Saccharide Effect on the Lower Critical Solution Temperature of Poly(organophosphazenes) with Methoxy-poly(ethylene glycol) and Amino Acid Esters as Side Groups

  • The lower critical solution temperature (LCST) of thermosensitive poly(organophosphazenes) with methoxypoly(ethylene glycol) (MPEG) and amino acid esters as side groups was studied as a function of saccharide concentration in aqueous solutions of mono-, di-, and polysaccharides. Most of the saccharides decreased the LCST of the polymers, and the LCST decrease was more prominently observed by saccharides containing a galactose ring, such as D-galactose, D-galactosamine and D-lactose, and also the polysacccharide, 1-6-linked D-dextran effectively decreased the LCST of the polymers. Such an effect was discussed in terms of intramolecular hydrogen bonding of saccharides in polymer aqueous solution. The saccharide effect was found to be almost independent on the kinds of the amino acid esters and MPEG length of the polymers. Such a result implies that the polymer-saccharide interaction in aqueous solution is clearly influenced by the structure of sacchardes rather than by that of the polymers. The acid saccharides such as D-glucuronic and D-lactobionic acid increased the LCST, which seems to be due to their pH effect.

  • 병원성 곰팡이에 대해 강한 길항성을 보이는 토양미생물 Bacillus subtilis HJ927을 Phytophthora capsici에 감염된 고추밭으로부터 분리해냈다. B. subtilis HJ927을 P. capsici와 함께 고추에 처리한 결과 P. capsici만을 접종한 처리구에 비해 크게 식물을 보호하는 것을 root mortality 측정결과 확인하였다. B. subtilis HJ927는 항곰팡이성 물질로 3-methylbutyric acid, 2-methylbutyric acid, 그리고 methyl 2-hydroxy, 3-phenyipropanoate를 분비해 내는 것을 HPLE와 GC-MS를 통해 분리 동정하였다. 또한 B. subtilis HJ927는 가수분해효소인 ${\beta}$-1,3-glucanase를 분비해 냄으로서 위 화합물과 함께 식물을 보호하는 것으로 분석되었다.

전분의 겔화와 노화에 미치는 당류의 영향 (Effect of Saccharides on the Gelation and Retrogradation of Starch)

  • DSC를 이용하여 acom starch와 com starch 및 starch-saccharide-water system 의 겔화와 노화에 관한 열적 메카니즘을 알아보았다. 전분에 fructose와 maltose를 첨가한 starch-saccharide-water 계의 엔탈피를 측정한 결과, 당을 첨가하지 않은 경우의 엔탈피 값보다 컸으며 겔화 온도 역시 증가하였는데 이는 당이 물과 상호 작용하여 비결정성 영역에 흡수된 자유수가 감소하고 결정부분이 안정화되어 겔화가 일어나는 것을 지연시키기 때문이라고 생각되었다. 노화 엔탈피는 acorn starch 와 com starch에 대해 1일 ∼ 14일까지 저장시간에 따르는 변화를 관찰한 결과, 저장시간이 길어짐에 따라 엔탈피 값이 유의성 있게 증가하는 경향을 나타내었으며 이는 amylopectin 의 재결정화가 서서히 일어나기 때문으로 보였다. 또한 s-s-w system의 노화과정을 관찰한 결과, 저장시간이 길어짐에 따라 엔탈피 값이 증가하다가 7일이 지나면서부터는 일정해졌다. 이는 당이 amylopectin의 재결정화를 정지시켜서 노화를 지연시키기 때문으로 판단되었다. 당의 첨가가 노화에 미치는 영향은 fructose와 maltose 중에서 maltose의 노화지연 효과가 더 컸는데 이는 전분 겔 계를 안정시키는 junction zone의 수와 equatorial OH 수 및 활동적인 수화상태가 증가되는 요인을 maltose가 fructose보다 더 많이 갖고 있기 때문인 것으로 설명할 수 있었다.

