Sawdust Media Affecting the Mycelial Growth and the Fruiting Body Formation of Sparassis crispa

  • Six strains of Sparassis crispa such as S. crispa DUM-01, DUM-02, DUM-03, DUM-04, DUM-05, and DUM-06 were tested for their mycelial growth on 6 different kinds of sawdust media and primordial formation on 10 different compositions of larch sawdust media. The highest mycelial growth was recorded on the larch sawdust. Of the 6 strains of S. crispa, S. crispa DUM-04 recorded the favorable formation of primordia. The primordial formation of S. crispa DUM-04 was more favorable on L-3 medium than 9 kinds of larch sawdust media. When S. crispa DUM-04 was cultured on the media of larch sawdust+pine sawdust, the formation of its fruiting bodies was more outstanding on the media of larch sawdust+pine sawdust than those of larch sawdust.

Steam Treated Sawdust as Soilless Growing Media for Germination and Growth of Horticulture Plant

  • The major substrates used in soilless growing media are peat moss and perlite, where peat moss is a limited and expensive natural resource. Determination of appropriate substrates based on technical and economic feasibility is the vital aspect of research and the key to success in any soilless production system. This research work was performed to evaluate different low-cost and sustainable alternative substrate as soilless growing media for horticulture plant. The objective of this study was to compare the effect of sawdust species and steam treatment, for physico-chemical properties and growth of horticultural plant. This study involves the physical and chemical characterization and growth test of four substrate (pine sawdust, oak sawdust, steamed pine sawdust and steamed oak sawdust) in order to evaluate their use as components of growing media. Steamed oak sawdust ($121^{\circ}C$, 30 min) showed adequate physical and chemical properties compared to peat moss for their use as growing media. The growing media were prepared using different mixture proportion to grow Brassica campestris L., Festuca arundinacea and Lespedeza cyrtobotrya Miq. The highest germination, stem length and leaf area of Brassica campestris L., Festuca arundinacea and Lespedeza cyrtobotrya were observed in 30 minute steamed oak sawdust mixture growing media. The steam treatment condition of sawdust used in the growing media significantly positive affected the germination, the stem length and the leaf area.

  • 소나무잔나비버섯의 톱밥재배를 위한 가장 적정한 종균 배양적 특성을 요약하면 적정온도는 $30^{\circ}C$이며, 적정 pH가 소나무잔나비버섯균은 pH 5이다. 톱밥배지의 적정 수분함량은 60%이며, 주재료는 미송톱밥, 첨가재료는 맥주박이었다. 또한 적정 첨가재료인 맥주박의 첨가량은 20%, calcium carbonate의 첨가함량은 0.1%, sucrose의 첨가함량은 0.1%이었다. LVD의 첨가함량은 0.01%이었다.

Calcium Absorption by the Fruitbody of Saesongi (Pleurotus eryngii) Mushroom

  • Saesongi (Pleurotus eryngii) was cultivated in both potato dextrose agar (PDA) and sawdust media supplemented with Ca salts. The addition of Ca phosphate and Ca carbonate to sawdust media did not affect the growth, whereas Ca sulfate addition suppressed the mycelial growth appreciably. The efficiencies of Ca accumulation in the fruiting were studied based on mycelial growth experiments on Ca-supplemented sawdust media. Supplementation with 0.1 to 5% Ca phosphate increased the Ca content in the fruiting body by 4.5-6.5 fold, to a level of $314.6{\pm}22.7$ to $449.7{\pm}29.3$.

표고버섯 톱밥 인공재배(人工栽培)에 관(關)한 연구(硏究) (Studies on the Artificial Cultivation of Lentinus edodes on sawdust media)

  • 표고버섯의 배양적 특성과 톱밥수종 및 첨가재료를 이용(利用)한 인공재배(人工栽培) 가능성(可能性)을 검토한 결과(結果)는 다음과 같다. 1. 표고버섯의 톱밥인공재배(人工栽培)에 적합한 균주(菌株)는 ASI3046 균주(菌株)이며 균계생육(菌系生育) 최저온도(最低溫度)는 $22{\sim}26^{\circ}C$, pH는 $5.5{\sim}6.5$ 이였다. 2. ASI3046 균주(菌株)에 대한 균계생육(菌系生育)이 양호(良好)한 탄소원(炭素源)은 glucose이며 질소원(窒素源)은 peptone과 ammonium tartrate 였다. 3. 표고버섯 통밥인공재배시(人工栽培時) 배지(培地)의 수종 및 첨가재료(添加材料)는 혼합(混合)통밥(참나무톱밥+포플라톱밥)에 상수리 분말 5% (v/v)에 미강 10% (v/v)를 첨가(添加)한 구에서 군계생장(菌系生長) 및 균계밀도(菌系密度)가 양호(良好)하고 자실본수량도 높았다. 4. 표고버섯 통밥인공재배(人工栽培)에 적합한 균주(菌株)는 ASI3046 균주(菌株) 이였다.

