Substitution effect of white radish (Raphanus sativus L.)' by-product and tunic of sea squirt (Halocynthia rorentzi, von Drasche) for Undaria pinnatifida in feed of abalone (Haliotis discus, Reeve 1846)

  • Substitution effect of white radish' by-product (WRB) and tunic of sea squirt (TSS) for Undaria in feed on growth and carcass composition of abalone was determined. Eight-hundred forty juveniles were distributed into 12 containers. Three formulated diets were prepared in triplicate. A 15% Undaria was included in the control diet. The 15% Undaria was substituted with a same amount of WRB and TSS, referred to as the DRG and TSS diets, respectively. Finally, dry Undaria was also prepared to compare effect of the formulated diets on performance of abalone. The experimental diets were fed to abalone once a day at a satiation level with a little leftover. The feeding trial lasted for 16 weeks. Crude protein, crude lipid, and ash content of the experimental diets changed over all period of time. Higher crude protein and lipid and ash contents retained in all formulated diets compared to the Undaria were observed at 24 and 48 h after seawater immersion. Survival of abalone was not affected by the experimental diets. Weight gain and specific growth rate (SGR) of abalone fed the TSS diet was greater than all other diets. Weight gain and SGR of abalone fed the control diet was not different from those of abalone fed the WRB diet. Proximates of the soft body of abalone was affected by the diets. In conclusion, WRB and TSS are the promising feed ingredients to replace Undaria in abalone feed; especially, TSS is superior to either Undaria or WRB. The formulated diets produced improved growth performance of abalone over the single Undaria.

Effect of Temperature and Dissolved Oxygen on the Survival Rate and Physiological Response of the Warty Sea Squirt Styela clava (수온과 용존산소 변화에 따른 미더덕 Styela clava의 생존율 및 생리적 반응)

  • Decrease in dissolved oxygen concentrations associated with temperature fluctuation is an important criteria to evaluate the mortality rate of the species. Based on this parameter, we investigated the survival rate, physiological response and histological change of warty sea squirt. It was found that the survival rate of the warty sea squirt species was 63.3% at $23^{\circ}C$ and 56.6% at $26^{\circ}C$ respectively. However, exposure of six days at $29^{\circ}C$ caused deaths among species, which indicated the 6day-$LT^{50}$ of the tested species to be $24.58^{\circ}C$ ($19.48{\sim}35.48^{\circ}C$). Further, after 11 day of exposure, the dissolved oxygen concentration has been found to decrease, with the survival rate of 20% at $4.0mg\;L^{-1}$ and deaths at $2.0mg\;L^{-1}$, thus 11day-$LC^{50}$ calculated to be $3.88mg\;L^{-1}$ ($3.29{\sim}4.57mg\;L^{-1}$). In addition, decrease in rate of oxygen consumption and excretion of ammonia was also noted at this critical water temperature and dissolve doxygen concentration. Moreover, there has been common histopathological changes were observed in warty sea squirt's gill pouch, digestive tract, and tunic as follows such as: proliferation of epithelial cells, condensation and necrosis, permeation of phagocyte and blood cell, loss of cilium and muscular fiber degeneration. Based on our study results, we suggest that these parameters can also be useful to evaluate the survival rate and physiological response in other species.

Starvation Effects on Occurrence of Tunic Softness Syndrome in Sea Squirt Halocynthia roretzi (양식 멍게(Halocynthia roretzi)의 물렁증 발생에 미치는 절식의 영향)

  • We examined the effect of starvation on the occurrence of tunic softness to determine the cause of mass mortality of cultured Halocynthia roretzi (Drasche) in the Tongyeong region, Korea. In terms of the survival rate of H. roretzi and the occurrence rate of tunic softness, H. roretzi starved for 35 days at water temperatures of 8, 12, and $15^{\circ}C{\pm}0.5^{\circ}C$ (room temperature of $15^{\circ}C{\pm}1^{\circ}C$) did not exhibit tunic softness at water temperatures of either $8^{\circ}C$ or $12^{\circ}C$. for morphological changes, although the tunic of H. roretzi was shrunken and became visibly smaller with a darkening color in all experimental groups, as compared to the state prior to starvation, its tunics bulbs continuously. The ratio of RNA/DNA concentrations and protein contents for each of the tunic sections were lower in the starved group. Our results indicate that tunic softness is not related to feeding deficiency, as no histopathological symptoms were apparent in the digestive gland or tunic of H. roretzi due to starvation.

