Stem cell attached 3-dimentional printed polycarprolactone scaffold (줄기세포 탑재 3차원 프린팅 polycarprolactone 스캐폴드)

  • Stem cell therapy is not expected to bestow any therapeutic benefit because of the low engraftment rates after transplantation.Various cell-carrying scaffolds have been developed in order to overcome this problem. When the scaffold is formed by 3-dimensional (3D) printing, it is possible to create various shapes of scaffolds for specific regions of injury. At the same time, scaffolds provide stem cells as therapeutic-agents and mechanically support an injured region. PCL is not only cost effective, but it is also a widely used material for 3D printing. Therefore, rapid and economical technology development can be achieved when PCL is printed and used as a cell carrier. Yet PCL materials do not perform well as cell carriers, and only a few cells survive on the PCL surface. In this study, we tried to determine the conditions that maximize the cell-loading capacity on the PCL surface to overcome this issue. By applying a plasma treated condition and then collagen coating known to improve the cell loading capacity, it was confirmed that the 3% collagen coating after plasma treatment showed the best cell engraftment capacity during 72 hours after cell loading. By applying the spheroid cell culture method and scaffold structure change, which can affect the cell loading ability, the spheroid cell culture methods vastly improved cell engraftment, and the scaffold structure did not affect the cell engraftment properties. We will conduct further experiments using PCL material as a cell carrier and as based the excellent results of this study.

Numerical Simulation of Nonlinear Interaction between Composite Breakwater and Seabed under Irregular Wave Action by olaFlow Model (olaFlow 모델에 의한 불규칙파 작용하 혼성방파제-해저지반의 비선형상호작용에 관한 수치시뮬레이션)

  • For the design of composite breakwater as representative one of the coastal and harbor structures, it has been widely discussed by the researchers about the relation between the behavior of excess-pore-water pressure inside the rubble mound and seabed caused by the wave load and its structural failure. Recently, the researchers have tried to verify its relation through the numerical simulation technique. The above researches through numerical simulation have been mostly applied by the linear and nonlinear analytic methods, but there have been no researches through the numerical simulation by the strongly nonlinear mutiphase flow analytical method considering wave-breaking phenomena by VOF method and turbulence model by LES method yet. In the preceding research of this study, olaFlow model based on the mutiphase flow analytical method was applied to the nonlinear interaction analysis of regular wave-composite breakwater-seabed. Also, the same numerical techniques as preceding research are utilized for the analysis of irregular wave-composite breakwater-seabed in this study. Through this paper, it is investigated about the horizontal wave pressures, the time variations of excess-pore-water pressure and their frequency spectra, mean flow velocities, mean vorticities, mean turbulent kinetic energies and etc. around the caisson, rubble mound of the composite breakwater and seabed according to the changes of significant wave height and period. From these results, it was found that maximum nondimensional excess-pore water pressure, mean turbulent kinetic energy and mean vorticity come to be large equally on the horizontal plane in front of rubble mound, circulation of inflow around still water level and outflow around seabed is formed in front of rubble caisson.

Garden Construction and Landscape Characteristics of the Seochulji Pond Area in Gyeongju during the Middle of the Joseon Dynasty (조선 중기 경주 서출지(書出池) 일원의 정원 조영과 경관 특성)

  • This study examined the background of Gyeongju Seochulji Pond (world heritage, historic site No. 138), a historic pond in Sam-guk-yu-sa (三國遺事), and its landscaping period when it served as the garden of the Pungcheon Lim clan (豊川 任氏) in the middle of the Joseon dynasty. For this study, a literature review of poetry, prose, and a personal anthology, and a field survey were conducted. Changes in the landscape were analyzed by comparing the landscape appearing in the literature of the Joseon period with past photographs. The results were as follows: First, even though the function and landscape at that time cannot be guessed as the objective ground from Silla to the early part of the Joseon dynasty is insufficient, it has been managed as a Byeolseo (別墅) garden as Pungcheon Lim's family resided in the area of Eastern-Namsan Mountain during the Joseon dynasty. At that time, Seochulji Pond was recognized as a historic place. It functioned as the garden of Pungcheon Lim's family as Lim Jeok (任勣, 1612~1672) built the Yiyodang pavilion (二樂堂). Second, in the literature, the Yiyodang pavilion has been called Gaekdang (客堂), Jeongsa (精舍), Byeolgak (別閣) and Byeolseo, etc. It can be seen as Nu and Jeong (樓亭), utilized for various uses. Because of this, the name Bingheoru Pavilion (憑虛樓) has mostly been in common use. Third, Seochulji Pond was positioned where the scenery is beautiful, with Gyeongju Mt. Namsan (Mt. Geumo) in the background and with a wide field and the Namcheon River flowing in the front. This was typical of Byeolseo gardens of the Joseon dynasty, combining human environments with natural environments. Fourth, the relationship with the Byeolseo garden disappeared as the head of Pungcheon Lim's family added a temple, lotus flowers, pine trees, and a bamboo forest as described in the old poetry and prose. Currently, the landscape does not appear to be significantly different from that as development has not occurred in the area of Seochulji Pond. Also, crape myrtle (Lagerstroemia indica), which now symbolizes the Seochulji Pond, was not identified in the old poetry or past photographs and is not old enough to confirm whether it was prominent at the time. Through this study, it is necessary to reconsider the spatial meanings of the gardens of the Joseon dynasty period and not to highlight the area of Seochulji Pond as a place in the legend. This is a cultural asset in the area of Eastern-Namsan Mountain and has an important meaning in terms of garden history.

