A Study on the Water Impermeability and Growth Inhibitors of Leguminoseae Seeds (콩과식물(科植物) 종자(種子)의 부투수성(不透水性) 및 생장억제물질(生長抑制物質)에 관(關)하여)

  • In the present paper, we intented to analyze and to compare the different seed dormancy of two legume plants, Albizzia and Cercis. We found two different types of seed dormancy: one type of dormancy in Albizzia seed was caused by the water impermeability, and other type of seed dormancy was found in Cercis and it was induced by inhibitors. We confirmed that the extract substances from Cercis seed seemed to have strong regulational functions on the inhibition of seed germination and primary root growth of lettuce seed.

Physiological Activity and Antiproliferation Effects of Citron Seed Extracts on Cancer Cells (유자씨 추출물의 생리활성과 암세포 성장 억제효과)

  • This study was carried out to investigate the total polyphenol, total flavonoid content, antioxidant activity, angiotensin I-converting enzyme (ACE) inhibitory activity, $\alpha$-glucosidase inhibitory activity, and antiproliferation activity of the citron seed. The citron seed were separated to hull and embryo, and extracted with n-hexane and 70% ethanol. Antioxidant activity of ethanol extract was higher than that of n-hexane extract. IC50 value for DPPH radical scavenging activity of ethanol extract of hull (CSE1) and embryo (CSE2) were 3.18 and 8.43 mg/mL, and those of total antioxidant activity were 19.96 and 11.28 mg AA eq/g, respectively. ACE inhibitory activity and $\alpha$-glucosidase inhibitory activity on CSE1 showed the highest values of 31.61 and 45.17%, respectively. Antiproliferation effects on the MCF7, HepG2, H460, HCT-116, and PC3 cell line showed the highest values of 14.09, 19.12, 12.29, 9.78, and 9.12% in extract concentration of 5 mg/mL, respectively. These results suggested that citron seed can be used for development of functional food material which have biological activities.

Anti-Oxidant and Anti-Inflammatory Activities of Various Organ Extracts from Trichosanthes kirilowii Maxim. (하늘타리(Trichosanthes kirilowii Maxim.) 부위별 추출물의 항산화 및 항염증 활성)

  • In this study, antioxidant and anti-inflammatory activities of the extract of different organs such as seed, fruit and root from Trichosanthes kirilowii Maxim. (TKM) were investigated in vitro. Among organs of TKM, the methanol extract of seed showed weak 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities with $10.4{\pm}0.1%$ and $25.5{\pm}1.1%$ at $50{\mu}g/mL$ and $100{\mu}g/mL$, respectively, and strong 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) radical scavenging activities with $72.4{\pm}6.3%$ and $96.4{\pm}2.3%$ at $50{\mu}g/mL$ and $100{\mu}g/mL$, respectively. Anti-inflammatory effects of the extracts were investigated by measuring nitric oxide (NO) production, mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in LPS-induced Raw 264.7 macrophage cells. The methanol extracts of seed and fruit at $50{\mu}g/mL$ potently suppressed LPS-stimulated production of NO and reduced the expression of iNOS mRNA. The methanol extracts of seed and fruit also reduced the expression of COX-2 mRNA remarkably. Therefore, seed and fruit of TKM may be utilized for a wide range of health benefits associated with antioxidant and anti-inflammatory activities.

Allelopathic Effect of Ganghwa mugwort (Artemisia spp.) on Seed Germination and Seedling Growth of Plants (강화약쑥 추출물이 종자발아 및 유식물 생장에 미치는 알레로파시 효과)

  • This study was conducted to identify allelopathic effect of Ganghwa domestic Artemisia spp., named Sajabalssuk and Ssajuarissuk, for various receptor plants including clover (Trifolium repens L.), alfalfa (Medicago sativa L.), lawn grass (Zoysia japonica Steud.), dandelion (Taraxacum platycarpum Dahlst.), and dahurianpatrinia (Patrinia scabiosaefolia Fisch. ex Trevir). Receptor plants were treated with the aqueous and essential oil extract of Artemisia plants. In consequence, their allelopathic effects were evaluated by measuring seed germination rates, seedling growth, and dry weights of the receptor plants. The seed germination and seedling growth of the receptor plants were inhibited by all treatments of both aqueous and essential oil extracts of the Artemisia plants, and, in addition, the inhibitory effects were increased according to the higher concentration. Among the donor plants, A. $sp.^*III$ showed most effective allelopathic effect. Comparing the alleopathic effect among the receptor plants, seed germination was most inhibited in lawn grass while inhibitory effect of seedling growth was comparatively higher in dandelion. Although inhibitory effects were comparatively lower, the allelopathic effects of Artemisia plants were identified in clover and alfalfa since the seedling growth of these plants were inhibited more than 70%. Thus, in result, Ganghwa domestic Artemisia spp. could be possibly used for weed control since natural products of the plants showed inhibitory effects on seed germination and seedling growth of various receptor plants.

