Elsinoe araliae에 의한 두릅나무 더뎅이병 (Scab Disease of Aralia elata Caused by Elsinoe araliae)

  • Scab disease of Aralia elata (Miq.) Seemann occurred severely in Kangwon province, Korea. First, brown spots were formed on leaves and enlarged along vein. The infected leaves including petiole and midrib were twisted forming scab and died eventually. Also the disease infected twigs and trunks of the tree forming scab. Conidia, ascospores and asci developed in locule were observed in the infected tissue. The causal organism of the scab disease of Araliae elata was isolated and identified as Elsinoe araliae Yamamoto (imperfect stage: Sphaceloma araliae Jenkins) based on the morphological and cultural characteristics, and pathogenic test.

Occurrence of Potato Powdery Scab Caused by Spongospora subterranea in Korea

  • Potato tubers with scab lesions were found in several regions of Gangwon province in Korea during disease surveys in 2000 and 2001. The lesions appeared as shallow scabs with torn and raised skin around the periphery of the lesion. Sometimes the lesions expanded on tuber surface and formed hollowed-out areas. The causal organism was identified as Spongospora subterranea based on morphological characteristics using light and electron microscope. Ten cultivars of potato were evaluated to select resistant cultivars against powdery scab. Among the 10 cultivars, Jowon, Superior, and Atlantic were resistant, while Sephody, Gawon, and Irish Cobbler were susceptible to powdery scab. In the pathogenicity test, typical symptoms of powdery scab were formed on potato tubers, and galls developed on root hairs. This is the first report of powdery scab of potato caused by S. subterranea in Korea.

  • Occurrence of white scab of tea (CAmellia sinensis) showing numerous, small, circular, reddish or yellowish brown spots on young tea leaves was observed at a tea plantation of Boseung, Chonnam, from May to June in 1992-1994. At the late growth stages, the center of the spot became light gray. The causal fungus of the white scab was identified as Sphaceloma theae Kurosawa. Symptoms were produced 5∼6 days after inoculation by the artificial inoculation of s. theae conidia, which was similar to those on naturally infected leaves.

우리나라 감귤 더뎅이병균의 형태적 특성과 병원형 (Morphological Characteristics and Pathotype of Sphaceloma fawcettii Causing Citrus Scab in Korea.)

  • Citrus scab caused warty and scabby lesions on the surface of leaves, twigs and fruits of Satsuma mandarin. Warty lesions were mainly developed before July but scabby ones were developed during summer season in Cheju island, Korea. The casual organism of scab was morphologically identified as Sphaceloma fawcettii and was thought to be Tryon's pathotype of Elsinoe fawcettii, because it was pathogenic on rough lemon and Cleopatra mandarin but non-pathogenic on sour orange, grapefruit and sweet orange among the 5 differential hosts of E. fawcettii.

First Report of Citrus Scab on Trifoliate Orange (Poncirus trifoliata)

  • Citrus scab symptoms were observed on leaves of trifoliate orange (Poncirus trifoliata) in open field of Citrus Research Institute, National Institute of Horticultural and Herbal Science. Typical scab pustules were formed and they were elevated protuberances form and light brown to grey. The pathogens were isolated from the symptomatic leaves and then were identified to Elsinoë fawcettii by morphological characteristics, pathogenicity and PCR assay. The morphological characteristics of colonies formed very slow-growing, pulvinate, or raised and deeply fissured, gummy to mucoid or tomentose colonies on potato dextrose agar medium. The pathogens were pathogenic to leaves of Satsuma mandarin, lemon, sour orange and grapefruit with typical scab symptoms. In PCR assay, specific amplified of products of 717 bp with Efaw-1 and 384 bp with Efaw-2 were observed from trifoliate isolates and E. fawcettii as reference but not from E. australis. This is first report of trifoliate orange being positive for citrus scab disease.

