Profiling of glucose-induced transcription in Sulfolobus acidocaldarius DSM 639

  • Sulfolobus species can grow on a variety of organic compounds as carbon and energy sources. These species degrade glucose to pyruvate by the modified branched Entner-Doudoroff pathway. We attempted to determine the differentially expressed genes (DEGs) under sugar-limited and sugar-rich conditions. RNA sequencing (RNA-seq) was used to quantify the expression of the genes and identify those DEGs between the S. acidocaldarius cells grown under sugar-rich (YT with glucose) and sugar-limited (YT only) conditions. The functions and pathways of the DEGs were examined using gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses. Quantitative real-time PCR (qRT-PCR) was performed to validate the DEGs. Transcriptome analysis of the DSM 639 strain grown on sugar-limited and sugar-rich media revealed that 853 genes were differentially expressed, among which 481 were upregulated and 372 were downregulated under the glucose-supplemented condition. In particular, 70 genes showed significant changes in expression levels of ${\geq}$ twofold. GO and KEGG enrichment analyses revealed that the genes encoding components of central carbon metabolism, the respiratory chain, and protein and amino acid biosynthetic machinery were upregulated under the glucose condition. RNA-seq and qRT-PCR analyses indicated that the sulfur assimilation genes (Saci_2197-2204) including phosphoadenosine phosphosulfate reductase and sulfite reductase were significantly upregulated in the presence of glucose. The present study revealed metabolic networks in S. acidocaldarius that are induced in a glucose-dependent manner, improving our understanding of biomass production under sugar-rich conditions.

Exploring parenting variables associated with sweetness preferences and sweets intake of children

  • BACKGROUND/OBJECTIVES: Excessive sugar intake is one of the causes associated with obesity and several chronic diseases prevalent in the modern society. This study was undertaken to investigate the effect of parenting variables based on the theory of planned behavior, on the sweetness preferences and sweets intake of children. SUBJECTS/METHODS: Parents and their children (n = 103, aged 5-7 years) were enrolled to participate in a survey for this study, after providing the required informed consent. Parents were asked to fill out a self-administered questionnaire at their residence. The sweetness preference test for children was conducted at a kindergarten (or daycare center) by applying the one-on-one interview method. RESULTS: The children were divided into two clusters categorized by the K-mean cluster analysis: Cluster 1 had higher sweetness preference (0.42 M sugar, 35%; 0.61 M sugar, 65%); Cluster 2 exhibited lower sweetness preference (0.14 M sugar, 9.5%; 0.20 M sugar, 9.5%; 0.29 M sugar, 81%). Cluster 1 had a higher frequency of sweets intake (P < 0.01), and lower sweets restriction (P < 0.05) and nutrition quotient score (P < 0.05). Sweets intake was negatively correlated with the nutritional quotient (r = -0.204, P < 0.05). The behavioral intention of parents was higher in cluster 2 (P < 0.05), while affective attitude, feeding practice, and reward were higher in cluster 1 (P < 0.001, P < 0.05, and P < 0.01, respectively). Furthermore, behavioral intention of parents showed a negative correlation with affective attitude (r = -0.282, P < 0.01) and feeding practice (r = -0.380, P < 0.01), and a positive correlation with subjective norm (r = 0.203, P < 0.05) and parenting attitude (r = 0.433, P < 0.01). CONCLUSIONS: This study indicates that the sweetness preferences and sweets intake of children is related to the parent's affective attitude, feeding practice and reward. We suggest that to reduce the sugar consumption of children, guidelines for access to sweets and pertinent parenting practices are required.

고등학생의 스트레스, 당류섭취행동 및 주관적 구강건강상태와의 융합적 관계 (The Convergence Relationship on Stress, Sugar Intake Behaviors, and Oral Health Status in High school Students)