Analysis of Polycyclic Aromatic Hydrocarbons (PAHs) in Shellfishes from Korean Coastal Area (우리나라 연안해역 생산 패류 중 다환방향족탄화수소 분석)

  • Polycyclic Aromatic Hydrocarbons (PAHs) were analyzed and safety evaluation was carried out in Korean coastal area. The target congeners were benzo[a]pyrene, benzo[a]anthracene, benzo[b]fluoranthene, and chrysene. The method of analysis was significant. The highest PAHs 4 congener concentrations (12.2 ㎍/kg) was found in the mussel of Gangwon area. Comparison among the average concentration of PAHs for each congener showed that, benzo[a] anthracene was found in mussel (0.276 ㎍/kg), oyster (0.463 ㎍/kg), ark shell clam (1.92 ㎍/kg), and chrysene was found in mussel (0.848 ㎍/kg), oyster (1.36 ㎍/kg), scallop (0.489 ㎍/kg), sea squirt (3.07 ㎍/kg), and ark shell clam (0.449 ㎍/kg). In addition, benzo[b]fluoranthene was found in mussel (0.253 ㎍/kg), scallop (0.244 ㎍/kg), and sea squirt (1.64 ㎍/kg). The most hazardous benzo[a]pyrene was found in mussel (0.147 ㎍/kg), and scallop (0.244 ㎍/kg), it was not detected in the other shellfishes. However, all the PAHs levels recorded in this study did not exceed international MRLs. There was no significant difference among the PAHs concentration in shellfish based on sampling area or species. Body exposure was calculated based on PAHs concentration and intake rate. The results of this study indicated that PAHs in shellfish were within acceptable safe levels.

Oxygen Consumption of Sea Squirt Halocynthia roretzi Depending on the Water Temperature and Body Size (수온과 크기에 따른 멍게(Halocynthia roretzi)의 산소 소비)

  • The oxygen consumption rate (OCR) based on the water temperature and body size of the sea squirt Halocynthia roretzi was examined to provide quantitative information about the metabolic response of the species. OCRs were measured using a closed flow-through respirometer at four different water temperatures (10, 15, 20 and 25℃) and two different body sizes (21.4±1.1 g and 150.5±1.3 g, wet weight) with triplicates of each treatment. OCR increased as water temperature increased at both body sizes, but decreased as body size increased regardless of the water temperature (P<0.001). The effect of body size evaluated as a power function ranged from 0.8055 to 0.8884. The highest Q10 values in the small and large size groups ranged from 15 to 20℃ and 20 to 25℃, respectively. The metabolic daily energy loss rate via respiration at all tested temperatures ranged from 56.2 to 106.1 J g-1 d-1 in the small-size group and from 44.5 to 92.0 J g-1 d-1 in the large-size group. Our results indicate that the metabolic response of H. roretzi highly depends on fluctuating water temperature at a given life stage.

Accumulation and Depuration of Paralytic Shellfish Poison in Marine Organisms (수산생물 종류별 마비성 패류독소 축적 및 정화)

  • To compare the accumulation of paralytic shellfish poison (PSP) in different marine organisms, the occurrence and variation of PSP were surveyed in blue mussel Mytilus edulis, oyster Crassostrea gigas, short neck clam Ruditapes philippinarum, bay scallop Argopecten irradians, and warty sea squirt Styela clava collected from Jinhae Bay, Korea, in 2005 and 2006 year. We also investigated the ability of the blue mussel to detoxify PSP by relaying and depuration (via the water flow or water circulation system). In the marine organisms examined, PSP levels were the highest in blue mussel, followed in order by bay scallop, oyster, short neck clam, and warty sea squirt. Comparing the maximum PSP levels in the bivalve species examined in 2005 and 2006, PSP in blue mussel was 1.6-2.0, 4.0-5.9, and 5.1-6.0 times higher than in bay scallop, oyster, and short neck clam, respectively. Therefore, blue mussel could be useful as a bioindicator for PSP monitoring. With the increasing PSP levels in blue mussel in 2006, the proportion of PSP in its digestive gland increased to 95.1% when the maximum level was detected from the whole tissues of blue mussel on May 29. Subsequently, the PSP proportion in the digestive gland decreased as the PSP level in whole tissue decreased. The detoxification of PSP in blue mussel was greatest with relaying, followed by the water flow, and water circulation systems. Relaying decreased the PSP level below the regulatory limit of $80{\mu}g$/100 g after 2 days in low toxic sample with $124{\mu}g$/100 g, and after 7 days in high toxic sample with $401{\mu}g$/100 g. During depuration in the blue mussel with $401{\mu}g$/100 g via the water flow system, the PSP amounts in the digestive gland decreased by about 50% after 1 day, and about 77% after 7 days. In contrast, the PSP amounts in the soft body, gill, and mantle did not change significantly with depuration.

