The Effect of Saururus Chinensis Extracts on Antioxidant Activity and Melanin Synthesis (삼백초 추출물이 항산화활성과 멜라닌 합성에 미치는 영향)

  • Saururus chinensis has white roots, leaves, and flowers and is known to have antibacterial activity and anti-cancer efficacy. The aim of this study was to investigate the effect of the ethyl acetate fraction of a methanol extract of Saururus chinensis (SCEA) on antioxidant activity and melanin synthesis. SCEA at 64 ㎍/ml showed 62% of the DPPH radical scavenging activity of vitamin C, and its reducing power was 33% greater than that of vitamin C. Tyrosinase activity was 26% higher and melanin synthesis was 44% higher in the presence of SCEA at 64 ㎍/ml than in a blank group in a dopa oxidation assay. MTT assay showed that SCEA displayed cytotoxicity above 0.5 ㎍/ml, and SCEA at 1 ㎍/ml increased melanin synthesis by 69% in live B16F1 cells. SCEA was also separated into 13 fractions by silica column chromatography, and fraction 2 (Fr. 2) showed the highest DPPH radical scavenging activity, reducing power, and melanin synthesis. SCEA also promoted melanin production in live cells. LC-MASS analysis showed that Fr.2 had a molecular weight of 239, and these findings suggest that SCEA could be available for the promotion of melanin synthesis in black hair.

Hepatoprotective Constituents of Saururus chinensis Roots Against Tacrine-induced Cytotoxicity in Human Liver-derived Hep G2 Cells (삼백초근의 타크린으로 유발한 간 세포독성 보호 성분)

  • Five liglans, sauchinone (1), di-0-methyltetrahydrofuriguaiacin B (2), manassantin A (3), manassantin B (4) and saucerneol B (5), have been isolated from the MeOH extract of Saururus chinesis roots. The evaluation for protective effect of compounds 1-5 against tacrine-induced cytotoxicity in human liver-derived Hep G2 cells was conducted. Compounds 1,2, and 5 showed significant protective effects with the EC$_{50}$ values of74.2${\pm}$0.9, 111.3${\pm}$0.8,64.3${\pm}$0.8 ${\mu}$M, respectively. Silybin, one of the well-known hepatoprotective agents, used as a positive control, and also showed protective effect with an EC$_{50}$ value of 86.2${\pm}$0.5 ${\mu}$M.

Optimization of Dressing Preparation from Yogurt Added Saururus chinensis (Lour.) Bail Extract (삼백초 추출물 첨가 요구르트를 이용한 드레싱 제조의 최적화)

  • Yogurt base was prepared from milk powder $(14\sim18%)$ to which was added 0.4% Saururus chinensis (Lour.) Bail water extract (SCE) and fermented with lactic acid bacteria (a mixed strain of Streptococcus thermophilus and Lactobacillus bulgaricus) at $37^{\circ}C$ for 24 hr. The viscosity of the yogurt added SCE (SCE yogurt) made from 18% milk was higher than that of SCE yogurt containing $14\sim16%$ milk, whereas the pH and titratable acidity of the SCE yogurt were not significantly different on the range of milk contents. The optimal milk concentration for SCE yogurt manufacture was 18%. In order to optimize the preparation of dressing from SCE yogurt, the central composite design was conducted in terms of the yogurt (30, 40, 50, 60, 70 g), the mayonnaise (6, 12, 18, 24, 30 g) and the salt (0.1, 0.3, 0.5, 0.7, 0.9 g) contents. Sensory evaluation was performed and evaluated using a response surface methodology. The optimum ingredient ratio for SCE yogurt dressing was determined to be 61.2% of yogurt, 23.5% of mayonnaise, 0.58% of salt, 0.58% of honey, 1.75% of mustard, 0.23% of Tabasco pepper sauce, 0.94% of wine and 0.04% of white pepper.

Effect of the Samwondan Ethanol Extract on Obesity Inducer (三元丹 에탄올 추출물이 비만유도인자에 미치는 영향)

  • Objectives : In this study, we investigated the biological activities such as anti-obesity using Samwondan ethanol extract (SWD). SWD is a complex with Salicornia herbacea Linnaeus, Saururus chinensis Baill and Houttuynia cordata Thunberg as the main raw material.Methods : The SWD was extracted 80% ethanol. 3T3-L1 preadipocytes were induced adipogenesis by differentiation media with SWD at 1 μg/mL, 10 μg/mL, and 100 μg/mL. Effect of SWD performed using MTT assay, oil red O staining (observation by microscope), and reverse transcription polymerase chain reaction. Also we measured production of triglyceride (TG), fatty acid, and acetyl-CoA carboxylase (ACC).Results : Non-cytotoxicity was in all test group from range of 1 μg/mL to 100 μg/mL on pre-adipocyte. The droplet and production of lipid were decreased significantly by the SWD. And TG was decreased by approximately 89%, 85% and 82%, upon the SWD treatment at concentration of 1 μg/mL, 10 μg/mL, and 100 μg/mL. Moreover, the SWD showed inhibitory effects on the expression of the C/EBP (CCAAT/enhaner binding protein)β, C/EBPα, and peroxisome proliferator-activated receptorγgenes in adipocytes. The SWD at 100 μg/mL concentration showed inhibitory effect on fatty acid production by 79%. Also ACC production were decreased dose-dependently.Conclusions : From the results above, we concluded that the SWD indicated significantly anti-obesity effects.

