토양조건이 퉁퉁마디의 생육에 미치는 영향 (Effects of Different Soil on the Growth of Salicornia herbacea)

  • 해안지대에 서식하는 염생식물인 퉁퉁마디의 염처리와 인공토양에 따른 생육반응을 조사하여 시설 내 인공재배상에 적합한 토양을 선발하고자 본 연구를 실시하였다. 토양은 가볍고 인공재배상에서 이용이 간편한 난석, 버미큘라이트, 펄라이트 및 혼용처리로 하였으며 염처리는 NaCl 농도를 200mM로 처리하였으며 복합비료를 시비하여 4개월간 재배하였다. 초장, 생체중, 건물중에 있어서 식물 생육은 다공성 난석과 펄라이트를 혼합한 토양에서 생육이 좋았으며 NaCl 처리 시 생육과 엽록소 함량은 토양종류에 관계없이 감소하였다. 프롤린 함량은 보습성이 낮은 난석과 펄라이트의 경우 대조구 값이 염처리에 비해 높은 것으로 나타났다. 항산화활력은 처리별 그 차이가 거의 없었고 퉁퉁 마디의 경우 염처리가 항산화물질의 증가에 영향을 미치지는 않는 것으로 나타났다.

함초의 부위별 항산화 및 항혈전 활성 (Anti-oxidation and Anti-thrombosis Activities of Different Parts of Salicornia herbacea L)

  • 함초를 이용한 고부가가치 기능성 음료 개발 연구의 일환으로, 성숙한 함초의 잎, 줄기, 씨를 대상으로 ethanol 추출물을 조제하고 이들의 항산화 및 항혈전 활성을 평가하였다. 추출물의 총 폴리페놀 함량은 잎 > 줄기 > 씨의 순으로, 총 플라보노이드 함량은 잎 > 씨 = 줄기 순으로 나타났다. 총당 및 환원당 함량은 잎과 줄기에서 유사하게 나타났으며, 씨 추출물은 잎 추출물의 1/10 정도의 낮은 총당 및 환원당 함량을 나타내었다. 추출물의 항산화 활성을 RC50로 계산한 결과, DPPH 음이온 소거능, 환원력과 nitrite 소거능은 잎 > 씨 > 줄기, ABTS 양이온 소거능은 잎 = 씨 > 줄기 순으로 나타났다. 항응고 활성 평가결과, 씨 추출물에서 우수한 트롬빈 저해, 혈액응고인자 저해가 나타났으며, 모든 추출물에서 혈소판 응집저해활성 및 인간 적혈구 용혈활성은 나타나지 않았다. 본 연구는 함초 잎과 씨를 이용한 항혈전제 개발이 가능함을 제시하고 있다.

함초 보충시 당뇨유발 흰쥐의 지질과산화와 무기질 함량에 미치는 영향 (Effects of Salicornia herbacea L. Supplementation on Lipid Peroxidation and Mineral Levels in Streptozotocin-Induced Diabetic Rats)

  • This study was designed to examine the effects of Salicornia herbacea L. (glasswort: GW) on the lipid peroxidation and mineral levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of glasswort powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-GW 10% and N-GW 20% treated groups, STZ-control, STZ-GW 10% and STZ-GW 20% treated groups. The rats' liver and muscle glycogen, liver and kidney protein, cholesterol and triglyceride (TG) in liver, malondialdehyde (MDA) in liver and kidney values were measured, along with the hepatic of chromium (Cr), iron (Fe), and zinc (Zn) content. The liver glycogen levels was significantly affected in N-GW 20% group among all the experimental groups. The liver MDA levels of the STZ-GW 10% and STZ-GW 20% groups were significantly lower than for the STZ-control group. There were significant differences between the N-control group and the STZ-control group in the hepatic of Zn levels. The hepatic of Cr levels in the N-GW 20% and STZ-GW 10% and STZ-GW 20% groups were significanly higher than for the each control groups. These results exhibited dose related effect of glasswort and it may have favorable influence on lipid peroxidation in the liver.

Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of glasswort powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-GW 10% and N-GW 20% treated groups, STZ-control, STZ-GW 10% and STZ-GW 20% treated groups. The rats' body weights, aminotransferase activities and hematocrit (Hct) values were measured, along with plasma levels of glucose, protein, cholesterol, HDL-cholesterol, triglyceride (TG) and free fatty acids (FFA). The non-diabetic rats gained weight, while the diabetic rats lost weight. There were significant differences between the control group and the diabetic groups in the weight of the kidney, liver and pancreas. Asparate aminotransferase activity was lower in the non-diabetic control group compared to diabetic experimental groups, even though the difference was not significant. The plasma protein of N-GW 20% group was lower among all experimental groups but it was not significantly different. The blood glucose levels of the STZ-GW 10% group and STZ-GW 20% group were significantly lower than for the diabetic-control group. There were no significant difference of cholesterol levels among diabetic groups. The normal rats of 20% glasswort group in FFA and TG levels showed significant changes among all groups. These results exhibited dose related effect of glasswort and it may contain antihypoglycemic compounds.

