Micro-Determination of D-Amino Acids in Food by Using Achirai/Chiral Coupled Column Method (Achiral/Chiral Coupled Column법에 의한 식품 중의 D-아미노산의 정량분석)

  • Detectable levels of several free D-amino acids were found in some food. This was accomplished by using a column switching method. The determination of total amount of D- and L-amino acids was based on an achiral separation with a $C_{18}$ column. The level of D-amino acids to L-amino acids was determined by the column switching system including the postcolumn reaction detection of the amino acids derivatized with O-phthalaldehyde. The chiral separation of the postcolumn detection system was carried out with chiral crown ether column. This system was applied for the micro-determination of D-amino acids in food such as soy sauce, fermented soy bean and beans. It turned out that the sampling process is critical for the trace analysis of D-amino acids under this achiral / chiral coupled-column system. It was found that commercial soy sauce contained 42ppm, conventional soy sauce 102ppm, fermented soy bean 8.34mg per 1g and bean 2.87mg per 1g sample for phenylalanine. D-phenylalanine was found 0.67% in commercial soy sauce, 0.34% in conventional soy sauce, less than 1.81% in fermented soy bean, and Jess than 2.82% in bean.

  • Volatile organic compounds (VOCs) are an important public health issue in Korea and many important questions remain to be addressed with respect to assessing exposure to these compounds. Because they are ubiquitous and highly volatile, special techniques must be applied in their analytic determination Valid Personal exposure assessment methods are needed to evaluate exposure frequency, duration and intensity, as well as their relationship to personal exposure characteristics. Biological monitoring is also important since it may contribute significantly in risk assessment by allowing the estimation of effective absorbed doses. This study was on ducted to establish the environmental measurement, personal dosimetry and biological monitoring methods for VOCs. These methods are needed to compare blood, urinary and exhalation breath VOC levels and to provide tools for risk assessment of VOC exposure. Passive monitors (badge type) and a active samplers (trap) for the VOCs collection were used for air sampling. Methods development included determining the minimum detectable amounts of VOCs in each media, as well as evaluating collection methods and developing analytical procedures. Method reliability was assessed by determining breakthrough volumes and comparing results between laboratories and with other methods. A total capacity of trap used in this study was 60ι. Although variable by compound, the average breakthrough was 20%. Also, there was no loss of compounds in trap even if keep for 45 day in -7$0^{\circ}C$. The recovery of active and passive methods was 69% ~ 126% and method detection limit was 0.24 $\mu\textrm{g}$/trap and 0.07 $\mu\textrm{g}$/badge. There was no statistical difference (P > 0.05) between active and passive methods.

  • Micropore filtration of dilute red blood cell (RBC) suspensions is a widely known method for determining red blood cell deformability. Use of this method for cells from various laboratory animal species does require considering the effects of the cell size to pore size ratio and of suspension hematocrit. In general, previous animal studies have utilized 5% hematocrit suspensions and five micron pores, and thus conditions similar to human clinical laboratory practice. However, when used for repeated sampling from small laboratory animals or for parallel multiple samples from different sites in large laboratory animals, the volume of blood sampled and hence the hematocrit of the test suspension may be limited. Our results indicate that hematocrit levels yielding stable values of RBC pore transit time are pore size and species specific: three micron pores = $2{\sim}5%$ for dog and $3{\sim}5%$ for rat; five micron pores $3{\sim}5%$ for dog and $1{\sim}5%$ for rat. An analytical approach using a common expression for calculating transit time is useful for determining the sensitivity of this time to hematocrit alterations and hence to indicate hematocrit levels that may be problematic.

A Study on Evaluation for the Han River Water Quality Index (한강의 수질지수 산정에 관한 연구)