Evaluation of Abelmoschus Esculentus Seed for Antioxidant and Phytochemical Analysis Using In vitro Assays

  • The aim of this study was to assess the in vitro potential of methanolic seed extract of Abelmoschus esculentus as a natural antioxidant. The DPPH activity of the Ethyl acetate soluble fraction (10, 20, 40, 80, and $160{\mu}g/mL$) was increased in a dose dependent manner, which was found in the range of 18.97-90.47% as compared to ascorbic acid 26.44-93.71%. The $IC_{50}$ values of Ethyl acetate soluble fraction (EAES) and ascorbicacid in DPPH radical scavenging assay were obtained to be 28.12 and $18.43{\mu}g/mL$, respectively. Measurement of polyphenol content of the EAES of A. esculentus seed was achieved using Folin-Ciocalteau reagent containing 53.80 mg/g of total phenolic content, which was found signicantly higher when compared to reference standard gallic acid. Similarly total flavonoids and proabthocyanidis of EAES and chloroform soluble fraction (CAES) were found significantly 147 mg/g and 14.24 mg/g respectively when both compared to reference standard quercetin. EAES exhibited high significant lipid peroxidation inhibition effects in a dose-dependent manner, with $IC_{50}$ values of $38.08{\mu}g/mL$, whereas, standard quercetin, with $IC_{50}$ value of $36.67{\mu}g/mL$. All extract/fractions showed dose dependent reducing power ability and these differences were statistically significant (p<0.001). The results obtained in this study clearly indicate that A. esculentus seed has a signicant potential to use as a natural antioxidant agent.

Protective Effects of Helianthus annuus Seed Extract against Chemical-Induced Neuronal Cell Death (해바라기씨 추출물의 뇌세포에 대한 사멸 보호 효과)

  • To develop an anti-dementia agent with potential therapeutic value in the protection of neuronal cells, we selected a water extract of Helianthus annuus seed for analysis. We measured acetylcholinesterase inhibitory activity in the extract, and analyzed the protective effect of the extract on neuronal cell death induced by hydrogen peroxide, or amyloid ${\beta}-peptide$, of SH-SY5Y neuroblastoma cells. The result showed that the extinct exerted protective effects of 83%, 72% and 53% respectively, on cell death induced by 100M, 200M, and 500M hydrogen peroxide. Also, when 50M of amyloid ${\beta}-peptide$ was added to the cells, the extract showed a protective effect (up to 80%) on cell death. Overall, the results showed that the H. annuus extract inhibited acetylcholinesterase activity in a dose-dependent manner, and the extract also strongly protected against cell death induced by hydrogen peroxide or amyloid ${\beta}-peptide$.

Antioxidant Effects and Physiological Activities of Pumpkin (Cucurbita moschata Duch.) Extract from Different Aerial Parts (늙은 호박 부위별 추출물의 항산화 효과 및 생리활성에 관한 연구)

  • This study investigated the antioxidant effects and physiological activities of extract from various parts of pumpkin (Cucurbita moschata Duch.) such as leaf, flesh, skin and seed. Different dried parts of C. moschata were boiled in ethanol for three hours, evaporated, and lyophilized with a freeze-dryer. Total polyphenol contents were the highest in leaf ($29.62{\pm}0.88mg$ GAE/g DM). The same result was observed in the measurement of the flavonoid content. The leaf extract of C. moschata had stronger DPPH radical-scavenging activities and ABTS-radical scavenging activities than any other part. Leaf extract of C. moschata was found to have the highest ferric-reducing antioxidant power (FRAP) value. Meanwhile, at 0.1 mg/mL of all extracts higher degree of tyrosinase inhibitory effects than kojic acid was observed (leaf; 49.58%, flesh; 40.20%, skin; 41.30%, seed; 36.79% and kojic acid; 20.70%). The glycation inhibitory activity decreased in the order leaf>skin>flesh>seed. The ${\alpha}$-glucosidase inhibitory activity of seed extract was the highest with a value of 63.58% at the concentration of 1 mg/mL.

