Transformation of Orchardgrass (Dactylis glomerata L.) with Heat Shock Protein Gene (Heat Shock Protein 유전자를 이용만 오차드그래스의 형질전환)

  • An experiment was carried out to introduce OsHSP17.9, a low molecular HSP gene isolated from rice plant to orchardgrass (Dactylis glomerata L.) using Agrobacterium. Mature seed-derived calli of orchardgrass were co-cultured with Agrobacterium tumefaciens EHA101 harboring the plasmid pIG-HSP17.9 for transformation. Calli selected by hygromycin were transferred to N$_{6}$ medium containing 1 mg/L NAA, 5 mg/L kinetin, 250 mg/L cefotaxime and 50 mg/L hygromycin and several hygromycin resistant plants were obtained. Stable incorporation of the introduced OsHSP17.9 to the genome of the hygromycin resistant plants was confirmed by PCR and Southern blot analysis. Transformation efficiency was variable between cultivars in which it was 16.5% in Potomac and 8.0% in Frontier. Constitutive expression of the transgene in the transformed orchardgrass tissues was identified by Northern blot analysis but transcript levels were different among individual plants.s.

  • RFID systems are being studied and developed in the area of the industry and marketplace. Recently RFID systems are core element of the ubiquitous technologies in individual life and industry. However, RFID systems often cause some serious problems such as violation of privacy and information security because their contactless devices communicate each other by radio frequency In this paper, we propose multiple objects RFID tag scheme including tag structure and authentication protocol. The proposed RFID tag structure maintains several object IDs of different applications in a tag memory. The tag structure allows those applications to access object IDs simultaneously. The authentication protocol for multiple objects tag is designed ta overcome the problems of security and privacy. The protocol has robustness against various attacks in low cost RFID systems. We evaluate the efficiency of proposed scheme and compare security of our scheme with several traditional schemes.

  • A cultivar 'Yellow Cream' of Phalaenopsis(P. Blume) was developed by cross breeding and line selection at the Flower Research Institute, Gyeongnam ARES, from 2000 to 2008. Characteristic trials for this cultivar were carried out three times from 2006 to 2008. 'Yellow Cream' was developed from a crossing between P. 'Sogo Firework' and P. 'Sogo Gold'. 'Yellow Cream' exhibits light yellow flower color with pink stripe and favorable flower shape. Individual flowers are formed with parallel arrangement and have long life with more than 55 days. 'Yellow Cream'(Grant No.3232) was registered to the Korea seed and variety Service(KSVS) for commercialization in 2010 and suitable for the cultivation under greenhouse conditions in Korea.

Genome-wide identification and expression profiling of the pectin methylesterase gene family in Citrus sinensis (L.) Osbeck

  • Pectin methylesterase (PME) plays an important role in vegetative and reproductive development and biotic/abiotic stress responses by regulating the degree of methyl-esterification of pectic polysaccharides in the plant cell wall. PMEs are encoded by a large multigene family in higher land plant genomes. In general, the expression of plant PME genes shows tissue- or cell-specific patterns and is induced by endogenous and exogenous stimuli. In this study, we identified PME multigene family members (CsPMEs) from the sweet orange genome and report detailed molecular characterization and expression profiling in different citrus tissues and two fruit developmental stages. We also discussed the possible functional roles of some CsPME genes by comparing them with the known functions of PMEs from other plant species. We identified 48 CsPME genes from the citrus genome. A phylogenetic tree analysis revealed that the identified CsPMEs were divided into two groups/types. Some CsPMEs showed very close phylogenetic relationships with the PMEs whose functions were formerly addressed in Arabidopsis, tomato, and maize. Expression profiling showed that some CsPME genes are highly or specifically expressed in the leaf, root, flower, or fruit. Based on the phylogenetic relationships and gene expression profiling results, we suggest that some CsPMEs could play functional roles in pollen development, pollen tube growth, cross incompatibility, root development, embryo/seed development, stomata movement, and biotic/abiotic stress responses. Our results shed light on the biological roles of individual CsPME isoforms and contribute to the search for genetic variations in citrus genetic resources.

Analysis of the Effects of Stocking Density and Individual Size on the Growth and Movement Characteristics of Juvenile Abalone Haliotis discus hannai (사육 밀도와 개체 크기에 따른 북방전복(Haliotis discus hannai) 치패의 성장과 이동 특성)

  • Behavior serves as an important indicator of the effects of environmental stressors on organisms. In this study, one-year-old abalone Haliotis discus hannai (15.1±0.8 mm) were used and each 0.34 m2 tank was stocked with 100, 150, 200, 250, and 300 juvenile abalone. After 328 days, we found that the low stocking density treatment showed significantly higher growth, whereas the high stocking density treatment showed significantly lower growth. In addition, abalone showed a lower rate of movement, distance moved, and velocity at higher stocking densities, but only velocity differed significantly. In contrast, In the Supplied the feed tanks significant difference between the moving distance and velocity. When the two groups of different sizes were mixed in tank, the rate of movement, distance moned, and velocity showed significant differences between differently-sized indivisuals. Our study indicates that breeding abalone at high densities has a negative effect on their growth and survival, which is thought to primarily be caused by spatial competition, and secondarily by differences in body size, ultimately affecting growth and productivity.

