Preparation and Bioavailability of Oriental Medicine containing Baicalin (II) : Gastro-Intestinal Absorption and Antibacterial Effect of Coprecipitated Product of Scutellariae Radix and Coptidis Rhizoma (바이칼린 함유 생약의 제제화 및 생체 이용률 (제 2보) : 황금 및 황련 공침물의 장내 흡수 및 항균 효과)

  • Precipitation was formed during the preparation of decoction from a mixure of Scutellariae Radix and Coptidis Rhizoma or Phellodendri Cortex according to the prescription of Hwang-ryean-hae-dog-tang. Baicalin and berberine, the active ingredients of the two herbal medicine were identified in coprecipitated product. Pills were prepared using the coprecipitated product and various binders. The dissolution rate of baicalin and berberine from pills was increased in at pH1.2 when acacia or tragacanth was used. The absorption rate of baicalin from the coprecipitated product was faster than that from Scutellaria extract, but the absorption of berberine from CPP was slower in stomach, duodenum and jejunum of rats compared with Coptis extract. The time required for the maximum serum concentration (Cmax) of baicalin and berberine from CPP in mice were 150 and 200 min after oral administration, respectively. The maximum serum concentration of baicalin from CPP in mice was higher than Scutellaria extract, but the concentration of berberine was lower compared with Coptis extract. The minimum inhibitory concentration of CPP was below $50\;{\mu}g/ml$ against gram positive bacteria, and was higher than that against gram negative bacteria. The antibacterial activity of CPP was lower than that of herberine, but was more potent than Scutellaria extract. It was found that the inhibition rates of growth by CPP against S. epidermidis, K. pneumoniae, B. cereus and S.aureus were 60.0, 51.1, 45.4 and 39.9%, respectively.

  • The Literatual Study on Treatment of Toothbleeding Through the literatual study on treatment of toothbleeding, we concluded as follows, 1. The causes of the toothbleeding are divided into heat of stomach (胃熱) and deficiency of renal (腎虛). If caused by heat of stomach, the treatment methods are Ryang-Hyul-Ji-Hyul(凉血止血), Cheong-Yul-Sa-Hwa(淸熱瀉火). If caused by deficiency of renal, the treatment methods are Ryang-Hyul-Ji-Hyul(凉血止血), Ja-Eum-Gang-Hwa(滋陰降火). 2. The medications are several - taking a medicine for internal use(內服法), keeping with one's mouth full of an infusion and spoutting it Haamsoobup(含漱法), and others like taking a medicine for external use(外用法) only. 3. The treatment drugs divided by medication are as follows : Taking a medicine for internal use : Cheongwesan(淸胃散), Gamroeum(甘露飮), Joweseunggitang(調胃承氣湯), Seogagjihwangtang(犀角地黃湯), Yugmijihwangtang(六味地黃湯), Palrnihwan(八味丸), Soansinhwan(小安腎丸) etc... Haamsoobup : Gudogeum(救毒飮), Jihwangtang(地黃湯), Mangchosan(莽章散) etc... Taking a medicine for external use : Pilseungsan(必勝散), Rogposan(綠包散, Hyunggeousan(荊槐散), Bingoksan(氷玉散), Sahyangsan(麝香散), Ulgeumsan(鬱金散), Yongnoisan(龍腦激) are used. 4. A coposition of the medication : Cheong-Yul medicine(淸熱藥), Bo-IK medicine(補益藥), Ji-Hyul medicine(止血藥), Gae-Gyu medicine(開竅藥), Su-sap medicine(收澁藥) are mainly used. Cheong-Yul medicine : Rehmanniae Radix(鮮地黃), Moutan Cotex(牧丹皮), Coptidis Rhizoma(黃連), Scutellariae Radix(黃芩) Bo-Ik medicine : Glycyrrhizae Radix(甘草), Angelicae gigantis Radix(當歸), Rehmanniae Radix Preparat(熟地黃), Ginseng Radix(人蔘) Ji-Hyul medicine : Biotae Cacumen(側柏葉), Sophorae Flos(槐花), Typhae Pollen(蒲黃) Gae-Gyu medicine : Moschus(麝香), Bomeolum(氷片) Su-sap medicine : Alumem(白礬) 5. A result of the observation on the medication : Taking a medicine for internal use : Glycyrrhizae Radix, Rehmanniae Radix Preparat, Rehmanniae Radix, Moutan Cortex, Scutellariae Radix are used. Yugmijihwangtang and Cheong-Yul medicine are mainly used. Taking a medicine for external use : Alumem, Moschus, Salt(鹽), Asari herba cum Radice(細辛) are mainly used. Especially there is a method that uses a burnt drug. Haamsoobup and taking a medicine for external use : Medicines of Cheong-Yul-Ryang-Hyul(淸熱凉血) and Su-Ryum-Ji-Hyul(收斂止血) are used.

