Breeding of new variety Pleurotus pulmonarius using protoplast fusion technique (원형질체융합 기법을 이용한 산느타리 계통육성)

  • In this study, monokaryons of "Heukari" (Pleurotus ostreatus) and "Hosan" (Pleurotus pulmonarius) were separated to remove the cell wall, and a cross-species protoplast fusion was developed through chemical treatment with polyethylene glycol. The protoplast-fused PF160306 and PF160313 strains have a culture period of 10 and 2 days shorter than that of the "Heuktari" and "Hosan" cultivars, respectively. Furthermore, the growth of the strains was faster than that of the existing cultivars. The yield was 135.9 g per bottle, which was approximately 8% higher than that of the commercially available "Hosan" cultivar; however, it was not statistically significant. A growth survey was conducted after treatment at five temperatures (15, 18, 21, 23, and 25℃). The growth of the strains accelerated with the increase in temperature. However, at 21℃, the yellow color of pileus was the brightest. Band pattern, assessed using URP Primer 7, was similar to that of the "Hosan" cultivar. The DPPH radical scavenging capacity and polyphenol content were 62.5% and 43.5 mg/mL, respectively, for "Sunjung" and 65.7% and 49.9 mg/mL, respectively, for PF160313. Furthermore, the antihypertensive activities of the "Sunjung" cultivar and PF160313 were similarly high at 74% and 75%, respectively. In conclusion, cross-species hybridization via the protoplast fusion technique can be used for obtaining primary data for mushroom breeding to develop new varieties. In addition, the protoplast fusion technique might aid in expanding the market for yellow mushrooms.

Effects of Light Intensity and Electrical Conductivity Level on Photosynthesis, Growth and Functional Material Contents of Lactuca indica L. 'Sunhyang' in Hydroponics (수경재배에서 광도와 양액 농도가 베이비 산채 왕고들빼기 '선향' 광합성과 생육 및 기능성 물질 함량에 미치는 영향)

  • This study was conducted to examine the changes of photosynthesis, growth, chlorophyll contents and functional material contents in light intensity and EC concentration of wild baby leaf vegetable, Indian lettuce (Lactuca indica L. cv. 'Sunhyang') in DFT hydroponics. The cultivation environment was 25±1℃ of temperature and 60±5% of relative humidity in growth system. At 14 days after sowing, combination effect of light intensity (Photosynthetic Photon Flux Density (PPFD 100, 250, 500 µmol·m-2·s-1) and EC level (EC 0.8, 1.4, 2.0 dS·m-1) of nutrient solution was determined at the baby leaf stage. The photosynthesis rate, stomatal conductance, transpiration rate and water use efficiency of Indian lettuce increased as the light intensity increased. The photosynthesis rate and water use efficiency were highest in PPFD 500-EC 1.4 and PPFD 500-EC 2.0 treatment. The chlorophyll content decreased as the light intensity increased, but chlorophyll a/b ratio increased. Leaf water content and specific leaf area decreased as light intensity increased and a negative correlation (p < 0.001) was recognized. Plant height was the longest in PPFD 100-EC 0.8 and leaf number, fresh weight and dry weight were the highest in PPFD 500-EC 2.0. Anthocyanin and total phenolic compounds were the highest in PPFD 500-EC 1.4 and 2.0 treatment, and antioxidant scavenging ability (DPPH) was high in PPFD 250 and 500 treatments. Considering the growth and functional material contents, the proper light intensity and EC level for hydroponic cultivation of Indian lettuce is PPFD 500-EC 2.0, and PPFD 100 and 250, which are low light conditions, EC 0.8 is suitable for growth.

Impact of East Asian Summer Atmospheric Warming on PM2.5 Aerosols (동아시아 지역의 여름철 온난화가 PM2.5 에어로졸에 미치는 영향)

  • This study analyzed the effect of warming on PM2.5 aerosol production in mid-latitude East Asia during June 2020 using PM2.5 aerosol anomalies, which were identified by incorporating meteorological and climate data into the Weather Research Forecasting model coupled with Chemistry (WRF-Chem) model. The decadal temperature change trend over a 30-year period (1991-2020) in East Asia showed that recent warming has been greater in summer than in winter. Summer warming in East Asia generated low and high pressure in the lower and upper troposphere, respectively, over China. The boundary between the lower tropospheric low and upper tropospheric high pressure sloped along the terrain from the Tibetan Plateau to Korea. The eastern China, Yellow Sea, and Korean regions experienced a convergence of warm and humid southwesterly airflows originating from the East China Sea with the development of a northwesterly Pacific high pressure. In June 2020, the highest temperatures were observed since 1973 in Korea. Meanwhile, enhanced warming in East Asia increased the production of PM2.5 aerosols that travelled long distances from eastern China to Korea. PM2.5 anomalies, which were derived solely by inputting meteorological and climatic data (1991-2020) into the WRF-Chem model and excluding emission variations, showed a positive distribution extending from eastern China to South Korea across the Yellow Sea as well as over the Pacific Northwest. Thus, the contribution of warming to PM2.5 aerosols in East Asia during June 2020 was more than 50%. In particular, PM2.5 aerosols were transported from eastern China to Korea through the Yellow Sea, where the warm and humid southwesterly airflows implied wet scavenging of sulfate but promoted nitrate production.
