Direct Intratumoral Injection of Ethanol in the Patients with Obstruction of Major Bronchus (주기관지 폐색환자에서 종양내 ETHANOL 주입치료 효과)

  • Background: For the relief of the occlusion of major bronchi, laser therapy, radiation therapy or combined therapy is generally used. But the effect of radiation therapy is very slow and not consistent and laser therapy requires expensive equipments and technical expertise with occasional serious side effects. Direct ethanol injection has been widely used for the control of bleeding in gastrointestinal lesion, esophageal varices or renal cell carcinoma with good results. So we tried direct injection of ethanol into the tumor to relieve the obstruction of major bronchus in 11 patients. Method: All procedures were done under the fiberoptic bronchoscopy with continuous oxygen supplement and aliquoted 0.5-1.0 ml of absolute ethanol directly into the tumor through the endobronchial aspiration needle. The tumor was endoscopically removed with a biopsy forceps immediately after ethanol injection. The whole procedure was repeated 3-4 days interval until the lumen opens. Usually after 2-3 trials of ethanol injection, the lumen opened up. Results: The immediate effect of ethanol injection was whitening of the mucosa and prompt cessation of bleeding. The late effect was necrosis of the tumor. The final results of this procedure were improvement of symptoms and reexpnasion of the lung in all patients. $FEV_{1.0}$ and FVC were improved and $PaO_2$ was increased from $68.1{\pm}9.2$ mmHg to $83.9{\pm}8.1$ mmHg, $SaO_2$: from $94{\pm}8.5%$ to $96.6{\pm}1.1%$, and $AaDO_2$ was reduced from $26.5{\pm}8.5$ mmHg to $10.9{\pm}9.1$ mmHg. Conclusion: Direct ethanol injection into the tumor tissue is a rapid, cheap and relatively safe method of relieving the complete occlusion of major bronchus.

Inhibitory Effect of Rice Extract on the Chemically Induced Mutagenesis (쌀 추출물의 돌연변이 억제효과)

  • The inhibitory effects of rice extract on mutagenicity induced by 3-amino-1,4-dimethyl-5H-pyrido [4,3-b]indole(Trp-P-1), 3-amino-1-methyl-5H-pyrido [4,3-b]indole(Trp-P-2), sodium azide(SA), 2-nitrofluorene(2NF), mitomycin C(MMC), aflatoxin $B_1(AFB_1)$ and 4-nitroquinoline oxide(4-NQO) were investigated using Salmonella typhimurium reversion assay, SOS chromotest and spore rec-assay. In Salmonella typhimurium reversion assay, methanol extract from brown rice (Illpumbyeo, Japonica variety) showed the highest inhibitory effect among other extracting solvent including hexane, chloroform and water. Methanol extract showed stronger inhibitory effect, above 85%, on indirect-acting mutagens(Trp-P-1, Trp-P-2 and $AFB_1$) than those on direct-acting mutagens(4-NQO, 2NF). In SOS chromotest, methanol extracts showed $77.6{\sim}88.9%$ effects on SOS function induced by Trp-P-1, Trp-P-2, $AFB_1$ and 4-NQO. In spore rec-assay, methanol extracts inhibited the mutagenicity induced by $AFB_1$ and MMC. As the concentration of methanol extract increased, inhibitory effect on mutagenicity increased but reached at steady state as inhibition rate of 90% when the concentration was above 5 mg/plate. In inhibitory effects of methanol extracts by various rice varieties, all of 11 varieties turned out to have inhibitory effect on mutagenicity. There was no significant difference (p>0.05) in inhibitory effect of methanol extracts between brown and white rice against Trp-P-1, but showed difference (p<0.05) against 4-NQO.

