Quantitative Analysis of Cinnamic Acid and Cinnamic Aldehyde in Traditionally Preparations by High Performance Liquid Chromatography (고속액체(高速液體)크로마토그라피에 의한 생약제제중(生藥製劑中)의 계피산(桂皮酸) 및 계피(桂皮)알데히드의 정량(定量))

  • The decoctions and extracts were prepared on the traditionally prescribed four kinds (Kejigamcho-tang, Kejibanha-tang, Keji-tang, Kejiinsam-tang) of preparations containing Cinnamoni Raulus. The contents of cinnamic acid and cinnamic aldehyde in these preparations were determined by high performance liquid chromatography using reverse phase partition column, and 12% MeOH as eluting solvent. The sample was determined using the two wave length, 254nm and 280nm. This method was sucessfully applied to the analysis of cinnamic acid and cinnamic aldehyde of various preparations containing Cinnamons.

  • Postabsorptive serum iron level was determined after oral administration of the compounds to human. In serum and whole blood, $Fe^{3+}$ was measured by ion chromatography (IC) using a pyridine-2,6-dicarboxylic acid (PDCA) as an eluent. The serum sample solutions were pretreated with I N HCI and 50% TCA. The whole blood sample solutions were treated with 3 N HCI for 30 min at $125^{\circ}C$. The limit of detection (LOD) of the IC technique is $0.2 {\mu}M$ for$Fe^{2+}$and 0.1 $\mu$M for $Fe^{3+}$. The area under concentration (AUC) can be obtained by the above analytical condition. In addition, to compare the stability of $Fe^{2+}$ to that of $Fe^{3+}$ in pharamaceutical preparations, accelerated stability test was carried out. After storing the samples under $40^{\circ}C$, 75%RH in light-resistant container for various time intervals, the contents of iron of different valencies were determined separately by the IC technique and the change and/or the interchange of among those iron species in preparations was investigated. Iron raw materials are stable, but $Fe^{2+}$ in$Fe^{3+}$ source materials was slightly converted to $Fe^{3+}$ by oxidation. $Fe^{2+}$ in$Fe^{3+}$ source raw materials and $Fe^{3+}$ in $Fe^{2+}$ raw materials are determined as impurities. Therefore, IC technique is found to be an appropriate method for comparative evaluation of dissimilar bioavailability of $Fe^{2+}$ and $Fe^{3+}$, stability of $Fe^{2+}$ and $Fe^{3+}$ raw materials and preparations.

  • A new application of high pressure liquid chromatography for the determination of fenitrothion and cis- and trans-neopynamin in insecticidal preparations was investigated. Optimum conditions for a good separation and determination were determined; solvent system: dichloromethane + n-hexane = 17 + 83; Bow rate: 0.5ml/min; column: u-porasil ($4mm{\times}3Ocm$); absorbance wavelength: 254nm; 0.05 AUFS and sample size: 30 ul. Recovery of fenicrothion, cis- and trans-neopynamin from mixed artificial preparations was 99.6%, 99.7% and 99.8% respectively. Also reproducibility tests showed that the coefficient of variation was 0.89% for fenitrothion, 0.74% cis-neopynamin and 1.1% for trans-neopynamin. There was no interference with insecticidal preparation containing DDVP, allethrin, S-421 and kerosene. HPLC method was rapid, accurate and it gave better reproducibility and higher sensitivity than any other analytical method. It was considered that HPLC could be greatly applied to the analysis of fenitrothion and neopynamin in insecticidal preparations.

Application of near-infrared spectroscopy for hay evaluation at different degrees of sample preparation

  • Objective: A study was conducted to quantify the performance differences of the near-infrared spectroscopy (NIRS) calibration models developed with different degrees of hay sample preparations. Methods: A total of 227 imported alfalfa (Medicago sativa L.) and another 360 imported timothy (Phleum pratense L.) hay samples were used to develop calibration models for nutrient value parameters such as moisture, neutral detergent fiber, acid detergent fiber, crude protein, and in vitro dry matter digestibility. Spectral data of hay samples prepared by milling into 1-mm particle size or unground were separately regressed against the wet chemistry results of the abovementioned parameters. Results: The performance of the developed NIRS calibration models was evaluated based on R2, standard error, and ratio percentage deviation (RPD). The models developed with ground hay were more robust and accurate than those with unground hay based on calibration model performance indexes such as R2 (coefficient of determination), standard error, and RPD. Although the R2 of calibration models was mainly greater than 0.90 across the feed value indexes, the R2 of cross-validations was much lower. The R2 of cross-validation varies depending on feed value indexes, which ranged from 0.61 to 0.81 in alfalfa, and from 0.62 to 0.95 in timothy. Estimation of feed values in imported hay can be achievable by the calibrated NIRS. However, the NIRS calibration models must be improved by including a broader range of imported hay samples in the modeling. Conclusion: Although the analysis accuracy of NIRS was substantially higher when calibration models were developed with ground samples, less sample preparation will be more advantageous for achieving rapid delivery of hay sample analysis results. Therefore, further research warrants investigating the level of sample preparations compromising analysis accuracy by NIRS.