Nutrient Value of Saltwort (Salicornia herbacea L.) as Feed for Ruminants

  • Saltwort (Salicornia herbacea L.), a kind of halophytes in Japan, is under investigation as a feed source for livestock as well as potential oilseed crop in salt marshes and salt fields. The present experiment was undertaken to analyze the nutritive value of saltwort as feed for ruminants. To determine the apparent digestibility and nutritive value of saltwort, five Japanese native goats were fed the diets consisting of alfalfa hay cubes with 15 or 20% (n=3 and n=2, respectively) inclusion levels of saltwort in the total diet on a DM basis. All the animals were randomly offered alfalfa hay cubes as a base diet or a mixed diet of alfalfa hay cubes and saltwort at maintenance level, thereafter, alternative feed (a base or mixed diet) was offered to the experimental animals (the incomplete crossover design). Analysis of the chemical composition of saltwort showed that the plant contained high levels of total ash (40.2% DM), sodium (12.7% DM) and chlorine (19.7% DM), and relatively high levels of CP (11.7% DM) and NDF (40.4% DM). Contents of TDN and digestible CP (DCP), DE and ME of saltwort were 33.5% DM, 8.4% DM, 7.4MJ/DM kg and 5.0MJ/DM kg, respectively. These results indicate that saltwort is rich in DCP and minerals (mainly sodium chloride), but poor in energy, suggesting that saltwort could be used as a mineral or CP supplement for ruminants raised around salinized areas in which high quality feed may not be available.

함초 추출액을 첨가한 브라운 소스의 품질 특성 (Quality Characteristics of Salicornia herbacea L. Extract Added Brown Sauce)

  • This study performed physicochemical characteristics and sensory tests on brown sauce with different Salicornia herbacea L. (saltwort) content to find the best brown sauce recipe with saltwort. Moisture content increased with greater saltwort extract content. There was significant (p<0.001) pH difference with saltwort content. There was also significant (p<0.001) sweetness difference in the samples and sweetness increased with greater saltwort content. Viscosity measured at $20^{\circ}C$ and $60^{\circ}C$ was significantly (p<0.001) lower with greater saltwort content. The brightness of brown sauce containing saltwort extracts significantly (p<0.001) increased with greater saltwort content and its redness and yellowness usually decreased. SPS4 with the greatest saltwort content showed the highest value of 4.25% and DPPH radical removal increased significantly (p<0.001) with greater saltwort content. In the sensory test, the color value was lowest at 4.82 with BSL1 and the flavor value was lowest at 5.00 with BSL0, the control group. The taste and the flavor values were highest with BSL2 with 2% saltwort content. Overall acceptance was highest with BSL3, the brown sauce with 3% saltwort content, at 6.09. As a result, it was concluded that the brown sauce with 3% saltwort content was most suitable. Therefore, this study concluded that saltwort with various functions could be used for food and may be used to replace salt and have other functions in brown sauce, a popular sauce consumed in Korea.

함초발효액으로부터 항산화 활성 물질의 분리 및 동정 (Isolation and Identification of Antioxidative Compounds in Fermented Glasswort (Salicornia herbacea L.) Juice)

  • 함초발효액의 기능성 해명과 유용 기능성 성분 탐색연구의 일환으로 함초발효액으로부터 항산화 활성물질을 분리하여 구조해석 하였다. 함초발효액을 용매분획 하여 얻어진 용매분획물들 중 $CHCl_3$층이 높은 항산화 활성을 나타냈다. 그래서 $CHCl_3$층을 Sephadex LH-20 column chromatography의 분자체 효과 및 흡착능의 특성을 각각 이용하여 정제함으로써 2종의 항산화 활성물질을 분리하였다. 단리한 이들 화합물을 대상으로 MS 및 NMR 분석을 통하여 cirsiumaldehyde(1)와 chrysoeriol(2)로 각각 동정하였다. 이 화합물들은 $ABTS^+$ 및 DPPH radical-scavenging 활성을 나타냈으며, 화합물 2는 화합물 1에 비해 더 높은 활성을 나타냈다.