DPPH Radical Scavenging Effect and Antimicrobial Activities of Some Herbal Extracts (주요 허브 추출물의 항산화성 및 항균활성)

  • Seven native and four introduced herbs namely $Thymus$ $quinquecostatus$, $Chrysanthemum$ $zawadskii$ var. $latilobum$, $Rosmarinus$ $officinalis$, etc. were selected for analysis of the DPPH radical scavenging and anti-microbial activity of their extracts. These perennial herbs are classified as $Labiatae$ and $Compositae$ except for $Saururus$ $chinensis$ and can be propagated through seedling and cuttage. These edible herbs are used as medicinal as well as ornamental plants. Their extract has electron donating ability which ranges from 69.7 to 78.7% for native herbs and 67.4 to 75.4% for introduced herbs. Native herbs have higher (3.54%) average DPPH radical scavenging than introduced herbs. In native herbs, maximum DPPH radical scavenging activity was observed in $Agastache$ $rugosa$ (78.7%) followed by $Saururus$ $chinensis$ while $Chamaemelum$ $nobile$ showed highest activity among the introduced herbs. Many herbs viz. $Saururus$ $chinensis$, $Chrysanthemum$ $zawadskii$ var. $latilobum$ and $Solidago$ $virga-aurea$ var. $gigantean$ showed excellent anti-microbial activity against gram positive $Enterococcus$ $faecalis$, maximum (80.0%) by $Saururus$ $chinensis$. Other herbs viz. $Solidago$ $virga-aurea$ var. $gigantea$, $Chrysanthemum$ $zawadskii$ var. $latilobum$, $Salvia$Salvia $plebeia$, $Chrysanthemum$ $indicum$, $Rosmarinus$ $officinalis$, $Chamaemelum$ $nobile$ and $Lavandula$ $stoechas$ showed anti-microbial activity against gram negative $Citrobacter$ $freundii$. Especially, the inhibition of colony growth of $Citrobacter$ $freundii$ was highest in the extract of $Chrysanthemum$ $zawadskii$ var. $latilobum$, and $Chamaemelum$ $nobile$.

The Efficacy of Saururus chinensis on Cervical Cancer Cells : The Inhibitory Effect on the Function of E6 and E7 Oncogenes of HPV Type 16 (삼백초 추출물의 자궁경부암세포 억제 효능)

  • Cervical cancer is one of the leading causes of female death from cancer worldwide with about 500,000 deaths per year. A strong association between certain human papilloma viruses (HPV type 16 and 18) and cervical cancer has been well known. An extract of Saururus chinensis, named as PE-46, has been used to investigate whether this agent has the ability of inhibiting the oncogenes E6 and E7 of HPV type 16. PE-46 inhibited the proliferation of human cervical cancer cell lines in a dose response manner. PE-46 also inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53. In addition, PE-46 inhibited the in vitro binding of E7 and Rb which is essential tumor suppressor for the control of cell cycle. The levels of mRNA for E6 and E7 were also decreased by PE-46. SiHa cells treated with PE-46 induced G0/G1 arrest, resulting in inhibition of growth. Our study showed that the PE-46 can inhibit the cervical carcinomas via both inhibition of bindings between oncogenes and tumor suppressors, and inhibition of G1longrightarrowS transition. PE-46 inhibited the oncogenecity of E6 and E7 of HPV 16 type, thus could be used as a putative modulating agent for the treatment of cervical carcinomas caused by HPV.

Preparation and Quality Characteristics of Yogurt Added with Saururus chinensis (Lour.) Bail (삼백초를 첨가한 요구르트의 제조와 품질 특성)

  • Yogurt base was prepared from whole milk and skim milk added with 0.2∼1.0% (w/v) of Saururus chinensis(Lour.) Bail water extract (SCe) and fermented with lactic acid bacterias (the mixed strain of Streptococcas themophilus and Lactobacillus bulgaricus) at 37$\^{C}$ for 24 hr. Quality characteristics of the yogurt were evaluated in terms of acid production, number of viable cells, viscosity and sensory property during lactic acid fermentation. The composition of organic acids was also measured by HPLC. Addition of SCe stimulated the growth of lactic acid bacteria and remarkably enhanced the acid production. The viscosity and lactic acid content of yogurt were also increased by addition of SCe. The sensory score of yogurt added with 0.4% of SCe was significantly higher than other groups in taste and overall acceptability The storage abilities of yogurts added with SCe were relatively good at 5$\^{C}$ for 15 days.

