Analysis of Functional Components in Roasted Okra (Abelmoschus esculentus L. Moench) Seeds (볶음 오크라 종자의 주요 기능성분 분석)

  • This study was conducted to investigate the general characteristics of raw okra seeds and the functional components of roasted okra seeds. The number of okra seed per pod was 78 in 'Greensod' and 88 in 'Beny'. The weight of okra seed per pod of 'Greensod' and 'Beny' were 4.4 g and 6.3 g, respectively. Free amino acid contents of the stir-fry and fresh okra seeds were measured as $2.69mg{\cdot}g^{-1}$ and $0.31mg{\cdot}g^{-1}$. Total polyphenolic compound content of the stirfry okra seeds was estimated as $12.61mg\;CGA{\cdot}g^{-1}$, compared to $2.54mg\;CGA{\cdot}g^{-1}$ fresh okra seeds, Thus, free amino acid and total polyphenolic compound contents in the stir-fry okra seeds were higher than fresh one, Antioxidant activities, such as DPPH and ABTS radical scavenging in the stir-fry okra seeds was the higher than fresh okra seeds.

Influence of Polyethylene Film Mulch and Seedling Types on Growth and tuber Yield of Fall-Grown Potato (폴리에틸렌필름 멀칭 및 종묘종류가 가을감자의 생육과 수량에 미치는 영향)

  • Normal seed tubers and plug seedlings raised from stem cutting and mini-tubers ($10\pm3$ g) were planted or transplanted on August 20,2001 in bare soil or under transparent and black polyethylene film mulches to evaluate the effects of mulch material and seedling type on the growth and tuber yield of fall-grown potato (Solanum tuberosum L. cv. Dejima) in Jeju region. Total tuber number per plant was 4.6 regardless of mulch treatment, and direct planting seed potatoes resulted in the greatest tubers (5.14)/plant and followed by transplanting seedlings raised from cutting and mini-tubers (4.15 and 5.15 tubers/plant). Transparent and black polyethylene film mulches increased total tuber yield by 16 and 8%, respectively, and average tuber weight by 14 and 12%, respectively, compared with no mulch (21.16 t/ha and 72.2 g/tuber). Transplanting plug seedlings raised from cutting resulted in the greatest tuber yield, followed by transplanting plug seedlings raised from mini-tubers and direct planting seed potatoes.

Influence of roasting conditions on the flavor quality of sesame seed oil (참깨 볶음조건이 참기름의 향미에 미치는 영향)

  • Sesame seeds were roasted for 30, 60, 90, and 120 min at different temperatures (100, 200, and $300^{\circ}C$) and extracted to investigate an adequate condition for producing the high quality sesame oil. Sesame seeds roasted at $200^{\circ}C$ for 90 min gave the high yield of oil. The oil contained the low content of brownish-black precipitates and exhibited an excellent organoleptic quality when judged by descriptive sensory analysis. Thirty one volatile flavor compounds, which are the largest number of volatiles among the oil samples prepared, were identified from the oil sample. The oil contained relatively high concentrations of furfurals (sweet candy-like flavor) and pyrazines (roasted-like flavor), that are considered as good contributors to sesame seed oil flavor, and low concentations of aldehydes $(C5{\sim}C10)$ and ketones, which are considerd as bad contributors (oxidized fat-like and painty-like flavors). These results suggest that the roasting condition of $200^{\circ}C$ for 90 min was the best for the oil production in terms of the overall aroma and taste quality under the test conditions (Received July 13, 1993; accepted November 4, 1993).

Studies on Protein Profiles and Isozymes in Germinating Seeds (종자발아에 있어서 Protein Profile과 Isozyme에 관한 연구)

  • The purpose of this experiment was to study one side of germination physiology based on that protein profiles and protease relating to protein metabolism, that peroxidase, catalase, $\alpha$-amylase, $\beta$-amylase, and malate dehydrogenase involved in the carbohydrate metabolism of seed germination. All these experiments were divided into the two groups with and without acetone treatment, and were carried out. The protein bands of each germinating stage between the groups treated with and without acetone showed certain basic pattern in polyacrylamide gel disc electrophoresis. However, there was a little difference in the number of protein band, optical density, and migration velocity between two groups. The isozyme bands of peroxidase, and catalase between two groups in polyacrylamide gel disc electrophoresis did not show the numeral difference, but the optical density of certain germinating stage treated with acetone was higher than the group untreated with it and it showed their enzyme activity. The $\alpha$-amylase and $\beta$-amylase activities which involved in starch metabolism of seed germination were higher in the treated group than the other. On one hand, the protease activity of hydrolase occurred in the seeds for germination was also higher, more or less in the treated group than in the other. The isozyme band pattern of malate dehydrogenase in TCA cycle of energy metabolism pathway was very different between two groups growing for 72 hours with and without acetone treatment in cellulose acetate electrophoresis. It indicated that two isozyme bands of malate dehydrogenase was high. Consequently these experimental results mentioned above indicated that acetone treatment before sowing had an effect on dissolving certain complexed lipid substance involved in the seed coats, the activity of carbohydrate hydrolase increased with water absorption which was most comfortable in its germination, dissolved glycerin and fatty acid became certain energy source, and they stimulated the acceleration of respiration metabolism.