Protective effects of Scutellariae Radix on impairments in learning and memory induced by brain ischemia in rats (뇌허혈로 인한 흰쥐에서의 기억력 및 학습효과 저해에 대한 황금의 보호효과)

  • Objectives : It has been reported previously that the roots of Scutellaria baicalensis (known as Huang-Gum in Korean, henceforth referred to as S. baicalensis) could prevent neuronal cell death after global cerebral ischemia. In Genuine Korean medicine, S. baicalensis is known to relieve fever in upper body, and it was thus thought to be able to alleviate deteriorations in brain function. Methods : The protective effects of S. baicalensis against post-stroke memory retardation using 4-vessel occlusion model were examined in the present study. Results : S. baicalensis was shown to significantly alleviate the deficits in learning and memory by increasing the fraction of time spent in the quadrant in which the platform was initially placed ($34.9\;{\pm}\;3.2%$, p < 0.05) compared to that of the ischemia group ($28.0\;{\pm}\;2.5%$). The cytoprotective effect of S. baicalensis on CA1 hippocampal neurons was evaluated by measuring the neuronal cell density. Neuronal cell density in S. baicalensis extracts-treated ischemia group ($138.0\;{\pm}\;13.6\;cells/mm^2$) was significantly increased compared to saline-treated ischemia group ($22.1 \;{\pm}\;9.3\;cells/mm^2$, p < 0.05). In the study of OX-42 immunohistochemistry, S. baicalensis could decrease the micrgial activation in hippocampus after brain ischemia. Conclusion : These results may provide experimental support for the use of S. baicalensis in treating post-stroke memory impairment.

  • To beef up natural immunity, we have used Bojoongikgitang which has been known to treat enervation in the oriental medicine. This study is analyzed out structure material and the chief virtue of a prescription through Literature Study on Bojoongikgitang and Clinical Application. And this study is investigated to make sure of the necessity and additional symptoms in using Bojoongikgitang. The results are as follows : 1. It is regarded that the structure materials of Bojoongikgitang consist of Astragali Radix one jeon(錢) five poon(分), Ginseng Radix, Atractylodis Macrocephalae Rhizoma, Glycyrrhizae Radix one jeon for each, Citri Pericarpium, Angelicae Gigantis Radix five poon for each, Cimicifugae Rhizoma, Bupleuri Radix three poon for each. 2. The necessity symptoms in using Bojoongikgitang are a pale complexion, drowsy eyes(目無精光), vigorless, lethargy, sluggish talk. 3. The fittest prescriptions prior to the necessity symptoms in using Bojoongikgitang are Bojoongikgitang added Paeoniae Radix Alba, Scutellariae Radix in fever, exterior heat, mild fever, Bojoongikgitang added Ephedrae Radix, Tritici Fructus Levis, Aconiti Iateralis Preparata Radix in spontaneous sweating, spontaneous sweating by yang deficiency, Bojoongikgitang added Paeoniae Radix Alba, Scutellariae Radix in feeling the pulse like a flood, largeness and weakness, scatter and largeness, flood and largeness for diagnosis respectively. Bojoongikgitang Entering the heart channel by culturing the blood prescription in vexation, vexation and anxiety, Soongihwajoongtang in headache, DossiBojoongikgitang in rigor, Bojoongikgitang annexed Saengmaecsan in thirst, Daninsamtang or Jojoongikgitang in asthma, asthma by congestion of the upwardness, Eeegongsan in light eating, eschewing food, losing one's appetite, Ikweeseungyangtang in deficiency failing to control blood and blood collapsey. 4. To treat a functional disease is superior to organic one in using Bojoongikgitang.

