Effects of Shiryung-tang Extract on the Liver Injury induced by Ethanol in Rats (시령탕(柴苓湯)이 에탄올 투여로 유발된 흰쥐의 간손상에 미치는 방어효과)

  • Alcoholic liver disease (ALD) is a major cause of morbidity and mortality around the world. Although much progress has been made in understanding the pathogenesis of ALD, there remains no effective therapy for it. Accumulated evidence indicates that oxidative stress is the main pathological factors in the development of ALD. Ethanol administration causes accumulation of reactive oxygen species (ROS), including superoxide, hydroxyl radical, and hydrogen peroxide. ROS, in turn, cause lipid peroxidation of cellular membranes, and protein and DNA oxidation, which results in hepatocyte injury. In addition to pro-oxidants formation, antioxidants depletion caused by ethanol administration also results in oxidative stress. The objective of this study is to investigate the effects of Shiryung-tang extract on the chronic alcoholic liver injury induced by EtOH. Male Sprague Dawley rats were used in this study. All rats were maintained under standard laboratory conditions ($23{\pm}1^{\circ}C$, 12h light/12h dark cycles). All animals (n=30) were randomly divided into following groups: (1) Normal group, treated with distilled water (n=10); (2) Control group, treated with ethanol (n=10); (3) Sample group, treated with ethanol + pharmacopuncture (n=10). For oral administration of ethanol in Control and Sample group, the ethanol was dissolved in distilled water in concentrations of 25%(v/v). Throughout the experiment of 8 week, the rats were allowed free access to water and standard chow. Sample group were administrated by Shiryung-tang extract daily for 8 weeks. Control group were given normal saline for same weeks. As a results, the oral administration of ethanol for 8 weeks leads to hepatotoxicity. The levels of hepatic marker such as HDL-cholesterol, triglyceride, aspartate aminotransferase and alanine aminotransferase were altered. The ethanol also increased lipid peroxidation and depletion of antioxidant enzyme activities as well as hepatic tissue injury. However, the treatment of Shiryung-tang extract prevented all the alterations induced by ethanol and returned their levels to near normal. These data suggest that Shiryung-tang extract could have a beneficial effect in inhibiting the oxidative damage induced by chronic ethanol administration. Therefore, Shiryung-tang extract can be a candidate to protect against EtOH-induced liver injury.

Hepatoprotcetive Effects of Oyster (Crassostrea gigas) Extract in a Rat Model of Alcohol-Induced Oxidative Stress (알코올로 유도된 간 손상 동물모델에서 굴 추출물의 간 보호 효과)

  • This study was conducted to investigate the protective effects of water extract from Crassostrea gigas (CGW) against ethanol-induced hepatic toxicity in rats. Seventy-two male Wistar rats (6-week-old) were divided into six groups of 12 animals each: control group (1 mL saline/d), ethanol-treated group, positive control group (ethanol+Hovenia dulcis Thunb extract), CGWL group (ethanol+low dosage of CGW), CGWM group (ethanol+medium dosage of CGW), and CGWH group (ethanol+high dosage of CGW). All groups except the control group received ethanol (40% ethanol 5 g/kg) orally. CGW administration with ethanol resulted in prevention of ethanol-induced hepatotoxicity by increasing levels of serum alanine aminotransferase and ${\gamma}-glutamyltransferase$. CGW supplementation significantly reduced formation of malonaldehyde and inhibited reduction of hepatic glutathione and peroxidase levels, as compared with the ethanol-administration group. Further, CGW suppressed expression of CYP2E1, which was elevated by ethanol administration. Consequently, our results indicate that Crassostrea gigas may exert hepatoprotective effects against alcohol-induced hepatocyte injury by intensifying the anti-oxidative defense system.

Intramuscular Administration of Zinc Metallothionein to Preslaughter Stressed Pigs Improves Anti-oxidative Status and Pork Quality

  • This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.