Development of Self-Repairing Smart Concrete Using Micro-Biologically Induced Calcite Precipitation (미생물의 방해석 석출 작용을 이용한 자기보수 스마트 콘크리트 개발에 관한 연구)

  • This paper presents a study on the development of next generation smart concrete in an eco-friendly manner using micro-biologically induced calcite precipitation (MICP) via microbial biomineralization. It seems that currently, the reformation and functional improvement of concrete using MICP can be achieved using Sporosarcina pasteurii, which is a representative microorganism that produces calcite precipitation. Based on previous studies on MICP the biochemical tests and crystallinity evaluation of cement using sporoasrcina pasteurii and four additional micro-organisms from the concrete structures as identified by 16S rDNA sequence analysis were conducted. Also by applying the Sporosarcina pasteurii and separated four effective micro-organisms from the concrete structures to mortar, the compressive strength improvement by varying curing conditions, repair of crack were examined, and plans for future study were suggested. The effect of the application of effective micro-organisms can lead to the development of a new material that will contribute to resolution of environmental problems and facilitate repair work, and this can also serve as a new research theme in the future. In addition, the importance of this study is to use micro-organism, which is found common in concrete structures, this new microbial is not only environmentally safe but also persists in the natural environment for an extended period of time. Therefore, it seems to have a great potential to became a new environmentally low-burdened functional material.

Effects of Cumulus Cells and Reactive Oxygen Species (ROS) on Plasminogen Activator Activity during In Vitro Maturation of Porcine Oocytes

  • Plasminogen activators (PAs) are serine proteases that convert plasminogen to plasmin. The PA/plasmin system has been associated with a number of physiological processes such as fibrinolysis, ovulation and fertilization. Although correlations have been reported between reactive oxygen species (ROS) and oocyte maturation, the relationship between PA activity and ROS is unknown. The present study was undertaken to determine the effects of cumulus cells on PA activity in matured porcine oocytes under xanthine (X)-xanthine oxidase (XO) system. When oocytes were matured under the X-XO system, the proportion of oocytes remaining GV stage was higher (p<0.05) in oocytes without cumulus cells. The incidence of degenerated oocytes was higher (p<0.05) in the X+XO ($11.1{\pm}6.1$ and $21.6{\pm}3.4%$) than in the control group ($2.9{\pm}1.8$ and $4.0{\pm}1.6%$). The proportion of TUNEL-positive oocytes and activity of caspase-3 were higher (p<0.05) in cumulus-free oocytes and oocytes exposed to ROS. Tissue-type plasminogen activator-plasminogen activator inhibitor (tPA-PAI) and tissue-type plasminogen activator (tPA) activity were detected in oocytes that were separated from cumulus-oocytes complexs (COCs) at 44 h of maturation culture, and only tPA was produced in oocytes that were denuded before the onset of maturation culture. On the other hand, the activities of PA were increased (p<0.05) when oocytes were cultured under the X-XO system. The higher activity of tPA was observed in denuded oocytes (DOs) underwent apoptotic changes by oxidative stress. In COCs, however, tPA-PAI as well as tPA activity was detected and apoptotic changes such as DNA cleavage or caspase-3 activation were not observed. These results suggest that tP A may be relevant to apoptotic cell death in porcine oocytes by oxidative stress.

Screening of Extraction Solvent Condition of Fermented Rhus verniciflua Stem Bark by Antioxidant Activities (항산화활성 비교를 통한 발효 칠피의 추출용매 조건 탐색)

  • In this study, we investigated antioxidant capacity and determined the phenolic and flavonoid contents using each of various solvent conditions from fermented Rhus verniciflua stem bark (F-RVS). Each extracts displayed markedly similar content of extraction yield. However, M80 extract showed a significantly higher antioxidant activity in comparison to other extract investigated. M80 exhibited strong DPPH radical scavenging activity with $RC_{50}$ value of $10.5{\pm}1.4{\mu}g/m{\ell}$, reducing power value 1.04 Abs (concentration of 1 mg/$m{\ell}$), and anti-lipid peroxidation activity value of 94.6% (concentration of 10 mg/$m{\ell}$). M80 extract showed the high content of total phenolic (319.7 mg GAE/$m{\ell}$ extract) and total flavonoid (111.6 mg QE/$m{\ell}$ extract). Phenolic and flavonoid compounds showed significant relationship in DPPH radical scavenging ($R^2$=0.911 and 0.912, each extract) and reducing power ($R^2$=0.903 and 0.837, each extract) from fermented R. verniciflua stem bark. However, antilipid peroxidation activity ($R^2$=0.589 and 0.441, each extract) was not significant like DPPH radical scavenging and reducing power. Therefor the result indicated that the potential antioxidant activities and functional values were ovserved significantly at M80 solvent condition from the fermented R. verniciflua stem bark.