  • In order to establish utilijzation of medicinal herb as vegetable by Hadroponcis, the present studies were conducted to inverstigate the experiment on year-round Production of medicinal herb. The results obtained are summaried as follows: Among Planting date, April 5 Planting date obtained the highest yield of Scutellariae Radix and Agastache rugosa Folium. Treatment of $10\times5cm$(200 Plant/$m^2$) among Planting densities showed the highest yield of Scutellariae Radix and Agastache rugosa Folium. Plant height was thick in spacious planting, and was thin dense planting. Stem diameter was thick in spacious planting, and was thin in dense planting. Quality and yield of fresh weight ware highest in optimum planting densities($10\times5cm$ : 200 plants/$m^2$).

Antioxidative Activity and Therapeutic Effect of the Hydrogel Preparations of Scutellariae Radix and Zingiberis Rhizoma on Dermatitis (황금 및 생강 복합 수용성겔 제제의 항산화 활성 및 피부염 치료효과)

  • This study was performed to investigate the effects of treatment for dermatitis using the herbal gel preparations. Scutellariae Radix(SR) and Zingiberis Rhizoma(ZR) were used for the purpose of herbal preparations. Baicalin, baicalein are the ingredients of SR, having biological effects like anti-inflammatory, anti-oxidative, anti-bacterial and antiallergic action. 6-Gingerol is one of the ingredients of ZR having biologicai effects like anti-inflammatory and analgesic action. The three types of hydrogels(SRE, SRH, SZH) were formulated with Carbopol 940, Labrasol, Triethanolamine etc. Baicalin was hydrolysed to baicalein by $\beta$-glucuronidase for the purpose to increase rate of skin permeation. Content of ingredients by HPLC determination, rate of skin permeation using Franz type diffusion cell, anti-oxidative activity for the free radical, hydroxyl radical, superoxide, anti-inflammatory by using carrageenan injection, efficacy on the dermatitis induced by 2,4-dinitro-chlorobenzene(DNCB) were experimented. Baicalein showed higher permeability than baicalin, so it is considered that baicalein was more suitable form than baicalin for transdermal absorption by its lipophilic property. In the anti-oxidative experiments, SZH gel was the most effective scavenging activity than the other gels in all experiments. In anti-inflammatory test, SRM and SZH gel more decreased edma rapidly than SRE gel. In case of using SZH gel, treatment period for the dermatitis was more declined than that of other gel groups. These results suggests that the SZH hydrogel could be suitable preparations for the treatment of dermatitis.

Anti-hyperlipidemic Effects of Scutellariae Radix, Aucklandiae Radix and Bupleuri Radix (SAB) extract in FL83B cells (지방간 유도 세포모델에서 황금(黃芩), 목향(木香), 시호(柴胡) 복합 추출물의 이상지질혈증에 대한 효과)

  • Objectives : This study was conducted to evaluate the anti-hyperlipidemia effect of Scutellariae Radix, Aucklandiae Radix and Bupleuri Radix(SAB). Methods : FL83B cells were mouse liver hepatocytes, and we used this cell line. FL83B cells were treated with 0.5 mM oleic acid(OA) for 24 h, SAB extract was treated. After OA treatment, intracellular triglyceride (TG) and free fatty acid contents were measured with AdiopoRed™ assay and Free Fatty Acid Quantitation assay kit, respectively. Further, we evaluated several lipogenesis and metabolic markers such as sterol regulatory element-binding transcription factor-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), 3-hydroxy3-methyl-glutaryl CoA reductase (HMGCR), hormone-sensitive lipase (HSL), carnitine palmitoyltransferase (CPT-1), peroxisome proliferator activated receptor alpha (PPARα), and cluster of differentiation (CD36) using RT-PCR and Western-blot analysis. Results : OA markedly increased intracellular TG and free fatty acid, which plays a key role in reducing hepatic lipid accumulation, in FL83B cells. These increases were alleviated by SAB extract. The mRNA and protein expression of Fatty acid(FA) oxidation factors (CPT-1, PPARα), lipolysis factor(HSL), FA transporter(CD36), cholesterol synthesis factors (HMGCoA) and Lipodenesis (SREBP-1c, FAS, and ACC-1) were significantly increased by treatment of SAB extract in the OA-induced fatty liver cell model. Conclusions : In summary, the treat of SAB extract showed a significant reduction of the influx of fatty acids into hepatocytes, promoted the oxidation of fatty acids, and regulated fat synthesis-related factors, thereby regulating the accumulation of TG and free fatty acids.

