Estimation of the Through Car Equivalent for Left-Turn Movement According to the Left-Turn Curve Radius in the Signalized Intersection (신호교차로에서 곡선반경에 따른 좌회전의 직진환산계수 산정)

  • This study is to estimate the through car equivalent for left-turn movement according to left turn radius in the signalized intersection. The saturation flow rate for left-turn movement in 92' KHCM(Korea Highway Capacity Manual) is the same as that for through movement in the signalized intersection where the left turn radius is over 20m. This study uses Duncan 'Test's Test in order to find out the saturated position for queued vehicle in the signalized intersection. It found that the saturated flow rate of left-turn movement in the signalized intersection is less than that of through movement where the left-turn radius is less than 20m. It found that the through car equivalents for left-turn movement according to left-turn radius(8m-20m) are between 1.05 and 1.14.

Establishment of Ultrasonic Measurement Method for Stone Cultural Heritage Considering Water Content and Anisotropy (함수율과 이방성을 고려한 석조문화유산의 초음파 측정방법 설정)

  • This study was focused on measurement methods for stone cultural heritages by analyzing Ultrasonic (P-wave) velocity variations according to the water content and anisotropy of rocks. As a result of analyzing of rock properties, the water content and saturation degree were rapidly changed at the beginning of drying and then showed exponential curve which their rates of change gradually decreased. However, P-wave velocity and its rate of change maintained constant values after natural drying of 10 hours. Therefore, the ultrasonic measurement for stone cultural heritages should be performed after natural drying of 10 hours considering the weather and moisture conditions. In addition, the highest values of anisotropy coefficient exhibited in granite and limestone, and indirect method was insensitive to anisotropy compared to direct method. However, all rocks remained anisotropy by indirect method. Accordingly, ultrasonic measurement considering various directions is required. The research results will contribute to customized non-destructive testing and precise diagnosis for lithological characteristics of stone cultural heritage.

Evolution of Magnetic Property in Ultra Thin NiFe Films (나노두께 퍼말로이에서의 계면효과에 의한 자기적 물성 변화)

  • We prepared ultra thin film structure of Si(100)/ $SiO_2$(200 nm)/Ta(5 nm)/Ni$_{80}$Fe$_{20/(l~15 nm)}$Ta(5 nm) using an inductively coupled plasma(ICP) helicon sputter. Magnetic properties and cross-sectional microstructures were investigated with a superconduction quantum interference device(SQUID) and a transmission electron microscope(TEM), respectively. We report that NiFe films of sub-3 nm thickness show the B$_{bulk}$ = 0 and B$_{surf}$=-3 ${\times}$ 10$^{-7}$(J/$m^2$). Moreover, Curie temperature may be lowered by decreasing thickness. Coercivity become larger as temperature decreased with 0.5 nm - thick Ta/NiFe interface intermixing. Our result implies that effective magnetic properties of magnetoelastic anisotropy, saturation magnetization, and coercivity may change abruptly in nano-thick films. Thus we should consider those abrupt changes in designing nano-devices such as MRAM applications.

  • The muscarinic antagonist 1-[benzilic 4, 4'-$[^3H]$ QUINUCLIDINYL BENZILATE $([^3H]$ QNB) bound to a single class of muscarinic receptors with high affinity in rabbit ileal membranes. The $K_D\;and\;B_{ max}$ values for $([^3H]$ QNB calculated from analysis of saturation isotherms were 52.5 pM AND 154 fmol/mg, respectively. Chlopheniramine (CHP), histamine $H_1$ blocker, increased $K_D$ vlue for $([^3H]$QNB without affecting the binding site concentrations and Hill coefficient. The $K_i$ value of CHP for inhibition of $([^3H]$QNB binding in ileal membranes was 1.44\mu{M}$ and the pseudo-Hill coefficient for CHP was close to unit. In the functional assay carbachol, muscarinic agonist, increased the contractile force of ileum with $ED_{50}$ value of $0.11\mu{M}$. CHP caused the rightward shift of the dose-response curve to carbachol. The $pA_2$ value of CHP determined from Schild analysis of carbacholinduced contraction was 5.77 and the slope was unity indicating competitive antagonism with carbachol. The dissociation constant $(K_i)$ of CHP obtained in competitive experiments with $([^3H]$ QNB was similar to the $K_A$ value (1.69 \mu{M)}$ of CHP as inhibitor of carbachol induced contraction in rabbit ileum. This result suggest that the binding of $H_i$ blocker. CHP, vs $([^3H]$QNB to muscarinic receptors in ileal membranes represents an interaction with a receptor of physiological relevance.

  • In this study, modified triaxial compression tests were carried out to investigate the characteristics of shear strength of unsaturated dredged soils. The variation of shear strength generally depends on more matric suction than drained conditions, and then is gradually converged in matric suction 100kPa. It indicates that the effective angle of internal friction and effective cohesion in unsaturated conditions increase due to degree of saturation, namely, matric suction than those of saturated conditions. Therefore, it shows that apparent friction angle, ${\phi}^b$ due to the variation of matric suction to evaluate reasonable shear strength parameters in unsaturated soils should be considered.

