Purification and Properties of Isocitrate Lyase from Saccharomycopsis lipolytica (Saccharomycopsis lipolytica Isocitrate Lyase의 정제와 성질)

  • Isocitrate lyase from crude extract of Saccharomycopsis lipolytica ATCC44601 and MX9-11RX8 temperature-sensitive mutant was purified about 54 times and 87 times, respectively by ammonium sulfate fractionation, Toyo peal HW-55F gel filtration and DEAE-Cellulose ion exchange chromatography, The molecular weight of the purified isocitrate lyase from this yeast was estimated to be 230, 000 by gel filtration on Sephadex G-200, and SDS-polyacrylamide Eel electrophoresis showed that the enzyme consisted of four identical or similar subunits with a molecular weight of 59, 000 and the enzyme showed optimum activity at pH 6.9.

  • The effects of media composition on citric acid fermentation using surface immobilized Saccharomycopsis lipolytica were studied. The use of the standard medium for these organisms resulted in rapid decrease of citric acid production and a transformation of immobilized cell morphologies from a yeast-type to a mycelium-type. When the standard medium was enriched with vitamins, trace minerals, a growth factor and ammonium to form a Vigorous Stationary Phase (VSP) fermentation type medium, relatively stable citric acid production (10 mg/lㆍh) was obtained. Using the VSP type medium, the surface immobilized cells also retained their yeast-type form.

  • Effect of itaconate upon citrate and isocitrate production of Saccharomycopsis lipolytica were investigated. The percent inhibition of isocitrate lyase activity was about 80% at 0.8 mM itaconate concentration, with Ki value of 0.17 mM in the cleavage reaction. Inhibitory effect of itaconate at 20 mM on S. lipolytica growth was significant on n-hexadecane medium, whereas almost no inhibitory effect on glucose medium. Increasing itaconate concentration in n-hexadecane medium improved isocitrate production up to 80% but no difference was found in glucose medium.

  • Saccharomycopsis fibuligera is an amylolytic yeast that exhibits raw starch-degrading activity. In this study, adaptive laboratory evolution was performed to improve the tolerance of S. fibuligera to lactic acid by prolonged repeated batch fermentation in which the lactic acid concentration was gradually increased. The evolved S. fibuligera strain exhibited a significantly enhanced tolerance to lactic acid at concentrations up to 2.5% (w/v), as assessed by determining its specific growth rate using a plate assay. Scanning electron microscopy revealed an elongated and perforated morphology of the parent strain under lactic acid stress, indicating that its membrane might be more prone to damage caused by lactic acid than that of the evolved strain.

  • Sexually compatible heterothallic haploids and diploids therefrom of Saccharomycopsis lipolytica were compared with respect to production. Diploids constructed through mating were confirmed by random spore analysis, whereas those constructed through protoplast fusion were confirmed by haploidization. ATCC 44601 and IFO 1209 produced larger amount of citrate and isocitrate than IFO 1550 and IFO 1551. A mode of citrate production by diploids was intermediate of the parental haploid strains. The specific activities of citrate synthase and isocitrate lyase in IFO 1550 and IFO 1551 were higher than those in ATCC 44601 and IFO 1209, indicating little correlation between citrate production and specific activities of these enzymes.

  • Twenty-seven strains of starch utilizing yeasts were isolated from 30 samples of ‘Nuruk’, a traditional starter in Korea. These strains were identified as ten species belonging to four genera; Hansenula anomaly (six strains), Hansenula sydowiorum (two strains), Saccharomycopsis fibuligera (four strains), Schwanniomyces occidentalis (two strains), Candida fabianii(two strains), Candida famata(one strain), Candida hydrocarbofumarica (three strains), Candida silvicola (one strain), Candida steatolytica (four strains) and Candida tropicalis (two strains). Saccharomycopsis fibuligera Nu-01, Nu-08, Nu-12 and Nu-27 produced much amylase, and one of these, Saccharomycopsis fibuligera Nu-12 showed the highest amylase activity (16.9 IU/ml). Among the isolates, the strains of Hansenula anomala were exhibited relatively high specific growth rate in the medium used starch as a carbon source, and ethanol fermentation by the strains of Candida hydrocarbofumarica was not observed.

