Isolation, characterization and expression of transcription factor ScDREB2 from wild, commercial and interspecific hybrid sugarcane in salinity condition

  • Dehydration Responsive Element Binding (DREB) gene is one of the essential transcription factors plants use for responding to stress conditions including salinity, drought, and cold stress. The purpose of this study was to isolate the full length and characterize the DREB gene from three different genotypes of sugarcane, wild, commercial cultivar, and interspecific hybrid sugarcane. The length of the gene, designated ScDREB was 789 bp, and coding for a putative polypeptide of 262 amino acid residues. Sequences of the gene were submitted to the GenBank database with accession numbers of KX280722.1, KX280721.1, and KX280719.1 for wild sugarcane, commercial cultivar (KPS94-13), and interspecific hybrid (Biotec2), respectively. In silico characterization indicated that the deduced polypeptide contains a putative nuclear localization signal (NLS) sequence, and a conserved AP2/ERF domain of the DREB family, at 82-140 amino residues. Based on multiple sequence alignment, sequences of the gene from the three sugarcane genotypes were classified in the DREB2 group. Gene expression analysis indicated, that ScDREB2 expression pattern in tested sugarcane was up-regulated by salt stress. When the plants were under 100 mM NaCl stress, relative expressions of the gene in leaves was higher than those in roots. In contrast, under 200 mM NaCl stress, relative expressions of the gene in roots was higher than those in leaves. This is the first report on cloning the full length and characterization, of ScDREB2 gene of sugarcane. Results indicate that ScDREB2 is highly responsive to salt stress.

Arabidopsis Transcription Factor ANAC032 Enhances Salinity and Drought Tolerance

  • The plant-specific NAC transcription factors control various biological processes, including plant development and stress responses. We have isolated an ANAC032 gene, one of the NAC transcription factor family, which was highly activated by multi-abiotic stresses, including high salt and drought in Arabidopsis. Here, we generated transgenic plants constitutively expressing ANAC032 and its knockout to identify the functional roles of ANAC032 in Arabidopsis under abiotic stress responses. The ANAC032-overexpressing plants showed enhanced tolerance to salinity and drought stresses. The anac032 knockout mutants were observed no significant changes under the high salt and drought conditions. We also monitored the expression of high salt and drought stress-responsive genes in the ANAC032 transgenic plants and anac032 mutant. The ANAC032 overexpression upregulated the expression of stress-responsive genes, RD29A and ERD10, under the stresses. Thus, our data identify that transcription factor ANAC032 plays as an enhancer for salinity and drought tolerance through the upregulation of stress-responsive genes and provides useful genetic traits for generating multi-abiotic stress-tolerant forage crops.

Expression of a rice DREB1 gene, OsDREB1D, enhances cold and high-salt tolerance in transgenic Arabidopsis

  • OsDREB1D, a special DREB (dehydration responsive element binding protein) homologous gene, whose transcripts cannot be detected in rice (Oryza sativa L), either with or without stress treatments, was amplified from the rice genome DNA. The yeast one-hybrid assay revealed that OsDREB1D was able to form a complex with the dehydration responsive element/C-repeat motif. It can also bind with a sequence of LTRE (low temperature responsive element). To analyze the function of OsDREB1D, the gene was transformed and over-expressed in Arabidopsis thaliana cv. Columbia. Results indicated that the over-expression of OsDREB1D conferred cold and high-salt tolerance in transgenic plants, and that transgenic plants were also insensitive to ABA (abscisic acid). From these data, we deduced that this OsDREB1D gene functions similarly as other DREB transcription factors. The expression of OsDREB1D in rice may be controlled by a special mechanism for the redundancy of function.

A novel WD40 protein, BnSWD1, is involved in salt stress in Brassica napus

  • Genes that are expressed early in specific response to high salinity conditions were isolated from rapeseed plant (Brassica napus L.) using an mRNA differential display method. Five PCR fragments (DD1.5) were isolated that were induced by, but showed different response kinetics to, 200 mM NaCl. Nucleotide sequence analysis and homology search revealed that the deduced amino sequences of three of the five cDNA fragments showed considerable similarity to those of ${\beta}$-mannosidase (DD1), tomato Pti-6 proteins (DD5), and the tobacco harpin-induced protein hin1 (DD4), respectively. In contrast, the remaining clones, DD3 and DD2, did not correspond to any substantial existing annotation. Using the DD3 fragment as a probe, we isolated a full-length cDNA clone from the cDNA library, which we termed BnSWD1 (Brassica napus salt responsive WD40 1). The predicted amino-acid sequence of BnSWD1 contains eight WD40 repeats and is conserved in all eukaryotes. Notably, the BnSWD1 gene is expressed at high levels under salt-stress conditions. Furthermore, we found that BnSWD1 was upregulated after treatment with abscisic acid, salicylic acid, and methyl jasmonate. Our study suggests that BnSWD1, which is a novel WD40 repeat-containing protein, has a function in salt-stress responses in plants, possibly via abscisic acid-dependent and/or -independent signaling pathways.

