Avicelase, CMCase and salicinase of A.phoenicis K.U. 175 were purified from wheat bran culture by salting out with ammonium sulfate, dialysis and successive column chromatography Sephadex G-100. Optimum pH and temperature of avicelase were pH 3.8-4.8, $35-55^{\circ}C$ and that of CMCase, salicinase were pH4.5-5.5, $45-60^{\circ}C$ and pH 4.5-6.0, $45-60^{\circ}C$ respectively. These enzymes were relatively thermostable, alkali unstable and inhibited by $Ca^{++},\;Mn^{++},\;Cu^{++},\;and\;Hg^{++}$. Km values of avicelase, CMCase and salicinase were calculated to be $1.5{\times}10^{-4}M,\;5.5{\times}10^{-4}M\;and\;2.75{\times}10^{-5}M$ and Vmax values $1.66{\times}10^{-4}mM/min,\;3.33{\times}10^{-4}mM/min\;and\;1.14{\times}10^{-4}mM/min$, respectively.
The HACCP (hazard analysis critical control point) concept becomes an important aspect of Good Manufacturing Practices (GMP) in safe food production. The HACCP plan was developed with food safety objective. The authors already carried out hazards analysis on Changran Jeotgal manufacturing process in a previous report. In this study we developed a HACCP plan on the manufacturing process of Changran Jeotgal. We could decide two CCPs those were salt concentration and eliminations of foreign materials before packaging. Foreign materials should be certainly checked out with laser detector just before packaging. Salt should be maintained more than $8\%$ to inhibit pathogenic bacteria in end product. On the Jeotgal process free water is removed after salting and Jeotgal is seasoned with several subsidiary materials such as com syrup. In result end products have $8\%$ salt with water activity 0.82. Most bacteria are inhibited but some can live in this condition. Materials on the working may be contaminate by pathogens, chemicals or physical hazards. It will be decreased by SSOP (standard sanitary operating program). The SSOP needed in Jeotgal plants is nearly same as other general food manufacturing plants but essential to fulfill HACCP program.
This study was conducted to figure out the nutritional nature of seafoods categorized according to the medicinal effects and the usages introduced in Lim-Won-Sib-Rheuk.-Ji, one of the well-known encyclopedias published during the Chosun Dynasty. They were compared with those introduced in other cooking books in order to find out how they were used in the diet of real-life. The seafoods of the book were first categorized into the fish 20, the mollusk 5, the crustacea 4, and the shellfish 7, and reptile according to the biological taxonomy, and then each one was reviewed in terms of the taste and the food-nature. Generally, the fishes had the sweet taste; the crustacea and the shellfish the salty or sour in addition to the sweet taste; none of them were bitter or hot. Most of them had the mild nature; seven of them were regarded as warm; four as cool; one as cold from the standpoint of the traditional medicine. The food-nature of seafoods was closely related with the medicinal effects on the human body. These seafoods were characteristically used in daily dietary life as well as the medical applications; used as the side-dishes rather than the main-dishes. The cooking methods of boiling, steaming, and baking were preferred to drying and stewing. The preservation methods favored were the fermentation, drying and salting. These cooking and preservation methods were believed to reflect the nature of the seafoods to manifest the medicinal effects.

Laboratory experiments were conducted to study the effects of pH and salt level on the soil sorption equilibrium of pentachlorophenol (PCP) which is hydrophobic and ionogenic. Experimental results indicated that the sorption equilibrium constant (Kp) of PCP increased with decreasing pH. A quantitative sorption model involving linear isotherms was estabilished to predict the pH effect on the PCP sorption equilibrium over the pH range from 3 to 8. The model prediction was in good agreement with the experimental data. Also, the Kp increased with salt concentration over the entire pH range. At added salt levels less than 0.1M, increase in Kp was larger than when the added levels were higher than 0.1M. Salt might increase the PCP sorption by inducing 'salting out-effect' or by forming deprotonated PCP-cation ion pairs such as PCP$\^$-/K$\^$+/. Taking the pH range (5-8) and the salt content (up to 50 g/L) in the groundwater of Metropolitan landfill sites into consideration, the results indicated that the retardation factor of PCP in this area might range from 3 to 550 depending upon pH and salt content.