Effect of Salinity Stress on Growth, Yield, and Proline Accumulation of Cultivated Potatoes (Solanum tuberosum L.) (염 스트레스에 따른 감자 품종 (Solanum tuberosum L.) 간 생육, 수량 및 proline 함량 변이)

  • This study evaluated the responses of 18 potato cultivars to three levels of salinity stress (electrical conductivity, EC: 1.0, 4.0, and $8.0dS{\cdot}m^{-1}$). Stem, leaf, root, chlorophyll, tuber yield, and proline content were investigated and statistically analyzed using analysis of variance (ANOVA) and correlations. Stem number and stem diameter were not affected by salinity, but stem length and aerial weight showed highly significant responses to salinity. Aerial weight decreased with increasing salinity levels in most cultivars, while it increased in some the cultivars 'Daejima', 'Goun', 'Haryeong', and 'LT-8'. Leaf number, leaf area index, and leaf weight were most significantly affected by salinity and the cultivar ${\times}$ salinity interaction. Root length, root weight, total chlorophyll and chlorophyll a were affected by salinity, but not by the cultivar ${\times}$ salinity interaction. The opposite trend was shown in chlorophyll b. Although there was great variability among cultivars, tuber yield decreased in all cultivars, and was most significantly influenced by salinity and the cultivar ${\times}$ salinity interaction. 'Superior', 'Kroda', 'Romana', and 'Duback' gave better tuber yields under salinity at EC 4.0 and $8.0dS{\cdot}m^{-1}$ than the cultivars with better aerial weights. Proline content was increased by salinity in all cultivars, and was more remarkable in the cultivars with better aerial weights than in cultivars such as 'Superior' and 'Kroda' with better tuber yields. Leaf number, leaf area index, leaf weight, and root length parameters were considered to be useful criteria in the evaluation of salt tolerance because of their high positive correlation with tuber yield; however, given its negative correlation with tuber yield under high salinity, proline content was not. Salinity tolerances varied greatly among potato cultivars. The low correlation between growth and yields of aerial parts under high salinity suggests that, in commercial agriculture, it might be more practical to compare relative yields to controls. Additionally, 'Superior', 'Kroda', 'Romana', and 'Duback' might be very useful cultivars to use in breeding programs to develop salinity-tolerant potatoes, as well as for sustainable potato production in saline areas.

An Unusual Coastal Environment and Cochlodinium polykrikoides Blooms in 1995 in the South Sea of Korea

  • Cochlodinium polykrikoides bloom in 1995 was studied with a focus on an unusual coastal environment in the South Sea of Korea. Data on temperature, salinity, and zooplankton biomass during 1965-1998 and nutrients during 1990-1998 and chlorophyll-a during 1995-1998 were used in this study. These data were obtained from the serial oceanographic observations in Korean waters carried out by the National Fisheries Research and Development Institute. In 1995 the C. polykrikoides bloom began in the coastal area around Narodo Island in August and consequently occurred to the whole coastal area of the South and East Seas of Korea. During June-October 1995, the coastal environment was unusual compared with the long-term means during 1965-1998. In June 1995, sea surface temperature was 1-2$^{\circ}C$ warmer than in other years in all coastal areas, while salinity was high only to the east of Jeju Island. In August 1995, a strong coastal front appeared inshore of a line between Jeju and Tsushima Islands. In particular, a strong coastal front which showed the characteristics of upwelling front occurred in the coastal area around Narodo and Sorido Islands, not only because of a strong intrusion of the Tsushima Warm Current but also because of the upwelling of cold bottom water. Salinity was low in the neighboring waters of western side of Jeju Island. Nutrients and chlorophyll-a were high in the inshore area between Narodo and Sorido Islands in 1995 in contrast with the other years and areas. Zooplankton showed an unusually high abundance in the coastal area in October 1995. We conclude that the Tsushima Warm Current strongly influenced the South Sea of Korea in 1995 and created strong upwelling front bordering cold upwelled water in the coastal area around Narodo and Sorido Islands. It leads us that these physical structures introduce the favorable environment for the development of C. polykrikoides blooms. We suggest that C. polykrikoides has a bio-physical tolerance of high shear and stress and prefers frontal and upwelling relaxed areas as its habitat. We also find that nutrients were not supplied to the coastal area from the offshore where a low salinity water mass with high nutrients appeared around Jeju Island. Because the strong upwelling front protect the reach of offshore low saline water mass. The main source of nutrients was the upwelled water mass in the coastal area of Wando-Narodo-Sorido.