배추에서 신규 염 저항성 관련 유전자 분리 및 검정 (Isolation and Identification of a New Gene Related to Salt Tolerance in Chinese Cabbage)

  • 본 연구는 배추에서 염 저항성 관련 유전자를 발굴하기 위해 수행되었다. 우선 염처리(250mM NaCl)된 순계배추 'Chiifu'를 이용한 KBGP-24K oligo chip 데이터[BrEMD(B. rapa EST and microarray database)]를 분석하였다. 그 결과, 염처리 시 크게 반응하는 202개의 unigene들을 1차 선발하였고, 이들 중 기능이 정확히 알려지지 않았으나 완전장을 갖추고 있는 1개의 유전자를 최종선발하여 BrSSR(B. rapa salt sensitive resistance)로 명명하였다. BrSSR은 94개의 아미노산으로 번역되는 총 285bp의 오픈리딩프레임을 가지고 있으며, DUF581 도메인을 지니고 있다. 염 저항성을 분석하기 위하여 BrSSR이 과발현된 pSL94 vector를 제작하여 담배에 형질전환시켰다. BrSSR이 과발현된 $T_1$ 세대 담배 형질전환체들은 PCR과 DNA blot 분석에 의해 선발하였다. Quantitative real-time RT PCR 분석 결과, 형질 전환된 담배에서 BrSSR의 발현이 대조군 보다 약 3.8배까지 높게 발현되었다. 이는 RNA blot 분석 결과와도 일치했다. 또한 표현형 분석에서 5일간 250mM NaCl 염 처리 후 BrSSR이 과발현된 형질전환체들이 대조군보다 우수한 염 저항성을 보여 주었다. 위 결과들에 근거하여 염 스트레스 환경 하에서 BrSSR 유전자의 과발현은 식물의 염 저항성을 향상과 매우 밀접한 관계가 있는 것으로 판단된다.

Heterogeneous overexpression of Oryza sativa salt induced RING Finger protein OsSIRF1 positively regulates salt and osmotic stress in transgenic Arabidopsis

  • Plants suffer from various abiotic stresses among them; soil salinity is one of major adverse factor in declining agricultural productivity. So, development of salt stress tolerance crops have potential role to increase crop production. The RING finger proteins are known to play crucial roles in abiotic stress environment to plants. In this study, we identified one Salt-responsive Really${\underline{I}nteresting}$ ${\underline{n}ew}$ ${\underline{g}ene}$ (RING) E3 ubiquitin ligase gene OsSIRF1 from rice root tissues during salt stress and studied its molecular function. Expression of OsSIRF1 was induced under various abiotic stress conditions, including salt, heat, drought, and ABA. Result of an in vitro ubiquitination assay clearly showed that OsSIRF1 Possess an E3 ligase activity. Moreover, OsSIRF1 was found to be localized to the nucleus within the cell. Heterogeneous overexpression of OsSIRF1 in Arabidopsis improved seed germination and increased root length under salt and Manitol stress conditions. Taking together, these results suggested that OsSIRF1 may be associated with plant responses to abiotic stressors and positively regulates salt and osmotic stress environment.

애기장대에서 activation tagging system을 이용한 새로운 고염 스트레스 반응 유전자의 동정 (Identification of Novel Salt Stress-responsive Genes Using the Activation Tagging System in Arabidopsis)

