Use of Dietary Salt to Rainbow Trout (Oncorhynchus mykiss) for Increasing Seawater Adaptability (무지개송어(Onchorhynchus mykiss)의 해수 적응 능력 개선을 위한 식염사료의 적용)

  • One 30-day feeding trial was conducted to examine the effects of dietary salt on seawater adaptability of rainbow trout (Onchorhynchus mykiss) fed three experimental diets containing 0% (control), 4% and 8% salt. The experimetal period included 30 days of feeding trial in freshwater, 3 days of the step by step seawater acclimation with-out feeding diets, and 21 more days of seawater adaptation period (not with all experimental fish) with feeding the basal diet. Growth rates from triplicate groups were determined fur 30 days of feeding trial. Blood samples were taken at the begining and at the end of feeding trial, and 3 times (on 1st, 4th and 8th day) of the seawater adaptation period. Daily survival rates of duplicate groups from three experimetal treatments were recorded for 21 days of the seawater adaptation period. Total average initial and final fish weight were $149.5{\pm}7.6\;and\;187.1{\pm}7.6g$. Feed efficiency of fish fed diets containing 4% and 8% salt were significantly better than those of fish fed the control diet. Average cumulative survival rates were 72, 80 and 88% from the control, 4% and 8% salt diets, respectively. Pulse rate per minutes decreased with dietary salt level. Serum $Na^+\;and\;Cl^-$ concentrations of fish fed 4% and 8% salt diets were significantly higher than those of fish fed the control diet (P<0.05), however, the concentrations were stabilized after 8 days of seawater adaptation. Serum cortisol, glucose, cholesterol and tryglyceride concentrations, and the osmorality of fish decreased with dietary salt level, these values were significantly lower than those of fish fed the control diet. These results indicated that the dietary supplementation of salt could have advantages for seawater adaptability of rainbow trout.

Differential Seawater Adaptability in Three Different Sizes of Under-yearling Steelhead Trout

  • Seawater adaptability of steelhead trout increases along with the increase in the size of the fish, independent of parr-smolt transformation. Three 96 h seawater challenge tests were conducted to determine the size at which seawater adaptability of steelhead trout develops. Plasma Na+ and Cl- levels, moisture content, gill Na+/K+ ATPase activity, and mortality during the 96 h after direct transfer to seawater (32 ppt) were determined. Plasma Na+ and Cl- levels in 50 g fish continuously increased during the 96 h after the transfer to seawater (p<0.05), but the levels in 100 and 150 g fish leveled off after 24 h (p<0.05). Both 100 and 150 g size steelhead trout maintained muscle moisture content (%) better than 50 g size fish (p<0.05). Gill Na+/K+ ATPase activity in the 100 g size group increased in a time-dependent manner after transfer to seawater (p<0.05), whereas activity in the 50 and 150 g sizes did not increase (p>0.05), for which a possible explanation was discussed. A mere 2.6% mortality in both the 50 and 150 g size groups was observed. In conclusion, the current results indicate that 50 g size steelhead trout did not show development of a high level of hypoosmoregulatory capacity, whereas fish in the 100 and 150 g size groups showed a high level in our experimental conditions. Therefore, the steelhead trout larger than a 100 g size is recommended for transfer to seawater culture.

  • According to increasing demand of water mainly due to the growth of population and increased water consumption, many countries either face or worry about the shortage of fresh water. Proportionately, importance and efforts of each country to develop the potable water has been gradually increasing as well. Among others, desalination of seawater has been developed to one of the solutions mainly from the middle east and other arid regions to produce large quantity of fresh water from seawater. We installed beach seawater collector wells to develop the filtered seawater supply for desalination in a refinery. We came to a conclusion that the beach seawater collector well is one of the recommendable alternatives of seawater pre-treatment for desalination applications with lower operating cost and higher efficiency.

Seawater Adaptability of Land-locked Masu Salmon Oncorhynchus masou masou by Acclimation (순치 기간에 따른 육봉형 산천어(Oncorhynchus masou masou)의 해수 적응능력)

