Antioxidant activity analysis of Catechin compounds in Korean green tea using HPLC On-line $ABTS^{+}$ Antioxidant screening system (HPLC On-line $ABTS^{+}$ Antioxidant screening 시스템을 이용한 한국산 녹차로부터 Catechin compounds의 황산화 활성분석)

  • In this work, we describes analysis of the antioxidant potential of Korean green tea phenolics using an high-performance liquid chromatography (HPLC) on-line $ABTS^{+}$ antioxidant screening method. In conjunction with the analysis of their 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+) radical scavenging ability, the extraction of catechine compounds from Korean green tea were performed by various temperature and time. The optimum operating conditions were experimentally determined to analyze the catechine compounds in the pretreatment extracts. From the results, the extraction temperature $60^{\circ}C$, time 3 min was selected as an optimal antioxidant activity condition. The analysis by $C_{18}$ column was performed, the flow rate of mobile phase and UV wavelength was fixed at 1.0 ml/min and 254 nm, respectively. the mobile phase was composed from acetonitrile and water, and the gradient elution mode were applied.

Application of Health Behavior Theories to Breast Cancer Screening among Asian Women

  • Background: Although breast cancer is a major public health worry among Asian women, adherence to screening for the disease remains an obstacle to its prevention. A variety of psycho-social and cultural factors predispose women to delay or avoidance of screening for breast cancer symptoms at the early stages when cure is most likely to be successful. Yet few interventions implemented to date to address this condition in this region have drawn on health behavior theory. Materials and Methods: This paper reviews the existing literature on several cognitive theories and models associated with breast cancer screening, with an emphasis on the work that has been done in relation to Asian women. To conduct this review, a number of electronic databases were searched with context-appropriate inclusion criteria. Results: Little empirical work was found that specifically addressed the applicability of health theories in promoting adherence to the current breast cancer prevention programs Among Asian women. However, a few studies were found that addressed individual cognitive factors that are likely to encourage women's motivation to protect themselves against breast cancer in this region of the world. The findings suggest that multi-level, socio-cultural interventions that focus on cognitive factors have much promise with this issue. Conclusions: Interventions are needed that effectively and efficiently target the personal motivation of at-risk Asian women to seek out and engage in breast cancer prevention. Concerning implications, personal motivation to seek out and engage in individual preventive actions for breast cancer prevention among Asian women is a timely, high priority target with practical implications for community development and health promotion. Further studies using qualitative, anthropologic approaches shaped for implementation in multi-ethnic Asian settings are needed to inform and guide these interventions.

Effective Screening Condition for Low-Temperature Germinability of Rice (벼 저온발아성의 효율적 검정 조건)

  • This study was carried out to determine the desirable screeing condition for low-temperature germinability of rice seeds. The germination test was performed with fifty rice varieties from three countries, mainly including the recent Korean rice cultivars. The analyzed result of germination test under three temperature conditions(13$^{\circ}C$, 16$^{\circ}C$, $25^{\circ}C$) pointed out that 11 days after seeding at 13$^{\circ}C$ was the most efficient testing condition for low-temperature germinability in terms of mean and variation in germination rate. Highly significant correlations were observed between germination rates of 11 days after seeding at 13$^{\circ}C$ and germination coefficients, average days of germination at 13 for 20 days. Low-temperature germinability were highly positively associated with days to heading or amylose content, but negatively correlated with grain width.

  • Wolff Parkinson White (WPW) syndrome is a condition in which there is an aberrant conduction pathway between the atria and ventricles, resulting in tachycardia. A 42-year-old patient, who was treated for WPW syndrome previously, presented with chronic somatic pain. With her cardiac condition in mind, she was thoroughly worked up for a recurrence of disease. As part of routine screening of all patients at our pain clinic, she was found to have severe depression as per the Patient Health Questionnaire-9 (PHQ-9) criteria. After ruling out sinister causes, she was treated for depression using oral Duloxetine and counselling. This led to resolution of symptoms, and improved her mood and functional capability. This case highlights the use of psychological screening tools and diligent examination in scenarios as confusing as the one presented here. Addressing the psychological aspects of pain and adopting a holistic approach are as important as treatment of the primary pathology.

