GC/MS에 의한 tropane alkaloids의 분석 및 흰쥐의 생체내 분포 (Tissue Distribution of Tropane Alkaloids in Rats and its Determination by GC/MS After the Oral Administration of Scopolia Rhizome)

  • Scopolia rhizome is mistaken as an atractylodes rhizome because of their similarities in shape. That is why atractylodes rhizome imported from China sometimes contain scopolia rhizome, which is very toxic. 8 persons were intoxicated atractylodes after taking imported atractylodes rhizome which is tainted. In kampo medicine prepared with such imported atractylodes rhizome, the level of tropane alkaloids ranged from 1.12∼4.34 mg/dose. In this study, we tried to investigate the tissue distribution of scopolia rhizome in rats. The extracts of scopolia was administered orally to rats (a single dose of 10mg/kg, 20mg/kg and 7 days repeated dose of 10mg/kg). Their blood was collected at 0.5, 1, 2, 4, 6 hrs, and liver, kidney, lung and spleen were collected after 6 hrs. The tissue homogenate was applied to solid phase extraction column for the determination of tropane alkaloids. After the oral administration of 20mg/kg scopolia extracts, l-hyoscyamine was detected in rat blood to 2 hrs after dosing. The concentration of tropane alkaloids was the highest in liver followed by lung, kidney and spleen. However, lung, kidney and spleen were similar in amount.

광의의 미치광이풀속(Scopolia Jacq. s.l., 가지과-Hyoscymeae족)의 잎표피 미세구조와 이의 계통분류학적 중요성 (Leaf epidermal microstructure of the genus Scopolia Jacq. s.l. (Solanaceae-Hyoscymeae) and its systematic significance)

  • 세계산 Scopolia Jacq. s.l.에 포함되는 세 개의 속인 Scopolia s.s., Anisodus, Atropanthe와 근연 군외군으로 선택된 Przewalskia 식물의 잎표피 미세구조를 조사하기 위하여 4속 10종(37개체)의 잎을 광학현미경 및 주사전자현미경을 이용하여 관찰하였다. 연구 분류군의 기공은 향축면과 배축면 모두에 존재하는 양면기공엽 (amphistomatic type)으로 밝혀졌다. 공변세포의 크기는 $18-64{\times}11-48{\mu}m$로 속과 종에 따라 다소 차이를 보이는데, 가장 작은 공변세포는 군외군으로 포함시킨 단형속인 Przewalskia tangutica($24-27{\times}16-17{\mu}m$)이었고, 가장 큰 공변세포는 Anisodus carniolicoides ($62-64{\times}43-48{\mu}m$)에서 나타났다. 기공복합체의 형태는 대부분 불규칙형 (anomocytic:Scopolia s.s., Anisodus taxa:A. luridus, A. carniolicoides, A. acutangulus)과 불균등형 (anisocytic:Anisodus tanguticus, Przewalskia, Atropanthe)이 나타난다. 공변세포의 모양은 대부분 초생달 모양의 타원형이나, Przewalskia tangutica에서는 거의 원형으로 나타난다. 부세포의 수충벽(anticlinal wall)은 대부분 파상형 (undulate)이며, Przewalskia tangutica에서는 다소 아치형을 이루었다. 표피세포의 형태는 거의 불규칙한 다각형 (polygonal)으로 수충벽은 다소 또는 심하게 굴곡을 이루는 undulate/sinuate형이었다. 표피세포의 표면은 왁스층이 대부분의 분류군에서는 잘 발달되지 않았고, 일부의 분류군(e.g., Anisodus tanguticus)에서만 기공 주변으로 발달한 양상을 보였다. Scopolia s.s.와 Przewalskia 분류군에서는 독특하게 다세포로 구성된 'elongate-headed' 선모가 존재하였고, Atropanthe 및 Anisodus의 대부분의 분류군은 모용이 존재하지 않았고, 단지 1종(A. luridus)에서는 단순(simple) 또는 가지를 친(branched:dendritic-type) 비선모가 존재하는 것으로 나타났다. 끝으로 조사된 Scopolia s.l.(Przewalskia 포함)의 분류군내의 속간 그리고 종간의 동정과 식별을 위한 잎의 미세형태학적 형질(기공복합체, 모용, 등)의 계통분류학적 또는 생태학적 가치를 검토하였다.

Distribution and morphological characteristics of Scopolia japonica Maxim. in Korea

  • The morphological characteristics of the Korean Scopolia japonica Maxim., commonly known as 'Mi-chi-gwang-i-pul' in Korean, were re-evaluated in comparison with results from previous research, while the distribution map was formulated according to existing flora and voucher specimens. The result of the re-evaluation has shown that there are some distinctive discrepancies compared to the previous reports of some characteristics such as the color of the basal stem, numbers of lateral veins, apex shape of calyx and corolla including the color of corolla. The Korean Scopolia japonica Maxim is found throughout Korea except Jeju-do province. The intra-specific species such as S. parviflora (Dunn.) Nakai, S. lutescens Y.N. Lee and S. kwangdokensis Y.N. Lee were treated as synonyms of S. japonica Maxim. in this research.

The complete plastid genome of Scopolia parviflora (Dunn.) Nakai (Solanaceae)

  • Scopolia parviflora of the family Solanaceae is an endemic species of Korea and a traditional Korean medicinal plant. The plastid genome was sequenced by next-generation sequencing (NGS) method. The characterized cp genome is 156,193 bp in size; the large single-copy (LSC) region is 86,364 bp, the inverted repeat (IR) is 25,905 bp, and the small single copy (SSC) region is 18,019 bp. The overall GC content of the plastid genome amounts to 37.61%. The cp genome contains 113 genes and 21 introns, including 80 proteincoding genes, four RNA genes, 30 tRNA genes, 20 group II introns, and one group I intron. A phylogenetic analysis showed that Scopolia parviflora was closely related to Hyoscyamus niger.

