(X) as $Y_2$=21.6151 + 1.3724X.

Effects of Temperature and Moisture Level during Preconditioning on Germination and Seedling Elongation of Soybean Seeds with and without Osmoconditioning (온도, 종자수분 및 삼투처리가 대두의 발아와 묘신장에 미치는 영향)

  • Germination and seedling length of soybean seeds (cultivar: Williams) with and without polyethylene glycol-8000 (PEG) treatments were measured following preconditioning at two temperatures (15 and 30$^{\circ}C$) and two moisture levels 〔low (30 percent) and high (50 percent seed water content)〕 for 0, 2, 4, or 8 days. A split-split plot in time was used with four replications. Observations were made after two days of germination at 30$^{\circ}C$ Seedling growth accelerated with two days of preconditioning at 30$^{\circ}C$, but was reduced as preconditioning duration increased up to eight days at the same temperature. PEG treated preconditioned seeds exhibited reduced moisture uptake and seedling growth. Preconditioning at a high moisture level increased seedling moisture content and also increased seedling length until four days of preconditioning duration. Seedling dry weight decreased when preconditioning temperature was 30$^{\circ}C$ and when the high moisture level of preconditioning was continued for eight days.

Comparison of Characteristics Related with Soybean Sprouts between Glycine max and G. soja (콩의 재배종과 야생종의 콩나물관련 특성)

  • Soybean sprout trails were evaluated for soybeans for sprout (Glycine max) and wild soybeans (Glycine soja). Soybeans for sprout are characterized with small seed size of 10-14 grams per 100 seeds and are traditionally used only for producing sprouts. No difference was observed in hypocotyl length and sprout yield between two types of soybean. (However total length, hypocotyl hypocotyl, body weight, and the rate of good quality were greater in soybeans for sprout.) Number of lateral roots and root length were greater in wild soybeans. Contents of crude oil, oleic acid, and sucrose were higher in soybean for sprout, while that of protein and linoleic acid were higher in wild soybeans. There was no difference in sprout isoflavone content. Out of 17 amino acids analyzed, 10 amino acids such as aspartic acid, lysine, and arginine. etc. showed significantly Higher content in wild soybeans. Hypocotyl color of sprouts estimated by color meter based on the degree of L(brightness), a(red), and b(yellow) values were better in wild soybeans than soybean for sprout.

Response of colchicine for the efficient chromosome doubling in Codonopsis lanceolata

  • Polyploidy has opened a new horizon for selection to sculpt a variety of new gene functions, traits, and lineages. The aim of this study was to investigate the efficacy of the colchicine concentration, temporal changes, and suitable material for inducing effective tetraploid plants of Codonopsis lanceolata. A total of 180 individuals from 16 treatment groups were germinated, and exposed to different concentrations of Colchicine. The plant height of the diploid (18.1 cm) was slightly shorter than that of the tetraploid (13.4 cm). The fresh weight of the main root in the diploid (0.5 g) was 4-fold higher than the tetraploid (2.2 g). The colchicine-treated plant regeneration rate in C. lanceolata was decreased at the elevated concentration of colchicine. A total of 126 individual plants were regenerated in the entire treatment group and tetraploid (2n=4x=32) plants were obtained. In particular, 5 individuals of the tetraploid plant were induced in the 0.05% colchicine for 6h, which is a higher rate (29.4%) than other regenerated plants. As in the seed treatment result, the plant height of the diploid was significantly higher (10.4 cm) than tetraploid. The root length of the tetraploid (10.1 cm) was longer than the diploid, and the root was also thicker. Taken together, the results obtained from the present study may be helpful for the efficient recovery of such polyploid plants through the in vitro application of colchicine, and may improve the productivity and breeding of C. lanceolata.