Antioxidant and Anticancer Activities of Methanol Extracts Prepared from Different Parts of Jangseong Daebong Persimmon (Diospyros kaki cv. Hachiya) (장성 대봉감의 부위별 메탄올 추출물의 항산화 활성 및 항암 활성)

  • The antioxidant activity of methanol extracts from five different parts (flesh, peel, core, seed, calyx) of Jangseong Daebong persimmon (Diospyros kaki cv. Hachiya) were evaluated by determining total phenol content (TPC), DPPH radical scavenging activity (RSA), ABTS RSA, and reducing power (RP). The flesh extract gave the highest yield (92.93%) while the lowest yield was obtained from the seed (5.17%). The seed extract showed the highest total phenolic content ($76.47\pm0.009$ mg GAE/g extract), DPPH RSA ($IC_{50}=52.05\pm1.61\;{\mu}g/mL$), ABTS RSA ($IC_{50}=30.94\pm0.41\;{\mu}g/mL$) and RP ($IC_{50}=87.94\pm0.37\;{\mu}g/mL$). In addition, the calyx extract also showed high antioxidant activity. On the other hand, the core extract gave the lowest TPC and all antioxidant assays. In particular, HT-29 cells showed extensive cell death when treated with $500\;{\mu}g/mL$ of calyx extracts. Thus, these results suggest that methanolic extracts of Jangseong Daebong persimmon seed and calyx may serve as a potential source of natural antioxidant for food and nutraceutical application.

Extraction Yield and Anti-Yeast Activity of Extract from Green Tea Seeds by Pretreatment and Extraction Conditions (녹차씨 전처리와 추출 조건에 따른 녹차씨 추출물의 추출 수율 및 항효모 활성)

  • Green tea (Camellia sinensis) seed extract (GTSE) was prepared under various pretreatment conditions and used to investigate its extraction yield and anti-yeast activity. Anti-yeast activity of GTSE from seeds with or without the coats was the same, whereas the extraction yield was slightly higher in extract from seeds without the coat. Anti-yeast activity of GTSE from seeds with different water contents or particle sizes was the same, whereas the extraction yield was highest in extract from seeds with 7.3% moisture or a smaller particle size. Anti-yeast activity of defatted green tea seed extract (DGTSE) was the same as that of GTSE. Extraction yield was higher in DGTSE from defatted seeds by the oil press machine compared to hexane extraction. Defatted green tea seed (DGTS), a by-product from the oil extraction process, is a good natural source of anti-yeast preservative. The extraction yield and anti-yeast activity of DGTSE were investigated using various extraction solvents, temperatures, and times. The results show that water was an economic extraction solvent, and anti-yeast activity of DGTSE was unstable at $90^{\circ}C$. These results suggest that water, extraction temperature of $50^{\circ}C$, and extraction time of 4 h were the most efficient for extraction of anti-yeast compounds from DGTS.

Effects of Feral Haw (Crataegus pinnatifida BUNGE) Seed Extracts on the Antioxidant Activities (야생산사(Crataegus pinnatifida BUNGE)씨 생리활성물질의 산화억제 효과)

  • Haw (Crataegus pinnatifida $B_{UNGE}$) is a medicinal plant commonly used in Korea due to its beneficial health effects. This study was performed to investigate the antioxidative activity of bioactive components from haw seed. Freeze-dried seed was processed with various solvents such as 70% methanol, 70% ethanol, a mixture of chloroform and methanol (2:1, v / v), n-butanol and ethyl acetate (EA). DPPH, reducing power, ferric reducing antioxidant power (FRAP), and ABTS radical scavenging activity decreased in this order: 70% methanol, 70% ethanol, EA, CM, n-butanol extracts. NO radical scavenging activity was high in the n-butanol extract (36.73% in 0.8 mg/ml concentration) and low in the 70% ethanol extract (24.96% in 0.8 mg/ml concentration). $OH^-$ radical scavenging activity of n-butanol extract (40.42% in 0.8 mg/ml concentration) was higher than the other extracts, but EA extract was the lowest (13.39%) at the same concentration. The results show that each extract contained various antioxidative substances and it seemed that different influence at each antioxidative activity.
