The Aurora Kinase Inhibitor CYC116 Promotes the Maturation of Cardiomyocytes Derived from Human Pluripotent Stem Cells

  • Human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) have great potential in applications such as regenerative medicine, cardiac disease modeling, and in vitro drug evaluation. However, hPSC-CMs are immature, which limits their applications. During development, the maturation of CMs is accompanied by a decline in their proliferative capacity. This phenomenon suggests that regulating the cell cycle may facilitate the maturation of hPSC-CMs. Aurora kinases are essential kinases that regulate the cell cycle, the role of which is not well studied in hPSC-CM maturation. Here, we demonstrate that CYC116, an inhibitor of Aurora kinases, significantly promotes the maturation of CMs derived from both human embryonic stem cells (H1 and H9) and iPSCs (induced PSCs) (UC013), resulting in increased expression of genes related to cardiomyocyte function, better organization of the sarcomere, increased sarcomere length, increased number of mitochondria, and enhanced physiological function of the cells. In addition, a number of other Aurora kinase inhibitors have also been found to promote the maturation of hPSC-CMs. Our data suggest that blocking aurora kinase activity and regulating cell cycle progression may promote the maturation of hPSC-CMs.

Prediction of tenderness in bovine longissimus thoracis et lumborum muscles using Raman spectroscopy

  • Objective: This study was conducted to evaluate Raman spectroscopy technique as a noninvasive tool to predict meat quality traits on Braford longissimus thoracis et lumborum muscle. Methods: Thirty samples of muscle from Braford steers were analyzed by classical meat quality techniques and by Raman spectroscopy with 785 nm laser excitation. Water holding capacity (WHC), intramuscular fat content (IMF), cooking loss (CL), and texture profile analysis recording hardness, cohesiveness, and chewiness were determined, along with fiber diameter and sarcomere length by scanning electron microscopy. Warner-Bratzler shear force (WBSF) analysis was used to differentiate tender and tough meat groups. Results: Higher values of cohesiveness and CL, together with lower values of WHC, IMF, and shorter sarcomere were obtained for tender meat samples than for the tougher ones. Raman spectra analysis allows tender and tough sample differentiation. The correlation between the quality attributes predicted by Raman and the physical measurements resulted in values of R2 = 0.69 for hardness and 0,58 for WBSF. Pearson's correlation coefficient of hardness (r = 0.84) and WBSF (r = 0.79) parameters with the phenylalanine Raman signal at 1,003 cm-1, suggests that the content of this amino acid could explain the differences between samples. Conclusion: Raman spectroscopy with 785 nm laser excitation is a suitable and accurate technique to identify beef with different quality attributes.

  • 근원섬유단백질의 생화학적 성질에 대하여서는 아직도 불명한 점이 많고, 특히 식품으로써의 근원섬유단백질의 저장 중의 변화에 대하여서는 규명되어야 할 점이 너무나 많다. 본 연구는 근원섬유단백질의 저장 중의 변화를 추구하기 위한 기초작업으로서 subcelular structure인 근원섬유를 재료로 하여 그 형태학적 측면과 생화학적 측면의 상관성을 비교 검토하였다. 근원섬유의 조제방법에 따라서 근원섬유의 근절(筋節)(sarcomere)의 길이는 변화하고 있었고 근절의 길이의 변화는 생화학적 성질. 즉 ATPase활성에 현저한 변화를 일으켰다. 근원섬유를 저농도의 trypsin으로 처리하면 근원섬유의 ATPase활성은 현저히 증가하나, 근원섬유의 위상차(位相差)현미경 상은 수축상태의 상을 나타내었다가 처리시간의 연장에 따라 sarcomere가 fragmentation을 나타냄을 보여주었다. 얻어진 결과로 근원섬유 중에는 ATPase활성을 저해하는 factor가 존재하며, 이 factor는 단백질의 inhibitory action은 아니고 steric effect임을 추정하였다. 또한 Z-line의 구성물질 중에 troponin의 관여가 추정되었다.

Effects of Sous-vide Cooking Temperature on Triceps Brachii of Black Goats

  • The aim of this study was to determine the effects of sous-vide cooking temperature on the triceps brachii of black goats. Triceps brachii of black goats (12 months) were sous-vide cooked at 55℃, 60℃, and 65℃. The samples were examined for color, scanning electron microscope photographs, sarcomere length, fiber cross-sectional area, cooking yield, shear force, sensory evaluation, and aromatic profile. The results showed that CIE a*, CIE b*, and chroma increased with increasing sous-vide cooking temperature. However, the cooking yield significantly decreased with increasing sous-vide cooking temperature, and the shear forces of the 60℃ and 65℃ samples showed no significant differences. For sensory evaluation, the 60℃ sample showed the highest scores for flavor, texture, and off-flavor. Furthermore, the 60℃ sample showed the significantly lowest value of octadienone (aroma characteristics of metallic) intensity (p<0.05). Therefore, sous-vide cooking of triceps brachii of black goats at 60℃ is effective in reducing off-flavor and improving tenderness.