Effect of the Sowing and Harvesting Dates on the Agronomic Characteristics and Feed Value of Corn and Sorghum×Sorghum Hybrid in Youngnam Mountain Area (영남산간지역에서 옥수수와 수수×수수 교잡종의 파종시기와 수확시기가 조사료 생육 특성과 사료가치에 미치는 영향)

  • This study was carried out from 2015 to 2016 to identify the suitable sowing and harvesting dates of summer crops in the mountain of Yeongnam, South Korea. The experimental design consisted of the different sowing and harvesting dates as follows: corn hybrid (Z. mays, Kwangpyeongok) of sowing (May 8, 19, and 27) and harvesting (August 10, 20, and 30); sorghum${\times}$sorghum hybrid (Sorghum bicolor ${\times}$ Sorghum bicolor, SS405) of sowing (May 27, June 19 and June 27) and harvesting (August 10, 20, and 30). In corn hybrid, ear rate and dry matter (DM) yield decreased (p<0.05) with the postponement of sowing date. Otherwise, ear rate and DM yield increased (p<0.05) with the postponement of harvesting date. Crude protein content decreased (p<0.05) with the postponement of sowing date, but neutral detergent fiber content increased (p<0.05). In sorghum${\times}$sorghum hybrid, plant length and DM yield with the postponement of harvesting date increased (p<0.05), while crude protein content with the postponement of harvesting date decreased (p<0.05). This study concluded that sowing corn hybrid in early May and sorghum${\times}$sorghum hybrid in early June then harvest in the middle of August was recommend to increase dry matter yield and feed value.

Experiments on Flow Characteristics of Asphalt Seal Composite Waterproofing Method for Underground Concrete Structure Exterior Wall Waterproofing (지하 콘크리트 구조물 외벽 방수용 아스팔트 씰재 복합방수 공법의 흘러내림 특성에 관한 실험)

  • With the changing trend of the building construction to high rising and large scaling, the underground structure has been increased, and its usage also increased and variety. Hence, to protect the underground structure against underground water, various water proofing methods has been developed. Among the many water proofing methods, the combined water proofing method using both asphalt seal and sheet has been widely used to secure the sufficient performance and decrease the construction failure. However, during the summer period of extremely high temperature conditions, the asphalt sealing materials were separated and leaked into the structure. Therefore, the aim of the research is to provide the quality standard of asphalt sealing material based on the various temperature changes depending on seasons. According to the experimental results, the temperature of the sealing materials applied on the slab was increased up to $54^{\circ}C$ which was $3^{\circ}C$ higher than the structure temperature of $51^{\circ}C$. Based on the melting test for asphalt sealing materials applied on the outside wall of the structure, in the case of water-dispersing typed materials showed significant melting down due to its slow evaporation and low viscosity. Furthermore, from the accelerated test conducted indoor conditions, a solvent-type and water-dispersing typed materials showed significant melting down due to their low viscosity and melting point in ambient conditions. Based on these results, viscosity and melting point are found as the important factors on asphalt sealing materials' quality, and it is necessary to designate the quantitative level of the viscosity and melting point for quality control.

