Effect of Organic Compost on Growth and Yield in Scutellaria baicalensis (유기질 퇴비 시용이 황금의 생육 및 수량에 미치는 영향)

  • This experiment was carried out to investigate the effect of several organic compost on the growth and root yield of Scutellaria baicalensis G. with organic cultivation from 2008 to 2009. Four organic compost were used ; Rice bran compost (RC : 1.1-3.6-1.4), Wheat bran compost (WC: 1.1-3.4-1.4), Hangawi compost (HC : 3.7-4.0-1.0), Cheonnyeonjigi compost (CC : 12.4-3.4-5.4) and Fermented chicken droppings compost (FC : 2.9-6.1-3.2). Organic matter, available phosphate and $K_2O$ of soil increased in all plots with the addition of organic compost, specifically in HC, CC and FC application. Treatment of HC, CC and FC application increased emergence rate and improved the growth characters of shoot, root parts of plants compared to those of RC application. Subterranean part yields by different organic compost was high 149~156 kg per 10a as by growth increment of dried root yield. length of main root and diameter of main root. respectively, 17.4~18.4 cm, 12.65~12.94 mm from HC, CC and FC application. There were significantly positive correlation between the growth characters of shoot, root parts of plants, dry root yield in Scutellaria baicalensis G. grown under organic compost application.

Inhibitory effects of herbal medicine on coccidiosis (한약재의 첨가 급여에 따른 콕시듐 억제 효과)

  • This study examined the efficacy of natural ingredient formulated herbal medicine as feed additives against coccidiosis in broiler chicken. Medicinal herbs used in this study were Artemisia annua, Pulsatilla chinensis, Clematis chinensis and Scutellaria baicalensis. Two week old chicks were administered with 1% ingredient formulated herbal medicine. Observations of the efficacy were confirmed by examining the lesion score, histopathological appearance of cecum, shedding of fecal oocyst and production index in farm. S. baicalensis of the most effective substance were analyzed by UPLC. The results are as follows; among various types of Chinese medicine, the most efficacious ingredient was found to be pure bicalein. The major index substance is bicalein and $13{\mu}g/ml$ per gram was found. Feeding of chickens with S. baicalensis induced the coccidiostat effect and reduced the fecal shedding of E. tenella oocyst by 14%. Experimental results on a farm, feed conversion ratio and weight gain and production rate in the group with natural ingredient formulated additives than in control group increased. The rate of weight gain (%) in each experimental group showed 108% and 126% increase while the mortality rate was 1.4% lower than the control group. The investigation showed that the most efficacious ingredient was found to be S. baicalensis among herbal medicine products for coccidiostats. However, as it was found in the farm experiment that pure S. baicalensis alone is economically unviable. The additives with cocktailed of efficacious ingredient are suggested to be implemented for effective protection against coccidiosis.

Free Radical Scavenging Activity and Ascorbate Content in Various Plant Cell Lines (다양한 식물배양세포주에서 자유라디칼 포착활성과 Ascorbate 함량)

  • We investigated the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in methanol extracts of 64 cultured cell lines, which were derived from various plant species, and the ascorbate content in cell lines, which showed a high radical scavenging activity. Thirteen cell lines revealed the antioxidative activity ($IC_{50}$) by methanol extracts of less than 50 mg in cell fresh wt. Of them, six cell lines showed the same Rf value as ascorbate on the DPPH sprayed silica gel TLC. The ascorbate content in cell lines of Rosa multiflora, Scutellaria baicalensis, and Achyranthes japonica showed 48.5, 30.3, and $16.8\;\mu\textrm{g}$ per g cell fresh wt by HPLC analysis, respectively. In callus cultures of S. baicalensis, the concentration of ascorbate reached a maximun ($39{\pm}3.4\;\mu\textrm{g}/g$ cell fresh wt) on 30 days after subculture, which corresponded to the stationary growth phase, and subsequently decreased by successive culturing.

