  • The agar plate titrations of antibiotic sensitivities of Salmonella and Shigella isolated from human during 1967 to 1972 were studies. 1. The most effective antibiotics against Salmonella and Shigella were chloramphenicol, tetracycline, kanamycin, minomycin, and gentamycin. 2. All strain of Salmonella typhi were resistant to cloxacilline. 3. The most effective antibiotics against Shigella were kanamycin, gentamycin and minomycin.

Drug Resistance Profiles of Salmonella sp. and Shigella sp. Isolated from Diarrheal Patients in Pusan, Korea (부산지역의 설사환자로부터 분리한 Salmonella sp. 및 Shigella sp.의 약제내성 유형에 관한 연구)

  • A total of 34 Salmonella sp. an d25 Shigella sp. were isolated from 311 patients with diarrhea. The isolation rates of Salmonella sp. ad Shigella sp. were 10.9% and 8%, respectively. The serogroups of 34 Salmonella sp. were in order of group D(50%), group B(38.25), group E(8.8%) and group C 92, 9%0. the serogroups of 25 Shigella sp. were group D(96%) and group B(4%). Seasonal distribution of isolated Salmonella sp. and Shigella sp. were shown the most high at July, 17.65% and 64%, respectively. Age group distribution of isolated Salmonella sp. and Shigella sp. were shown the most high at twenties and thirties (23.5%), and teens(76%), respectively. The Salmonella isolates were resistant in order of prevalence use of streptomycin(SM) (100%), erythromycin (EM) and movobiocin (NB)(90.6%), penicillin G(PG) (65.6%) and cephalexin (CPX)(46.9%). the isolates of Shigella sp. were resistant in order of prevalence use of EM (95.8%), NB(91.7%), SM(87.5%). Eighteen kinds of resistant patterns of Salmonella ioslates were detected. The multiple resistance patterns of Shigella isolates were mostly SM, EM, NB type (79.2%). The minimum inhibitory concentration of Salmonella sp. and Shigella sp. and Shigella sp. isolated from patients with diarrhea were tabulated.

  • The clinical specimens used in this study were collected during the period from March 4, to December 30, 1983, from children's hospitals in Seoul area. They came from clinically apparent cases of diarrheal disease in hospitals. Many specimens were taken from rectal Swabs. During this period, 2166 stool cultures were streaked onto MacConkey plate and were them deposited in selenite broth. Colonies resembling pathogens on MacConkey medium were picked to KIA, Urea agar, malonate broth, ONPG broth, SIM. Reaction on those media cultures were identified biochemically with using API 20E test kit and confirmed serologically with commercially avabile Salmonella antisera(Difco) or Shigella antisera(Denka, Japan). The sensitivity of Salmonella and Shigella tested to ampicillin cephalosporin, chloramphenicol, colistin, gentamicin, tetracycline, streptomycin, nalidixic acid, neomycin, polymyxin B was performed by means of disc diffusion method recommended by Bauer-Kirby, using the discs prepared in BBL Laboratory. 1. There were 34 (1.6%) isolations of Salmonella cultures and 52(2.4%) isolations of Shigella from the 2,116 specimens. Only 53%of Salmonella were isolated by direct streaking on MacConkey plating media, by contrast, 80% of the Shigella were isolated directly. 2. Shigella flexneri types comprised 56% of the Shigellae isolate from 52 Shigellae identified 24% of Salmonella enteritidis ser typhimurium were identified. 3. Concerning to Salmonella and Shigella occurance according to month and sex, They shows relatively higher for the male than in case of female, and 2-3 age were shown the highest group. 4. October is the month with highest incidences. 5. In the sensitivity patterns of Shigellae, most of them were appeared to be resistant ampicillin, streptomycin, tetracycline, in case of Salmonella, 15% of them were resistant to chloramphenicol.

