  • Recombinant baculoviruses are widely used to express heterologous genes in cultured insect cells. Recombinant baculoviruses can serve as gene-transfer vectors for expression of recombinant proteins in a wide range of mammalian cell types. Baculovirus system has significant benefits in view of safety, large-scale, and high level of gene expression. In this study, baculoviral vectors which were reconstructed from pOPINEneo-3C-GFP vector, were recombined with cytomegalovirus (CMV) promoter, green fluorescent protein (GFP), and p53 with NcoI and XhoI. These recombinant vectors were infected with various cells and cell lines. The baculovirus vector thus developed was analyzed by comparing the metastasis and expression of the recombinant genes with conventional vectors. These results suggest that the baculovirus vector has higher efficiency in metastasis and expression than the control vector. • Objectives : Practice-Based Research Networks (PBRNs), collaborations of practitioners and academic researchers, have provided platforms for conducting research to address clinical questions generated from daily routine care. This review aimed to critically analyse articles from PBRNs that are related to complementary and integrative medicine (CIM) and to suggest future directions for a PBRN which is appropriate for Korean Medicine (KM). Methods : PubMed, PBRN registries in Agency for Healthcare Research and Quality and relevant PBRN websites were searched up to November 2019 for research articles from PBRNs that focused on CIM regardless of study design. Methodological quality of the included studies was assessed. The included studies were read in full, classified and summarised according to their topics. Results : A total of 51 articles published from 1998 through 2020 were included in this review. They were categorised into three principal themes based on research questions and findings: health services research (embracing researches examining characteristics of patients and CIM practitioners/practices, and communication between patients and practitioners); effectiveness and safety of CIM practices/interventions; and feasibility studies of instruments and interventions in PBRN settings. The study designs varied including surveys (n=30), prospective observational studies (n=6), 2ndary analyses of existing studies (n=7), protocols (n=7), retrospective chart review (n=1) and qualitative study (n=1). Quality of the included studies greatly varied. Conclusions : PBRNs can serve as a feasible platform for conducting practice-relevant research on KM and CIM. Considering growing demands on evidence-base for routine practice of KM amid various stakeholders, a PBRN in KM community and further researches nested within PBRN designs are warranted.

  • C-ITS(Cooperative Intelligent Transport System) that pursues traffic safety and convenience uses various sensors to generate traffic information. Therefore, it is necessary to improve the sensor-related technology to increase the efficiency and reliability of the traffic information. Recently, the role of CCTV in collecting video information has become more important due to advances in AI(Artificial Intelligence) technology. In this study, we propose to identify and track dynamic objects(vehicles, people, etc.) in CCTV images, and to analyze and provide information about them in various environments. To this end, we conducted identification and tracking of dynamic objects using the Yolov4 and Deepsort algorithms, establishment of real-time multi-user support servers based on Kafka, defining transformation matrices between images and spatial coordinate systems, and map-based dynamic object visualization. In addition, a positional consistency evaluation was performed to confirm its usefulness. Through the proposed scheme, we confirmed that CCTVs can serve as important sensors to provide relevant information by analyzing road conditions in real time in terms of road infrastructure beyond a simple monitoring role.

  • Purpose: To examine whether the Patient Experience Assessment (PEA) has led to perceptible changes at the ground level of health care, as a way of evaluating PEA as a policy intervention for quality improvement. Methods: Four focus group discussions (FGDs) were conducted, each comprising six to eight participants who were employees responsible for patient experience at their respective hospitals. The primary focus of the FGDs was on questions such as: 1) How do hospitals respond to PEA? 2) What significant changes were observed after the implementation of PEA? 3) What were the unintended consequences of implementing PEA, if any? 4) What areas of improvement have been identified for maximizing the potential of PEA? Results: Two broad themes emerged out of the FGDs: changes observed post implementation of PEA, and areas for improvement of PEA. Four significant changes were reported by participants: changes in perception and attitude regarding patient experience in hospital employees, increased active involvement by the hospital leadership, enhanced efforts to improve patient experience, and increased cooperation between such activities. Furthermore, eight areas of improvement were identified, which have been grouped in three categories: improving the process of data collection for PEA, introducing additional catalysts to facilitate further changes, and paying attention to structure- and patient-level constraints that must be addressed in parallel. Conclusion: The implementation of PEA led to perceptible changes within hospitals, which implies that it can serve as an effective catalyst for improving patient experience. A number of areas of improvement that would aid in maximizing the potential of PEA were also identified.