Carcass characteristics and meat quality of lambs that are fed diets with palm kernel cake

  • Objective: The aim was to evaluate carcass characteristics, cut yield, and meat quality in lambs that were fed different inclusion levels of palm kernel cake. Methods: Forty-five woolless castrated male Santa $In{\hat{e}}s$ crossbred sheep with an initial average body weight of $23.16{\pm}0.35kg$ were used. The experimental design was a completely randomized design with five treatments, with palm kernel cake in the proportions of 0.0%, 7.5%, 15.0%, 22.5%, and 30.0% with nine replications per treatment. After slaughter, the gastrointestinal tract was weighed when it was full, after which it was then emptied. The heart, liver, kidney, pancreas perirenal fat were also collected and weighed. The carcass was split into two identical longitudinal halves and weighed to determine the quantitative and qualitative characteristics. Results: The empty body weight, carcass weight and yield, and fat thickness decreased linearly (p<0.05) as a function of palm kernel inclusion in the diet. There was no difference (p>0.05) for the rib eye area of animals that were fed palm kernel cake. There was a reduction in the commercial cut weight (p<0.05), except for the neck weight. The weights of the heart, liver, kidney fat, small, and large intestine, and gastrointestinal tract decreased. Nevertheless, the gastrointestinal content was greater for animals that were fed increasing levels of cake. For the other organs and viscera, differences were not verified (p>0.05). The sarcomere length decreased linearly (p<0.05), although an effect of the inclusion of palm kernel cake was not observed in other meat quality variables. It is worth noting that the red staining intensity, indicated as A, had a tendency to decrease (p = 0.050). Conclusion: The inclusion of palm kernel cake up to 30% in the diet does not lead to changes in meat quality characteristics, except for sarcomere length. Nevertheless, carcass quantitative characteristics decrease with the use of palm kernel cake.

Combined Effects of High Pressure and Heat on Shear Value and Histological Characteristics of Bovine Skeletal Muscle

  • Changes in shear force value, transverse sections, myofibrils and intramuscular connective tissue of bovine skeletal muscle exposed to the combination of high-pressure up to 400 MPa and heat (30 and $60^{\circ}C$) were studied. The shear force value decreased by pressure-heat treatment up to 200 MPa at 30 and $60^{\circ}C$, and then slightly increased over 200 MPa at $30^{\circ}C$. Shear force values of treated muscles were lower than those of untreated ones. Gaps between muscle fibers in the untreated muscle were a little clear, and then they became very clear in the treated muscles up to 200 MPa at 30 and $60^{\circ}C$. However, the gaps reduced significantly over 200 MPa at $30^{\circ}C$. The remarkable rupture of I-band and loss of M-line materials progressed in the myofibrils with increasing pressure applied. However, degradation and loss of the Z-line in myofibrils observed in the muscle treated at $60^{\circ}C$ was not apparent in the muscle treated at $30^{\circ}C$. The length of the sarcomere initially contracted by pressure-heat treatment of 100 MPa at $30^{\circ}C$ seemed to have recovered with increase of the pressure up to 400 MPa. In the muscle treated at $60^{\circ}C$, the length of sarcomere gradually decreased with increase of the pressure up to 400 MPa. In the treated muscles, changes in the honeycomb-like structure of endomysium were observed and accelerated with increase of the pressure. A wavy appearance clearly observed at the inside surface of endomysium in the untreated muscles gradually decreased in the treated muscles with increase of the pressure. Tearing of the membrane was observed in the muscles treated over 150 MPa at $30^{\circ}C$, as observed in the sample pressurized at 100 MPa at $60^{\circ}C$. The roughening, disruption and fraying of the membrane were observed over 200 MPa at $60^{\circ}C$. From the results obtained, the combination of high-pressure and heat treatments seems to be effective to tenderize tough meat. The shear force value may have some relationship with deformation of intramuscular connective tissue and myofibrils.

사후 저장온도 $15^{\circ}C$에서 한우적색근육의 생화학, 물리적 변화에 관한 연구 (Postmortem Biochemical and Physical Changes in the Red Musculature of Korean Native Beef Cattle Stored at $15^{\circ}C$)

  • 본 연구는 사후 저장온도 $15^{\circ}C$가 한우근육(M. sternomandibularis)의 생화학, 물리학적 변화에 미치는 영향을 조사하였다. 도살 1.5시간 후의 pH는 6.7/6.8이었으며, 저장온도 $15^{\circ}C$에서 $18{\sim}20$시간 후 최종 pH인 5.5에 도달했다. 한우육의 불가역적인 신전성의 상실 즉, 사후강직은 $15^{\circ}C$에서 도살 $9{\sim}10$시간 후 pH 6.1{\sim}6.0$, R-value $1.05{\sim}1.10$에서 시작하여 도살 $18{\sim}20$시간, pH 5.9에서 강직이 완료되어 신전성을 완전히 상실하였다. 저장온도 $15^{\circ}C$에서 한우근육은 약 20%, 근절은 15% 정도 수축하였다. 한우육은 도살 약 1일 후 전단력이 4.7 kg으로 가장 높았으며, 9일 숙성 후 3.0 kg으로 숙성기간이 경과함에 따라 연도가 증가하였다.