Effect of SiO2 on yield and phenolic profile of Tartary buckwheat

  • The aim of this study was to enhance the Tartary buckwheat growth and phenolic profile by the application of $SiO_2$ on potsin the glasshouse. The liquid of $SiO_2$ were applied at three times after three weeks of sowing at ten days interval. The doses of the $SiO_2$ was low (2.5 ml/5 L water), high (10 ml in 5 L water) and control experiment was done without $SiO_2$. In this study, it is clearly shown that high dose of $SiO_2$ increased the buckwheat plant growth including, plant height, leaf length and width, stem diameter, fresh weight and number of seed per plant compared to control treatment. In the same way, the total phenolic compound (1421 mg/100 g), total flavonoid (35.1 mg/100g), rutin (3,130 mg/100g) and DPPH (82%) in plant were also increased high dose of $SiO_2$ compared to control (1,175 mg/100g, 31.9mg/100g, 860 mg/100g, 59%, respectively). Moreover, higher phenolic compound (1,421 mg/100g) and DPPH (82%) was observed in plant compared to seed (196 mg/100mg, 72 %, respectively). In the contrary, total flavonoid (36.2 mg/100g) and rutin (1,400 mg/100g) was higher in buckwheat seed compared to plant (35.1 mg/100g, 3,130 mg/100g, respectively). Finally, it is concluded that higher dose of $SiO_2$ enhance buckwheat growth and phenolic profile. Further investigation is needed to evaluate the optimum dose of $SiO_2$ according to soil conditions in the field.

Effect of Grape Seed Proanthocyanidins on Tumor Vasculogenic Mimicry in Human Triple-negative Breast Cancer Cells

  • Vasculogenic mimicry (VM) refers to the unique ability of highly aggressive tumor cells to mimic the pattern of embryonic vasculogenesis, which was associated with invasion and metastasis. The grape seed proanthocyanidins (GSPs) had attracted much attention as a potential bioactive anti-carcinogenic agent. However, GSPs regulation of VM and its possible mechanisms in a triple-negative breast cancer cells (TNBCs) remain not clear. Therefore, we examined the effect of GSPs on VM information in HCC1937 cell model. In this study, we identified the VM structure via the three-dimensional (3D) matrix in vitro. Cell viability was measured using the CCK8 assay. The effects of GSPs on human triple-negative breast cancer cells (TNBCs) HCC1937 in terms of related proteins of VM information were determined using western blot analysis. In vitro, the tubular networks were found in highly invasive HCC1937 cells but not in the non-invasive MCF-7 cells when plated on matrigel. The number of vascular channels was significantly reduced when cells were exposed in GSPs ($100{\mu}g$/ml) and GSPs ($200{\mu}g/mL$) groups (all p<0.001). Furthermore, we found that treatment with GSPs promoted transition of the mesenchymal state to the epithelial state in HCC1937 cells as well as reducing the expression of Twist1 protein, a master EMT regulator.GSPs has the ability to inhibit VM information by the suppression of Twist1 protein that could be related to the reversal of epithelial-to-mesenchymal (EMT) process. It is firstly concluded that GSPs may be an p otential anti-VM botanical agent for human TNBCs.

Density and Species Composition of Soil Seed Bank in Rural Stream Topsoil (농촌하천 표토내 매토종자의 발아량 및 종구성)

  • Purpose of this study was to evaluate germination characteristics of soil seed bank in rural stream topsoil using seedling emergence method in order to provide data for future ecological restoration of stream utilizing topsoil. There were 24 families, 52 genera, 61 taxa of soil seed bank flora found in topsoil from 6 rural streams. The most frequently found taxa were Compositae (12 taxa) followed by Gramineae (8 taxa), Caryophyllaceae (5 taxa), Cruciferae (4 taxa), Scrophulariaceae, Labiatae, Polygonaceae and Cyperaceae. Plant with the most number of germination was Stellaria aquatica followed by Erigeron annuus, Imperata cylindrica var. koenigii, Poa annua, Cyperus microiria and Veronica undulata. Naturalized plants found were Erigeron annuus, Rumex crispus, Oenothera odorata, Cerastium glomeratum, Bidens frondosa, Erigeron philadelphicus, etc.

