Isolation and characterization of cholesterol-lowering lactic acid bacteria from kimchi (김치에서 분리된 콜레스테롤 감소능을 가진 젖산세균의 특성)

  • The objective of this study was to isolate and characterize lactic acid bacteria (LAB) exhibiting cholesterol-lowering activity from the Korean traditional fermented food, kimchi. The previously isolated LAB strains were assessed for cholesterol-lowering efficacy in the presence of 0.1% cholesterol. All LAB strains tested in this study were able to assimilate cholesterol at varying levels, ranging from 35.0 to 99.4%. Among them, the Lactobacillus plantarum FMB 31 strain exhibited the highest cholesterol-lowering effect with 99.4% cholesterol removal efficiency. The strain was stable in the presence of acid, bile, and salt stress, and showed high adherence on HT-29 cells, a human colon line. In addition, the LAB strain showed no pathogenic properties such as the production of hemolysin and biogenic amines. Thus, this study suggests that the L. plantarum FMB 31 strain isolated from kimchi can be a potential source of probiotic products with strong cholesterol-lowering effect.

Recovery Process of Vanadium from the Leaching Solution of Salt-Roasted Vanadate Ore (바나듐광 염배소물 수침출 용액으로부터 바나듐 회수공정 고찰)

  • In this study, the effects of solution components were investigated in the recovery of vanadium as ammonium metavanadate from vanadium-ore-salt roasting-water leaching solution. The vanadium-containing solution is strongly alkaline (pH 13), so the pH must be lowered to 9 or less to increase the ammonium metavanadate precipitation efficiency. However, in the process of adjusting the solution pH using sulfuric acid, aluminum ions are co-precipitated, which must be removed first. In this study, aluminum was precipitated in the form of an aluminum-silicate compound using sodium silicate, and the conditions for minimizing vanadium loss in this process were investigated. After aluminum removal, the silicate was precipitated and removed by adjusting the solution pH to 9 or less using sulfuric acid. In this process, the concentration and addition rate of sulfuric acid have a significant influence on the loss of vanadium, and vanadium loss was minimized as much as possible by slowly adding dilute sulfuric acid. Ammonium metavanadate was precipitated using three equivalents of ammonium chloride at room temperature from the aluminum-free, aqueous solution of vanadium following the pH adjustment process. The recovery yield of vanadium in the form of ammonium metavanadate exceeded 81%. After washing the product, vanadium pentoxide with 98.6% purity was obtained following heat treatment at 550 ℃ for 2 hours.

Assessment of 1,4-Dioxane Removal in Polyester Wastewater by Activated Sludge and Its Microbial Property by 16S rDNA (폴리에스테르 중합폐수의 활성슬러지 공정에서의 1,4-다이옥산 제거 및 16S rDNA에 의한 미생물 군집특성 평가)

  • 1,4-Dioxane($C_4H_8O_2$), which is used as a solvent stabilizer, could make harmful effects on ecosystem because of its higher solubility, toxicity and carcinogenic by US EPA. From 2011, its discharge limit to waterbody will be regulated at 5 mg/L by Ministry of Environment Republic of Korea. It was thus to investigate that the currently operating activated sludge in polyester manufacturing processes in Gumi can properly treat it to meet with the regulation standard. For that purpose, the removal rate of 1,4-dioxane and its microbial properties were assessed for a few companies(i.e. K, H and T). Its removal efficiency was the most highly recorded in H as 98% and then 77% for K, which met with the regulation standard. However, concentration of 1,4-dioxane of T was 23 mg/L in the effluent, which is more than the regulation standard. Aside from, microbial degradation test was done for 100 ppm of 1,4-dioxane in BSM (Basal salt medium) inoculated with each of activated sludge. After 7 days, 1,4-dioxane was completely removed in the test bottle inoculated with H sludge, 67% in T and 52% in K, which could confirm that the given activated sludge might have different biodegradability against the amount of 1,4-dioxane. Therefore, microbial diversity in each company was investigated by 16s rDNA cloning methods where a species, e.g. Methylibium petroleiphilum PM1, was the greatest observed from H and in lesser from K, but it was not detected from T. Methylibium petroleiphilum PM1 is known to efficiently degrade ether like methyl tertiary-butyl ether(MTBE). It is concluded that the activated sludge in H can be most effectively adopted for a biodegradation of 1,4-dioxane in the concern of industrial sector.

