Effect of Water Temperature on the Expression of Stress Related Genes in Atlantic Salmon (Salmo salar) Fry (수온이 대서양 연어(Salmo salar) 치어의 체내 스트레스 관련 유전자 발현에 미치는 영향)

  • The warming of water as a result of climate change affects fish habitat. Variations in water temperature affect fish physiology almost totally. The rise in water temperature due to climate change leads to hypoxia following decreased oxygen solubility and decreased binding capacity of oxygen-carrying hemoglobin. This study was conducted to evaluate the health status of Atlantic salmon (Salmo salar) fry at elevated water temperatures($20^{\circ}C$) compared with optimum water temperature ($15^{\circ}C$). The method facilitated the detection of biomarker genes using NGS RNAseq analysis and evaluation of their expression pattern using RT-qPCR analysis. The biomarker genes included interferon alpha-inducible protein 27-like protein 2A transcript variant X3, protein L-Myc-1b-like, placenta growth factor-like transcript variant X1, fibroblast growth factor receptor-like 1 transcript variant X1, transferrin, intelectin, thioredoxin-like, c-type lectin lectoxin-Thr1-like, ladderlectin-like and calponin-1. The selected biomarker genes were sensitive to changes in water temperature based on NGS RNAseq analysis. The expression patterns of these genes based on RT-qPCR were similar to those of NGS RNAseq analysis.

Oxidative Stability of Salmon (Salno salar) Mince as Affected by an Added Stabilizing Protein Ingredient and Storage Temperature (안정제 첨가와 저장온도에 따른 Salmon(Salno salar) Mince의 산화 안정성)

  • The oxidative stability of salmon mince to which milk protein concentrate(MPC) added was investigated. Salmon mince was stored at 4$\^{C}$ for 21 days, at -18$\^{C}$ for 20 days and at -18$\^{C}$ for 20 days after cooking in an electric omen. At each storage point, peroxide values(POV) were determined. Salmon mince with 4% MPC increased greater oxidative stability than control (without MPC). Sensory evaluation for measuring the oxidative stability of salmon mince was accomplished. Sensory scares of salmon mince with 4% MPC were higher than those of control. The results indicate that MPC could be useful for oxidative stability, as stabilizing protein ingredient of salmon mince.

  • Carboxypeptidase B from Salmo Salar eggs was purified by CM-cellulose, 0.5 ammonium sulfate saturation, DEAE-cellulose, and Sephadex G-75 gel filtration and its enzymatic properties were investigated. Optimum temperature was 55$^{\circ}C$, pH optima were 4.0 and 7.0 at 37$^{\circ}C$, and the enzyme was stable at pH 2.0∼3.0 and 5.5∼7.0 for 1.5h. This enzyme showed substrate specificity hydrolyzing the peptide bond of glycyl-L-arginine. Its K$_m$ values was 0.21mM, and the enzyme activity was stimulated by Cu$^{2+}$ and Fe$^{3+}$, while inhibited by Zn$^{2+}$. The lysine was found to be competitive inhibitor and its K$_i$ value was determined to be 4.3mM. Molecular weight of this enzyme was determined to be 36,400 daltons by SDS-PAGE and the enzyme was monomeric protein composed of 19 kinds of amino acid residues.

  • Carotenoprotein from Salmo Salar eggs was purified and characterized by CM-cellulose, 50% $(NH_4)_2SO_4$, DEAE-cellulose and sephadex G-75 column. The chromoprotein had a spectrum with ${\lambda}_{max}$ 409, 540 and 580nm in p-buffer (pH 7.0) at initial step. Molecular weights by sephadex G-200 gel filtration were 50, 200 and 26,000 daltons. SDS-PAGE analysis showed a structure with four identical subunits (12,500 daltons). Its sample retained a small amount of carbohydrates and lipids. Amino acids were analyzed, and mannose, galactose and glucosamine also were identified. Carotenoid extacted with acetone was found to be astaxanthin ester by partition test, epoxy test, iodine test, allylic test, reduction, acetylation, uv/vis, ir and nmr datas. Stearate (47.9%) and palmitate (21.4%) were predominant fatty acids in the astaxanthin ester.