Influence of Inclusion of Salicornia Biomass in Diets for Rams on Digestion and Mineral Balance

  • A metabolism trial was conducted with 28 Najdi rams allocated into seven dietary groups to evaluate the effect of dietary inclusion of Salicornia bigelovii Torr biomass on nutrient digestibility, rumina I fluid metabolites and nitrogen and mineral balances. Either the stems (ST) or spikes (SP) of this seawater-irrigated halophyte were incorporated into complete diets at rates of 0, 10, 20 and 30% levels, replacing equal amounts of rhodesgrass hay in a ground mixed control diet. Digestibility of DM, OM, EE, NFE and fecal and urinary nitrogen were not affected by increased level of ST in the diet. As level of ST increased from 0 to 20% in the diets, CP digestibility and nitrogen retention approached their maximum (p<0.01), whereas CF digestibility reached its minimum (p<0.01). On the other hand, except for EE, digestion of all nutrients and nitrogen retention were linearly depressed (p<0.01) as SP increased in the diets from 10 to 30% level. Concentration of ammonia-N, total VFA and pH values in the rumen fluid were lower (p<0.01) with the ST- or SP-fed diets than with the control diet. Increasing level of ST or SP in the diet was associated with an increase (p<0.01) in the proportion of acetate and a decline (p<0.01) in molar percentage of propionate in the ruminal fluid. Sodium absorption increased (p<0.01) with increased ST and SP in the diets up to the 10 and 20% level, respectively, followed by constant absorption values up to the 30% level. When the level of ST in the diet gradually increased to 30%, a concomitant increased (p<0:01) in Ca and P absorption were obvious; whereas, increased level of SP in the diets from 0 to 30% resulted in noticeable (p<0.01) depression in Ca and P apparent absorption.

Anti-Proliferative Effect of Polysaccharides from Salicornia herbacea on Induction of G2/M Arrest and Apoptosis in Human Colon Cancer Cells

  • In this study, we investigated the anti-proliferative effect of polysaccharides from Salicornia herbacea on HT-29 human colon cancer cells. Crude polysaccharides from S. herbacea (CS) were prepared by extraction with hot steam water, and fine polysaccharides from S. herbacea (PS) were obtained through further size exclusion chromatography. The anti-proliferative effect of CS and PS were measured using the MTS assay, apoptosis analysis, cell cycle analysis, and RT-PCR. HT-29 cells were treated with CS or PS at different dosages (0.5, 1, 2, 4 mg $ml^{-1}$) for 24 or 48 h. CS and PS inhibited proliferation and stimulated apoptosis of cells in a dose-dependent manner. Flow cytometric analysis after Annexin V-FITC and PI staining revealed that treatment with CS or PS increased total apoptotic death of cells to 24.99% or 91.59%, respectively, in comparison with the control (13.51 %). PS increased early apoptotic death substantially - up to 12 times more than the control. Treatment with CS or PS resulted in a concentration-dependent increase of the G2/M cell population of the cell cycle as determined by flow cytometry. G2/M arrest was induced significantly with the highest concentration (4 mg $ml^{-1}$) of PS. RT-PCR was performed to study the correlation between G2/M arrest and transcription of cell cycle control genes. The anti-proliferative activity of CS and PS was accompanied by inhibition of cyclin B1, and Cdc 2 mRNA. Moreover, both CS and PS induced expression of the p53 tumor suppressor gene and the Cdk inhibitor p21. These results suggest that polysaccharides from S. herbacea have anti-cancer activity in human colon cancer cells.

Effects of Salicornia herbacea Powder on Quality Traits of Sun-Dried Hanwoo Beef Jerky during Storage

  • The objective of this study was to evaluate the quality characteristics of sun-dried Hanwoo beef jerky added with Salicornia herbacea (SH). Sliced Hanwoo beef shank were marinated and sun-dried at $28-30^{\circ}C$, relative humidity (RH) 30-35% for 3.5 h. The physicochemical and microbiological traits of the Hanwoo beef jerky were analyzed during the aerobically packaged storage at $25^{\circ}C$. The water activities of beef jerky with 0.5% and 1.0% SH were lower than those of the control at 0 d (p<0.05). The pH values of beef jerky with SH were significantly higher than those of the control (p<0.05). The beef jerky with SH and ascorbic acid showed significantly lower TBARS values than the control (p<0.05). The beef jerky with SH showed a significantly lower redness ($a^*$) than the control (p<0.05). Total plate count (TPC) of beef jerky with 1.0% SH was significantly lower than that of the control during the storage of 20 d (p<0.05). Yeast/mold was detected in the control and beef jerky with SH after storage periods of 10 and 20 d, but was not detected in jerky with ascorbic acid. With regard to the sensory properties, beef jerky with SH showed significantly greater flavor scores than the others (p<0.05). The result shows that SH powder can be used to increase the sensory quality and microbial safety of beef jerky.