  • 본 연구는 고등학생들의 스트레스, 당류섭취행동 및 주관적 구강건강상태와의 상관성을 파악하여 당 섭취 저감화 실천 영양지도를 연계한 구강건강관리 프로그램을 위한 기초자료로 활용하고자 한다. 자료 수집은 전북지역의 고등학생 273명을 대상으로 조사하였고 SPSS 18.0 프로그램을 사용하여 일원배치분산분석, 피어슨의 상관분석을 하였다. 스트레스는 당류섭취행동과 양의 상관관계가 있는 것으로 나타났고, 주관적 구강건강상태와는 음의 상관관계가 있는 것으로 나타났다. 당류섭취행동과 주관적 구강건강상태는 음의 상관관계를 나타냈다. 고등학생들의 스트레스, 당류섭취행동과 주관적 구강건강상태는 서로 연관성이 있는 것으로 나타났다. 이에 올바른 생활습관과 건강한 식생활 형성을 위한 체계적인 당 저감화 영양교육이 갖추어진 구강건강프로그램이 마련되고, 지속적인 홍보와 지도가 필요할 것으로 사료된다.

Application of Decision Trees for Prediction of Sugar Content and Productivity using Soil Properties for Actinidia arguta 'Autumn Sense'

  • Environmental conditions are important in increasing the fruit sugar content and productivity of the new cultivar Autumn Sense of Actinidia arguta. We analyzed various soil properties at experimental sites in South Korea. A Pearson's correlation analysis was performed between the soil properties and sugar content or productivity of Autumn Sense. Further, a decision tree was used to determine the optimal soil conditions. The difference in the fruit size, sugar content, and productivity of Autumn Sense across sites was significant, confirming the effects of soil properties. The decision tree analysis showed that a soil C/N ratio of over 11.49 predicted a sugar content of more than 7°Bx at harvest time, and soil electrical capacity below 131.83 µS/cm predicted productivity more than 50 kg/vine at harvest time. Our results present the soil conditions required to increase the sugar content or productivity of Autumn Sense, a new A. arguta cultivar in South Korea.

반응표면분석에 의한 덖음 모시풀잎 가루 첨가 머핀의 품질특성 (Quality Characteristics of Muffins with added Dukeum (pan-fired) Ramie Leaf (Boehmeria nivea) Powder using Response Surface Methodology)

  • The objective of this study was to produce muffins with added dukeum ramie leaf powder and to compare them to a control. We used two different optimal composition ratios to produce muffins. The physical and sensory characters were varied by changing the ratio of dukeum ramie leaf powder, butter, and sugar to identify a critical value using response surface regression analysis. The ranges for the added dukeum ramie leaf powder, sugar, and butter were 15-35, 60-100, and 60-100 g, respectively. Muffins with added dukeum ramie leaves had comparable mineral and antioxidant vitamin compositions to those of dried ramie leaves. Muffins with added dukeum ramie leaf powder satisfied the suitability of the model, showing a p-value within 5% of significance level (p<0.05) for every aspect Volume and height differed without any association to ingredients. Lightness increased when less dukeum ramie leaf powder was added with more butter and sugar, redness increased when more dukeum ramie leaf powder and sugar were added with 70-80 g of butter, whereas yellowness increased when less dukeum ramie powder was added. Texture and toughness increased when less dukeum ramie leaf powder was added. The degree of gumminess and chewiness depended hugely on the amount of sugar. Cohesiveness, decreased with more dukeum ramie leaf powder and butter, whereas hardness increased with a medium range of ingredients. Sensory character increased with less dukeum ramie powder, and overall quality increased with more butter and sugar, but depended on the amount of added dukeum ramie power. The critical values from the sensory characteristics showed that 27 g of dukeum ramie powder, 72 g of sugar, and 86 g of butter was optimum.

대체당 첨가 고령자용 포도젤리의 품질 특성 (Quality Characteristics of Grape Jellies with Sugar Derivative Sweeteners for the Elderly)