Comparison of Cellular Features Diagnostic of Papillary Thyroid Carcinoma in Liquid-Based (Cell Scan 1500TM) Preparations and Conventional Smears

  • The study compared the cytological features of papillary thyroid carcinoma (PTC) in liquid-based preparations (LBPs) and conventional Pap (CP) smears from fine needle aspiration (FNA), and assessed the feasibility of LBP using the Cell Scan $1500^{TM}$ processor on thyroid FNA samples. Thyroid FNA samples were obtained from 883 consecutive patients. Each sample was divided into two and used for LBPs and CP smears. All were screened independently in a double-blind manner. From the 883 cases, 95 cases were diagnosed as PTC in one or both types of preparation (10.8%). PTC was diagnosed via CP smears in 83 cases (87.4%) and via LBPs in 70 cases (73.7%). However, there were differences in categorization between the paired preparations: Twelve (12) PTCs were misinterpreted in CP smears and 25 PTCs in LBPs. There was a significant discrepancy in the rate of detection of the diagnostic features, with LBPs having a lower detection rate. One (1) case (1.2%) of CP smears and 16 cases (22.9%) of LBPs were categorized as unsatisfactory/nondiagnostic in a total of the 95 PTCs. To conclude, the detection rate of the diagnostic features of PTC is lower in Cell Scan 1500TM samples than in CP smears. However, there are some cases in which a diagnosis of PTC is made in LBPs, but not in CP smears. Therefore, definitive cancer diagnosis in thyroid FNA preparations is likely to result from agreement between direct smears and Cell Scan 1500TM preparations.

Survey on the Korean Medicine Doctor's Awareness of the Extension and Activation of Herbal Medicine Preparations Insurance Benefits (한약제제 보험급여 확대 및 활성화 방안에 대한 한의사 인식도 조사)

  • Objectives : The aim of this study was to investigate the Korean medicine doctor's perception of the extension and activation of herbal medicine preparations insurance benefits. Methods : The sample of Korean medicine doctors' panel pool which is extracted by examining the square root quota of city location of Korean medicine clinics was used. In case of a vacancy in some participants, the number of Korean medicine doctors were supplemented and placed for investigation. Results : It has 54.0% awareness of the separation of prescribing and dispensing herbal preparations, 87.0% agreed on the need to be insured, but the separation as a necessary condition for being insured was only 60.3% agreed. When a large number of herbal preparations are being insured, it was expected that 30% to 50% of patients(30.3%) will be prescribed herbal preparations and the number of new patients will increase by 5 to 15 for the purpose of prescriptions only(49.3%). Increase amount of medical expense(?3,000) as a result of the separation of prescribing and dispensing herbal preparations are insufficient(54.3%). Conclusions : From the results of this study, extension of national health insurance to cover herbal preparations must be needed to activate herbal medicine preparations and improve Korean medical services.

Determination of Selenium in Dried Yeast Preparations (셀레늄 함유 건조효모제제 중 셀레늄 분석방법에 관한 연구)

  • In order to improve the sensitivity of the current assay methods of selenium in dried-yeast preparations, atomic absorption spectrophotometry (AAS), high performance liquid chromatography (HPLC) and UV-Vis spectrophotometry were employed. The sample was prepared with the digestion by acid mixture of hydrochloric acid, nitric acid and perchloric acid after elimination of ether-soluble substances. The range of quantitation of selenium was $1.0{\sim}6.0\;{\mu}g/ml$ by UV-Vis spectrophotometry, $5.0{\sim}20.0\;{\mu}g/ml$ by HPLC and $0.03{\sim}0.10\;{\mu}g/ml$ by AAS.

Quality Characteristics of Sulgiddeok with Different Commercial Rice Flours (시판 쌀가루를 이용한 설기떡의 품질 특성)

  • The study assessed the quality characteristics of Sulgiddeok with self-made and commercially-available rice flours during a 1-day storage at ambient temperature($20\sim22^{\circ}C$). The nutritional components, color value, physical tests, internal structure as revealed by scanning electron microscopy SEM, and sensory evaluation of Sulgiddeok were determined. Sulgiddeok prepared with self-made rice flour and two commercial rice flour were designated as sample A, B, and C, respectively. Moisture content, crude protein, crude lipid, and pH did not differ significantly among the preparations. Sweetness was highest in sample B(p<0.01) and crude ash was highest in sample A(p<0.001). L color, a value, and b value was highest in sample B, C, and C, respectively. Texture property analysis showed that hardness, gumminess, and chewiness tended to decrease during the 1-day storage, while adhesiveness, springiness, and cohesiveness tended to increase. Sensory characteristics of color, flavor, chewiness, gumminess, externals, and overall quality of sample A were superior to samples B and C. However, softness was highest in sample B. The internal structure of Sulgiddeok was preserved in sample A. It is concluded that rice flour B can make Sulgiddeok comparable to that prepared using self-made rice flour.

  • The advantage of internet survey compared to the traditional survey methods are speedy in data collection, cost-effective, high performed design and able to data process and analysis at the same time. The other side are difficult to select sample, come from serious nonresponse errors. We suggest the new internet survey method to the questionnaire design that have the high response rate, enough to advanced preparations and system stability.