  • Vaccination against Newcastle disease (ND) is the most effective means of controlling the disease, and these vaccines are commercialized only after their safety and effectiveness have been verified through tests that comply with Korean Standards of National Lot Release for Veterinary Biologics. This study investigated whether a relatively convenient and safe serological test can be used in place of the challenge test using highly virulent ND virus. Hemagglutination inhibition (HI) assay and enzyme-linked immunosorbent assay (ELISA) were considered positive of log2 2 or more and cutoff value of 200 or more, respectively, in both live and inactivated vaccines. However, when the antibody levels of the live and inactivated vaccines induced using the Ulster 2C, KBNP-C4152R2L, and K148/08 strains were compared, the antibody titers for inactivated vaccines were significantly higher than those for live vaccines in both the HI assay and ELISA. A strong positive correlation was observed between HI and ELISA antibody titers. The live vaccines corresponded to a survival rates of ≥ 80% and the inactivated vaccines corresponded to 100% survival rates. This study confirmed that standard efficacy tests can serve as serological tests, and can replace the challenge test and that the vaccine approval process can be improved.

  • Chlorine dioxide (ClO2) is widely used for post-harvest sterilization treatment. However, there are limitations in the retail application of ClO2 due to difficulties in handling, expensive facilities, and safety concerns. Therefore, it is necessary to develop a ClO2 technology that can be easily applied and continuously released for a long period. In this study, a series of ClO2 self-releasing sachets were developed. First, poly(ether-block-amide) (PEBAX) and polyethylene-glycol (PEG) composite films containing different ratios of citric acid (CA) were prepared using the solution casting method. The as-prepared PEBAX/PEG-CA composite films were evaluated using FT-IR, DSC, and TGA to confirm chemical structure and thermal properties. Subsequently, PEBAX/PEG-CA composite films were designed in the form of a sachet and NaClO2 powder was transferred into the sachet to achieve a ClO2 self-releasing system. The ClO2-releasing behavior of the sachet was investigated by measuring the release amount of the gas using UV-vis. The release amount of ClO2 increased with increasing CA contents owing to the existence of higher protons (trigger) in the polymer matrix. Further, ClO2 gas was released for a longer time. Therefore, the as-prepared smart sachet can be tuned according to applications and packaging sizes to serve an optimal sterilization effect.

  • Lane management with a central variable lane(s) (or reversible lane) where the traffic flow is temporarily reversed in one or more lanes during peak periods has been evaluated as an effective strategy to alleviate congestion caused by tidal traffic flows. However, due to traffic safety issues, such a movable barrier system can be considered as an alternative to supplement the existing its operation facilities such as static and/or dynamic signs and special pavement markings. In addition, when combined with a bus exclusive lane strategy, its effectiveness could be greatly increased. The objective of this study is to propose a feasibility analysis procedure for operational strategies of a bus-exclusive lanes using a barrier transfer system (BTS) for urban expressways. To this end, a case study was conducted on two urban expressways on the west side of the Han River in Seoul. As a result, temporary operation during rush hour in the morning was found to be most effective. The results presented in this study are expected to serve as a basis for establishing bus-exclusive lane operation strategies using similar systems in the future.

  • The demolition of domestic infrastructures mirrors other significant construction initiatives in presenting a markedly high accident rate. A comprehensive investigation into the origins of such accidents is crucial for the prevention of future incidents. Upon detailed inspection, the causes of demolition construction accidents are multifarious, encompassing unsafe worker behavior, hazardous conditions, psychological and physical states, and site management deficiencies. While statistics relating to demolition construction accidents are consistently collated and reported, there exists an exigent need for a more foundational cause categorization system based on accident type. Drawing from Heinrich's Domino Theory, this study classifies the origins of accidents(unsafe behavior, unsafe conditions) and human errors(human factors) as per the type of accidents experienced during demolition construction. In this study, a three-step model of QFD-FMEA(Quality Function Deployment - Failure Mode Effect Analysis) is employed to systematically categorize accident causes according to the types of accidents that occur during demolition construction. The QFD-FMEA method offers a technique for cause classification at each stage of the demolition process, including direct causes(unsafe behavior, unsafe environment), and human errors(human factors) through a tri-stage process. The results of this accident cause classification can serve as safety knowledge and reference checklists for accident prevention efforts.