  • This study was conducted to determine the quality characteristics of grape jellies for the elderly. The jellies contained sugar (control) or sugar derivative sweeteners (erythritol, isomaltooligosaccharide, sorbitol, and xylitol). Agar (0.31%) and $\kappa$-carrageenan (0.27%) were the gelling agents. The average age of the subjects participating in the acceptance test was 79. The lightness (L), redness (a), and yellowness (b) values of the agar gel with erythritol mostly decreased, indicating a darker and pale red color. The L and b values of the carrageenan gel with sugar derivative sweeteners increased, indicating brighter and yellowish color. The agar and carrageenan gels with sorbitol showed higher gelling and melting temperature, indicating that gelation occurred easily and did not easily melt. The agar and carrageenan gels with xylitol showed a low-melting temperature, indicating low stability with temperature change. The break-down rate of the agar and carrageenan gels with erythritol was low, whereas that of agar gel with sorbitol was relatively high despite its high melting temperature. Hardness, cohesiveness, gumminess, and chewiness of the gels with sugar derivative sweeteners decreased, and this tendency was most distinct with isomaltooligosaccharide in the agar gel and with sorbitol in the carrageenan gel. The rupture properties of the gel were the same as the hardness of the gel. Sensory acceptance of the agar gels with erythritol, sorbitol, isomaltooligosaccharide, and the carrageenan gel with erythritol was fairly high, whereas that of the agar gel with xylitol and the carrageenan gel with isomaltooligosaccharide and xylitol was low. The results show that sorbitol and erythritol are appropriate as sugar substitutes in grape jellies for the elderly about the acceptability and stability of the gels.

대체 감미료를 활용한 저당 유자 시럽 제조 및 이화학적 특성 조사 (Production Process and Physiochemical Characterization of Low-sugar Yuzu Syrup)

  • Yuzu (Citrus junos) undergoes traditional processing to create preserved yuzu with a sugar content of 50%. This study aimed to produce low-sugar yuzu syrup using artificial or natural sweeteners instead of sugar. Among artificial sweeteners, maltitol showed minimal browning and lack of deposition at high temperatures. The addition of xanthan gum (0.2%) and carboxymethyl cellulose (0.2%) resolved the issue of layer separation and viscosity decrease of yuzu syrup. Alternatively, grain syrup, a natural sweetener, improved viscosity and homogeneity without the additives. Yuzu syrups were developed using yuzu juice and preserved yuzu, with maltitol (20~40%) or grain syrup (50~70%) as the sweetener. Yuzu syrups containing 35% maltitol (M35) or 55% grain syrup (G55) had less than 5% and 10% free sugar, respectively. These syrups exhibited taste patterns similar to commercial yuzu syrup in analysis using an electronic tongue. Furthermore, M35 and G55 contained yuzu flavonoids at concentrations of 19.82 mg/g and 24.09 mg/g, respectively. Antioxidant activity (DPPH, ABTS radical scavenging) of M35 and G55 was equivalent to 10.55, 17.59 mg/100 g of Vitamin C and 97.39, 33.92 mg/100 g of Vitamin C, respectively. Consequently, M35 and G55 offer promising alternatives to preserved yuzu, providing low-sugar yuzu syrups enriched with functional ingredients.

서울, 경기지역의 초등학생 대상 당 저감화에 관한 소비자 인식 및 관련 교육 효과 분석 (Analysis of the consumer perception and related education effect on the reduction of sugar for elementary school students in Seoul and Gyeonggi-do)