Physicochemical properties of dried Saururus chinensis and the antioxidative activities of water and 70% ethanol extracts (덖음온도를 달리하여 전저리한 삼백초 건조물의 이화학적 특성 및 물과 70% 에탄올 추출물의 항산화효과)

  • Purpose: This study was conducted to evaluate the physicochemical properties of different batches of Saururus chinensis using different roasting temperature that were dried at different using different roasting temperatures and their were determined the antioxidative activities of water and 70% ethanol extracts. Methods: Extracts were examined for the total phenolic acid content, the and flavonoids contents and the antioxidant properties, including DPPH radical scavenging activity, ABTs scavenging activity and, the reducing power. Results: Moisture content was significantly higher in the LSC and the crude ash content was significantly higher in the HSC. The crude protein content was higher in the LSC (although not significantly), and the crude fat and carbohydrate contents were higher in the HSC (although not significantly). The total phenolic content was lower in the samples extracted with water, but there was no significant difference. However, the extracts extracted with 70% ethanol at a high drying temperature were significantly higher. The low temperature and high drying temperature batches of Saururus chinensis were significantly higher in the samples extracted with 70% ethanol than those extracted with 70% ethanol. The total phenolic acid content, the total flavonoid content and the electron donating ability were highest in the ethanol extract of Saururuschinensis treated at a high temperature. However, the ABTs radical activity was highest in the water extracted, high-temperature treated Saururuschinensis. The 70% ethanol extract of high temperature roasted Saururuschinensis had the highest antioxidative activities of all the Saururuschinensis batches. Conclusion: The total phenolic acid contents, total flavonoid contents, electron donating activity and reducing power activity were highest in all the 70% ethanol extraction batches of the high-temperature treated samples.

Antibacterial and Intracellular Clearance Effect of Saururus chinensis Baill Water Extract against Salmonella typhimurium (삼백초 수용성 추출물의 Salmonella typhimurium 균에 대한 항균 및 숙주세포 내 사멸 효과 규명)

  • Salmonella spp. are one of major pathogens for zoonosis in worldwide, and can replicate within host cells and generally cause enterocolitis and foodborne poisoning which represents a considerable public health burden. The present study was designated to investigate the safty for host cells, antibacterial effects of Saururus chinensis Baill water extract (SCWE) on pure culture and infection with Salmonella enterica serovar Typhimurium (S. typhimurium) in murine derived macrophage RAW 264.7 cells. The different treatment of SCWE concentration (1, 10 or $100{\mu}g/ml$) did not show any cytotoxic effect to RAW 264.7 cells for 24 h incubation. In determination of antibacterial activity of SCWE against S. typhimurium, bacterial viability was markedly decreased compared to SCWE-untreated control. In RAW 264.7 cells, SCWE significantly induced morphological change (p<0.05). In infection assay of S. typhimurium in RAW 264.7 cells pretreated with $100{\mu}g/ml$ of SCWE, which are non-cytotoxic concentration, bacterial uptake ability of macrophage was increased corresponding with morphological change, whereas bacterial survival rates within macrophage was markedly reduced comparing to that of SCWE-untreated control. Furthermore, nitric oxide (NO) production in SCWE-treated cells was slightly decreased until 24 h post infection. Taken together, these findings demonstrated that SCWE have the antibacterial activity for S. typhimurium and the protective effects against S. typhimurium infection through activating murine macrophage independent on NO, suggesting that SCWE were beneficial on the disease caused by intracellularly replicating pathogens as a safe alternatives of conventional chemotherapies.

Antioxidative effects of traditional medicinal plants on lipid peroxidation (지질 과산화에 대한 전통약용 식물의 항산화 효과)

  • To assess the antioxidative activity of 12 medicinal plants on lipid peroxidation, twelves traditional medicinal plants extracted with 95% methanol were investigated the antioxidative activity using DPPH, thiocyanate acid method, and thiobarbituric acid (TBA) methods. Out of 12 medicinal plants extracted with methanol, the extraction yields of Sedum kamtschaticum was the highest values (49.46%) among them and Geranicum sibiricum, Saururus chinensis root (R), Agrimonia pilosa leaf (L), Agrimonia pilosa root was the lowest value (9.97%). Radical scavenging effect of the selected traditional medicinal plants extracted from different extract solution were examined by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical method. Antioxidative activity of methanolic extracts was higher than those of ethanol and n-hexane extracts. Scavenging effects in Sedum kamtaschaticum (R) determined by DPPH radical showed the highest among the 12 plants. The antioxidative effects of the first four medicinal plants were similar to those of butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT), but higher than that of tocopherol, which was used as a handled control. Antioxidative effects of each indicated concentration of the methanolic extracts on linoleic acid by thiocyanate method was the highest in Sedum kamtschaticum and followed by Geum japonicum and Agrimonia pilosa and their antioxidative effect were similar to those of BHA, and BHT, but higher than that of tocopherol. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by thiocyanate acid method were examined for 15 days. Peroxidation of control and tocopherol group occurred on days 5 and 9, respectively, but BHA, BHT, selected medicinal methanolic extract group did not occur until on day 15. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by TBA method were examined for 15 days. Antioxidative activity was similar to those obtained by thiocyanate acid method.