  • The glucoamylase was purified from the induced culture filtrate of hybrid between Saccharomycopsis sp. and Saccharomycopsis sp. made by nuclear transfer and characterized for some enzyme properties. The enzymewas purified 76-fold in an overall yield of 16% from the culture medium by ammonium sulfate fractionation,Sephadex G-150 gel permeation chromatography and DEAE-Sephadex A-50 ion exchage chromatography.The molecular weight of the purified glucoamylase was estimated to be 57.5 KDa on SDS-polyacrylamidegel electrophoresis and Sephadex G-150 gel permeation chromatography. The purified enzyme was active atpH-5.0 and $40^{\circ}C$. The Km value for soluble starch was 2.6 mg/ml. The enzymatic activity was stimulated inthe presence of TEX>$Ca^{2+}$, EDTA, $Co^{2+}$, $Mg^{2+}$, and $Mn^{2+}$

  • A yeast strains, CJ 2, CJ 7 and CJ 8, isolated from soil and contaminated choose, mated with authentic strains of Saccharomycopsis lipolytica and were identified as Saccharomycopsis lipolytica with mating A, B and B, respectively. The strain CJ 7 produced large amount of isocitric acid in glucose and n-hexadecane medium as compared with another strains. All strains produced larger amount of citric acid in n-hexadecane medium as compared with glucose medium, and citric acid production of diploids was greater than that of the parental haploid strains. The specific activity of isocitrate lyase in n-hexadecane grown cells was $15{\sim}20$ times greater than that in glucose-grown cells, but the specific activity of citrate synthetase was not so influenced by carbon source. Little correlation between citric acid production and the specific acitivity of these enzymes was noticed irrespective of strains and ploidy.

A study on anti-inflammatory activity of fermented rice bran of Saccharomycopsis fibuligera isolated from Nuruk (누룩에서 분리한 Saccharomycopsis fibuligera 미강 발효물의 항염활성에 대한 연구)

  • This paper aims to increase functionality and competitiveness through fermentation of rice bran, which is used as a raw material for alternative foods and cosmetics. Cytotoxicity and anti-inflammatory effects were confirmed using rice bran extract fermented with Saccharomycopsis fibuligera A8 isolated from Nuruk. In the case of cytotoxicity, cytotoxicity was shown at 100 ㎍/mL for rice bran extracts, but cytotoxicity was not shown for fermented rice bran extracts. Meanwhile, as a result of confirming anti-inflammatory effects through inflammatory indicators such as nitric oxide, TNF-α, IL-1β, and IL-6, rice bran extracts showed anti-inflammatory effects at concentrations of 100 ㎍/mL or higher only nitric oxide and TNF-α. And fermented rice bran extracts exhibited anti-inflammatory effects at concentrations of 25 ㎍/mL, 25 ㎍/mL, 50 ㎍/mL, and 50 ㎍/mL, respectively, exhibiting anti-inflammatory effects in lower concentration. Meanwhile, as a result of testing the cytotoxicity and anti-inflammatory effects of heat-killed S. fibuligera, it was found that the heat-killed S. fibuligera showed anti-inflammatory effect in fermented products.

A Study on Enzyme Activity and Antioxidant Activity of Fermented Rice Bran of Saccharomycopsis fibuligera Isolated from Nuruk (누룩에서 분리한 Saccharomycopsis fibuligera 미강발효물의 효소활성 및 항산화능에 대한 연구)

  • This paper aimed to increase the functionality and competitiveness of rice bran, which is used as an alternative food and cosmetic raw material, through fermentation. As a result of comparing the enzyme activity of 𝛼-amylase, CMCase, 𝛽-glucosidase, protease, etc. of fermented rice bran using standard strains with 6 strains with excellent enzyme activity among 50 strains of Saccharomycopsis fibuligera isolated from 5 types of Nuruk, A8 strain showed excellent enzyme activity of 13.7%, 21.1%, 50.3%, and 10.0% compared to the KCTC 7806 strain. As a result of the ABTS and DPPH tests as a result of fermentation, it was confirmed that rice bran fermented with A8 had 1.12 times and 1.28 times more antioxidant activity than rice bran fermented with KCTC 7806 strain. In this paper, it was possible to confirm the S. fibuligera A8 strain superior to the standard S. fibuligera strain KCTC 7806.