Antimicrobial Activity of Whangkumtang Extract and Scutellariae Radix Extract on the Food-Borne Pathogens (황금탕 및 황금추출물의 식중독 세균들에 대한 생육억제 효과)

  • The effects of Whangkumtang extract and Scutellariae radix extract on the microbial growth were investigated. The antimicrobial activities and cell growth inhibiting effects were investigated to selected strains with different concentrations of Whangkmtang and Scutellariae radix extracts. Whangkmtang and Scutellariae radix extracts showed the antimicrobial activities on Staphylococcus aureus, Bacillus cereus, Shigella sonnei, Shigella flexneri, Salmonella Typhimurium, Salmonella Enteritidis, Vibrio parahaemolyticus, Escherichia coli O111 and Escherichia coli O126. Whangkumtang and Scutellariae radix extracts did not show the antimicrobial activity on Listeriamonocytogenes. Scutellariae radix extract showed the antimicrobial activity on Escherichia coli O157 but Whangkumtang extract did not, Minimum inhibitory concentrations (MIC) of Whangkumtang extract for each strain appeared to 40 mg/mL on Staphylococcus aureus and Bacillus cerus, 100 mg/mL on Shigella Flexneri and Salmonella enteritidis. The MICs of Sutellariae radix extract appeared to 10 mg/mL on Bacillus cereus, 20 mg/UL on Staphylococn aureus, Shigellanexneri and Salmonella Enteritidis. Scutellariae radix extract showed the higher antimicrobial activity than Whangkumtang extract. The cell growth inhibitions by Whangkumtang and Scutellariae radix extracts were observed from Staphylococcus aureus and Bacillus cereus, repectively, during 48-hr incubation period.

  • This study was attempted to see more clear relationships among the enterobacteria, especially between the intestinal normal flora and pathogenic bacteria. It has been known that some intestinal normal flora produce the bactrial metabolites that are harmful to other enteric bacteria. One of the metabolites is known as colicin, the protein fraction, which possesses certain degree of inhibitory effect against other bacterial growth fraction, whih possesses certain degree of inhibitory effect against other bacterial growth. As a preliminary study for a colicin purification, the antagonistic effect of E, coli to groups of Salmonella and Shigella has been studied by means of in vitro quantitative culture method. 1. E.coli showed definite inhibitory effects aganist both Salmonella and Shigella groups in the mixture of two organisms. 2. The inhibitory effects of E.coli in the E.coli-Salmonella and the E.coli-Shigella mixture occurred from 4 hours incubation following the inoculation. 3. Even the complete inhibition of pathogenic enteric bacterial growth was noticed in the E.coli-Salmonella mixture at overnight incubation. 4. Among the diluted mixtures, 1:100, 1:1,000, and 1:10,000, survival rate of pathogenic enteric bacteria in the mixtures with E.coli showed least affected at the 1:1,000 dilution. 5. It was found that the antagonistic effect aganist groups of Salmonella-shigella was depending upon the groups of the genera.

Antimicrobial Effect of Forsythiae Fructus Extracts on Several Food-Borne Pathogens (연교 추출물의 항균활성 검색 및 일부 식중독균의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effect of the Forsythiae Fructus extracts against food-borne pathogens. First, Forsythiae Fructus was extracted with methanol at room temperature and the methanol extracts were fractionated by using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Forsythiae Fructus extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Forsythiae Fructus showed the highest antimicrobial activity against Salmonella paratryphimurium and Salmonella typhimurium. A synergistic effect in inhibition was observed when Forsythiae Fructus extract was mixed with Ulmus davidiana Japonica extract as compared to each extract alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of Forsythiae Fructus against Shigella flexneri and Salmonella paratyphimurium. The aqueous extract of Forsythiae Fructus had strong antimicrobial activity against Staphylococcus epidermidis at the concentration of 10,000 ppm. At this concentration, the growth of Shigella fexneri was retarded for more than 24 hours and for up to 12 hours for Staphylococcus epidermidis. In conclusion, the methanol extracts of Forsythiae Fructus efficiently inhibited Staphylococcus epidermidis and Shigella flexneri.