  • (1) 자연상태(自然常態)에서 활엽수(闊葉樹) 기주(寄主) 선택성(選擇性)인 목재(木材) 부후성(腐朽性) 식용균(食用菌) 6종(種)을 골라 소나무 톱밥을 재료(材料)로 하여 배양(培養) 실험(實驗)했다. (2) 소나무톱밥만을 단용(單用)한 배지(培地)에서는 pholiota squarrsa를 제외(除外)한 다른 균류(菌類)의 균계(菌系) 신장(伸長)은 보다 조해(阻害)되고 있다. (3) 광당전즙(光糖煎汁) 가용(加用)톱밥 배지(培地)에서는 추출(抽出) 처리구(處理區) 및 무처리구(無處理區) 어느 경우(境遇)나 균계(菌系) 신장(伸長)은 거의 차이(差異) 없이 호양(好良)하다.

표고버섯(JNM3022) 톱밥재배에 적합한 배지조성과 배지크기 (The optimum media size and composition for sawdust culture of Lentinula edodes (Strain: JNM3022))

  • 표고버섯(J NM3022) 의 톱밥재배시 적절한 배지크기 및 배지조성을 구명하기 위하여 '05년부터 2년동안 시험결과 다음과 같은 결과를 얻었다. 표고버섯 톱밥재배를 위하여 배지길이와 직경이 다른 12가지 크기의 배지를 이용하여 배양기간, 배양완성률을 시험한 결과 가장 적절한 배지크기는 길이 25 cm ${\times}$ 직경 14cm, 무게 2.5 kg 배지였다. 소나무톱밥의 포화수분함량은 495 $m{\ell}/100g$으로 침 나무동밥 251 $m{\ell}/100g$보다 약 2배 많았다. 참나무톱밥 100% 등 10조합의 배지조성 중 균사생장속도와 균사일도가 가장 좋았던 배지조성은 참나무톱밥 80%+밀기울 20%와 참나무톱밥 80%+미강 20% 처리였으며, 가장 적합한 배지수분함량은 55%였다. 배양기간을 단축하기 위하여 배지중앙에 유공관을 설치하였을 때 무처리에 비하여 길이 20 cm 배지는 11일, 30 cm 배지는 32일을 단축할 수 있었다. 배지표면 갈변화에 가장 좋았던 처리는 $10^{\circ}C$ 정도의 변온과 봉지천공으로 갈변 소요기간은 35일로 동일조건의 무천공처리에 비하여 54일을 단축시킬 수 있었다.

  • 노루궁뎅이버섯균의 균사생장과 밀도에 가장 적절한 종균배양적 특성을 요약하면 적정온도는 $25^{\circ}C$, pH는 5, 톱밥 배지의 적정 수분함량은 $65{\sim}70%$이며, 주재료는 참나무톱밥, 첨가재료는 밀기울이었다. 또한 적정 첨가재료인 밀기울의 첨가량은 20%, calcium carbonate의 첨가함량은 0.1%이며, magnesium sulfate의 첨가함량은 0.2%, sucrose의 첨가함량은 0.2%이었다.

The Artificial Cultivation of Oudemansiella mucida on the Oak Sawdust Medium

  • To produce fruiting bodies of Oudemansiella mucida, porcelain fungus, on the oak sawdust medium, additives suitable for the mycelial growth and fruiting body formation were screened. In general, the mycelial growth of the three strains of O. mucida used in this study have been good on oak sawdust mixed rice bran of $20{\sim}30%$. The mycelia incubated in potato dextrose broth for 7 days were inoculated on oak sawdust medium supplemented with various ratios of rice bran and incubated for 30 days at $25^{\circ}C$ in the dark condition until the mycelia of O. mucida fully colonized the media from top to bottom. Then, top surface of the media in the bottles were horizontally scratched with a spatula and filled with tap water for 3 hours. To induce the primordial formation of O. mucida, the bottles were transferred to the mushroom cultivating room under 12 hrs of light (350 lux) and dark condition with relative humidity of 95% at $17^{\circ}C$. The primordia of O. mucida were formed on the surface of oak sawdust media after 7 days of incubation. The mature fruiting bodies were observed 5 days after primordial formation. The fruiting bodies O. mucida were formed on oak sawdust medium mixed with 5 to 30% rice bran. However, abundant fruiting-bodies of O. mucida were produced in oak sawdust medium supplemented with 20% rice bran. This is the first report associated with an artificial fruiting body production of O. mucida in Korea.

퇴비화 암모니아 가스의 톱밥 바이오 필터 매체에 의한 생물학적 탈취 (Biofiltration of Ammonia Gas from Composting Using Sawdust as Biofilter Media)

  • Dairy manure amended with crop and forest residues (moisture 69% wet basis, C/N 22) was composted in a 605 L pilot-scale vessel using continuous air flow (56 L/min) for 19 days. Three pilot-scale sawdust biofilters (moisture 63%, pH 5.0) were built to clean biological waste gas from the composting process. For each methods, two replicated experiments were monitored over a period of three weeks. The system was evaluated to determine the biofilter media depth that would be adequate for compost odour reduction. The compost air cleaning was measured based on ammonia gas concentration before and after passing through the biofilter. Ammonia gas removal efficiency over 3 weeks was 42, 75 and 87% at sawdust biofilter media depth levels of 202, 400 and 600 mm, respectively. Each sawdust biofilter was operated at a moisture content in the range of 60~62% (wb), a temperature from 15 to $25^{\circ}C$, an average pressure drop from 240 to 340 Pa and a detention time from 60 to 180 seconds during the biofiltration process.