포장조건에서 주요 배 유전자원과 종간교배 집단의 검은별무늬병 저항성 (Scab Resistance of Some Pear Genetic Resources and Inter Specific Hybrid Seedlings in Field Condition)

  • 본 연구는 주요 배유전자원 43 accession을 대상으로 육종소재로 활용할 수 있는 배 유전자원 선발과 종내 또는 종간 5개 교배집단 609 seedling에서 배 검은별무늬병 저항성 발현을 확인하고자 수행하였다. P. pyrifolia, P. bretschneideri, P. ussuriensis는 검은별무늬병 병든 과실률이 각각 59.5, 73.4, 63.0%, P. pyrifolia와 P. bretschneideri 종간 교잡으로 얻어진 품종은 77.7%로 병든 과실률이 비교적 높았으나 P. communis, P. pyrifolia ${\times}$ P. communis종간교잡에 속하는 품종들은 15% 미만의 병든 과실률을 보였다. 검은별무늬병에 대한 저항성을 종별로 구분하여 Duncan 검정한 결과, P. bretschneideri, P. pyrifolia, P. ussuriensis, pyrifolia ${\times}$ P. bretschneideri 종이 P. communis, P. pyrifolia ${\times}$ P. communis두 개 종 그룹보다 검은별무늬병 병든 과실률이 유의하게 높았고 이러한 경향은 교배집단에서도 동일하였다. P. pyrifolia 종내 교배집단은 80% 이상의 검은별무늬병 감염률을 보인 반면, P. pyrifolia ${\times}$ P. communis의종간교배 집단은 2% 수준의 낮은 배 검은별무늬병 감염률을 보였다. P. pyrifolia 와 P. communis를 종간교배 하면 서양배의 저항성 인자가 우성으로 작용하여 그 후대에서 배 검은별무늬병에 강한 개체들이 대부분 얻어지기 때문에 P. communis는 배 검은별무늬병저항성 품종 육종소재로 적합하다.

바이오 황을 이용한 감귤 더뎅이병 발병 억제 효과 (Suppressive Effects of Bio-Sulfur on Citrus Scab)

  • 본 연구에서는 친환경 농가에서 사용되고 있는 바이오 황(Bio-S, bio-sulfur)을 이용하여 감귤 더덩이병(Citrus scab)에 대한 발병 억제 효과를 알아보고자 수행되었다. 바이오 황이 감귤 더뎅이병의 발아관 생장 억제 효과는 감귤 더뎅이병균을 PDB와 Agar 배지에서 배양하여 발아관을 관찰한 결과 접종 40시간과 88시간 모두 바이오 황 500배, 1,000배, 2,000배 처리구에서 발아관 형성이 억제되었으며, 4,000배 이상 희석배수가 높아질수록 발아관은 형성되었으나 무처리구에 비해 생장이 억제되었다. 포장에서 감귤 봄순 잎에 대한 더뎅이병의 발병 억제 효과는 무처리구 이병율은 40.3%였으며, 화학농약인 Imibenconazole 수화제 이병율이 5.3%을 보였으며, 친환경 농업에 사용하는 석회보르도액 2-4식과 6-6식은 모두 10.3%, 바이오 황 500배 12.3%, 석회유황합제 15.3%로 비슷한 경향을 나타냈으며, 반면 바이오 황 1,000배에서는 24.0%로 비교적 높은 이병율을 보였다. 감귤 과실에 대한 시험결과 무처리구 이병율은 79.3%였으며, Imibenconazole 수화제 이병율은 4.0%을 보였으며, 석회보르도액 2-4식 33.8%, 6-6식 42.0%, 바이오 황 500배 43.3%, 석회유황합제 44.8%로 비슷한 효과를 나타냈으며, 반면 바이오 황 1,000배에서는 78.0%로 비교적 높은 이병율을 보였다. 따라서 감귤 더뎅이병은 봄순이 전개되는 5월부터 감귤의 잎에 발생하기 때문에 봄순이 전개되기 전인 4월 중하순부터 예방방제를 시작하여 예찰을 통해 방제하면 높은 방제율과 노동력 및 영농비용 등을 절감할 수 있을 것으로 생각된다.