Growth Variation with Culture Depth, Interval Between Suspended Lines and Locality of Seedling of Sea Squirt, Halocynthia roretzi (von Drasche) in Suspended Culture in the East Sea (동해안 지역 우렁쉥이, Halocynthia roretzi의 양식 수심, 수하연 간격 및 종묘생산지에 따른 성장 특성)

  • We examined the growth variation with water depth, interval between suspended lines and locality of seedling production of sea squirt. Halocpthia roretzi, measuring approximately $10.1\~15.3$ mm in tunic height, was cultured at the farm near Guryongpo, Pohang city from 7 January to 6 December 1998 (333 days). Growth condition depending on culture depth was most favourable at the depth of $10\~12$m, It was concluded that nutritional conditions were better at the depth since there was a greater mount of phytoplankton than at any other depth in the water column. Growth condition depending on distance between suspended lines was most favourable at the interval of 2 m and 2.5 m than at the interval of t m and 1.5 m, There was no significance difference in growth variation with locality of seedling. The seasonal variations in growth of tunic height, meat and total weight were correlated with temperature and chlorophyll a, In the depth of $10\~12m$ the rate of increase in tunic height was generally greatest in the growth period II ($244{\mu}m/day$) and IV ($338{\mu}m/day$), lower in the period I ($108{\mu}m/day$), and slightly increased during the period III ($185{\mu}m/day$). The water column have showed a typical seasonal pattern with temperature ranged between $12.07{\sim}24.14^{\circ}C$ and over $20^{\circ}C$ between July and September.

Survey on Waste Rates of Foods for Menu Planning (합리적인 식단작성을 위한 식품폐기율 조사 연구)

  • The waste rates of 98 food items after pre-preparation were surveyed to provide database for good menu planning. The waste rates turned out 0-64.27% in vegetables, 6.38-7.03% in potatoes, 6.25-68.75% in fishes and shellfishes, 16.61% in eggs, and 16.00-56.84% in fruits. Foods with high waste rates were vegetables, fishes and shellfishes. Survey results were compared with other food composition tables. Foods with 30% higher waste rate than other food composition tables were squash leaf, pacific ocean perch, sweet potato stalk, water cress, green peas, alaska pollack, bluefin tuna, beka squid, crown daisy, dodok, amaranth, beef ribs. Food which waste rates turned out to be decreased by about 30% in this study were corb shell, pomfret, sea mussel, warty sea squirt. For the menu planning, reasonaly exact waste rates for each food items are essential. Since survey results show significant deviations, there should be more studies for exact waste rates for each food.

Population Ecological Study of Cultured Sea Squirt (Halocynthia roretzi) and Management Implications (양식 우렁쉥이(Halocynthia roretzi)의 자원 생태학적 분석 및 적정관리 방안)

  • A population ecological study was carried out to estimate survival and growth rates, biomass, biological production and turnover ratio of cultured sea squirt, Halocynthia roretzi, by growth stages, using data from in situ culture experiment off Hansando in the southern part of Korea from February 1985 to July 1986. The squirt population followed an exponential decay function and the instantaneous coefficient of total mortality (Z) was estimated to be 0.0614 $month^{-1}$(Var (Z) = 0.000126). Growths in total weight and meat weight of squirts were expressed as linear functions during the period of culture experiment. The growth of squirts showed a negative correlation with the water temperature. The mean biomass per string ranged from 2.14 kg for March of the first year to 16.26 kg for March of the next year. The biological production per string was estimated to range from 3.28 kg for the first summer (June - July) to 6.46kg for the first late winter (February-March). The peak of turnover ratio occurred in the late winter (February-March) as 3.013 and the ratios sharply declined thereafter. Based on the results of this study, management implications for culturing sea squirts were also suggested. The optimum harvest time ($t_{mb}$) when the peak biomass in terms of total weight occurred was estimated to be late June of the second year, which corresponded to 16.7 months after the main hanging. However, the time when the peak biomass in terms of meat weight was occurred was early July of the second year. The maximum harvest biomass was 17.4 kg per string in terms of total weight and 6.3 kg per string in terms of meat weight. In conclusion, the process of culture should be conducted on the basis of the knowledge of population ecological theories as shown in this study.