  • The effects of water temperature, salinity and irradiance on the growth of harmful algae Chattonella marina isolated from Gamak Bay in South Sea, Korea were investigated. C. marina was able to grow in temperatures of $15-30^{\circ}C$ and salinities of 10-35 psu. Maximum specific growth rate (0.64/day) was observed with combination of $25^{\circ}C$ and 25 psu. Optimal growth (${\ge}70%$ of maximum specific growth rate) was obtained with all salinities of the above $20^{\circ}C$. This result indicated that C. marina is a stenothermal of the high water temperature and euryhaline organism. C. marina was did not grow at irradiance ${\le} 10{\mu}mol$ photons/($m^2\;s$). Photoinhibition did not occur at $300{\mu}mol$ photons/($m^2\;s$), which was the maximum irradiance used in this study. The irradiance-growth curve was described as ${\mu}=0.78(I-11.4)/(I+34.1)$ at $25^{\circ}C$ and 25 psu. The half-saturation photon flux density (PFD) ($K_s$) was $56.9{\mu}mol$ photons/($m^2\;s$) and compensation PFD ($I_c$) was $11.4{\mu}mol$ photons/($m^2\;s$). The result of the present study indicate that C. marina has advantage physiological characteristic to the interspecific competition at the embayment and costal areas of South and West Sea, Korea in summer.

Unusual Allosteric Property of L-alanine Dehydrogenase from Bacillus subtilis

  • Kinetic studies of L-Alanine dehydrogenase from Bacillus subtilis-catalyzed reactions in the presence of $Zn^{2+}$ were carried out. The substrate (L-alanine) saturation curve is hyperbolic in the absence of the metal ion but it becomes sigmoidal when $Zn^{2+}$ is added to the reaction mixture indicating the positive cooperative binding of the substrate in the presence of zinc ion. The cooperativity of substrate binding depends on the xinc ion concentration: the Hill coefficients ($n_H$) varied from 1.0 to 1.95 when the zinc ion concentration varied from 0 to $60\;{\mu}m$. The inhibition of AlaDH by $Zn^{2+}$ is reversible and noncompetitive with respect to $NAD^+$ ($K_i\;=\;5.28{\times}10^{-5}\;M$). $Zn^{2+}$ itself binds to AlaDH with positive cooperativity and the cooperativity is independent of substrate concentration. The Hill coefficients of substrate biding in the presence of $Zn^{2+}$ are not affected by the enzyme concentration indicating that $Zn^{2+}$ binding does not change the polymerization-depolymerization equilibria of the enzyme. Among other metal ions, $Zn^{2+}$ appears to be a specific reversible inhibitor inducing conformational change through the intersubunit interaction. These results indicate that $Zn^{2+}$ is an allosteric competitive inhibitor and substrate being a non-cooperative per se, excludes the $Zn^{2+}$ from its binding site and thus exhibits positive cooperativity. The allosteric mechanism of AlaDh from Bacillus subtilis is consistent with both MWC and Koshland's allosteric model.

  • The effects of light quality and irradiance on the growth of Cochlodinium polykrikoides were investigated in the laboratory. At 25°C and 30 psu the irradiance-growth curve was described as μ = 0.34 (I-9.76)/(I+12.5), (r=0.98). This suggests half-saturation photon flux density (PFD) (Ks) of 32.0 μmol photons m–2 s–1, and a compensation PFD (Ic) of 9.76 μmol photons m–2 s–1. Because the Ic equates to a depth of ca. 15.4 m, these responses suggest that irradiance at the depth around and below the thermocline in Yeosuhae Bay would provide favorable conditions for C. polykrikoides. Photoinhibition did not occur at 300 μmol photons m–2 s–1, which was the maximum irradiance used in this study. Blue (450 nm), yellow (590 nm) and red (650 nm) light had different effects on the growth of C. polykrikoides: it grew well under blue light, but not under yellow light. This implies that C. polykrikoides is more likely to cause an outbreak of red tide in the open sea where blue-green wavelengths predominate, rather than in enclosed water bodies where suspended particles absorb most of the blue wavelengths, and yellow-orange wavelengths predominate.

Supralinearity of UV Irradiated Magnesium Aluminum Spinel (자외선 조사된 Magnesium Aluminum Spinel의 Supralinearity)

  • Three-dimensional thermoluminescence (TL) spectra from $MgAl_2O_4$ irradiated with UV light were measured over 300${\~}$600 K and 300${\~}$800 nm. The peak positions of TL glow curves were shifted to lower temperature with increasing the exposure time of UV light. The 476 K TL glow curve is due to the second kinetics and its activation energy and escape frequency factor are calculated to be 0.85 eV and $1.92{\times}10^6 sec^{-1}$, respectively The TL spectra were split into 530 nm and 700 nm emission bands which were associated with $V^{2+}$ and $Cr^{2+}$, respectively. The linearity range of 700 nm omission band is smaller than that of 530 nm emission band, but the saturation time of 700 nm emission band is longer than that of 530 nm omission band.

Estimation of WEPP's Parameters in Burnt Mountains (산불지역의 WEPP 매개변수 추정)

  • Fire-enhanced soil hydrophobicity often increases runoff and erosion in the mountain hillslope following severe wildfires. Estimation techniques for WEPP's parameters were studied in burnt mountain slopes. In burnt mountain slopes, the model over-predicted runoff in the small runoff and under-predicted runoff in the great runoff, and in the lower sediment runoff it had a tendency to over-predict soil loss. The effective hydraulic conductivity was most sensitive in the WEPP's runoff and its sediment runoff was mainly effected by the effective hydraulic conductivity, initial saturation, rill erodibility, and interrill erodibility. To improve the applicability of the WEPP, the adjustment coefficient of effective hydraulic conductivity was defined for runoff and the adjustment coefficient of rill erodibility and interrill erodibility was presented for sediment runoff. The adjustment coefficient of effective hydraulic conductivity in wildfire mountain slopes increased with maximum rainfall intensity of single storm and the vegetation height index. The adjustment coefficients of rill erodibility depended on soil components of size distribution curve and total rainfall depths in single storm. The adjustment coefficients of interrill erodibility decreased with increases of maximum rainfall intensity and vegetation height index. These results may be used in the application of WEPP model for wildfire mountain slopes.