  • In emergency department, the clinicians occasionally meet some patients with toxicity caused by ingestion of unknown wild edible greens or herbal agents. Due to there was few quantitative or qualitative studies about the mechanism or clinical features of these materials, it' s hard to approach to the patients and start the initial management in emergency department. Authors experienced the case of poisoning by Scopolia Japonica ingestion. We report this case with review of Scopolia Japonica.

  • The prevalence rate of plant poisoning is growing up gradually. However, it could be difficult to get proper treatment without detail medical history taking due to the diversity of poisonous plants and their various symptoms. Scopolia Japonica, which grows naturally in Korea, is reported to cause severe toxicity which could be lethal. A few therapeutic measures have been reported in Traditional Korean Medicine (TKM), but it has not been reported yet that those measures have been successfully proven in the clinical practice in modern times. This is a case report of toxicity caused by scopolia japonica ingestion. A 65years-old male patient with cognitive impairment, behavioral disorder, gait difficulty, dysarthria, dysphagia and abnormal pupil reflex recovered from those symptoms during taking Gamigamdu-tang and getting acupuncture. This study shows the clinical aspects of Scopolia Japonica toxicity and the possibility of TKM treatments as one of plant poisoning treatments.

미치광이풀(Scopolia parviflora)의 모상근 배양에 의한 Tropane Alkaloid 생산 (Production of Tropane Alkaloids by Hairy Root Cultures of Scopolia parviflora)

  • Transformed hairy roots of Scopolia parviflora, producing tropane alkaloids and native to Korea, were obtained following infection of rhizome segments with Agrobacterium rhizogenes A4. Each root tip induced from inoculum sites excised and cultured in MS agar or liquid medium. About seventy of hairy root clones were established. Among them, several fast growing hairy root clones were examined for alkaliod content. Two dimensional TLC analysis showed that the tropane alkaloid pattern of hairy root was more complicated than that in the rhizome of mother plant. On the other hand, some hairy root clones did not produce scopolamine and hyoscyamine. In HPLC analysis, some hairy root clones yield higher levels of scopolamine and hyoscyamine than those of mother plant rhizome which used for infection. Scopolamine and hyoscyamine were identified by comparison of their retention times and of their spectra data with those of authentic compounds.

ITS 염기서열에 의한 미치광이풀속의 계통 (Phylogeny of Scopolia Jacq. s. str. based on ITS sequences)

  • Scopolia s. str.(미치광이풀속) 및 그 근연속인 Anisodus, Atropanthe 등을 대표하는 8 분류군으로부터 얻은 14개체를 대상으로 핵 ribosome DNA의 ITS 염기서열 결정을 실시하였다. 그 결과 한반도 고유종인 S. parviflora(미치광이풀)은 ITS 전체 길이와 염기서열 변이에 있어서 근연종인 S. japonica와 매우 큰 차이가 있음이 밝혀졌다. 이와 같은 결과는 두 종을 동일시했던 대부분의 분류학적 견해와 배치되는 것이다. 한편, S. parviflora는 유럽에 격리분포하는 종인 S. carniolica와 더 가까운 계통유연관계를 나타냈다. S. japonica가 S. parviflora 및 S. carniolica와 높은 유전적 차이를 나타내는 것과는 대조적으로 매우 높은 형태적 유사성을 보이는 것은 이들이 오랜 격리기간에도 불구하고 유사한 생육환경에 처함으로 인해 형태진화가 지연되었거나 비슷한 형질을 갖는 방향으로 진화가 일어났을 것이라는 형태정체(morphological stasis) 개념으로 이해되었다. 다른 한반도 고유종인 S. lutescens(노랑미치광이풀)은 S. parviflora와 ITS 염기서열이 거의 동일하였고 계통수 상에서도 두 종에 속하는 개체들이 전혀 구분되지 않는 것으로 밝혀졌다. 자연개체군 내에서 이들 두 종을 구분하는 주요 식별형질들의 유용성도 결여되어 S. lutescens는 S. parviflora의 품종 혹은 단순한 개체 변이로 이해되었다. 한편, Scopolia속으로 최초 기재되었다가 화분의 형질에 의해 Anisodus속으로 이전되었던 A. carniolicoides는 A. luridus 및 A. tanguticus와 단계통군을 형성하였으며 Anisodus는 단형속인 Atropanthe와 자매군 관계를 형성하였다.

Production of Monoclonal Antibody about Specific Key Enzyme of Hyoscyamine $6{\beta}-Hydroxylase$ (H6H) in Scopolia parviflora

  • Total RNAs were isolated from cultured roots of Scopolia parviflora, $poly(A)^+$ RNA was obtained through the mRNA purification, cDNA library of Hnh6h was constructed. Recombinant baculoviruses in Spodoptera frugiperda (Sf) cells were constructed by use of the transfer vector pBacPAK, which has the AcNPV sequence under the polyhedrin promoter. The expression vector carrying Hnh6h gene was transferred to S. parviflora and obtained transgenic hairy root lines. Our results confirmed the over expression of the H6H protein was used by anti-pBacPAK about cDNAs of S. parviflora. This study will served for production of tropane alkaloids by metabolic engineering.