Effects of Reactive Oxygen Species (ROS) on Sperm Function and Plasminogen Activator Activity in Porcine Spermatozoa

  • Plasminogen activators (PAs) are serine protease that cleave plasminogen to form the active protease plasmin and may participate in mammalian fertilization. Although correlations have been reported between reactive oxygen species (ROS) and sperm function, the relationship between PA activity and ROS is unknown. We determined the effects of ROS on sperm function and PA activities in boar spermatozoa preincubated under the X-XO system. When spermatozoa were treated with the X+XO group, a significant increase (p<0.05) was observed in the percentage of acrosome reacted spermatozoa compared with that of the control group. However, when antioxidants were added to the medium with X+XO, the rate of acrosome reaction tended to decrease. Also, a significantly lower percentage of acrosome reacted spermatozoa was observed in the X+XO+catalase group at 6 hr of incubation compared with that of X+XO group. The density of malondialdehyde (MDA) was higher in the X+XO group than in different treatment groups. In another experiment, incubation of spermatozoa in medium with X+XO was associated with a significant (p<0.05) increase in activity of tPA-PAI and tPA compared with the control group. Antioxidants decreased the increased activity of tPA-PAI and tPA by preincubation in the X-XO system. Also, a significantly lower (p<0.05) activities of tPA-PAI and tPA were observed in the X+XO+catalase group compared with the X+XO group. No significant differences, however, were observed in the activity of uPA. These results suggest that the increase of acrosome reaction by the X-XO system resulted in increase of PAs activity in the sperm incubation medium.

Relationship between In Vitro Maturation and Plasminogen Activator Activity on Porcine Cumulus-Oocytes Complexes Exposed to Oxidative Stress

  • This study was undertaken to evaluate the relationship between in vitro maturation and plasminogen activators (PAs) activity on porcine cumulus-oocytes complexes (COCs) exposed to oxidative stress. When COCs were cultured in maturation medium with hydrogen peroxide ($H_2O_2$), the proportion of the germinal vesicle breakdown (GVBD) and oocytes maturation were decrease with addition of $H_2O_2$, and were significantly (p<0.05) lower in medium with 0.1 mM $H_2O_2$ than control group. Also, the rate of degenerated oocytes was increased in as $H_2O_2$ concentration in eased. When COCs were cultured for 48 h, three plasminogen-dependent lytic bands were observed: tissue-type PA (tPA); urokinase-type PA (uPA); and tPA-PA inhibitor (tPA-PAI). PA activity was quantified using SDS-PAGE and zymography. When $H_2O_2$ concentration was increased, tPA and tPA-PAI activities also increased in porcine oocytes cultured for 48 h, but not uPA. In other experiment, embryos were divided into three groups and cultured in (1) control medium, (2) control medium with 1.0 mM $H_2O_2$ and (3) control medium with 1.0 mM $H_2O_2$ along with catalase in concentrations of 0.01, 0.1, and 1.0 mg/ml, respectively. $H_2O_2$ decreased the rate of GVBD and maturation in porcine COCs but catalase revealed protective activity, against oxidative stress caused by $H_2O_2$. In this experiment, tPA and tPA-PAI activities were higher in media with 1.0 mM $H_2O_2$ alone. Increasing concentration of catalase decreased tPA and tPA-PAI activities in porcine oocytes. These results indicate that the exposure of porcine follicular oocytes to ROS inhibits oocytes maturation to metaphase-II stage and increase the oocytes degeneration. Also, we speculated that increased ROS level may trigger tPA and tPA-PAI activities in porcine oocytes matured in vitro.