The Current Status and Prospect of Presidential Records Management (대통령기록관리의 현황과 전망)

  • Legislation and enforcement of the Presidential Records Management Law was an important turning point in Korean archival management history. In the past, the notion of presidential records was vague. The law was a starting point of establishing presidential records management. The Presidential Records Management Law provides the definition of presidential records and its scope, and establishes the protection of presidential records through restricted access to the records. The key to the law is to enable a president freely to produce records and transfer them to the next administration without omission. In other words, it aims to stop the practice that presidential records are produced but never be left. But, 'disputes over the release of presidential records' and the disclosing of access-restricted presidential records presented a crisis to national records management as well as the prospect of presidential records management, even if they were 'legal procedures.' The instability of presidential records management could give a serious impact on the national records management and its operation. Amid this situation, it is required to review the presidential records management system and provide recommendations for improvement, even if the enforcement of law has just started. The most urgent things in improving presidential records management are to secure its independence, specialty, and to complement restricted access to presidential records. For securing independency, presidential records management should be done by a separate organization other than the National Archives of Korea while for promoting specialty, a newly established organization could serve as a professional archive. And for complementing restricted access to the presidential records, the access should be more limited. In other words, more discretion is needed in permitting access. And more specific regulations should be applied to the permitted records. However, these regulatory actions may not have effects unless independency is not secured. Thus, more fundamentally, independency of the National Archives of Korea should be first established.

Osteoblastogenic Activity of Locusta migratoria Ethanol Extracts on Pre-Osteoblastic MG-63 Cells (풀무치 에탄올 추출물이 MG-63 조골세포 분화에 미치는 영향)

  • Insects have been investigated as a novel source of food and biomaterial in several recent studies. However, their osteoblastogenic cell activity has not been sufficiently researched and so, to investigate the potential of this natural material for promoting osteoblastogenesis, we studied the activity of Locusta migratoria ethanol extract (LME) on MG-63 pre-osteoblast cells. The cytotoxicity and proliferation effects of LME on MG-63 cells were measured by MTS assay, and there was no cytotoxicity up to $1,000{\mu}g/ml$. With LME treatment of 500 and $1,000{\mu}g/ml$ for 48 hr, cell proliferation increased to 105% and 116% versus control, respectively. The osteoblastogenic activity of the LME was measured through alkaline phosphatase (ALP) staining at three and five days. As a result, both 500 and $1,000{\mu}g/ml$ LME concentrations were seen to increase ALP activity by more than three times compared with control at three and five days. In addition, the expression level of the osteogenic markers ALP and RUNX2 was markedly increased after LME treatment. These results demonstrate that Locusta migratoria ethanol extract promotes osteoblastogenesis as evidenced by the increased osteogenic markers and suggest that LME may be a potential agent for bone formation and osteoporosis prevention.

Site Competition of Ca2+ and Cs+ Ions in the Framework of Zeolite Y (Si/Al = 1.56) and Their Crystallographic Studies (제올라이트 Y (Si/Al = 1.56) 골격 내의 Ca2+과 Cs+ 이온의 자리 경쟁 및 그들의 결정학적 연구)

  • The present work was performed in order to study the effect of competing cation of $Ca^{2+}$ ion on ion exchange of $Cs^+$ on zeolite Y (Si/Al = 1.56). Three single-crystals of fully dehydrated and partially $Cs^+$-exchanged zeolites Y (Si/Al = 1.56) were prepared by the flow method using mixed ion-exchange solutions. The $CsNO_3:Ca(NO_3)_2$ molar ratios of the ion exchange solution were 1 : 1 (crystal 1), 1 : 100 (crystal 2), and 1 : 250 (crystal 3) with a total concentration of 0.05 M. The single-crystals were then vacuum dehydrated at 723 K and $1{\times}10^{-4}Pa$ for 2 days. The structures of the crystals were determined by single-crystal synchrotron X-ray diffraction technique in the cubic space group $Fd{\bar{3}}m$, at 100(1) K. The unit-cell formulas of crystals 1, 2, and 3 were ${\mid}Cs_{21}Ca_{27}{\mid}[Si_{117}Al_{75}O_{384}]-FAU$, ${\mid}Cs_2Ca_{36.5}{\mid}[Si_{117}Al_{75}O_{384}]-FAU$, and ${\mid}Cs_1Ca_{37}{\mid}[Si_{117}Al_{75}O_{384}]-FAU$, respectively. In all three crystals, the $Ca^{2+}$ ions preferred to occupy site I in the D6Rs, with the remainder occupying sites I', II', and II. On the other hand, the significant differences in the fractional distribution of $Cs^+$ ions are observed depending on the intial $Cs^+$ concentrations in given ion exchange solution. In Crystal 1, $Cs^+$ ion are located at sites II', II, III, and III', and in crystal 2, at sites II, IIIa, and IIIb. In crystal 3, $Cs^+$ ions are only located at sites IIIa and IIIb. The degree of $Cs^+$ ion exchange decreased sharply from 28.0 to 2.7 to 1.3 % as the initial $Ca^{2+}$ concentration increases and the $Cs^+$ content decreases.