  • One of the important things in low level radioactivity measurement is determination of the optimized counting time. Counting strategy has to be established, in order to count the radioactivity of the sample by condition of optimized measurement. There were three kinds of counting strategies in this report ; about fixed time, about fixed count, to compared sample, background, and reference level. The best of them was satisfied rendition to give about condition of instrument and process, as an example, efficiency of detector, counter capacity, maximum and average background count rate of counter, reference level and limit of derision and detection, etc. Therefore, we can decide the optimized counting time in the screening and monitoring. And we can save the time for courting the sample of course the data of count will be counted by optimized accuracy finally, in rountine measurement of radioactivity these strategies will be used available.

A Novel Bioassay System for Screening of Compounds Affecting Anthocyanin Biosynthesis Pathway in White Corn Leaf Segment (백화옥수수 잎절편을 이용한 안토시아닌 생합성 조절제 탐색용 신규 검정법 확립)

  • This study was carried out to establish a novel bioassay system for screening of compounds affecting aromatic amino acid or anthocyanin biosynthesis through investigating a degree of sucrose-induced anthocyanin formation such as size of plant material, buffer conditions, light intensity and irradiated duration, incubation temp., etc were determined and standard procedure (suitable experimental condition) was set up as follows. The second leaf blade of white corn seeding induced by fluridone treatment were segmented into a size of 5${\times}$5 min. The segments were floated on the solution of 1% sucrose in 1.0mM MES buffer (pH6.0∼6.5) and incubated at 26$^{\circ}C$ for 2days under the continuous light condition(70∼100$\mu$mol m$^{-2}$ s$^{-1}$ ). Anthocyanin in the purpled tissues was extracted with methanol containing 1% HCl and the optical density of the clear supematants was determined at 528mm. Influences of some chemicals were tested using this system. Glyphosate, 5-enolpyruvylshikimate 3-phosphate synthase inhibitor, showed most sensitive response with I$_{50}$ value at 3.3$\mu$M. Dicyclohexylcarbodiimide(DCCD) and parachloromercuribenzenesulfonic acid(PCMBS) had a relatively strong ingibition with I50 value at 7.1$\mu$M and 10.2$\mu$M, respectively. These results show that sucrose-induced anthocyanin formation in white com leaf segment provide a very simple and rapid system for searching new compounds affecting aromatic amino acid or anteocyanin biosynthesis by screening at less than 10$\mu$M.

Screening methods for drought and salinity tolerance with transgenic rice seedlings

  • Abiotic stress is one of the major serious limiting factors in rice (Oryza sativa) and caused rice production losses. It is important to precisely screen valuable genetic resources for improving stress tolerance and understanding tolerance mechanism to abiotic stresses. Because there are differences of experiment designs for screening of tolerant plant in several studies related to abiotic stress, this study has performed to provide the rapid and efficiency screening method for selection of tolerance rice to drought and salinity stresses. Two week-old rice seedlings that reached about three leaf stage were treated with drought and salinity stresses and examined tolerant levels with tolerant and susceptible control varieties, and transgenic plants. To determine the optimum concentration for the selection of drought and salinity condition, tolerant, susceptible and wild-type plants were grown under three soil moisture contents (5, 10 and 20% water contents) and three NaCl concentrations (100, 200 and 250 mM) for 10 days at seedling stage. 200 mM NaCl concentration and 5% moisture content soil were determined as the optimum conditions, respectively. The described methodologies in this study are simple and efficiency and might help the selection of drought and salinity tolerance plants at the 3,4-leaf-seedling stage.

Development and physiological assessments of multimedia avian esophageal catheter system

  • We developed multimedia esophageal catheters for use with birds to measure and record ECG and angular velocity while anesthesized, at rest, and in flight. These catheters enable estimates of blood pressure based on readings given by an angular velocity sensor and by RR intervals of ECG affected by EMG. In our experiments, the catheters had the following characteristics: 1. Esophageal catheters offer a topological advantage with 8-dB SNR improvement due to elimination of electromyography (EMG). 2. We observed a very strong correlation between blood pressure and the angular velocity of esophageal catheter axial rotation. 3. The impulse conduction pathway (Purkinje fibers) of the cardiac ventricle has a direction opposite to that of the mammalian pathway. 4. Sympathetic nerves predominate in flight, and RR interval variations are strongly suppressed. The electrophysiological data obtained by this study provided especially the state of the avian autonomic nervous system activity, so we can suspect individual's health condition. If the change of the RR interval was small, we can perform an isolation or screening from the group that prevent the pandemics of avian influenza. This catheter shall be useful to analysis an avian autonomic system, to perform a screening, and to make a positive policy against the massive infected avian influenza.