Effects of soil covering materials on the overwintering and the growth of Cynanchum wilfordii Hemsley

  • This study was performed to select the soil covering materials for winter safety of Cynanchum wilfordii Hemsley and to investigate the effect on the growth after overwinter. Soil covering materials such as vinyl, rice straw, lagging cover, vinyl+lagging cover, and non-covering were investigated how effect on overwintering rate, growth and yield of Cynanchum wilfordii Hemsley. In changes of soil temperature according to the soil covering materials, non-covering treatment was recorded the highest to $26.7^{\circ}C$, and lagging cover and rice straw showed little change of soil temperature with 9.2 and $9.3^{\circ}C$, respectively. Regrowth rates of early growth after overwinter were lowered in the order as rice straw > vinly+lagging cover > lagging cover > vinyl > non-covering. From the above-ground growth by soil covering materials, vine length showed similar growth in all treatments except vinyl covering and stem diameter exhibited the thicker pattern in vinyl and lagging cover, and there was no significant difference among treatments in number of branch. Flowering period is recorded July 30 in all treatments and ripening period is October 16. Yield of seed production is showed the best in vinyl+lagging cover with $25g/m^2$. Root weight and root length for subterranean growth showed no significant differences among all treatments. Yield is produced the highest in vinyl+lagging cover with 433kg/10a and the next to lagging cover with 431kg/10a. In comparison with incomes, lagging cover earned to 9,882 thousand won/10a, showed the effect on income enlargement to 21%, compare to that of the non-covering.

The Effect of Different Concentration of Glyphosate on the Growth of Coconut Seedlings

  • Coconut (Cocos nucifera L) is one of the predominant plantation crops in Sri Lankan economy which is known to have existed for over thousands of years. During the past decades coconut production had been reduced by a significant quantity. The usage of poor quality planting materials is a major reason for the low coconut production. Thus much attention needs to be paid in coconut nurseries. Weed management is a critical management practice in the nursery. Though glyphosate application is becoming popular in nurseries it can affect weeds as well as coconut seedlings growth. Therefore the effects of glyphosate were evaluated by determining the growth of shoot and root of coconut seedlings. Poly bag nursery was prepared and three treatments were used. Treatments were no glyphosate and manual weeding ($T_1$), application of glyphosate 1.08 ai kg $ha^{-1}$ at 2 monthly interval ($T_2$) and application of glyphosate 1.44 ai kg $ha^{-1}$ at 2 monthly interval ($T_3$). Application of glyphosate at early stage of seedling growth had a no significant effect on growth parameters tested. However, the concentrations of glyphosate negatively affected numbers, volumes and dry weights of secondary, tertiary and quaternary roots at the latter stage of seedling growth. The leaf area and the height of seedling were significantly reduced by the highest concentration of glyphosate. Among the growth parameters tested, seedling girth and shoot dry weight were not affected by the application of glyphosate. These results revealed that the usage of glyphosate at both concentrations negatively affected root growth of coconut seedlings. Based on these results, the both concentration levels of glyphosate should be applied to coconut nurseries before sprouting the seed nuts.

Effect of Arbuscular Mycorrhizal Fungi on Growth of Korean Ginseng (Panax ginseng C. A. Mey.) Seedlings (수지상균근균이 인삼 (Panax ginseng C. A. Mey.)의 생장에 미치는 영향)

  • The purpose of this study was to investigate effects of inoculation of arbuscular mycorrhizal fungi (AMF) on growth of Korean ginseng (Panax ginseng C. A. Mey.) seedlings. Five species of AMF (Glomus mosseae, Glomus intraradices, Glomus etunicatum, Acaulospora longula, Acaulospora trappei) were inoculated to ginseng seedlings at 60 days after ginseng seed germination. Dry weights, heights of the plants and chlorophyll fluorescence were measured at 120 days after inoculation of AMF. Dry weights and shoot heights were increased in seedlings inoculated with AMF compared to the controls without AMF. However, chlorophyll fluorescence of seedlings with AMF showed no significant difference compared to the control.

Quality Characteristics of Vegetable Soybeans at Different Harvesting Time (풋콩의 수확시기에 따른 품질관련특성의 변화)

  • The changes in major characteristics of vegetable soybean at different stages were investigated. The 100-seed weight of vegetable soybean had a tendency to increase before 43 days after flowering, but decrease slightly after then. After 37 days after flowering, the sucrose content of Douya had a tendency to decline and in case of Shirofusa, total sugar content was sharply decreased at 40 days after flowering. The total vitamin C content of Douya and Shirofusa was dramatically increased at 37 days after flowering and then got the plateau, otherwise the total vitamin C content of Shirofumi and Hwaeomputkong was the highest level at 43 days after flowering. The hardness changes of vegetable soybeans were tend to increase according days after flowering. In sensory evaluation, preference score was significantly associated with harvesting stages. Correlation coefficient between sweetness and beany taste was significantly negative.