Evaluation of Horticultural Characteristics on Water Dropwort (Oenanthe stolonifera DC.) Genetic Resources for Various Utilization

  • Water dropwort (Oenanthe stolonifera DC.) is a perennial herbaceous plant that grows wild throughout Korea. As of 2020, 24,819 tons (51.7% of open field, 48.3% of facility) are produced in 1,278 ha (59.9% of open field, 40.1% of facility) nationwide. Water dropwort, which is rich in nutrients such as vitamins and iron, is mainly cultivated by vegetative propagation method using local traditional species, however, seed propagation and breeding of cultivars are insufficient so far. Since securing, propagating and continuous characterization of various genetic resources are required to breed new cultivars, this study was conducted to compare the main characteristics of domestic genetic resources and to improve their utilization. Growth characteristics such as plant height, fresh weight, plant type, and flowering date were investigated for the 89 varieties of genetic resources owned by Jeollabuk-do Agricultural Research and Extension Services after vegetative propagation in the individual pots. Also, the morphological image information of leaves and flowers was constructed. Genetic resources were collected nationwide and originated in 30 regions including Hwaseong, Siheung and Jeju. Their plant types could be classified into straight, intermediate and creeping types according to their morphological characteristics, and at this time, the number of intermediate types accounted for the largest proportion. Flowering was carried out under high-temperature and long-day conditions in summer. According to the flowering date, they could be classified into early, middle and late flowering varieties, and at this time, the middle flowering varieties occupied the largest proportion. As a result of the investigation of vegetative growth characteristics, varieties with long plant height, heavy fresh weight and thick stem thickness were identified. Along with this result, it is thought that classification and selection of genetic resources for various purposes will be possible through additional investigations such as analysis of components and antioxidant activity. Moreover, it is judged that such results can be used as basic data for breeding new water dropwort cultivars in the future.

Assessment of environmental impact of vitamin A-enhanced soybeans and hybrid soybeans

  • An understanding of safety problems pursuant to environmental release of GM (Genetically Modified) crops is considered important. Among the recognized safety problems, the possibilities of weediness and ecosystem invasion are constantly being validated. We herein compared the growth characteristics and germination rate of soybeans formed by hybridization with vitamin A-enhanced soybeans carrying an introduced gene that increases β-carotene content. We also examined overwintering, survival, and weed competitiveness to evaluate hybrid ecological impact on long-term unmanaged cultivatable land. These studies revealed that the hybrid soybeans exhibited intermediate growth characteristics and germination rate compared with the vitamin A-enhanced soybeans and wild soybeans, or exhibited traits similar to those of the maternal strain. Overwintering experiments were conducted by planting seeds at depths of 0, 5, 10, and 20 cm and recovering them after three or five months. After five months, all seeds at depths more than 5 cm lost viability. Among seeds recovered after three months, only wild soybeans retained viability at depths of more than 5 cm. Survival and weed competitiveness were assessed by sowing each type of seed and performing no irrigation, or pest or weed control. Quantitative assessment of numbers of individual soybean plants that appeared in the experimental plot revealed that all plants germinated after sowing, but only wild type plants survived overwintering. These studies suggest that both GM soybeans and hybrid soybeans cannot survive in uncultivated land even if they are released into the environment, which indicates less possibility of ecosystem invasion and weediness.

Changes in Polyamine and Tyramine Concentrations in Rice (Oryza sativa L.) during Maturation and Preharvest Sprouting (벼 등숙기와 수발아 기간동안 폴리아민과 티라민의 농도변화)

  • The levels of polyamines were measured to investigate the alternative nitrogen metabolism during maturation and sprouting in rice. The rice plants (cv. Ansanbyeo) were cultivated in 20-year-old non-fertilized field. The flag leaves and spikes were collected weekly after the earing stage and the seeds were harvested daily after lodging. Free, bound, and conjugated polyamines were analyzed using reverse phase HPLC. Putrescine, spermidine, spermine, agmatine and tyramine were the major amines found in rice. The level of stress-induced amine, putrescine increased during the preharvest sprouting confirming that the process was a stress to the plants. With all other polyamines, tyramine in free form decreased in flag leaves and panicles during seed maturation. However, agmatine in bound form showed a noticeable increase about 8-fold during 6 weeks period of maturation after which it declined to the bottom level. Among the individual amines, tyramine and spermine in conjugated form showed a marked change during matutation and sprouting. Interestingly, the level of tyramine with all conjugated polyamine decreased in spikes during seed maturation and increased during preharvest sprouting implying that tyrosine decarboxyation and conjugation to phenolic acids may play a key role in preharvest sprouting. Spermine in conjugated form was synthesized only at the early earing stage in the level of $3.4mole\;g^{-1}$ fresh weight, and then decreased to the level of nmole during maturation. Thereafter, it dramatically increased to 2.8 mole during preharvest sprouting. In this study we found the tyramine is a major amine in rice, and it would play a critical role in N-assimilation during seed maturation and sprouting.