  • The purpose of this study was to find the effect of honey on the sugar metabolism In Sprague-Dawley rats. All experimental rats were fed ad libitum, for seven weeks, 68% saccharide diet anti 10% or 20% honey from acacia, sumac and miscellaneous flower honey, respectively, and sucrose. The amount of glucose in whole blood was increased in all groups fed with honey except the group fed with 10% sumac honey solution. The amount of fructosamine in serum of rat taken diet with honey solution was increased in comparison with normal group, control group, sucrose group. The amount of fructosamine in serum of rat taken diet with honey solution was high in order of sumac honey > miscellaneous flower honey > acacia honey. The amount of lactate and pyruvate in whole blood of rat taken diet with sucrose group or honey group were decreased in comparison with the normal group and control group. The amount of $\beta$-hydroxybutyrate in serum of rat taken diet with sucrose or honey was increased in comparison with the normal group, control group.

Effect of Polysaccharide Elicitors on the Production of Decursinol Angelate in Agelica gigas Nakai Root Cultures

  • Root cultures of Angelica gigas Nakai were found to be sensitive to elicitation by poly-saccharide elicitors, such as methyl-$\beta$-cyclodextrin, glucan, carboxymethyl-$\beta$-chitin, chitosan, yeast extract and pectin. For the production of decursinol angelate, ca rboxymethyl-$\beta$-chitin and glucan were found to be the most efficient elicitors. The e nhanced accumulation of decursinol angelate was proportional to the increase of the phenylalanine ammonialyase (PAL) activity after the treatment with most of the elicitors. However, carboxymethyl-$\beta$-chitin treatment did not stimulate the PAL activity, despite the 1.6-fold increase in the decursinol angelate production.

감미료 소재로서 Di-D-Fructofuranose DianhydrideIII의 물리 및 생리적 특성

  • Some physical and physiological properties of di-D-fructofuranose dianhydrideIII (DFAIII), as a new sweetener, were investigated via in vitro experiments. The disaccharide was prepared by decomposing inulin with inulin fructotransferase (depolymerizing) from Arthrobacter sp. A-6. DFAIII had more excellent heat and acid stability than sucrose. This was one of the most desirable properties especially for the oligomer types of sweetener. DFAIII showed the least pH drop in the Streptococcus mutans culture, compared with the other saccharides examined. This indicates that the sugar will be fairly effective for preventing dental caries. The saccharide also had a selective Bifidus growth-promoting effect in PYF medium. Whereas, E. coli did not show growth promotion in the DFAIII-containing medium. In the co-culture of Bifidus longum and E. coli in the BL medium, Bifidus longum had a selective growth while the growth of E. coli appeared rather to be inhibited.

Antioxidant and Hypoglycemic Activity of Polysaccharide from Tea

  • Tea polysaccharide had high antioxidant activity and it could be used to cure diabetes. Antioxidant activity of tea poly-saccharide (TPS) from three kinds of tea (green tea, oolong tea and black tea) were compared, the result indicated that oolong tea polysaccharide (OTPS) had the highest antioxidant activity. In order to explicate the mechanism of antioxidant and hypoglycemic activity, the streptozotocin (STZ)-induced diabetes mice model (DM) was established. The influence of OTPS on blood-glucose, content of MDA and NO, and activities of GSH-PX, SOD, NOS in serum, kidney and liver were investigated. The result showed that after four weeks injection of OTPS to DM mice, the blood-glucose of three treatment group reduced by 14.5%,21.5% and 33.3%, respectively, comparing to the model control. The reduction effect of OTPS increased with the rise of dose. The activity of SOD and GSH-PX elevated significantly, while the activity of NOS decreased. The content of MDA and NO reduced significantly. The above results imply that antioxidant activity was enhanced. Comparing to XKW treatment, the effect of a dose of 300mg/(kg. bw) OTPS was much better. The research showed that the OTPS had a significant effect on reducing blood glucose, and could enhance the antioxidant activity of DM mice.

쥐의 비장세포로부터 $IL-1\beta$ 분비에 있어서 한국산 겨우살이 추출물 M11C (비렉틴 구성물질)의 효과 (Effect of M11C (Non-lectin Components) Obtained from Korean Mistletoe on the $IL-1\beta$ Secretion from Mouse Splenocytes)