Salting-out technique was adopted to crystallize dye crystals from dye solution. In this research solubility of dye solution and crystallization kinetics of Reactive dye (RB-8, RB-49, RR-218) was investigated. The empirical expressions of salting-out crystallization kinetics for Reactive dye (RB-8, RB-49, RR-218) in continuous MSMPR crystallizer was RB-8 in crystal growth kinetics $G=7.1{\times}10^{-4}{\Delta}C^{0.67}$ and nucleation kinetics $B^0=3.1{\times}10^{15}{\Delta}C[1.2{\times}10^{-8}+{\Delta}C^{0.7}M_T{^2}]$, RB-49 in crystal growth kinetics $G=5.2{\times}10^{-4}{\Delta}C^{0.3441}$ and nucleation kinetics $B^0=7.2{\times}10^{15}{\Delta}C[3.3{\times}10^{-8}+({\Delta}C)^{0.7}M_T{^2}]$, RR-218 in crystal growth kinetics $G=4.4{\times}10^{-4}{\Delta}C^{0.2361}$ and nucleation kinetics $B^0=6.3{\times}10^{15}{\Delta}C[7.9{\times}10^{-8}+({\Delta}C)^{0.7}M_T{^2}]$. Also, comparison of calculated crystal size distribution applying to characteristic curve method with experimental crystal size showed good agreement.
1,3-Propandiol is a promising chemical which can be produced from fermentation of glycerol because the application of 1,3-propanediol is mainly in the production of bio-polytrimethylene terephthalate (bio-PTT). However, the cost of downstream processes in the biological production of 1,3-propanediol can make a high portion in the total production cost due to the large amount of water and the by-produced carboxylic acids such as succinic, lactic and acetic acids in 1,3-propanediol fermentation broth. In this study, aqueous two-phases systems composed of hydrophilic alcohols and phosphate salts were applied to the recovery of 1,3-propanediol from its artificial aqueous solution. Formation of aqueous biphases in hydrophilic alcohols and phosphate salts was due to the salting-out effect of salts in bottom phase, thereby 1,3-propanediol in bottom phase was moved into top phase. Extraction efficiency for 1,3-propanediol was proportional to the polarity of hydrophilic alcohols and the basicity of salts and the maximum value of extraction efficiency was more than 98%. In the aqueous two-phases systems after extraction, there was no carboxylic acid in top phase. Therefore, it was concluded that the aqueous two-phases systems composed of hydrophilic alcohols and phosphate salts were effective for the selective recovery of 1,3-propanediol without any coextraction of carboxylic acids.
The analytical method of extracting compounds from human blood to examine accumulated organochlorine pesticides (OCPs) has been widely used the traditional liquid-liquid extraction (LLE) method and solid-phase extraction (SPE) method, yet these methods have certain limitations in purification and usafe of a large amount of sample. In order to overcome the se problems reside in these, solid-phase microextraction (SPME), known as a highly efficient extration method with less samples and relatively simple, was employed to collect 18 different kinds of OCPs in blood as extraction method in this study. To optimize extraction method, we examine various experimental SPME-parameters such as adsorption (fiber type, adsorption time, adsorption temperature, salting out effect), and desorption (desorption time, desorption temperature etc.). From the experimental results, the optimal conditions are as follows: fiber was polyacrylate with $85{\mu}m$, adsorption time was for 5 min, adsorption optimum temperature was at $280^{\circ}C$, and salting out effect was NaCl with 0.1 g. MDL, precision and accuracy was in the ranges of 0.05~0.20 ng/mL, 5.59~13.39%, respedively, and accuracy was -0.5% ~24.5% for all OCPs.