Effects of Leucine on in Vivo Protein Synthesis in Skeletal Muscles of Fed and Food-Deprived Rats (Leucine이 정상 또는 굶게 된 쥐의 골격근육의 단백질 생합성에 미치는 영향)

  • In vivo effects of leucine on skeletal muscle protein synthesis in fed and I-day food deprived young rats were examined. Animals assigned to leucine group were given a single i.p. injection of 80 or 160flmoles of leucine while control group animals were saline sham injected. The rate of protein synthesis was measured by the amount of $^{14}\textrm{C} incorporated into muscle protein after a single injection of $^{14}\textrm{C}-tyrosine, IO$\mu$ Ci/l00g B.W. Examined muscles were two different types of hind limb muscles. the oxidative solues and the glycolytic EDL and plantaris. Administered leucine elevated the concentration of free leucine in soleus muscles by 4-6.8 times the normal level. A massive dose of leucine, 160 flmoles, stimulated protein synthesis in the EDL and plantaris by 24 %, 29 % respectively of straved rats. The soleus of I-day food deprived rats and both types of muscles in fed rats did not respond to the injected leucine. The synthesis rate of the EDL and plantaris was supressed to one-half of the normal while the soleus that was not stimulated by leucine maintained a relatively normal rate, 78 %, of protein synthesis after I-day of food deprivation. Thus, in vivo stimulatory effect of leucine appears to be not a general phenomenon but to be related to the degree of catabolic condition developed by stress such as food deprivation. Although anabolic effects of leucine observed in this study was limited, any applicability of this special property of leucine to human subjects for the purpose of protein sparing in skeletal muscles remains to be examined.

Changes of Growth and Antioxidative Enzyme(SOD, APX, GR) Activities of Spinach Beet(Beta vulgaris var. cicla) Under Saline Condition (염 환경하에서 근대(Beta vulgaris var. cicla)의 생장과 항산화효소(SOD, APX, GR)의 활성변화)

  • Antioxidative enzymes (superoxide dismutase; SOD, ascorbate peroxidase; APX, glutathione reductase; GR) play major roles in scavenging mechanism of reactive oxygen species which were involved in various stress conditions including salt. In order to investigate the relation between their growth responses (dry weight) and the changes of antioxidative enzymes activity, salt-tolerant spinach beet having 15cm of shoot length were treated with various salt levels (0, 50, 200, 1000 mM NaCl) for 24 hours. Spinach beet exhibited an increase in the activity of antioxidative enzymes by salt, the maximal activity at 200 mM NaCl and the lowest activity at 50 mM NaCl in 2 hrs. after treatments. As a result of PAGE, it has been confirmed that spinach beet contained 3 isoforms (Fe-SOD, CuZn-SOD and Mn-SOD) of SOD and main isoform was CuZn- SOD form. In case of APX, isoforms of the low molecular weight(No. 7, 8) were showed strong expression especially at 200 and 400 mM NaCl treatment. Meanwhile, GR did not show specific pattern of isoforms among the salt treatments. Especially, in case of 50 mM treatment, plant showed the lowest activity of SOD with the best growth, a low enzyme activity was induced by inactivation of the Mn-SOD. Therefore, we suggested that the decrease of SOD activity at a low salt level (50 mM NaCl) or the increase of enzyme activity at a high salt level (200 mM NaCl) may be related to expression of the Mn-SOD isoform. These antioxidative enzymes showed the increase of activity in a short time by salt addition. So, it is considered that spinach beet copes effectively with a stressful condition such as salt by operating effective antioxidative defense mechanism rapidly under high salt level.

Technical Details Imaging Axillary Lymph Nodes in Breast-Specific Gamma Imaging (유방특이감마영상검사에서 액와부 영상 획득 방법에 대한 연구)