Effect of Atractylodes Rhizoma Alba, Houttuyniae Herba, Lonicerae Flos, Scutellariae Radix and Coptidis Rhizoma Extracts Combined with Metformin on the Antioxidant and Adipocyte Differentiate Inhibition (백출, 어성초, 금은화, 황금, 황련과 Metformin의 병용 투여 시 항산화 효과 및 전지방세포 분화 억제 효과)

  • Objectives: This study was to investigate the antioxiative capacity, antiobesity effects of Atractylodes Rhizoma Alba, Houttuyniae Herba, Lonicerae Flos, Scutellariae Radix, and Coptidis Rhizoma on Raw 264.7 and 3T3-L1 cell lines. Methods: Three different types of herb extracts (A. Rhizoma Alba, H. Herba, L. Flos, S. Radix, and C. Rhizoma; water 100%, ethanol 30%, ethanol 100%) were used in this study. Total polyphenol compound, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, reactive oxygen species (ROS) activity, NO production and cell proliferation were measured. Results: Total polyphenol compound measurement of L. Flos, A. Rhizogenes, and C. Rhizoma extracts were higher than A. Rhizoma Alba, H. Herba. DPPH radical scavenging activity, ROS activity and NO production of S. Radix, C. Rhizoma extracts were lower than L. Flos, A. Rhizoma, and H. Herba. Conclusions: Metformin and S. Radix, C. Rhizoma, A. Rhizoma Alba, and L. Flos extracts combination groups showed synergistic effect on adipocyte differentiation inhibition and antioxidative activity.

The Effect of Bacillus-Fermented Scutellariae Radix Acupuncture Solution on Chemokine and Growth Factor Production in Mouse Macrophage Stimulated by Lipopolysaccharide (바실러스균 발효황금약침액이 마우스 대식세포의 케모카인 및 생성인자 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of Bacillus-fermented Scutellariae Radix acupuncture solution (SB) on chemokine and growth factor production in RAW 264.7 mouse macrophages stimulated by lipopolysaccharide (LPS). Methods : Productions of chemokine and growth factor were measured by High-throughput Multiplex Bead based Assay with Bio-plex Suspension Array System based on xMAP$^{(R)}$ technology. Firstly, cell culture supernatant was obtained after treatment with LPS (1 ${\mu}g$/mL) and SB for 24 hours. Then, it was incubated with the antibody-conjugated beads for 30 minutes. Detection antibody was then added and incubated for 30 minutes. After incubated for 30 minutes, strepavidin-conjugated phycoerythrin (SAPE) was added. After another 30 minutes incubation, the level of SAPE fluorescence was analyzed in Bio-plex Suspension Array System. Results : The results of the experiment are as follows. 1. SB significantly inhibited the LPS-induced production of vascular endothelial growth factor (VEGF), interferon-inducible protein (IP)-10, and granulocyte-colony stimulating factor (G-CSF) at the concentration of 25, 50, 100, and 200 ${\mu}g$/mL in RAW 264.7 cells (P < 0.05). 2. SB significantly inhibited the LPS-induced production of Eotaxin at the concentration of 25, 100, and 200 ${\mu}g$/mL in RAW 264.7 cells (P < 0.05). 3. SB significantly inhibited the LPS-induced production of MIP-$1\alpha$ at the concentration of 25 and 100 ${\mu}g$/mL in RAW 264.7 cells (P < 0.05). Conclusions : These results suggest that SB has immuno-modulatory property related with its inhibition of VEGF, IP-10, G-CSF, and Eotaxin production in macrophages.