Germinability of Film-Coated Snap Bean Seed as Affected by Oxygen Diffusion Rate under Different Soil Moisture Contents

  • The film coated snap bean (Phaseolus vulgaris) seeds with five different coating materials treated with 3% increase in seed weight were planted at sandy loam soil controlled moisture content of 18, 19, 20 and 21 %. The oxygen diffusion rate (ODR) was calculated from the different moisture content soil. The number of normal seedlings, seedling vigor, and seedling capability in field (seed vigor x dry matter weight) were observed at 9 days after planting and compared to the changes of ODR. The germination rate and ODR were sharply decreased simultaneously in the seeds planted at 21 % soil moisture content. Seedling emergence did not occur at all as the soil moisture content increased above 22 %. Hence this value should be considered as the threshold of soil moisture content for seedling emergence. An ODR value under 20% did not influence the percent emergence significantly. The certain difference observing in the emergence at the same ODR was not related clearly to the condition of soil. So it can be assumed that the limit of soil moisture content for the emergence of snap bean was approximately 20%. The value of 18% soil moisture content may be considered as the optimum for snap bean emergence. There was close relationship between the mean value of ODR in different soil moisture contents and the emergence. The germination rates of the seeds coated with the different materials were quite different when the seeds were planted at 21 % soil moisture. Dry weight of the seedlings from film coated seeds was decreased slightly, but the germination rates were not much different from the non-treated control under relatively higher soil moisture content (21 %). Major factor lowering emergence rate was oxygen stress while film coating act as a minor constraint for snap bean sown in excess soil moisture condition.

Improvement of blood glucose control in type 2 diabetic db/db mice using Platycodon grandiflorum seed extract (도라지 종자 추출물의 처리가 제2형 당뇨 db/db 마우스의 혈당개선에 미치는 효과)

  • The biological activities of Platycodon grandiflorum (PG) root extracts have been studied intensively, whereas there are limited number of studies on PG seed extract (PGSE). PGSE was prepared by ethanol extraction, and its antidiabetic effect was evaluated in mice with type 2 diabetes (C57BLKS/J-db/db). Results indicated that the administration of high-dose PGSE (600 mg/kg, wb) significantly stabilized the blood glucose levels, as evidenced by the results of the oral glucose tolerance test. Mice treated with high-dose PGSE exhibited significantly lower serum hemoglobin A1c, insulin, and leptin levels after eight weeks of feeding trial (p<0.05). High-dose PGSE administration significantly improved glucose uptake in the femoral muscle of db/db mice by activating both IRS-1/PI3K/AKT/AS160 and AMPK phosphorylation pathways. GLUT4 translocation from the cytosol to the plasma membrane increased 1.7-fold in the PGSE high-dose group. These results suggest that PGSE has potential for development as an antidiabetic agent.

Management and Development of Seed Rearing in Red Spotted Grouper Epinephelus akaara (양식 붉바리(Epinephelus akaara) 종자의 개발과 사양관리)

  • We investigated seed management and feeding regime of larval red spotted grouper Epinephelus akaara using live food organisms. Fertilized eggs were produced in March 2020 by manipulating photoperiod and water temperature. Broodstock (F0) produced approximately 0.30×106 eggs, of which 0.25×106 were fertilized; first-generation broodstock (F1) produced 2.00×106 eggs, of which 1.90×106 were fertilized. Larvae were reared at temperatures of 21.0-24.5℃. The feeding regime consisted of rotifers Artemia, and commercial pellet feed. From 5-8 days after hatching (dah), rotifers collected from high-density incubation tanks were fed to grouper larvae at a density of 10-20 individuals/mL. From 9-30 dah, rotifers collected from low-density incubation tanks were fed to larvae at a density of 10-20 individuals/mL. Newly hatched Artemia were provided from 20-24 dah and enriched Artemia from 25-50 dah. Commercial pellet feed was given starting at 13 dah. The average mouth size in hatchlings was 263.4±3.7 ㎛ at 5 dah, 406.7±21.0 ㎛ at 11 dah, and 856.9±136.6 ㎛ at 21 dah. At 1 dah, larvae had a mean total length of 1.95±0.06 mm. At 66 dah, the mean TL was 4.0 cm. The final number of surviving red spotted grouper larvae was 218,380 fishes.