  • Purpose : The initial Breast-Specific Gamma Imaging (BSGI) protocol included bilateral breast imaging with 2 views of each breast-craniocaudal (CC) and mediolateral oblique (MLO). Furthermore, Axillary lymph nodes view can be acquired easily. The most meaningful prognosis factor for prediction of breast cancer is whether or not the breast cancer has metastasized to the lymph nodes. However, axillary view doesn't conduct in clinical. This article collates a diverse data of BSGI and describes technical details to acquire optimal imaging. Materials and Methods : A retrospective review was performed on 343 patients who had undergone BSGI between May 2011 and March 2012. Patients who had undergone BSGI received intravenous injection of 740 MBq (20 mCi) $^{99m}Tc$-sestamibi. Results : The following contents are the technical details for optimal axillary imaging. $^{99m}Tc$-sestamibi should be administered using an indwelling venous catheter or scalp needle followed by 10 cc of saline to flush to reduce extravasation and vascular trapping. After administration, patients raise their arm over their head and exercise with stress ball for 1 full minute. A lead shield attached to the gamma camera is removed and patients axilla is placed as close as possible to the camera at a $90^{\circ}$ angle. A lead apron is placed across the shoulder to reduce background from other organs. Acquisition time is enough for 120 sec~180 sec. Conclusion : If patients undergo bilateral axillary imaging as a standard with CC, MLO views, it could improve cancer treatment. Result of this study could maximize efficiency axillary imaging of breast cancer patients.

Effects of Exogenous Ghrelin on the Behaviors and Performance of Weanling Piglets

  • The objective of this study was to evaluate the effect of exogenous ghrelin on the behaviors, weight gain, and feed intakes of weanling piglets. A total of 25 pairs of Duroc$\times$Landrace$\times$Large White piglets weaned at 21 days of age were used in this experiment which finished on day 36. Each pair of healthy piglets from the same litter with similar body weight and of the same gender were selected and randomly arranged to ghrelin or control groups. Thus, there were 50 piglets (ghrelin 25; control 25) kept in 10 pens (ghrelin 5; control 5) and 5 piglets per pen. Initial body weight of the pigs did not differ between the control and ghrelin treatment ($7.43{\pm}0.17kg$; p = 0.81). Experimental pigs were infused with ghrelin ($1{\mu}g/d$ pig) via the marginal ear vein between 0750 and 0800 h at 22, 23, 24 days of age. Control pigs were infused with 0.9% saline. Feed consumption was measured on days 23, 24, 25, 29 and 36. Body weight was measured on days 22, 23, 24, 25, 29 and 36. Behavior data of individual piglets were collected by real-time observation from 0800-1500 h through remote supervisory equipment at 22, 23 and 24 days of age. The results indicated that ghrelin infusion could increase drinking (p<0.05) and lying behaviors (p<0.01) and decrease mounting behaviors (p<0.05). No significant influence of ghrelin was found on average daily weight gain (ADG) and average daily feed intake (ADFI) in this experiment (p>0.05). In conclusion, exogenous ghrelin by the method above and at the dosage of $1{\mu}g/d$ pig could cause a variety of behavioral effects, but not improve performance of weanling piglets.

Spermatogenic and Antioxidant Potential of Mucuna prureins (L.) in Epididymal Spermatozoa: A Dose Dependent Effect

  • The study aim is to investigate the free radicals scavenging and spermatogenic potentials, as well as to analyze any reproductive toxicity of ethanolic extract of Mucuna prureins (M. pruriens) Linn. in spermatozoa, under different dosages in normal male rat. Normal rats were randomly selected and suspension of the extract was administered orally at the dosages of 150, 200 and 250 mg/kg body weight of the different groups of male rats (n=6) once in a day for 60 days and grouped as group II, III and IV respectively. Saline treated rats served as control -group I. On the $60^{th}$ day the animals were sacrificed and the epididymal sperm were subjected to various analyses like level of ROS production, LPO, enzymatic and non enzymatic antioxidant, morphology, morphometry, chromosomal integrity and DNA damage. Results showed significant reduction in ROS production and peroxidation and significant increase in both enzymic and non-enzymic antioxidants in all concentration treated groups when compared with control. Results from all the drug treated groups showed good sperm morphology, increased sperm count and motility. There was no DNA damage and showed normal chromosomal integrity even in 250 mg/kg dose. When compared with control all the three extract treated groups showed increased ROS scavenging activity. However, group II (200 mg/kg) showed significant changes in all the parameters. From the present study it was confirmed that the M. pruriens has potential to improve the sperm qualitatively and quantitatively through scavenging the excess ROS with any adverse side effects. These observations suggest that ethanolic seed extract of M. pruriens may serve as anti-oxidant that can exploit to treat the oxidative stress mediated male factor infertility.
