토양의 형태 및 물리적 특성을 고려한 복분자 재배적지 기준설정 (Establishment of Soil Suitability for Korean Black Raspberry by Soil Morphological and Physical Properties)

  • The objective of this study was to establish the decision criteria of soil suitability for Korean Black raspberry using soil morphological and physical properties. The investigation was carried out in Gochang, Sunchang, Jeongeup, Pohang, and Hoengseong districts in Korea. The obtained results showed that factors related to the decision criteria of the soil suitability for Korean Black raspberry cultivation were soil texture, soil drainage class, land slope, and available soil depth. The criteria of the best suitability soil for Korean Black raspberry was valley/fan or hill geomorphology, well or moderately drainage class, B-slope(2-7%), coarse loamy soil texture family, less than <15% gravel contents, and available soil depth deeper than 100cm. The area criteria of soil suitability for Korean Black raspberry of Gochang was more than 50% including best and suitable areas.

Characterization and cDNA Cloning of a Defensin-Like Peptide, Harmoniasin, from Harmonia axyridis

  • We compared the mRNA expression profile of the Harmonia axyridis larvae that were either untreated or treated with LPS. The extracted mRNAs were subjected to ACP RT-PCR analysis using a combination of arbitrary primers and oligo (dT) primer. Among the 47 DEGs differentially expressed, we identified a cDNA showing homology with defensin-like antibacterial peptide. The cDNA showed a putative 32-residue signal sequence and a 50-residue mature peptide named harmoniasin. We also investigated the antibacterial activity of the harmoniasin analog, which exhibited potent antibacterial activities against Gramnegative and -positive bacteria strains and it also evidenced no hemolytic activity.

Application and Evaluation of LAMP-PCR for the Diagnosis of Silkworm Pebrine Disease

  • For stable silkworm breeding and high-quality sericulture product production, the detection of Pebrine disease in silkworm eggs is critical. Current diagnostic methods can be timeconsuming and complex. This study aimed to develop a simplified and rapid diagnostic method using loop-mediated isothermal amplification (LAMP) technology to detect pebrine infection in silkworm mother moths. Eight primer candidates targeting the ribosomal gene region of microsporidia were designed and evaluated for specificity and detection sensitivity. A simplified nucleic acid extraction method was established, and isothermal amplification was performed using the selected primers. Of these, primers ID30 and ID45 showed no polymerization, while ID5, ID18, and ID76 exhibited nonspecific reactions, making them unsuitable. Primers ID1, ID6, ID45, and ID82 successfully amplified DNA only in the presence of pebrine, with ID82 demonstrating the best reproducibility and sensitivity, detecting as low as 2.5 pg/ul of DNA through electrophoresis and 5 pg/ul via a colorimetric change with phenol red. The entire process, from nucleic acid extraction to detection, was completed within 60 min. The use of the ID82 primer set in LAMP technology offers a promising and efficient approach for the rapid diagnosis of pebrine disease, potentially enhancing quality control in sericulture.

  • Geographic information system (GIS) is being increasingly used for decision making, planning and agricultural environment management because of its analytical capacity. GIS and remote sensing have been combined with environmental models for many agricultural applications on monitoring of soils, agricultural water quality, microbial activity, vegetation and aquatic insect distribution. This paper introduce principles, vegetation indices, spatial data structure, spatial analysis of GIS and remote sensing in agricultural applications including terrain analysis, soil erosion, and runoff potential. National Academy of Agricultural Science (NAAS), Rural Development Administration (RDA) has a spatial database of agricultural soils, surface and underground water, weeds, aquatic insect, and climate data, and established a web-GIS system providing spatial and temporal variability of agricultural environment information since 2007. GIS-based interactive mapping system would encourage researchers and students to widely utilize spatial information on their studies with regard to agricultural and environmental problem solving combined with other national GIS database. GIS and remote sensing will play an important role to support and make decisions from a national level of conservation and protection to a farm level of management practice in the near future.

  • Through time-series CCD photometry using a 130mm refractor of Busan Science Academy, we found a new $\delta$ Scuti type variable star, esc 03665-00576. We performed following observations using a 1.8m telescope of Bohyunsan Optical Astronomy Observatory. Three frequencies have been derived by multiple-frequency analysis, which are 10.968, 7.156 and 15.182cycle $day^{-1}$, respectively. V-amplitude of esc 03665-00576 is ${\Delta}V$= 0.04.