  • 환경 스트레스는 식물의 성장을 저해하며 작물의 생산량을 감소시키는 주요 원인이다. 식물은 다양한 유전자의 발현 변화를 통해 스트레스에 대한 저항성을 나타낸다. 본 연구에서는 activation tagging system을 이용하여 기존에 밝혀지지 않은 새로운 고염 스트레스 반응 유전자들을 분리하였다. 애기장대의 발아 단계에서 고염 스트레스에 저항성을 보이는 9개의 activation tagging 라인을 선별하였다. 그 중 TAIL-PCR 방법을 이용하여 AT7508, AT7512, AT7527, AT7544, AT7548, AT7556의 6개 라인에서 T-DNA가 삽입된 위치를 확인하였으며 각 라인에서 T-DNA가 삽입된 주변 유전자의 발현을 RT-PCR로 분석하였는데 AT7508, AT7512, AT7527, AT7544, AT7556에서 각각 ClpC2/HSP93-III (At3g48870), plant thionin family (At2g20605), anti-muellerian hormone type-2 receptor (At3g50685), vacuolar iron transporter family protein (At4g27870), microtubule-associated protein (At5g16730)이 activation 된 것으로 밝혀졌다. 더불어 AT7548에서는 T-DNA가 삽입된 곳의 양쪽에 위치하는 두 유전자인 Arabinogalactan protein 13 (AGP13) (At4g26320)과 F-box/RNI-like/FBD-like domains-containing protein (At4g26340)이 모두 activation 되었다. Activation 된 7개 유전자는 기존에 고염 스트레스 저항성과 관련된 기능이 알려지지 않은 유전자로 본 연구를 통해 새롭게 고염 스트레스 반응에 대한 기능이 밝혀졌다. 7개의 activation된 유전자 중 ClpC2/HSP93-III, AGP13, F-box/RNI-like/FBD-like domains-containing protein의 3개 유전자는 고염 스트레스에 의해 발현이 증가하였다. 또한 AT7508과 AT7527, AT7544 라인은 발아 단계뿐만 아니라 유식물체 발달 과정에서도 고염 스트레스 저항성을 보여 activation tagging 라인의 선별 결과의 타당성을 뒷받침 하였다. 본 연구의 결과를 통해 activation tagging system이 새로운 스트레스 반응 유전자를 찾아낼 수 있는 유용한 기술임을 확인할 수 있었다.

Transcriptome Analysis in Brassica rapa under the Abiotic Stresses Using Brassica 24K Oligo Microarray

  • Genome wide transcription analysis in response to stresses is essential to provide the basis of effective engineering strategies to improve stress tolerance in crop plants. In order to perform transcriptome analysis in Brassica rapa, we constructed a B. rapa oligo microarray, KBGP-24K, using sequence information from approximately 24,000 unigenes and analyzed cold ($4^{\circ}C$), salt (250 mM NaCl), and drought (air-dry) treated B. rapa plants. Among the B. rapa unigenes represented on the microarray, 417 (1.7%), 202 (0.8%), and 738 (3.1%) were identified as responsive genes that were differently expressed 5-fold or more at least once during a 48-h treatment with cold, salt, and drought, respectively. These results were confirmed by RT-PCR analysis. In the abiotic stress responsive genes identified, we found 56 transcription factor genes and 60 commonly responsive genes. It suggests that various transcriptional regulatory mechanisms and common signaling pathway are working together under the abiotic stresses in B. rapa. In conclusion, our new developed 24K oligo microarray will be a useful tool for transcriptome profiling and this work will provide valuable insight in the response to abiotic stress in B. rapa.

염분 변화에 따른 무지개송어(Oncorhynchus mykiss)의 삼투조절 유전자 발현변화 (Gene Expression Profiles of Rainbow Trout Oncorhynchus mykiss after Salinity Challenge)

  • Euryhaline teleost have extraordinary ability to deal with a wide range of salinity changes. To study the seawater adaptability of rainbow trout Oncorhynchus mykiss (body weight 638±54 g, length 38.6±2 cm) to salinity increase fish were transferred from freshwater to 7, 14, 21, 28 and 32 psu and checked for mortality over 5 days. No mortality was observed in 0-32 psu. In fish transferred to 0-32 psu, blood osmolality was maintained within physiological range. The changes of serum enzyme activities (aspartate transaminase, AST and alanine transaminase, ALT) showed no significant level during experimental period. To explore the underlying molecular physiology of gill and kidney responsible for body fluid regulation, we measured mRNA expression of five genes, Na+/K+/2Cl- cotransporter1 (NKCC1), aquaporin3 (AQP3), cystic fibrosis transmembrane conductance regulator (CFTR), glucocorticoid receptor (GR) and growth hormone receptor (GHR) in response to salt stress. Based on our result, rainbow trout could tolerate gradual transfer up to 32 psu for 5 days without mortality under physiological stress. This study suggests to alleviate osmotic stress to fish, a gradually acclimation to increasing salinity is recommended.
