Difference of Catechins Extracted Level when Fermented Sun-dried Salt and Green Tea (천일염과 녹차를 발효시켰을 때 Catechin류의 추출량 변화)

  • In an experiment in which fermentation was done by adding fungal species that have antibiosis but do not have cellulase, the extraction amount of EGC, EC, EGCG, and ECG increased in all samples that fermented by adding sun-dried salt compared to those that fermented only with green tea after fermenting green tea by mixing it with sun-dried salt. In the analysis conducted according to the days of fermentation, the high extraction amounts of EGC(epigallocatechin), ECG(epicatechin gallate), EC(epicatechin), and EGCG(epigallocatechin gallate) were detected on the second and third day. Furthermore, when fermentation was done by adding ferment bacillus, all types of catechin(EGC, EC, EGCG, ECG) extraction increased in Paenibacillus spp but in Bacillus amyloliquefaciens, EGC and EC decreased while EGCG and ECG increased; whereas in Bacillus pumilus and Bacillus subtilis all types of catechin(EGC, EC, EGCG, ECG) decreased. The results of the above experiment reveal that the largest amount of catechin was extracted from the result which conducted fermentation for three days together with sun-dried salt and Paenibacillus spp in the green tea.

Salting-out Effects on the Partition of Proteins in Aqueous Two-phase Systems

  • The partition of proteins in the salt-rich phase of polyethylene glycol (pEG)/salt aqueous two-phase systems is limited by the salting-out effects of salt. The logarithm of the concentration of proteins partitioned in the salt-rich phase decreases linearly with increases in the concentration of salt in the salt-rich phase (salting-out). Therefore, the partition of a given protein in the salt-rich phase of aqueous two-phase systems can be estimated from the salting-out constant. The slope of the solubility line (salting-out con-stant) for a given protein is determined by the type of salt in the two-phase systems.

  • This study attempts to develop a mass production product standardized by the application of high pressure extraction cooking(HPEC) in order to suggest a desirable direction for the development of salt contained standardized chicken stock. In our experiment on chicken stock with varying its salt content, the total free amino acid content was highest in S3, which contained 0.3% of salt In addition, when the total content of free amino acid was divided into the contents of essential amino acid, palatable amino acid, and other types of amino acid, they showed the same distribution as the total content of free amino acid. In addition, the total content of palatable amino acid was highest among the specimens. In the results of investigating the palatability of chicken stock according to salt content, saltiness increased with the increase of the salt content, but no significant difference was observed in preference for saltiness. It is believed to have come from the difference in the sensory evaluators' preference for saltiness, and it shows that the salt content has an effect on sweet taste, delicate taste, fishy smell, and color. In particular, specimen 83, which contained 0.3% of salt, showed the highest content of palatable amino acid, and the highest level of delicate taste in sensory tests, suggesting the correlation between palatable amino acid and delicate taste.

  • In this study, an efficient ultrasound-assisted liquid-liquid extraction process was developed for recovering of paclitaxel from plant cell cultures. The optimal ultrasonic power and operating time were 250 W and 15 min at fixed ratio of bottom phase, methylene chloride to top phase, MeOH (25%, v/v). Under the optimal conditions developed in the present method, most of the paclitaxel (~92%) was recovered from crude extract by a single extraction step. Due to the synergistic effect of ultrasound by the addition of inorganic salt, an appropriate inorganic salt concentration and the ultrasonic power were found to be required for the effective recovery of paclitaxel using ultrasound-assisted liquid-liquid extraction.

  • Magnesium is widely used as a lightweight alloy for car engine components and case of cellular phone. Extraction technologies of magnesium are divided to fused salt electrolysis process and thermal reduction process. In this study, electrolysis magnesium is prepared by fused salt electrolysis process with magnesium chloride. We compared two kinds of mixed salt at 7V. As a result, 47% of current efficiency was obtained by electrolyzing KCl/NaCl/$MgCl_2$ mixed salt bath at $760^{\circ}C$, and purity of the prepared magnesium was over 98%. With this study, we can scale up fused salt electrolysis device and accumulate basic data which will be needed for designing an electrolysis cell.