사료의 영양수준에 따른 재래흑돼지의 산육능력 및 육질비교 (Effect of Nutritional Levels on the Growth and Meat Quality of Korean Native Black Pigs)

  • 본 연구는 재래흑돼지의 육질특성과 생산성 구명을 통해 소비 및 생산 활성화를 위한 기초자료로 활용할 목적으로 사료내 에너지(ME, kcal/kg)사이신(%)수준을 달리한 3처리구, 즉 상(H), 중(M), 하(L)로 나누어 사양시험을 한 후 성장 및 육질평가를 실시하였다. 본 연구결과, 사양시험기간 동안 재래흑돼지에 있어 에너지아이신 수준이 높을수록 일당증체량과 사료요구율이 향상되는 경향을 보이고 출하일령이 짧아지는 경향을 보였다(p<0.05). 도체성적의 경우 도체율, 등지방두께, 배최장근단면적, 살코기 생산량에서 처리구간에 유의적 차이를 보이지 않았으나(p>0.05), 거세돈이 암퇘지에 비해 등지방이 두꺼운 것으로 나타났다(p<0.05). 재래흑돼지 등심육의 일반조성분, 근내지방함량, 근절길이 및 근섬유 비율은 처리구별로 유의적 차이는 없었으나, 근내지방은 영양수준이 높은 H 처리구가 M, L 처리구보다 포화지방산은 높고, 불포화지방산은 낮게 나타났으며, n-3비율은 에너지/라이신 수준이 높아질수록 낮았으나, n-6/n-3 비율은 영양수준이 높아질수록 낮아지는 경향을 보였다. 근절길이 및 근섬유 비율도 유의적인 차이는 없었으나 영양수준이 증가됨에 따라 근절길이는 다소 증가하는 경향을 보였고, 적색근 비율은 L처리구가, 백색근 비율은 M 처리구가 각각 다른 처리구보다 높았다.

Chemical Components and Meat Quality Traits Related to Palatability of Ten Primal Cuts from Hanwoo Carcasses

  • To determine chemical components and meat quality traits related to palatability of 10 primal cuts, 25 Hanwoo carcasses were selected from 5 carcasses × 5 quality grades and used to obtain proximate data and meat quality characteristics. Significant differences (p<0.05) in chemical component and meat quality were found among the 10 primal cuts. The highest fat content was found in the kalbi, followed by dungsim, yangjee, chaekeut, ansim, abdari, suldo, moksim, udun, and satae. Protein and moisture contents in the 10 primal cuts were in reverse order of fat content. Moksim had the highest drip loss % and cooking loss % than all other primal cuts while kalbi showed the lowest (p<0.05) percentage of drip and cooking loss. Ansim had the longest sarcomere length but the lowest shear force values than all other cuts (p<0.05). The highest (p<0.05) score for overall acceptability was observed in ansim. Moksim, udun, abdari, and satae were rated the lowest (p<0.05) in overall acceptability among the 10 primal cuts from Hanwoo carcasses. In conclusion, ansim, dungsim, chaekeut, and kalbi had the highest overall acceptability due to their higher fat contents and lower shear force values.

Evaluation of Beef Carcass and Palatability Traits and Prediction of Tenderness in A Cross of Bos Indicus × Bos Taurus Cattle

  • Steers and heifers (N=490) were produced between 1991 and 1996 by reciprocal fiillsib backcross and $F_1$ crosses from Angus and Brahman to compare characteristics of carcass and palatability traits between Bos indicus and Bos taurus inheritance. Carcasses of 3/4Angus were heavier, fatter (p<0.05), more tender and higher in other palatability attributes (p<0.01) than those of 3/4Brahman. Reciprocal effects of parental cross breeds were found on some traits. Within 3/4Brahman inheritance group, Brahman sired progeny produced heavier and fatter carcasses with better palatability (p<0.05) than progeny with Brahman as a dam breed. Estimates of heritability were intermediate to high in most carcass and palatability traits. Genetic correlations of tenderness with marbling score (MARB), sarcomere length (SARC), fragmentation index (FRAG) and calpastatin activity (CALP) were moderate to high, suggesting potential use of the tenderness-influencing factors as indirect selection criteria to improve palatability attributes. MARB and SARC that were best predictors of tenderness explained 3.07 to 5.85% and 4.32 to 8.24% of variation in tenderness, respectively. However, there was no tenderness-influencing factor to dominantly explain large portion of variation in tenderness.