  • Twenty strains of Salmonella paratyphi A, 55 of S. typhi, 7 of Shigella flexneri, and 14 of Sh. sonnei which were isolated in Taegu area in 1977, were tested for the susceptibility to antimicrobial drugs. All strains of S. paratyphi A were resistant to sulfisomidine(Sa), but none was resistant to chloramphenical(Cm), tetracycline(Tc), streptomycin(Sm), ampicillin, nalidixic acid, kanamycin, gentamicin, amikacin, 1:20 mixture of trimethoprim and sulfamethoxazole, carbenicillin, cephaloridine, and rifampicin. Only one strain. of S. typhi was multiply resistant to Cm, Tc, Sm, and Sa, but all strains were susceptible to the other drugs tested. The resistant strain carried R plasmid; R(Cm Tc Sm Sa). All strains except one were highly resistant to Cm, Tc, Sm, and Sa, and all except one of multiply resistant strains carried R plasmid; R(Cm Tc Sm Sa).

The Pathogenic Role of Enterobacteria(Salmonella, Shigella and E. coli spp) Isolated in Korea (한국에서 분리된 장내세균(Salmonella, Shigella, E. coli 균속)의 병원적 역할에 관한 연구)

  • A total of 5,462 isolates suspicious of Salmonella, Shigella and E. coli which were isolated during 1983 to 1985 by 12 City Hygine Laboratories and General Hospital Laboratories were received and identified at the National Salmonella Center, Seoul, Korea. The result of identification of these strains were summarized as follows: 1. It was confirmed that the total organisms broke down into 2,014 strains of Salmonella 1,294 of which were S. typhi, 887 strains of Shigella and 2,561 strains of E. coli. 2. For seasonal distribution of enteric pathogens, July was the month with the highest out breaks of salmonellosis, May was the month of Shigellosis, and April was of the highest month it in the case of E. coli. 3. Salmonella typhi with the highest incidence of isolation was shown to belong to various phage types, especially with the strains detected in Seoul. M1 type was widely distributed all over the country, but the majority was E1 type in 1983. 4. For age distribution of patients, the 20-29 age group had the highest incidence of salmonellosis whileas the 1 to 9 age group had the highest incidence of Shigellosis. 5. For sexuly distribution of Salmonella and Shigella infections seemed to be relatively higher in the female than in the male. However, E. coli. had no relationship to both sex. 6. The antibiotic sensitivity patterns of S. typhi cultures showed a tendancy to be resistant to colistin, gentamycin, neomycin, tetracycline and streptomycin. 7. The isolates of S. paratyphi-A, S. typhimurium and S. enteritidis seemed to have a tendency of multiple drug resistance. 8. 93.9 percent of 1,568 E. coli strains showed negative reactions to the antisera of enteropathogenic E. coli and 15.6 percent of them produced a heat-labile enterotoxin, but positive reaction to the antisera was 6.1 percent and 11.6 percent of them producled the enterotoxin.

  • In order to determine the virulence properties of eleven strains of Sh. flexneri isolated from diarrheal patients the congo red test, the $Ser{\acute{e}}ny$ test, the HeLa cell invasion test and electrophoresis of plasmids were carried out. The results were summarized as follows. 1. Virulent strains were not determined by the result of Congo red absorption test. 2. Virulent strains showed positive reaction by the Sereny test and the HeLa cells invasion, but avirulent strains revealed negative reaction at those tests. 3. The temperature condition of bacterial growth was a factor of virulent expression. 4. Virulent strains were mostly possessed of a 130.3 Mdal plasmid, but avirulent strains were not.

Antimicrobial Effect of Lonicerae Flos Extracts on Food-borne Pathogens (식중독 유발세균의 증식에 미치는 금은화 추출물의 항균효과)

  • Lonicerae Flos was extracted with methanol and successively fractionated with petroleum ether, chloroform, ethyl acetate, and methanol to investigate their antimicrobial effects against food-borne pathogens and food spoilage bacteria using paper disc method. Ethyl acetate extracts of L. Flos showed highest antimicrobial activity against Shigella dysenteriae. Synergistic effect in inhibition was observed when L. Flos extract was mixed with Artemisa capillaris extract as compared to using each extract alone. Growth inhibition curves were determined using ethyl acetate extracts of L. Flos against Staphylococcus epidermidis and S. dysenteriae. Ethyl acetate extract of L. Flos had antimicrobial activity against S. dysenteriae at 3,000 ppm, retarding growth of S. dysenteriae up to 12 hr.