Effect of Decay Rate Coefficients with Sulfur Denitrification due to Salt in Seafood Wastewater (수산물 가공폐수내 염분이 황 탈질 반응속도 상수에 미치는 영향)

  • This study examined the effects of the salinity of seafood wastewater on the sulfur denitrification process. An examination of the denitrification efficiency showed that the optimal EBCT was 1hr at an influent T-N concentration of 20mg/L or lower and 2-3hr at an T-N concentration of 30mg/L. An examination of the denitrification efficiency according to the nitrogen load showed that the legal effluent water quality criterion was satisfied when the influent load was maintained within $0.496kg/m^3/day$. On the other hand, the reactor volume increased when this was applied to the site. Therefore, the influent load should be within $0.372kg/m^3/day$ considering the denitrification and economic efficiency. At a load of $0.248{\sim}0.628kg/m^3{\cdot}day$, the k value was $0.0890{\sim}0.5032hr^{-1}$. The batch experimental results according to the $Cl^-$ concentration showed that at an influent nitrogen concentration of 30.0mg/L, the effect of the denitrification efficiency was not large below the salinity of $7,000mgCl^-/L$, but inhibition occurred above $9,000mgCl^-/L$. Calculations of the reaction rate constant according to the $Cl^-$ concentration showed that the reaction rate constant was $0.1049{\sim}0.2324hr^{-1}$ at a raw wastewater concentration of ${\sim}5,000mgCl^-/L$. In contrast, the k value was $0.1588hr^{-1}$ at $7,000mgCl^-/L$ and $0.1049hr^{-1}$ at $9,000mgCl^-/L$.

Study on Characteristics of Biogas Production and Liquid Fertilizer with Anaerobic Co digestion of Livestock Manure and Food Waste (혐기성소화에서 가축분뇨와 음식물쓰레기의 혼용에 따른 바이오가스 생산 및 소화액의 액비 특성 연구)

  • Objective of this research was to investigate the characteristics of biogas production in anaerobic digestion reactor with different mixing ratio of food waste and swine manure. It was observed that the highest removal efficiency of organic material was 80% at 60 : 40 of mixing ratio (livestock manure : food waste). And also biogas yield was varied due to different mixing ratio of them. The cumulative biogas yield was highest at 60 : 40 of mixing rate (livestock manure : food waste). For use of the liquefied fertilizer as effluent from anaerobic digester, it was the limited ratio for 30% of co-digested food waste based on its salt content.

Analysis of Soil Changes in Vegetable LID Facilities (식생형 LID 시설의 내부 토양 변화 분석)

  • The LID technique began to be applied in Korea after 2009, and LID facilities are installed and operated for rainwater management in business districts such as the Ministry of Environment, the Ministry of Land, Infrastructure and Transport, and LH Corporation, public institutions, commercial land, housing, parks, and schools. However, looking at domestic cases, the application cases and operation periods are insufficient compared to those outside the country, so appropriate design standards and measures for operation and maintenance are insufficient. In particular, LID facilities constructed using LID techniques need to maintain the environment inside LID facilities because hydrological and environmental effects are expressed by material circulation and energy flow. The LID facility is designed with the treatment capacity planned for the water circulation target, and the proper maintenance, vegetation, and soil conditions are periodically identified, and the efficiency is maintained as much as possible. In other words, the soil created in LID is a very important design element because LID facilities are expected to have effects such as water pollution reduction, flood reduction, water resource acquisition, and temperature reduction while increasing water storage and penetration capacity through water circulation construction. In order to maintain and manage the functions of LID facilities accurately, the current state of the facilities and the cycle of replacement and maintenance should be accurately known through various quantitative data such as soil contamination, snow removal effects, and vegetation criteria. This study was conducted to investigate the current status of LID facilities installed in Korea from 2009 to 2020, and analyze soil changes through the continuity and current status of LID facilities applied over the past 10 years after collecting soil samples from the soil layer. Through analysis of Saturn, organic matter, hardness, water contents, pH, electrical conductivity, and salt, some vegetation-type LID facilities more than 5 to 7 years after construction showed results corresponding to the lower grade of landscape design. Facilities below the lower level can be recognized as a point of time when maintenance is necessary in a state that may cause problems in soil permeability and vegetation growth. Accordingly, it was found that LID facilities should be managed through soil replacement and replacement.
