Study on the of the Correlation between Soil Chemical Properties and Bioactive Compounds of Acer tegmentosum Maxim.

  • This research was carried out to investigate the correlation between soil chemical properties and bioactive compounds of Acer tegmentosum Maxim. The methods of determining bioactive compounds were determined by high performance liquid chromatography, that contained (-)-gallocatechin (0.04±0.01 ~ 0.43±0.28%), salidroside (0.90±0.06 ~ 3.86±0.59%), tyrosol (0.03±0.00 ~ 0.43±0.00%), (-)-catechin (0.05±0.01 ~ 0.37±0.14%), 6'-O-galloylsalidroside (0.02± 0.01 ~ 0.31±0.06%), (-)-epicatechin-gallate (0.01±0.00 ~ 0.04±0.01%). The soil chemical properties analysis such as soil pH, electric conductivity (EC), organic matter (OM), total nitrogen (TN), available phosphate (Avail. P2O5), exchangeable cation and cation exchange capacity (CEC) were performed following the standard manual. The correlation analysis between soil chemical properties and bioactive compounds of A. tegmentosum, soil pH, available phosphate and exchangeable cation (Ca2+ and Mg2+) were negatively correlated with content of salidroside. On the other hand, soil exchangeable cation (Na+) showed positive correlation with content of salidroside. The results of this study was able to investigate the correlation between soil chemical properties and bioactive compounds of A. tegmentosum.

  • Rhodiola rosea L., a traditional medicinal plant in Eastern Asia, is widely used by astronauts, divers and mountaineers to improve their stress resistance. This experiment was conducted to investigate the optimum application rates of phosphate and potassium under the forcing cultivation for high-quality Rhodiola rosea L. production. The application rate of $8kg\; 10a^{-1}$ of phosphate and $30kg\;10a^{-1}$ of potassium showed the highest yield of Rhodiola rosea L. root. There was no significant difference between the application rates of phosphate or potassium fertilizer and the content of salidroside.

Inhibition of LPS induced iNOS, COX-2 and cytokines expression by salidroside through the $NF{-\kappa}B$ inactivation in RAW 264.7 cells (Salidroside의 RAW 264.7 세포에서 $NF{-\kappa}B$ 불활성화를 통한 LPS에)

  • In this study, we investigated the anti-inflammatory effects of salidroside (SAL) isolated from the MeOH extract of Acer tegmentosum Maxim heartwood in RAW 264.7 macrophage cells. SAL pretreatment significantly inhibited nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) productions in the lipopolysaccharide (LPS)-induced RAW 264.7 cells. Western blot and RT-PCR analyses revealed that SAL inhibited the LPS-induced expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the protein and mRNA levels in a concentration-dependent manner. In addition, SAL reduced the release and the mRNA expressions of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and interleukin-6 (IL-6). Furthermore, nuclear factorkappa B ($NF{-\kappa}B$) luciferase reporter assay was performed to know the involvement of SAL in the production of pro-inflammatory cytokines, we confirmed that LPS-induced transcription activity of $NF{-\kappa}B$ was inhibited by SAL. Taken together, our data indicate that anti-inflammatory property of salidroside might be the result from the inhibition of iNOS, COX-2, $TNF-{\alpha}$ and IL-6 expressions via the down-regulation of $NF{-\kappa}B$ activity.

HPLC and GC-MS Analysis of Phenolic Substances in Acer tegmentosum

  • The stem barks, heartwoods, and leaves of Acer tegmentosum (Aceraceae) are widely used in Korea to treat hepatic or cerebral disorders mainly due to alcohol poisoning. This study was aimed to analyze phenolic substances in A. tegmentosum. Quantitative analysis of the three phenolic substances (salidroside, (+)-catechin and scopoletin) was performed by HPLC and the identification of volatile phenolic substances were done by GC-MS. The contents of the three compounds in the three MeOH extracts were higher in the stem bark (salidroside: 80.22 mg/g, (+)-catechin: 23.31 mg/g, and scopoletin: 9.45 mg/g) compared to the heartwoods and leaves. And GC-MS analysis of the stem bark extract demonstrated that p-tyrosol is a main substance of twenty-one compounds identified.

Protective Mechanism of Salidroside Isolated from Acer termentosum Max on Cyclosporine-Induced Nephrotoxicity in Rats (흰쥐에서 Cyclosporine 유도가 산겨릅나무에서 분리한 salidroside의 신독성 경감기전에 관한 효과)

  • Cyclosporine(CsA) is an immunosupressant drug widely used in post-allogeneic organ transplant to reduce the activity of the patient's immune system and so the risk of organ rejection. It has been studied in transplants of skin, heart, kidney, liver, lung, pancreas, bone marrow and small intestine. Initially isolated from a Norwegian soil sample, Both kidney and liver dysfunction are prominent side effects of CsA. The present study was designed to determine the possible protective effect of salidroside(Sal) isolated from the BuOH extract of Acer termentosum Max against oxidative damage in CsA-treated(50 mg/kg, ip) nephrotoxicity rats. Results showed oral administration of methanol and butanol extact of Acer termentosum Max(200 mg/kg, po) significantly reduced activities of marker enzymes(BUN, Creatinine) and LDH activity in serum to CsA induced experimental kidney injured rats. And significantly decrcease of protein amount level in urine and activities of free radical formation enzyme were significantly improved by the treatment of Sal. And significantly decrcease of MDA level in kidney and activities of calalase, glutathione peroxidation and SOD were significantly improved by the treatment of Sal(20 mg/kg, po). But glutathione concentration and glutathione S-transferase actitity was not affected. Results of this study revealed that Sal could afford a significant protection in the alleviation of CsA-induced nephrotoxicity injury.