  • The theory and practice of water quality scoring and indexing are introduced. The monthly water analysis data are available for six stations long the down-stream Han River whthin the areal boundary of the Special City of Seoul. The data cover the period between 1975 and 1979 inclusive and contain the analytical findings on 37 water constituents including DO, BOD, temperature, total solids and etc. Sic parameters are selected form the 37 items, that, to the judgement of the writer, best reflect the water quality of the Han River. They are; dissolved oxggen saturation, pH, fecal coliform, total solids, BOD and nitrate+ammonia. For each of the six parameters, a subscore function is developed and graphically presented to facilitate the transform of a measurment of the arameter to a subscore on a common score(e.G. 0-100) The score of a sample is calculated as a fuction of the six subscores, using four different approaches; (1) the unweighted arithmetic water quality score, (2) the weighted arithmetic water quality score, (3)the unweighted multiplicative score and (4) the reduced (total) score. Independent of these calculated scores, the experts' score which is calculated by averaging the ratings of water quality experts is obtained and compared with each of the four calculated scores by means of the least square method. The experts' score compares most favorably with the "reduced" score with the correlation coefficient of 0.956 : therefore this method of water quality scoring is adopted to calculate the Han River water quality scores and indices. Water quality index data for Guiri, ukdo, Pokwangdong, Noryangjin, Yongdungpo and Kayang Stations, 1975-1979 are as follow: The overall water quality index data of the Han River between Guiri and Kayang Stations are found; 47.3 in 1976, 48.0 in 1977, 48.5 in 1978 and 54.7 in 1979, indicating the general trend towards water quality improvent in this part of the river, in terms of the increased water quality index by average 1.85 points per year during this period. Finally the optimum sampling frequencies distributed among the six stations, using an equation which takes into account the coefficients of variation of the water quality scores and indices arec calculated.alculated.

Distributional Characteristics of Volatile Organic Compounds in the Indoor Air of Various Office Environments (다양한 사무실 실내환경에서의 휘발성유기화합물의 농도분포 특성)

  • This study was carried out to evaluate the concentration variations of volatile organic compounds (VOCs) in the office environments located in a large urban area with respect to seasonality, smoking status, types of ventilation and heating. Indoor air sampling was undertaken in 37 and 30 offices in Daegu city during summer and winter, respectively. The VOC samples were collected using adsorbent tubes, and were determined by thermal desorption coupled with GC/MS analysis. The analytical method was validated for repeatability, method detection limits (MDL), and duplication precision. A total of 34 VOCs of environmental concern were determined, including 15 aromatics and 19 halogenated hydrocarbons. Average concentrations of BTEX appeared to 1.91 ppb, 22.98 ppb, 3.44 ppb, and 3.70 ppb, respectively. These values were relatively higher levels than those measured at homes and outdoor roadsides reported by other researches. In general, the concentrations of VOCs were higher in winter than summer, in smoking offices than non-smoking offices, in forced ventilation type than natural ventilation type, and in combustion heating than non-combustion heating offices. However, such differences were not always significant at a level of 0.05 by statistical tests (t-test and/or Mann-Whitney test) with some exceptions for BTEX and styrene. This study demonstrated that smoking status, ventilation type and presence of combustion sources indoors could be important factors on the elevated concentrations of some VOCs in the office environment.

The Study on the Comparative Analysis of the Aquaculture Production Efficiency Regarding Methods and Species (양식업의 양식방법별 어종별 생산효율성 비교분석에 관한 연구)

  • The purpose of this study is to investigate the production efficiencies of the Korean aquaculture fishery with respect to species and methods using a Data Envelopment Analysis. The study extracted the 8 fishes in each of the sea cage culture, aquarium basin, and enclosed aquaculture for the analytical purposes. First, the study estimated the technical, pure technical, and scale efficiencies of the total of 24 aquaculture fishes based on the traditional DEA under the assumptions of both CRS and VRS. 2 fishes were identified as the efficient DMUs under the CCR-model, and 6 fishes under the BCC-model. Second, we tested to see if there was any difference in production efficiencies regarding those three different methods of aquaculture. we could not find any evidence of the differences in efficiency using a rank sum test based on the traditional DEA. However, we could do find that the pure technical efficiency in the sea cage culture was lower than others at 1% level of significance and the pure technical efficiency in enclosed aquaculture was also lower than others at 5% level of significance using Bilateral-DEA, which could explicitly consider the heterogeneity in the 3 production methods of aquaculture. Finally, the study obtained the 95% confidence intervals of the efficiency scores for the 24 fishes under our study using the smoothed bootstraping method in the process of the re-sampling in cooperation with both a kernel density estimation and a reflection method. At the same time, we could estimate the bias-corrected efficiency scores while the traditionally estimated efficiency scores suffered from the biases in the process of solving a linear programming with the deterministic nature of a production frontier. And hence, we could distinguish the differences in production efficiencies of the 8 fishes with respect to those 3 methods of aquaculture.