Induction of Cell Cycle Arrest at G2/M phase by Ethanol Extract of Scutellaria baicalensis in Human Renal Cell Carcinoma Caki-1 Cells (황금 에탄올 추출물에 의한 인간 신장암 세포주 Caki-1의 G2/M arrest 유발)

  • Objectives : In the present study, we investigated the effects of ethanol extract of Scutellaria baicalensis (EESB) on the progression of cell cycle in human renal cell carcinoma Caki-1 cells. Methods : The effects of EESB on cell growth and apoptosis induction were evaluated by trypan blue dye exclusion assay and flow cytometry, respectively. The mRNA and protein levels were determined by Western blot analysis and reverse transcription-polymerase chain reaction, respectively. Results : It was found that EESB treatment on Caki-1 cells resulted in a dose-dependent inhibition of cell growth and induced apoptotic cell death as detected by Annexin V-FITC staining. The flow cytometric analysis indicated that EESB resulted in G2/M arrest in cell cycle progression which was associated with the down-regulation of cyclin A expression. Our results also revealed that treatment with EESB increased the mRNA and proteins expression of tumor suppressor p53 and cyclin-dependent kinase (Cdk) inhibitor p21(WAF1/CIP1), without any noticeable changes in cyclin B1, Cdk2 and Cdc2. In addition, the incubation of cells with EESB resulted in a significant increase in the binding of p21 and Cdk2 and Cdc2. These findings suggest that EESB-induced G2/M arrest and apoptosis in Caki-1 cells is mediated through the p53-mediated upregulation of Cdk inhibitor p21. Conclusions : Taken together, these findings suggest that EESB may be a potential chemotherapeutic agent and further studies will be needed to identify the biological active compounds that confer the anti-cancer activity of S. baicalensis.

Glutathione Contents in Various Plant Cell Lines (다양한 식물배양세포주의 Glutathione 함량)

  • We investigated the levels of glutathione (GSH) and its oxidized form (GSSG) in 24 cell lines derived from various plant species to understand the antioxidative mechanism in plant cell cultures. The total glutathione content was 98$\pm$27 $\mu$g/g cell fresh wt, showing a slight difference in plant species. The average contort of GSH and GSSG was 72$\pm$20 and 26$\pm$10 $\mu$g/g cell fresh wt, respectively. The average GSH content in plant cell lines occupies approximately 73% in total glutathione. During the suspension cultures of Scutellaria baicalensis, one of the plant species we tested, the GSH content decreased in proportion to the cell growth during the exponential growth stage, showing the low level at the stationary growth stage (84 $\mu$g/g cell fresh wt), whereas the GSSG content increased to the stationary growth stage (31 $\mu$g/g cell fresh wt). The results suggested that the ratio of GSH and GSSG should be involved in the cell growth and antioxidative mechanism in cultured cells.

Effect of Scutellaria baicalensis Georgi Extract on Oxidant-Induced Inhibition of Organic Cation in Rabbit Renal Cortical Slices (황금약침액(黃芩藥鍼液)이 가토(家兎)의 신피질절편(腎皮質切片)에서 Oxidant로 유발된 유기양이온의 이동장애에 미치는 영향(影響))

  • Objective : This study was undertaken to determine whether Scutellaria baicalensis Georgi extract (SbG) exerts the protective effect against oxidant-induced alterations in organic cation transport in the renal proximal tubule. Methods : Organic cation transport was estimated by examining alterations in tetraethylammonium (TEA) uptake in rabbit renal cortical slices. The slices were treated with 0.2 mM tBHP for 60 min at $37^{\circ}C$. tBl-IP caused an inhibition in TEA uptake by renal cortical slices. Such an effect was accompanied by depressed Na+-K+-ATPase activity and ATP depletion. Result : SbG prevented tBHP-induced inhibition of TEA uptake in a dose-dependent manner at the concentration ranges of 0.05-0.1%. SbG also prevented H2O2-induced reduction in TEA uptake. tBHP-induced inhibition of Na+-K+-ATPase activity and ATP depletion were significantly prevented by 0.05% SbG. Oxidants increased LDH release, which was blocked by SbG. Oxidants caused a significant increase in lipid peroxidation and its effect was prevented by SbG. Conclusion : These results suggest that SbG prevents oxidant-induced alterations in organic cation transport in rabbit renal cortical slices. Such protective effects of SbG may be attributed to inhibition of peroxidation of membrane lipid.

  • Present experiments were carried out to examine the effect of plant growthregulators for callus induction and plantlet regeneration through leaf tissue culture of Scutellaria baicalensis GEORGI. The results indicated that Callus was induced well on MS medium supplemented with 0.5mg/L NAA or 0.5mg/L NAA Plus 0.5mg/L zeatin. MS medium supplemented with 1.0mg/L BAP plus 0. 5mg /L NAA or 1. 0mg /L zeatin Plus 0.5mg /L NAA and 1.0mg /L NAA were the most effective for plant regeneration. Thin layer chromatogram of baicalin component (Rf 0.39) was observed from callus cultured on MS medium containing 0.5mg /L NAA plus 0.5mg /L zeatin.

