명아주(Chenopodium album)와 바랭이( Digitaria sanguinalis)의 경쟁이 에너지 분해에 미치는 효과 (The Effect of Energy Allocation on Competition of Chenopodium album and Digitaria sanguinalis)

  • The effects of density and soil nutrient regime on competition between individuals in pure and mixed populations of two annuals. Chenopodium album and Digitaria sanguinalis were investigated at the level of enegy allocation. Seedling emergence of two species was extremely high (>90%) in both pure and mixed culture irrespective of the density and nutrient regime, but percentage of seedling establishment or fertile plant became gradually low with increasing density owing to 'self-thinning'. The mean plant dry weight was significantly reduced with increasing density and decreasing soil fertility. The dry matter production of D. sanguinalis in mixture was markedly greater than in pure culture under medium and high density. Also, as considered the number of seed production as reproductive allocation, relationship between them and the number of seed production per plant showed a similar tendency. Therefore, these results indicated that D. sanguinalis had a greater competitive advantage than C. album and energy allocations to variious organs were regulated by plastic response rather than determined genetically.

Elicitor에 의한 baccatin III의 세포내 생합성 조절연구 (Elicitors for the Regulation of baccatin III Biosynthesis in Plant Cell Culture System)

  • The yeast extract, coconut water, safflower seed oil, arachidonic acid, linolenic acid, jasmonic acid and methyl jasmonate were added to Gamborg's B$_{5}$ medium. The changes on productivity of baccatin III were estimated every 30 minutes and the results were compared using the selected high yielding cell culture system of Taxus cuspidata. In most cases, the peaks of baccatin III productivity occured at 90~120 min after addition of elicitors. Among the compound elicitors, safflower seed oil showed the highest productivity of baccatin III. Also arachidonic acid and linolenic acid increased the baccatin III production.

Biocontrol Effect of Gliocladium virens G1 and Soil Amendment on Astragal Stem Rot Caused by Rhizoctonia solani

  • In order to find an environment-friendly method to suppress astragal stem rot caused by the isolates of Rhizoctonia solani AG 1 and AG 4, we tested an antagonistic fungus Gliocladium virens G1 was evaluated as a biocontrol agent and estimated inorganic compounds and organic materials were tested for their effect of the disease suppression. G. virens G1 effectively inhibited mycelial growth in a dual culture and caused mycelial lysis in the culture filtrate. No adverse effect was observed when examined for seed germination and seedling growth. Promoted seedling growth was observed with the seed treatment. Seeds of astragal plant were germinated higher in the sterile soil than the natural soil. Of 14 inorganics tested, alum, aluminum sulfate and calcium oxide significantly suppressed the mycelial growth and sclerotial germination. Milled pine bark and oak sawdust also suppressed the mycelial growth. Soil amended with 1% of G. virens G1 composted with pine bark (w/v) significantly controlled astragal stem rot in the glasshouse experiments.

  • The study was conducted to determine the optimal basal media and the concentration of plant growth regulator for germination of seeds and growth of plantlet from Dendrobium monile. The results obtained were summarized as follows. Germination was similar in light and dark condition, but the growth of plantlet after germination was better under dark than under light condition in several media. Germination was best in Hyponex and Kyoto solution medium among the 9 media tested. The number of roots/shoot was most in the Hyponex medium containing 0,1ppm NAA and 1.Oppm BA.

한국 및 일본의 굴 양식에 관한 연구 -억제종묘의 비교 양식- (A Comparative Study of Oyster Culture in Japan and Korea Culture of Hardened Seed Oyster)

  • 양식굴의 주요 생산지인 한국의 한산만 및 일본의 광도만산 억제종묘를 광도시 앞 바다의 이도 굴양식장에 수하양식하여 1972년 6월부터 1973년 6월까지 매월 수온$\cdot$염분등의 환경요인과 양억제 종묘에 대한 각고$\cdot$각장$\cdot$육중양$\cdot$함수율$\cdot$비만도 그리고 생식소의 발달과정을 비교 검토하였다. 1. 본 실험을 한 광도시 앞 바다의 이도 굴 양식장은 한국의 한산만 굴 양식장의 수온 및 염분에 비하여 다소 고수온$\cdot$저염분$\cdot$이었다. 2. 양억제종묘의 각고$\cdot$각장의 성장을 보면, 한산만산 억제종묘가 다소 우세하였고, 특히 육중양의 증량이 뚜렷하게 우세하였다 3. 양억제종묘의 생식소의 발달과정에 대한 관찰결과, 1972년 6월 이후 12월까지는 생식소의 변화는 큰 차가 없었으나 다음 해 생식소의 발달이 시작되는 시기인 2월 이후에는 한산만산 억제종묘의 생식소가 광도만산 억제종묘의 생식소보다 조기에 발달하였다. 종묘의 성장, 육질의 비만 및 생식소의 발달과정 등을 금후종묘 이용도의 기준으로 볼 때, 한도만산 억제종묘가 광도만산 억제종묘보다 생산적 유용성이 있는 것으로 관찰되었다.