Concentration Characteristics of Airborne Hexavalent Chromium in the Industrial Area (산업단지 대기 중 6가 크롬 농도 특성에 관한 연구)

  • This paper describes the field evaluation of a sampling and analytical method for the quantity of airborne hexavalent chromium ($Cr^{6+}$) in the industrial areas, such as Sihwa and Banwol. Ambient measurements were performed by using the cellulose filter during the four seasons (October 2006 to June 2007). The determination of hexavalent chromium was carried out by ion chromatography. Performance validations, including method detection limit, relative standard deviation, and recovery percent, were also investigated. The mean concentrations of $Cr^{6+}$ in Sihwa and Banwol were 0.767 and 0.796 $ng/m^3$, respectively, which are similar to those of other foreign industrial areas. The seasonal variations of $Cr^{6+}$ levels were not quite different, which implies that the chromium was continuously emitted from the industrial sources throughout the year. The concentration variations between total chromium and $Cr^{6+}$ have also shown the similar pattern, suggesting that these components originate from the same sources. The concentration of $Cr^{6+}$ was found to be 0.7 to 2.4% of the total chromium. From these results, the approach using the cellulose filter may be suitable to collect $Cr^{6+}$ in the ambient air.

Development of a New Analytical Instrument with Glow Discharge System and Studies on the Direct Solid Analysis Method (글로우방전을 이용한 새로운 정밀분석기기의 개발 및 금속시료의 직접분석법에 관한 연구)

  • A new sputtering chamber, as Atomsource, for atomic absorption measurements was designed and built to have maximum ground state atoms in view and minimum emission. The high velocity gas jets not only change the visual and electrical characteristics of the discharge, but also increase the amount of material atomized, as is apparent from the craters that are produced in the sample where the jets strike the surface. The use of gas jets in a sputtering cell makes this a direct sampling method practical for atomic absorption spectrometry. The factors influencing absorption sensitivity and reproducibility have been investigated for the newly developed atomizer.

Determination of Phthalates Compounds in the Ambient Atmosphere (I) - Evaluation of a Measurement Method and its Application to a Field Study - (환경대기 중 프탈레이트 화합물의 농도 측정 (I) - 측정방법 평가와 현장 적용 -)

  • Phthalate compounds are widely used as plasticizers in polyvinyl chlororide (PVC) resins and other industrial consumer products, and some of them are known to be endocrine disruptors. In Korea, a number of studies have been carried out for the measurement of phthalates in consumer products and drinking water. However, no data are available for those compounds in the ambient air where the general public are routinely exposed. In this study, we evaluated sampling and analytical methods for the determination of phthalates in the ambient atmosphere. A wide range of phthalates compounds were included in the target analytes, which are dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), di(2-ethylhexyl) phthalate (DEHP), and di-n-octyl phthalate (DOP). Most of samples were collected using a high volume sampler with a PUF/XAD-2 column/quartz fiber filter and then analyzed by GC/MS. Some of samples were simultaneously collected on XAD-2 using a low-volume sampler, together with high-volume samples. The analytical method applied in this study showed good repeatability and linearity. Quantitative detection limits were estimated from 0.60 to 17.84 ng/$m^3$ in air, depending on individual compounds. The field measurements were carried out at 3 sites located in Sihwa- Banwall industrial areas and a suburban area from January 2007 to November 2007. From the field experiments, DEHP, DMP and DBP appeared to be the most abundant compounds in the ambient air. It was also found that DMP, DEP and DBP were mainly distributed in the vapor phase, while BBP, DEHP and DOP were predominantly associated with the particulate phase. The concentrations of DEHP and DMP in the industrial areas ranged from 45.7 to 1,012.7 ng/$m^3$ and from 7.7 to 375.1 ng/$m^3$, respectively. Overall, the high-volume sampling method was demonstrated to be superior to the low-volume method for the determination of phthalates in the ambient atmosphere.

Free-Form Surface Reconstruction Method from Second-Derivative Data (형상이차미분을 이용한 자유곡면 형상복원법)

  • We present an algorithm for surface reconstruction from the second-derivative data for free-form aspherics, which uses a subaperture scanning system that measures the local surface profile and determines the three second-derivative values at those local sampling points across the free-form surface. The three second-derivative data were integrated to get a map of x- and y-slopes, which went through a second Southwell integration step to reconstruct the surface profile. A synthetic free-form surface 200 mm in diameter was simulated. The simulation results show that the reconstruction error is 19 nm RMS residual difference. Finally, the sensitivity to noise is diagnosed for second-derivative Gaussian random noise with a signal to noise ratio (SNR) of 16, the simulation results proving that the suggested method is robust to noise.