토양개량제시용에 따른 화산회토양 감자 연작지 더뎅이병 억제 효과 (Effect of Applying Soil Amendments on Potato Scab Prevention in Volcanic Ash Soil with Continuous Cropping System)

  • 화산회토양 감자 연작지 토양 pH 수준별로 고토석회, 유황, 황산칼륨, 패화석, 규산질비료, 석회질소, 유안비료를 이용하여 감자 더뎅이병 발생을 경감시키고 토양건전성을 유지하는데 적절한 대체농자재를 선발하고자 수행하였다. 토양 pH가 낮은 황산칼륨 처리구(24 kg/10a)에서는 더뎅이병 발병률이 84.4%, 발병도가 28.4%로 가장 낮았다. 방제효과는 12.3%, 상품율은 93.2%로 다른 처리구와 비교할 때 가장 높게 나타났다. 석회질소 처리구(60kg/10a)가 발병도 38.3%로 낮았고 방제효과 23.8%, 상품율 66.3%로 높았다. 토양 pH가 높은 시험구 중 더뎅이병 발병률은 석회질소 처리구(60 kg/10a)가 38.3%로 가장 낮았고 유안 처리구(48 kg/10a)는 62.6%로 높았다. 더뎅이병 방제효과는 석회질소 처리구(60 kg/10a)에서 23.8%로 유황 처리구의 6.1% 보다 4배 이상, 상품률은 66.3%로 가장 높게 나타났다. 감자더뎅이병 병원균 Streptomyces acidiscabies과 Streptomyces scabiei를 석회질소 2.5, 5 g/L, 10 g/L에 접종시 생장량은 다른 토양개량제 보다 상대적으로 억제되었다. 결론적으로 감자 파종 전에 토양 pH가 5 이하이면 황산칼륨을, 6 이상이면 석회질소를 시비하면 더뎅이병 발생을 경감시킬 수 있을 것으로 보인다.

  • Potato scab, an important disease that affects developing tubers, causes a major problem in potato cultivation. The major potato cultivation areas in Korea are located in two Northern provinces, Gangwon and Gyeonggi, and two Southern provinces, Jeju island, and South Jeolla. In these areas, potato scab is widely distributed and has caused severe problem in potato cultivation. Therefore, potato-growing areas were surveyed for identification and distribution of potato scab pathogens from 1996 to 1999. Pathogenic Streptomyces strains were isolated from potato scab lesions and six representative Streptomyces species were characterized based on their phenotypic and molecular characteristics including, pathogenicity, physiological and morphological properties, analyses of 16SrRNA genes and 16S-23S ITS region, DNA relatedness, production of thaxtomin A, and the presence of nec1 and ORFtnp gene homologs. Three species were identified as previously described Streptomyces scabies, S. turgidiscabies, and S. acidiscabies, while other three species having distinct phenotypics properties were identified as novel S. luridiscabiei, S. puniciscabiei, and S. niveiscabiei.

Scab of Balsam Pear (Momordica charantia) Caused by Cladosporium cucumerinum in Korea

  • During winter season of 2004 to 2006, a scab disease on balsam pear (Momordica charantia) caused by Cladosporium cucumerinum was observed in Daesan-myon, Changwon-city, Gyeongnam province, Koyea. The disease symptom started with small dark brown speck on the fruits and leaves then the lesions expanded and spreaded irregularly. The aggregated mycelial mass and conidia of the fungus formed sooty scab. The colony of purely isolated fungus grew in greenish black to velvety on potato dextrose agar (PDA). Conidia were ellipsoidal, fusiform or subspherical, mostly one-celled but occasionally septated and $3{\sim}32{\times}2{\sim}6{\mu}m$ in size. The conidiophores were erected and had long branch, chains pale olivaceous brown in color and $6{\times}280{\mu}m$ in size. Ramoconidia were $10{\sim}34{\times}3{\sim}8{\mu}m$ in size. The fungus was identified as Cladosporium cucumerinum based on the morphological characteristics. The pathogenicity of the fungus was confirmed according to Koch's postulate. The optimum temperature of the isolate was about $20^{\circ}C$. This is the first report on scab of balsam pear caused by C. cucumerinum in Korea.