We used polymers of alternating cationic and anionic nature to build up shells on fiber surfaces, strengthen the worn-out fibers and improve paper properties made from such fibers. OCC and ONP pulps were either dipped or salted out in the cationic polyallylamine, polyacrylamide and starch solutions. After centrifugal drying, these were followed by treatments in anionic polyacrylic acid, poly-acrylamide, and starch solutions, respectively. The shell-enhanced fibers were formed into handsheets and their physical properties evaluated. The results show that building multiple shells on worn-out fiber surfaces can strengthen the fibers and paper. The simpler and more practical impregnation-centrifuging treatment provided the desired effects, whereas salting out the polymers produced somewhat superior fibers. The latter process, were impractical, however. The first pair of polymeric shells imparted marked strength improvement, whereas later layers had diminishing efficacies. Overall, the methods can improve fiber quality, attaining paper strength requirements without resorting to expensive measures. Alternate cationic polymer and filler powders were also deposited on fiber surface based on the micriparticle system in an anticipation of stiffness gains. Platy minerals, such as montmorillonite, bentonite, sericite, clay and talc were added following cationic PAM. After dewatering of polymer-pigment shelled fiber of one to 3 pairs of layers, handsheets either calendered or uncalendered were evaluated. The results indicate that regardless of calendaring, stiffness of the handsheets did not improve appreciably while certain other strength properties showed gains. We also attempted the novel starch gel filler addition method wherein tapioca starch and filers (PCC, sericite or clay) were mixed at high solids content of 50% and cooked until gelatinized. The filled handsheets were dried under various conditions and then tested for their properties. Improvements in strengths of modified filled paper were observed.

An extracellular acid phytase from Pseudomonas fragi Y9451 was purified to homogeneity from the culture supernatant by salting-out, DEAE-Sepharose column chromatography, CM-Sepharose column chromatography, and Sephacryl S-300 gel filtration. The molecular weight of the purified enzyme was estimated to be 74 kDa on gel filtration and 54 kDa and 25 kDa on SDS-PAGE, suggesting that the native enzyme was a heterodimeric protein. The purified enzyme was most active at pH 4.5 and $70^{\circ}C$ and fairly stable from pH 4.0- 6.0. It was specific for phytate and exhibited a $K_{m}$ value of 27 mM (sodium phytate, pH 4.5, $50^{\circ}C$). The phytase activity was strongly inhibited (at maximum by 87%) by $Fe^{3+},\;Cu^{2+},\;Fe^{2+}$, and $Zn^{2+}$ at 5 mM concentration, and greatly inhibited by $Ca^{2+}$ at 10 mM concentration. However, EDTA notably stimulated the phytase activity at 10 mM concentration. With optimum pH and stability, Pseudomonas fragi phytase could be a potential candidate for animal feed applications.
An extracellular chitinase of the selected strong antifungal microorganism, Serratia sp. 3095, was purified by salting out, affinity adsorption, Sepadex G-100 gel fitration, Sepadex G-75 gel fitration and DEAE Sepadex A-50 chromatography. The molecular weight of the purified chitinase was estimated to be 62,000 dalton by SDS-PAGE. Optimal pH and temperature of the chitinase were pH 7.5 and 45, respectively. The enzyme retained more than 80% of the activity between pH 5.5 and pH 10.5, and below $50^{\circ}C$ but was unstable above $60^{\circ}C$, below pH 5.0. The activity of the chitinase was inhibited about 60% by $Sn^{2+}$, 40% by $Hg^{2+}$ and $Ag^+$, 70% by AHA, 40% by iodoacetate, 35% by thiourea and p-CMB, but stabilized by SDS. $K_m$ value of the purified chitinase was 3.68 mg/ml for colloidal chitin. The chitinase from Serratia sp. 3095 showed antifungal activity to Fusariurm solani.