Strategy for Bio-Diversity and Genetic Conservation of Forest Resources in Korea (생물종(生物種) 다양성(多樣性) 및 삼림유전자원(森林遺傳資源) 보존(保存) 전략(戰略))

  • Due to its topographic complexities and various climatical condition, Korea exhibits diverse forest types. Dominant tree species in this zone are Quercus spp., Betula spp., Zelkova spp., Fraxinus spp., Pinus densiflora, Pinus koraiensis, and Pinus thunbergii ete. Genetic conservation in forest species in Korea there are three ways ; one is in situ, other is ex situ and third is in-facility conservation. In situ conservation include that are the present status of conservation of rare and endangered flora and ecosystem, the reserved forest, the national and provincial park, and the gene pool of natural forests. Ex situ conservation means to be established the new forest from in situ forest stands, progeny and provenance test populations, seed orchard and clone banks, and gene conservation in-facility. As a tool for low temperature storage, several aspects on in vitro system were studied ; (1) establishment of in vitro cultures from juvenile and/or rejuvenated tissues, (2) induction of multiple shoots from the individual micropropagules, (3) elongation of the proliferated shoots. Studies on cold storage for short-and long-term maintenance of in vitro cultures under $4^{\circ}C$ in the refrigerator were conducted. For the cryopreservation at $-196^{\circ}C$, various factors affecting survivability of the plant materials are being examined. The necessity of gene conservation of forest trees is enlarged not only to increase the adaptability for various environments but also to gain the breeding materials in the future. For effective gene conservation of forest trees, I would like to suggest followings ; 1. Forest stands reserved for other than the gene conservation purposes such as national parks should be investigated by botanical and gene-ecological studies for selecting bio-diversity and gene conservation stands. 2. Reserved forest for gene pool should be extented both economically important tree spp. and non-economical species. 3. Reserved forest for progeny test and clone bank should be systematically investigated for the use of Ex situ forest gene conservation. 4. We have to find out a new methodology of genetic analysis determining the proper and effective size of subpopulation for in situ gene conservation. 5. We should develop a new tree breeding systems for successful gene conservation and utilization of the genetic resources. 6. New method of in-facility gene conservation using advanced genetic engineering should be developed to save time and economic resources. 7. For the conservation of species with short-life span of seed or shortage of knowledge of seed physiology, tissue culture techniques will be played a great role for gene conservation of those species. 8. It is are very useful conservation not only of genes but of genotypes which were selected already by breeding program. 9. Institutional and administrative arrangements including legistlation must be necessarily taken for gene conservation of forest trees. 10. It is national problems for conservation of forest resources which have been rapidly destroyed because of degenerating environmental condition and of inexperienced management system of bio-diversity and gene conservation. 11. In order to international cooperation for exchanging data of bio-diversity and gene conservation, we should connect to international net works as soon as possible.

Comparison of Growth and Survival Rate in Shortnecked Clams, Ruditapes philippinarum from Different Seedling Production Areas (산지별 바지락, Ruditapes philippinarum의 성장과 생존율 비교)

  • Growth and survival rate in shortnecked clam, Ruditapes philippinarum were studied from different seedling production areas, Hadong, Ulsan and Kochang to Kwangyang Bay, Korea, and three stocking densities (1,000, 1,500 and 3,000 individuals/$m^2$) of clam from Ulsan seedling production area were examined. During the experimental period, water temperature and specific gravity were ranged from 4.5 to $26.0^{\circ}C$ and from 1.0216 to 1.0248, respectively. Shell length was grown from $2.30{\pm}0.05\;to\;3,29{\pm}0.02$ cm in Hadong, $1.32{\pm}0.02\;to\;3.15{\pm}2.02$ cm in Ulsan, $1.24{\pm}0.03\;to\;3.18{\pm}0.02$ cm in Kochang. Kochang's seed was displayed fast growth rate than others. In different rearing densities, the lower density group was showed a rapid growth rate. Shell length from the 1,000 individuals per $m^2$ group grew longer than that from the 2,000 individuals group. Total weight from the 1,000 individuals group was higher than that from the 1,5()0 individual and the 2,000 individual groups. Survival rate by seedling production areas, Hadong, Ulsan and Kochang showed $69.4\%,\;63.8\%\;and\;41.7\%$, respectively. Survival rate by rearing density, 1,000, 1,500 and 2,000 individual groups showed $70.8\%,\;67.8\%\;and\;59.0\%$, respectively.