RPSA Gene Mutants Associated with Risk of Colorectal Cancer among the Chinese Population

  • The primary aim of this study was to evaluate the relationship of single nucleotide polymorphisms (SNPs) in ribosomal protein SA (RPSA) gene with colorectal cancer (CRC). A case-control study including 388 controls and 387 patients with CRC was conducted in a Chinese population. Information about socio-demography and living behavior factors was collected by a structured questionnaire. Three SNPs (rs2133579, rs2269349, rs7641291) in RPSA gene were genotyped by Illumina SnapShot method. Multiple logistic regression models were used for assessing the joint effects between tea consumption and SNPs on CRC. The subjects with rs2269349 CC genotype had a decreased risk for CRC (OR=0.60; 95%CI = 0.37-0.99), compared with TT/CT genotype after adjustment for covariates. A similar association of rs2269349 with rectal cancer was observed (OR=0.49; 95%CI=0.24-1.00). Further analyses indicated that this SNP could modify the protective effect of tea drinking on CRC. Among the subjects with rs2269349 TT/CT or rs2133579 AA/GA, there was a marginal significantly lower risk of CRC (OR and 95%CI: 0.63 and 0.39-1.01 for rs2269349; 0.64 and 0.40-1.02 for rs2133579) in tea-drinking subjects in comparison to non-tea-drinking subjects. Mutants in the RPSA gene might be associated with genetic susceptibility to CRC and influence the protective effect of tea consumption in the Chinese population.

Metabolism of $C^{14}$-acetate in the Ehrlich ascites tumor (에르릿히 복수암에 있어서 $C^{14}$-초산염 대사)

  • Tissue homogenates of Ehrlich ascites tumor tissues and several normal tissue of mice were incubated separately in medium maintaining $C^{14}$_acetate concentrations of 5, 10, 20, 30, 40, 50 and 60 mg%, in order to determine maximum oxidative rates of acetate. In every incubation experiments, respiratory $CO_2$ samples rapped by alkaline which was placed in the center well of the incubation blask were analyzed for total $CO_2$ Production rates and their radoactivies. The fractions of $CO_2$ from medium acetate to total $CO_2$ production rate were obtained with relative specific activities (RSA) which were calculated by ratio between specific activities (SA) of $CO_2$ and medium $CO^{14}$_acetate and $CO_2$ production rates from medium acetate were calculated from RSA and total $CO_2$ production rates. Maximum plateau values of oxidative rates described above were determined at incubation experiments of various concentrations of medium acetate and compared the oxidative rates of acetate of tumor with those of normal tissues such as kidney, brain and liver. Maximum plateau values of total $CO_{2}$ Production rates were obtained at acetate concentration of 20 mg% and represent $25.0{\pm}0.54\;{\mu}M/hr/gm$ in the brain, $16.3{\pm}2.5$ in the kidney, $9.1{\pm}1.78$ in the liver and $11.5{\pm}3.2\;{\mu}M/hr/gm$ in the ascites tuners. Substancial $CO_2$ yield was observed in the tumor tissues as in the normal tissues. On the other hand, plateau values of RSA were $25.7{\pm}1.04%$ in thee brain, $9.1{\pm}0.72%$ in the kidney, $2.5{\pm}0.73%$ in the liver and $0.51{\pm}0.12%$ in the tumor tissues. $CO_2$ yields from the medium acetate, were 4.19 in the kidney, 2.28 in the brain, 0.228 in the liter and $0.059\;{\mu}M/hr/gm$ in the tumor tissue. These show wide range even in the normal tissue but remarkable decrease in the tumor tissue. This fact means that further oxidation of acetate was inhibited remarkably in the tumor tissue.

  • This study was conducted to develop self-healing ability of concrete so that inspection could be available even in the event of minute cracks without complex works at any time for more economic concrete structure maintenance and longevity. A completely different method has been carried out in comparison with many of similar researches on self-healing concrete. This is a basic study on the development of self-healing concrete using microbial biomineralization. Compounds were generated except for cells by precipitation reaction of CaC$O_3$ during the microbial metabolism and we examined the use as a binder that hardens the surface of sand using biomineralization that Sporosarcina pasteurii precipitates CaC$O_3$. In result, the formation of new mineral and hardening of sand surface could be verified partly, and it was available for cracks to be repaired by calcite with organic (microorganism) and inorganic (CaC$O_3$) complex structure through the basic experiment a little bit. Therefore the use of biomineralization by this sort of microbial metabolism for concrete structure helps to develop absolute repair-concrete like this concrete with microorganism. The effect of microbial application will be one of the most important research tasks having influence on not only repair for concrete structure but also development of new materials able to reduce environmental problems.