  • The recent increase in demand for lithium has led to the development of new brine prospects, The brines are hosted in closed salar basin aquifers of two types that are mature halite salars and immature clastic salars. Salar brines also contain other elements of commercial interest, most notably potassium and boron. As a result, there has been a plethora of new exploration projects focused on the brines hosted in the aquifers of the intermontane-closed basins. The estimate of lithium resources and reserves in these salars depends on a detailed knowledge of aquifer geometry, porosity, and brine grade. Because the resource is in a fluid state, it has the propensity to move, mix, rearrange itself relatively rapidly during the course of a project lifetime, and lower recovery factors compared with most metalliferous and industrial mineral deposits due to reliance on pumping of the brine from wells for extraction. This is unlike any other type of metallic mineral resource and hence a different approach specially focusing on hydrogeology and brine hydrology is required for these prospects.

  • Variations in water temperature are known to affect almost every part of fish physiology. The rise in water temperature due to climate change can physically damage fish. This study was conducted to evaluate the health status of the Atlantic salmon (Salmo salar) at high water temperature (20℃) than the optimum water temperature (15℃). Liver tissue exerts important metabolic functions in thermal adaptation. Therefore, liver tissue was used in this study. The evaluation method is to develop the biomarker gene using NGS RNAseq analysis and to examine the expression pattern using RT-qPCR analysis. The NGS RNAseq analysis revealed 1,366 differentially expressed genes, among which 880 genes were increase expressed and 486 genes were decrease expressed. The biomarker genes are such as heat shock protein 90 alpha (Hsp90α), heat shock protein 90 beta (Hsp90β) and cytochrome P450 1A (CYP1A). The selected genes are sensitive to changes in water temperature through NGS RNAseq analysis. Expression patterns of these genes through RT-qPCR were similar to those of NGS RNAseq analysis. The results of this study can be applied to other fish species and it is considered to be useful industrially.

  • Recent development of lithium ion batteries for vehicles industries have led to a boom in lithium exploration and development for the new generation of batteries. One of the cheapest sources of lithium is the brines hosted in the aquifers of the arid intermontane-closed salar basins. Because the resource is a fluid, with the attendant problems of in-aquifer mixing, reorganization, and lower recovery factors compared with most metalliferous and industrial mineral deposits due to reliance on pumping of the brine from wells for extraction, existing codes for filing resource and reserve estimates require new approach for these prospects. Evaluation of brine resources is complex and requires participation of a variety of qualified experts such as hydrogeologists, geologists, geochemists and chemical engineers. The technical reports disclosing the results of these estimates should reflect the inputs of multi-disciplinary approaches. The requirements for brine resource and reserve evaluation, drawing on several examples from the experiences in the Central Andes are reviewed in this paper.

  • This study was conducted to obtain the basic data for designing the lithium recovery process from the "salar de Uyuni" in Bolivia. For this study, the mock brine which has the similar chemical composition with the brine of "salar de Uyuni" was prepared, and the effects of reaction factors such as temperature, time, pH and so forth on the precitation reaction of magnesium hydroxide were investigated.

  • This study investigated the effects of photoperiod (NL 12L:12D and LL 24L:0D) and body sizes (30 g and 50 g) on parr-smolt transformation, post-smolt growth and blood properties in the off-season parr-smolt stage of Atlantic salmon reared in a recirculating aquaculture system (RAS). Potential off-season salmon smolt were reared in a freshwater RAS for 80 days and then all experimental fish were transferred to seawater. In both LL groups (LL-30 and LL-50), we recorded and increase in specific growth rate and reduction in feed conversion, although there were no significant difference in body size. The values of osmolality, and serum Na+, Cl- and cortisol concentrations in the LL groups were maintained at lower levels than in NL group fish, and LL group fish were observed to recover to the pre-seawater adaptation state more rapidly than those in the NL group. ID chips were inserted in all smolts reared in freshwater. These fish were subsequently transferred to full-strength seawater and thereafter individual growth rates were monitored for 120 days. The results indicated that compared with smolt reared under natural photoperiodic condition, 24 h lighting in freshwater contributed to enhancing post-smolt specific growth rate in seawater.