  • 톳의 유성생식을 통한 인공종묘생산 방법을 확립하기 위하여 초기 생장에 미치는 조도, 광주기 및 온도의 효과를 연구하였다. 각기 다른 조도 (500, 1,000, 2,000, 4,000, 8,000 lux), 광주기 (8 : 16, 12 : 12, 16 : 8 (L : D))와 온도 (10, 15, 20, 25, $30^{\circ}C$) 조건에서 유배를 70일간 배양한 결과 초기 생장 단계의 최적 조건으로는 조도 2,000-4,000 lux, 광주기 16 : 8 (L : D)와 온도조건으로는 15-2$0^{\circ}C$가 효과적인 것으로 나타났다.

The Anticancer Properties of Lunasin Peptide from Aged Callus Induced by the Soybean Tissue Culture

  • Lunasin is small subunit peptide of coded from Gm2S-1 gene in soybean. It has been previously demonstrated that lunasin is a novel and promising cancer preventive peptide. Lunasin peptide is found only in the seed and not other tissues. And lunasin peptide starts to appear at 5 weeks after flowering and remains in the mature seed. We report here firstly lunasin peptide identified from soybean callus induced by the tissue culture and demonstrate its anticancer properties. The lunasin was identified and purified from soybean callus aged for 6 months. The callus lunasin($1{\mu}M$) inhibited the acetylation of histone H3 and H4 by 58.8% and 56.5%, respectively. And it fully inhibited foci formation compared to the values of the positive control(no lunasin) and negative control(no MCA). Purified lunasin was able to internalize into the cell and localized in the nucleus.

Repeated-batch Culture of Immobilized Gibberella fujikuroi B9 for Gibberellic Acid Production: An Optimization Study

  • The performance of immobilized fungal cells on celite beads for the production of gibberrelic acid was investigated in flasks and 7-L stirred-tank reactor. Repeated incubations of immobilized fungal cells increased cell concentrations and volumetric productivity. The maximum volumetric productivity obtained in the immobilized-cell culture was 3-fold greater than that in suspended-cell culture. The concentration of cotton seed flour (CSF), among the various nutrients supplied, most significantly influenced productivity and operational stability. Notably, insoluble components in CSF were found to be essential for production. CSF at 6 g/L with 60 g/L glucose was found to be optimal for gibberellic acid production and stable operation by preventing excessive cell growth.

Production of Virus Free Seeds using Meristem Culture in Tomato Plant under Tropical Conditions

  • Protocol was established for production of virus free healthy seeds using meristem ($0.3-0.5\;\cal{mm}$ in size) culture and field management under net house condition in tomato. The isolated meristem was found well established in MS liquid medium containing $0.1\;\cal{mg}\;1^{-1}\;of\;GA_3$. For shoot and root development either from primary meristem or from nodal segment of meristem derived plants, semisolid MS medium having $0.5\;\cal{mg}\;1^{-1}$ of IBA was found most effective. The elimination of the studied viruses (ToMV, CMV, ToLCV) in meristem-derived plants was confirmed by DAS-ELISA test. For field management of the virus eradicated meristem-derived plants, use of net house was found very effective measures to check viral vector visit and eventually infection. The meristem-derived plants were vigor and high yielder than the native seed derived plants and produced healthy seeds. Due to stop vector visit, no viral symptoms were observed in both $R_1\;and\;R_2$ plants cultivated in net house condition. Starting of viral infestation was observed in $R_2$ generation when they were planted in open house condition without control of vector visit. Therefore, for management of viral diseases, use of virus free meristem derived plantlets and their subsequent cultivation in soil under net house condition without using any vector killing insecticide can be recommended for producing healthy seeds in tomato. The developed protocol for environmentally healthy tomato seed production in Bangladesh may be used in the countries having similar tropical like environment conducive for viral vector visit.