  • The seawater adaptability of land-locked masu salmon Oncorhynchus masou masou via acclimation was examined for aquaculture purposes. The survival, blood chemistry, and histological changes of masu salmon (150 g) were measured after 7-, 15-, and 30-day acclimation periods. After a total of 60 days in seawater cultures that incorporated the various acclimation periods, survival was 83.5, 87.2, and 91.0% for the 7-, 15-, and 30-day periods, respectively; thus, survival increased with longer periods of acclimation. Feeding efficiencies were 32.1, 52.0, and 40.6% for the 7-, 15-, and 30-day periods, and specific growth rates were 0.14, 0.26, and 0.23%, respectively. Generally, masu salmon appeared to exhibit better growth performance after an acclimation period of 15 days. Cortisol concentrations [mean ${\pm}$ SD] for 7, 15, and 30 days of acclimation were $21.0{\pm}6.5$, $17.8{\pm}4.8$, and $21.2{\pm}5.4\;{\mu}g/dl$, with the lowest values occurring with 15 days of acclimation. Osmolarities were $359.2{\pm}26.1$, $350.4{\pm}29.2$, and $354.6{\pm}29.3$ mOsm/kg, and glucose concentrations were $60.7{\pm}20.7$, $72.9{\pm}17.3$, and $76.6{\pm}14.1$ mg/dl for the 7-, 15-, and 30-day acclimation periods, respectively (P < 0.05). The histological study revealed that both gills and both kidneys of the masu salmon exhibited middle- to end-stage and middle-stage lesions in the 7- and 15-day groups, respectively, whereas these organs only had early-stage lesions in the 30-day group in the final experiment. Therefore, the seawater acclimation of masu salmon should involve more than 30 days in seawater.

Experimental Design in Laboratory for Ecological Restoration in the Slag Dumping Area

  • Experimental design for ecological restoration approach was investigated from the P' company's slag dumping area with higher pH value. The degree of pH recovery was measured by the injection of carbon dioxide from the LNG exhaust gas, and the residual carbon, for example, calcium and carbonate which can be controlled by artificial seaweed beds. The degree of adaptability from 3 algaes (Undaria pinnatifida, Sargassum horneri and Ecklonia stolonifera) and uptake nutrient function of Ecklonia stolonifera chosen in the first treatment were measured in the laboratory to determine the transplanting algae in artificial seaweed beds. The higher value of pH was decreased to 7.0~8.5 by injection of LNG exhaust gas with flow rate $20\;m^3/min$. In the experiment design at laboratory, the upper part of frond of Undaria pinnatifida and Sargassum horneri began to decay, and the color changed after 10 days. However, those of Ecklonia stolonifera were after 14~20 days. The uptake rate of nutrient from Ecklonia stolonifera was higher than those of other algaes, and those was similar pattern in the control (e.g. seawater); The DIN concentration uptake of Ecklonia stolonifera was $1.88\;{\mu}g/L/hr$ avg. in leachate, but $2.19\;{\mu}g/L/hr$ avg. in seawater. However, the ${PO_4}^3$-P concentration uptake was $0.18\;{\mu}g/L/hr$ avg. in leachate, but $0.31\;{\mu}g/L/hr$ avg. in seawater. These result indicated Ecklonia stolonifera uptaked these nutrients in the leachate as well as in seawater, and it may suggested for this species to transplant for constructing artificial seaweed beds.

A Study on the Seawater Filtration Characteristics of Single and Dual-filter Layer Well by Field Test (현장실증시험에 의한 단일 및 이중필터층 우물의 해수 여과 특성 연구)

  • This study performs to evaluate adaptability of seashore filtering type seawater-intake which adapts dua1 filter well alternative for direct seawater-intake. This study varies filter condition of seashore free surface aquifer which is composed of sand layer then installs real size dual filter well and single filter well to evaluate water permeability and proper pumping amount according to filter condition. According to result of step aquifer test, it is analysed that 110.3% synergy effect of water permeability coefficient is happened compare to single filter since dual filter well has better improvement. dual filter has higher water permeability coefficient compare to same pumping amount, this means dual filter has more improved water permeability than single filter. According to analysis result of continuous aquifer test, it is evaluated that dual filter well (SD1200) has higher water permeability than single filter well (SS800) by analysis of water permeability coefficient using monitoring well and gauging well, it is also analysed dual filter has 110.7% synergy effect of water permeability coefficient. As a evaluation result of pumping amount according to analysis of water level dropping rate, it is analysed that dual filter well increased 122.8% pumping amount compare to single filter well when water level dropping is 2.0 m. As a result of calculating proper pumping amount using water level dropping rate, it is analysed that dual filter well shows 136.0% higher pumping amount compare to single filter well. It is evaluated that proper pumping amount has 122.8~160% improvement compare to single filter, pumping amount improvement rate is 139.6% compare to averaged single filter. In other words, about 40% water intake efficiency can be improved by just installation of dual filter compare to normal well. Proper pumping amount of dual filter well using inflection point is 2843.3 L/min and it is evaluated that daily seawater intake amount is about $4,100m^3/day$ (${\fallingdotseq}4094.3m^3/day$) in one hole of dual filter well. Since it is possible to intake plenty of water in one hole, higher adaptability is anticipated. In case of intaking seawater using dual filter well, no worries regarding damages on facilities caused by natural disaster such as severe weather or typhoon, improvement of pollution is anticipated due to seashore sand layer acts like filter. Therefore, It can be alternative of environmental issue for existing seawater intake technique, can save maintenance expenses related to installation fee or damages and has excellent adaptability in economic aspect. The result of this study will be utilized as a basic data of site demonstration test for adaptation of riverside filtered water of upcoming dual filter well and this study is also anticipated to present standard of well design and construction related to riverside filter and seashore filter technique.