  • A solvent extraction system of copper-thiocyanate complex into various types of alkylamines such as secondary, tertiary and quaternary ammonium salt for the determination of trace copper by atomic absorption spectrometry is presented. The maximum extraction of copper shows at 0.1 M-thiocyanate and single extraction with 10 ml of 1% amine-MIBK from 50 ml of aqueous solution is enough to be quantitative for micro amounts of copper. The effect of amine diluents and of diverse ions are also examined.

Extraction Characteristics of Polysaccharide from Fomitopsis pinicola Jeseng Mushroom (소나무 재생버섯(Fomitopsis pinicola Jeseng) 다당류의 추출 특성)

  • This study was conducted to investigate the extraction characteristics of the polysaccharide from Jeseng mushroom (Fomitopsis pinicola Jeseng). Yields of the polysaccharide extracted from powdered mushroom by autoclaving(120, 30 min) with water at different pH and salt concentration were 8.2~9.2% in pH 5~11, 4.7~5.5% in 1~5% salt solution, respectively. The yield by the 0.05~1.0 N KOH-extraction was ranged 3.45~13.20%, while that by HAS-extraction(homogenizing after KOH swelling) using 1~2.5 N KOH 73.6~78.4%. Content of carbohydrate, protein, lipid and ash of the crude polysaccharide extracted from fruits body and its cultured mycelium by method of water extraction, KOH extraction(0.005~1N) and HAS-extraction were ranged 86.5~92.6%, 2.3~13.1%, 0.1~4.2% and 0.1~1.7%, respectively. The polysaccharide were composed of 62.0~77.8 g/g of pentose, 138.0~187.8 g/g of hexose and 21.2~117.3 mg/g of protein. From these results, the polysaccharide extracted was supposed to be a protein-bound polysaccharide.

  • It is to review the current status of magnesium smelting. Raw materials for magnesium source, worldwide production and producers of metallic magnesium, Korean magnesium markets and some important extraction technologies were reviewed. The magnesium extraction technologies were described according to the two major reduction methods: the fused salt electrolysis and the thermal reduction method. Also, the research on the extraction of magnesium from magnesite which has been being carried out at KIGAM was briefly introduced with discussing the related topics on the recycling of the chlorine and the hydrogen chloride gas used in the process.

  • Liquid fueled molten salt reactors (MSRs) have seen renewed interest because of their inherent safety features, higher thermal efficiency and potential for efficient thorium utilisation for power generation. Thorium fluoride is one of the salts used in liquid fueled MSRs employing Th-U cycle. In the present study, ThF4 was prepared by hydro-fluorination of ThO2 using anhydrous HF gas. Process parameters viz. bed depth, hydrofluorination time and hydrofluorination temperature, were optimized for the preparation of ThF4 in a static bed reactor setup. The products were characterized with X-Ray diffraction and experimental conditions for complete conversion to ThF4 were established which also corroborated with the yield values. Hydrofluorination of ThO2 at 450 ℃ for half an hour at a bed depth of 6 mm gave the best result, with a yield of about 99.36% ThF4. No unconverted oxide or any other impurity was observed. Rietveld refinement was performed on the XRD data of this ThF4, and Chi2 value of 3.54 indicated good agreement between observed and calculated profiles.

  • Based on solid phase extraction, gas chromatography and mass spectrometry procedure for determining phenol and its derivatives in natural water was presented. In solid phase extraction, three types of techniques using solid phase adsorption material were treated with acid and salt, and converted second portion of acetyl derivatives. Under the these condition, extraction efficiency and detection ability dependent on extraction methods were discussed. Obtained results using optimized solid phase extraction techniques showed more convenience, simplifier and lower cost than the conventional analytical methods with holding wide dynamic range and lower detection limits.