The Study on Formation of Deoneum of 19th Century's Sugungga (19세기 수궁가의 더늠 형성에 관한 연구)

  • This study examined the process of the formation of Deoneum, Sugungga(水宮歌), and its tendency of the change through the trace of the Pansori masters who worked in 19th century. The time that the main group of the Sugungga masters appeared in the Pansory history is estimated in early 19th century. The Sugungga masters in this time could be divided in two groups, one is the early days masters who worked in early 19th century and the other is the latter days masters who worked in middle and late 19th century. Kwon Sam-deuk(權三得), Song Heung-lok(宋興祿), Yeom Gyeo-dal(廉季達), and Shin Man-yeob(申萬葉) are the Sugungga masters who worked in early 19th century. By the records they left, I could confirm that they used the description about the dramatic characteristics in the work and Soritjo(Pansori master's tonality), like the appearance of 'Bangge(crab)' and 'Tiger', and particularly they developed which has the scene of the Rabbit's deceiving the Dragon King and Rabbit's return, 'Goandaejangja(寬大長者)', 'Gaja-Eoseoga', 'Sojinowha(笑指蘆花)', 'Apnae-Beodeuleun' and the scene of 'Rabbit's curse(the part that Rabbit curses Byeljubu)' etc. I could understand that the interest of the early days Pansori masters about Sugungga is on the characters and the latter part of the work. The separation of Dongpyeonje(東便制) and Seopyeonje(西便制) of Pansori was done in the middle and late 19th century. As the Dongpyeonje master, Song Wu-ryong(宋雨龍), Park Man-sun(朴萬順), Song Man-gab(宋萬甲), Shin Hak-jun(申鶴俊) and Yu Seong-jun(劉成俊) worked. As the Seopyeonje masters, Park Yu-jeon(朴裕全), Kim geo-bok(金巨福), Kim Su-yeong(金壽永) and Baek Gyeong-jun(白慶順) etc. sang Sugungga. The Dongpyeonje masters developed the 'Toggigibyeon(토끼奇變)' related Deoneum paying attention on the latter part of Sugungga same as the early days masters. Meanwhile it looks like that they had interest in developing the parts which belong to the middle and early parts of Sugungga like 'Toggiwhasang(토끼畵像)', 'Gogocheonbyeon(皐皐天邊)' and 'Tobyeolmundab(兎鱉問答)'. The Seopyeonje masters developed the parts belong to the early part of Sugungga, which are related to Dragon King and the courtiers in Sugung palace, like 'YongwangTansik(龍王歎息)'. And I could confirm that they developed the parts influenced by Shin Jaehyo's editorials like 'Tosahobi(兎死狐悲)' and 'Goguksancheon(故國山川)' etc. In short, I could confirm the trend that the Pansori masters in 19th century had interest in from the latter part to the early part of Sugungga. If taking a look focusing on main characters, they moved their interest in from Rabbit to the courtiers including Byeoljubu(鱉主簿) and Dragon King in Sugung palace, and it could say that, in this trend, Sori part and Deoneum were developed.

Antioxidant, Anti-wrinkle and Antimicrobial Effects of Coffee Grounds Extract from Dutch Coffee (더치 Coffee Grounds 추출물의 항산화, 주름개선, 항균 효과)

  • This study confirmed possibility of cosmetic material for Espresso coffee grounds extracted at high temperature, high pressure, short time and Dutch coffee grounds extracted at low temperature, atmospheric pressure, long time. For this purpose, we evaluated the biological activities of antioxidant, anti-wrinkles and antimicrobial effects using ethanol extracts of Esproso and Dutch coffee grounds. The results of total polyphenolic compound contents was $90.39{\pm}0.04mg/g$ for Dutch coffee grounds extract, which was higher than $64.96{\pm}0.38mg/g$ for Espresso coffee grounds extract, based on $113.63{\pm}0.22mg/g$ for coffee beans extract as the reference one. DPPH radical scavenging activity and SOD-like activity of Dutch coffee grounds extract were found to be better than those of Espresso coffee grounds extracts, referenced on coffee bean extract. As a result of inhibition effect of Elastase activity, Dutch coffee grounds extract showed higher inhibition effect than Espresso coffee grounds extract, based on coffee bean extract. In addition, Dutch coffee grounds extract showed good anti-microbial effects at Escherichia coli, Bacillus, Propionibacterium acnes and there was little difference in the clear zone size between Dutch coffee grounds extract and coffee bean extract as a reference one. From the results of the experiments, it was confirmed that Dutch coffee grounds extract had excellent antioxidant, anti-wrinkle and antimicrobial effects and could be used as safe natural cosmetic material in the future.