Long-Term Investigation of Marine-Derived Aspergillus Diversity in the Republic of Korea

  • Aspergillus species play a crucial role in terrestrial environments as degraders and are well known for producing various secondary metabolites. Recently, Aspergillus species have been discovered in marine environments, exhibiting adaptability to high salinity and producing diverse secondary metabolites with valuable properties. However, limited research has focused on their marine diversity, leading to inaccurate species identification. The current study addresses this gap by investigating diverse marine habitats in the Republic of Korea, including sediment, seawater, seaweed, and marine animals. From three coasts of the Korean Peninsula, 472 Aspergillus strains were isolated from the various marine habitats. A total of 41 species were accurately identified using multigenetic markers: internal transcribed spacer, calmodulin, and b-tubulin. The findings underscore the importance of accurate identification and provide a basis for elucidating the functional role of marine-derived Aspergillus species in marine ecosystems.

Gene Expression Profiles of Rainbow Trout Oncorhynchus mykiss after Salinity Challenge (염분 변화에 따른 무지개송어(Oncorhynchus mykiss)의 삼투조절 유전자 발현변화)

  • Euryhaline teleost have extraordinary ability to deal with a wide range of salinity changes. To study the seawater adaptability of rainbow trout Oncorhynchus mykiss (body weight 638±54 g, length 38.6±2 cm) to salinity increase fish were transferred from freshwater to 7, 14, 21, 28 and 32 psu and checked for mortality over 5 days. No mortality was observed in 0-32 psu. In fish transferred to 0-32 psu, blood osmolality was maintained within physiological range. The changes of serum enzyme activities (aspartate transaminase, AST and alanine transaminase, ALT) showed no significant level during experimental period. To explore the underlying molecular physiology of gill and kidney responsible for body fluid regulation, we measured mRNA expression of five genes, Na+/K+/2Cl- cotransporter1 (NKCC1), aquaporin3 (AQP3), cystic fibrosis transmembrane conductance regulator (CFTR), glucocorticoid receptor (GR) and growth hormone receptor (GHR) in response to salt stress. Based on our result, rainbow trout could tolerate gradual transfer up to 32 psu for 5 days without mortality under physiological stress. This study suggests to alleviate osmotic stress to fish, a gradually acclimation to increasing salinity is recommended.

Effects of Salinity and Temperature on the Survival of Vibrio cholerae non-O1 and Vibrio mimicus (Vibrio cholerae non-O1과 Vibrio mimicus의 생잔에 대한 염도와 온도의 영향)

  • Vibrio cholerae non-O1 and Vibrio mimicus, food poisoning bacteria, have detected frequently in fresh water and brackish water. To estabilsh prevention measures of food poisoning outbreak by these bacteria, the adaptability and population changes were examined in fresh water, brackish water $(10\%o\;NaCl)$ and seawater $(30\%o\;NaCl)$. Both species poorly survived as temperature increased regardless of water types employed. However, survival time was the shortest in fresh water and longest in seawater at $4^{\circ}C$. In case of brackish water, the bacteria survived best at $15^{\circ}C$ and population were varied only in small numbers. Any significant difference was not observed to both species with respect to water types and temperatures except V. mimicus survived about 5-6 days longer in brackish water. In conclusion, V. cholerae non-O1 and V. mimicus were not likely to be recovered In normal fresh water, brackish water and seawater, and both biological and physicochemical factors could affect survival of these species.

Heat tolerance of goats to increased daily maximum temperature and low salinity of drinking water in tropical humid regions

  • Objective: The daily maximum temperature and seawater level continuously increase as global warming continues. We examined the adaptability and production performance of heat-stressed goats with a supply of low-saline drinking water. Methods: Twelve Kacang and Kacang Etawah cross goats were exposed to two climatic conditions (control, 25℃ to 33℃, 83% relative humidity [RH], temperature humidity index [THI]: 76 to 86; and hot environment, 26℃ to 39℃, 81% RH, THI: 77 to 94) and two salt levels in drinking water (0% and 0.4% NaCl). The experimental design was a Latin Square (4×4) with four treatments and four periods (28 days each). Results: Temperature of the rectal, skin, and udder, and respiration rate rose, reached a maximum level on the first day of heat exposures, and then recovered. Plasma sodium rose at 0.4% NaCl level, while the hot environment and salinity treatments increased the drinking water to dry matter (DM) intake ratio. Water excretion was elevated in the hot environment but lowered by the increase in salinity. Total lying time increased, whereas change position frequency decreased in the hot condition. Lying and ruminating and total ruminating time increased and explained the enhanced DM digestibility in the hot conditions. Conclusion: The goats exhibited a high level of plasma sodium as salinity increased, and they demonstrated physiological and behavioral alterations while maintaining their production performances under increasing daily maximum temperatures.