  • 한국산 겨우살이 (Viscum album L)는 면역조절작용이 있음이 밝혀졌다. 본 연구에서 한국산 겨우살이 열탕추출물 M11C (non-lectin components)가 비장세포를 활성화시켜 $IL-1\beta$를 생산 분비하게 하는지를 조사하였다. 비장세포를 M11C로 자극한 후, 배양액을 수집 혹은 세포 용해물을 수거해 $IL-1\beta$ 분비와 전사량을 ELISA, immunoblotting, RT-PCR로 검사했었다. 비장세포로부터 $IL-1\beta$ 분비와 전사 효과에 있어서 M11C는 농도 의존성과 자극시간 의존성을 보였다. 비장세포로부터 최대의 $IL-1\beta$ 분비를 위한 M11C의 최대의 농도와 자극시간은 각각 $200{\mu}g/m\ell$와 8시간 이었다. 그리고 최대 $IL-1\beta$ mRNA 전사를 위한 M11C의 최대의 농도와 자극시간은 각각 $200{\mu}g/m\ell$와 4시간 이었다. 최대의 전사시간은 최대의 분비시간보다 4시간 빨리 도달된 것으로 나타났다. 이러한 최대의 $IL-1\beta$ 분비효과가 당분해효소인 Viscozyme L에 의해 완전히 저해되었다. 이는 M11C (non-lectin components)의 구성물질 들 중 다당체 혹은 올리고당들이 $IL-1\beta$ 생산 분비를 유도하는 주된 물질임을 말해주고 있다.

  • The purpose of this study was to find an effect of honey on the lipid metabolism of Sprague Dawley rats. All experimental rats were fed ad libitum, for seven weeks, 68% saccharide diet and 10% or 20% honey from acacia, sumac and miscelllaneous flower honey, respectively, and sucrose. The food efficiency ratio of rat taken diet with honey and high fructose of control group was increased in comparison with the control group. The concentration of cholesterol in serum of rats take총 diet with and high fructose of control group was more increased in comparison with the control and normal group. The concentration of H DL-cholesterol in serum of rats taken sumac honey was increased 57.0% in comparison with the control group, but the concentration of VLDL, LDL-cholesterol in serum of rats taken diet 10PA sumac honey was decreased 48.36% in comparison with the control group. The concentration of phospholipid in serum of rats taken diet with 20% acacia or 10% miscellaneous honey was increased 24.7, 16.25%, respectively, in comparison with the control group. The concentration of free fatty acid in serum of rats taken Inlet with sumac or miscellaneous honey and high fructose was increased in comparison with the comparison with the control group. The concentration supplemented diet with acasia honey was increase in comparison with the control group. The concentration of triglyceride in serum of rat was increased by feeding of honey. The concentration of triglyceride in liver was increased, but the level of phospholipid was decreased by feeding of honey.

당류계 계면활성제로 제조된 O/W 에멀젼의 구조 (Structures of OH Emulsion Prepared with Saccharide Surfactants)

  • 당류계 계면활성제인 sucrose distearate(S110), sucrose monostearate(S160), POE(20) ethyl glucose stearate(SSE20)과 오일성분인 알칸 계열의 탄화수소 hexane( $C_{6}$), decane( $C_{10}$), tetradecane( $C_{l4}$), octadecane( $C_{8}$)과 squalane (SQ) , liquid paraffin(LP) , octylpalminate(OP) , octylstearate(OS), alkylbenzoate(AB), isostearylbenzoate(ISB)를 사용하여 에멀젼을 제조하였으며 에멀젼 입자의 구조 형태는 광산란법을 이용하여 fractal 차원으로 나타내었다. 에멀젼 입자의 fractal 차원은 오일의 농도 및 탄소수와 오일의 비극성이 증가함에 따라 크게 나타났다. fractal 차원은 대부분 1.7~2.8으로 입자의 구조 형태는 fractal 구조를 이루고 있으나, S110계와 S160계의 Cla 50, LP의 fractal 차원은 3.0~3.2으로 내부 구조가 비교적 조밀한 회합체 구조를 형성하였다. S160계와 S110계의 fractal 차원은 2.I~2.6으로 서로 비슷한 fractal 구조를 형성하며, SSE20의 fractal 차원는 1.5~2.1으로 S160계 및 S110계 보다 덜 조밀한 회합체 구조를 이루는 것으로 나타났다. 입자의 구조 형태는 오일과 계면활성제의 종류에 영향을 받으며 계면활성제의 친수부가 동일하면 비슷한 구조를 형성하였으며 S160의 친수부 구조인 sucrose보다 SSE20의 polyoxyethylene group에 의한 장애효과가 더욱 크게 나타났다.다..