  • Scutellaria baicalensis GEORGI(SB) is used in oriental medicine for the treatment of incipient strokes. Although it has been reported that SB is neuroprotective in a hypoxia model, its mechanism is poorly understood. Here, we investigated the effect of SB on the modulation of heme oxygenase-1(HO-1), which has important biological roles in regulating mitochondrial heme protein turnover and in protecting against conditions such as hypoxia, neurodegenerative diseases, or sepsis. Rat cerebrocortical day In vitro(DIV)12 cells were grown in neurobasal medium. On DIV12 cells were treated with SB($20{\mu}g/ml$) and given a hypoxic shock ($2%\;O_2/5%\;CO_2,\;3\;hr$) on DIV14. In situ hybridization results revealed that SB upregulated HO-1 mRNA in neuronal dendrites in both normoxia and hypoxia(38.5% and 59.2%, respectively). At the protein level, SB upregulated HO-1 in the neuronal soma in both normoxia and hypoxia(22.4% and 15.7%, respectively). Interestingly, most significant increase was associated with astrocytes, which increased HO-1 protein by 77.5% compared to SB-untreated culture. These results indicate that SB upregulates both neuronal and glial HO-1 expression, which contributes to the neuroprotection efficacy in hypoxia).

Root Extract of Scutellaria Baicalensis Increases Gefitinib Sensitivity in H1975 Human Non-small Cell Lung Cancer Cells (H1975 세포에서 황금추출물에 의한 gefitinib 저항성 억제 효과)

  • Gefitinib, a first generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR TKI), provides obvious clinical benefit in patients with EGFR-mutant non-small cell lung cancer (NSCLC). However, patients ultimately develop gefitinib resistance which mainly caused by EGFR T790M secondary mutation. In the current study, we investigated whether the root extract of Scutellaria baicalensis (SB) overcomes gefitinib resistance. Gefitinib-resistant H1975 human NSCLC cells (EGFR L858R/T790M double mutant) were treated with gefitinib and/or ethanol extract of SB (ESB) to evaluate the effect of ESB on the gefitinib sensitivity. The cell viability was measured by MTT assay and trypan blue exclusion assay. The colony-forming ability was evaluated by anchorage-dependent colony formation assay. Combined treatment with gefitinib and ESB markedly decreased the cell viability and colony formation than single treatment with gefitinib or ESB in H1975 cells. In addition, cells treated with both gefitinib and ESB exhibited a significant increase of sub-G1 DNA content which indicates apoptotic cells compared with those treated with gefitinib or ESB alone. As a molecular mechanism, combined treatment with gefitinib and ESB strongly downregulated the phosphorylation of ERK and JNK than single treatment with gefitinib or ESB. Taken together, our results demonstrate that ESB sensitizes H1975 cells to gefitinib treatment. We cautiously propose that ESB can be used in combination with gefitinib for the advanced NSCLC patients with acquired resistance to EGFR TKIs.

Effects of Mixed Scutellaria baicalensis Extracts as Natural Preservative on Efficacy and Storage of Lactic Acid-Fermented Garlic Extract (천연보존료 복합 황금추출물이 유산균발효 마늘추출물의 저장성 및 기능성에 미치는 영향)

  • The natural preservative "complex Scutellaria baicalensis extract (BHC)" contains Scutellaria baicalensis, Glycyrrhiza uralensis (liquorice), Zizyphus jujube (jujube), and Astragalus propinquus (milk vetch root). BHC has been used as a natural preservative for more than 10 years to increase storage duration and quality of food with strong antibacterial activity. BHC has been added into functional foods as a subsidiary ingredient. However, no studies have been performed to test whether or not BHC affects the activity of main functional ingredients. In this study, we tested whether or not BHC has any effect on the hepatoprotective activity of lactic acid-fermented garlic extract (LAFGE) when formulated in a clinical test supplement. $H_2O_2-induced$ oxidative damage in HepG2 cells was not attenuated by BHC, indicating that BHC had no influence on the protective effect of LAFGE against oxidative damage. Furthermore, BHC had no effect on the hepatoprotective effect of LAFGE against acetaminophen-induced acute liver injury in rats, as indicated by no changes in alanine transaminase and aspartate transaminase levels. In conclusion, BHC, formulated in the clinical test supplement with LAFGE, had no effect on hepatoprotective activity, indicating BHC could be considered as a suitable natural preservative for liquefied functional food materials.