  • 본 연구는 초등학교의 당 관련 영양교육이 당 섭취에 관한 영양지식과 영양태도, 식행동에 어떠한 영향을 미치는지를 평가하기 위해 당 섭취에 관한 설문조사를 실시하였고, 당 교육인지군과 비인지군으로 나누어 영양지식과 식행동에 어떠한 차이가 있는지를 분석함으로써 초등학생들의 당류 섭취에 영향을 미치는 요인을 평가하고자 하였으며, 본 연구결과는 다음과 같다. 본 조사 대상자 중 당 교육을 받았다고 인지한 교육 인지군은 270명 (49.8%) 이었고, 비인지군은 302명 (50.2%) 이었다. 당 교육 인지 여부에 따른 이론적 지식 점수를 보면 교육 인지군 3.67점, 비인지군 3.55점으로 교육 인지군이 비인지군에 비해 유의적으로 높았으나, 바른 행동에 대한 지식 점수는 두 군간에 유의적인 차이를 보이지 않았다. 당류 섭취 관련식행동 점수, 음료 선택 및 간식 구매 빈도는 당 교육 인지 여부에 따른 두 군 간에 유의적인 차이를 보이지 않았으나, 영양표시에서 당류 함량을 확인하는 비율은 교육 인지군이 31.2%로 비인지군 15.4%에 비해 유의적으로 높았다. 아이들이 즐겨 섭취하는 식품군별 섭취 빈도를 보면 교육 인지군이 비인지군에 비해 무가당 캔디나 젤리류, 생과일류의 섭취 빈도가 유의적으로 높았으며, 식품군별 당류 섭취량은 달지 않은 빵류, 흰 우유 및 생과일을 통한 당류 섭취량이 비인지군에 비해 교육 인지군에서 유의적으로 높았다. 당 관련 지식 점수와 식행동 사이의 상관성을 보면 교육 인지군의 경우 식행동 점수, 아이스크림, 초콜릿, 탄산음료를 덜 먹으려는 행동과는 양의 상관성을, 방과 후 간식 구입 빈도, 평균 당류 섭취량과는 음의 상관성을 보였으며, 비인지군의 경우 식행동 점수, 아이스크림, 빵을 먹을 때 잼을 덜 먹는 행동 및 과일 섭취 시 설탕을 덜 먹으려는 식행동과 양의 상관성을, 방과 후 간식구입 빈도 및 당류 섭취량과는 음의 상관성을 보였으나, 비인지군에 비해 교육 인지군에 좀더 높은 상관계수 값을 보였다. 또한 하루 총 당류 섭취량에 영향을 미치는 요인을 분석한 결과 교육 인지군에서만 영양(교)사의 교육 여부가 총 당류 섭취량을 감소시키는 요인으로 분석되었다. 본 연구결과 당 교육을 인지한 대상자들이 비인지군에 비해 당류 함량이 낮은 식품을 섭취하려고 노력하는 것으로 나타나고 있으며, 영양교사의 교육 여부가 당류 섭취량에 영향을 미치는 중요한 요인으로 분석되고 있다. 그러므로 초등학생들의 당류 섭취량 감소를 위해서는 학교의 영양 전문가인 영양(교)사에 의해 지속적이고, 행동으로 실천할 수 있는 실천 중심의 영양교육이 수행될 필요가 있으며, 학교 현장에서 손쉽게 활용할 수 있는 다양한 형태의 당 관련 영양교육 자료의 개발 및 보급이 수행되어야 할 것이다.

진흥(振興)과 IR 667의 엽초(葉鞘) 및 절간(節間)의 당(糖) 및 전분함량(澱粉含量)과 등숙환경(登熟環境) (Sugar and starch in leaf-sheaths and internode of Jinheung and IR667 rice under various ripening environments)

  • 진흥(振興)과 IR667-수원(水原) 214의 작기이동재배(作期移動栽培)에서 출수기(出穗期)와 출수(出穗)4주후(週後)에 절위별(節位別) 엽초(葉鞘) 및 절간(節間)의 당(糖) 및 전분함량(澱粉含量)에 따라 탄수화물대사(炭水化物代謝)에 있어 고당형(高糖形)(당(糖)>전분(澱粉)), 당경향(糖傾向)(당함량증가경향(糖含量增加傾向)), 고전분형(高澱粉形)(전분(澱粉)>당(糖)), 전분경향등(澱粉傾向等) 분류(分類)의 타당성(妥當性)을 재검토(再檢討)한 결과(結果) 다음과 같다. 1) 진흥(振興)보다 IR667이, 엽초(葉鞘)보다 절간(節間)이, 조기(早期)보다 만기(晩期)가 출수기(出穗期)보다 4주후(週後)가 저온(低溫)에서보다 고온(高溫)에서 당경향(糖傾向)이었다. 따라서 출수기(出穗期)에는 조기(早期) 및 만기재배(晩期栽培)의 진흥엽초(振興葉鞘)와 절간(節間)이 고전분형(高澱粉形)이고 조기(早期) IR667의 하위절간(下位節間) 및 만기(晩期) IR667의 엽초(葉鞘)와 절간(節間)은 고당형(高糖形)이었다. 극만기(極晩期)에서는 진흥출수기제(振興出穗期第)1절간(節間)을 제(除)한 두 품종의 모두 절간(節間)이 고전분형(高澱粉形)이었다. 출수(出穗)4주후(週後)에는 진흥(振興)의 제1(第) 및 4위엽초(位葉鞘)를 제외(除外)한 두품종의 모든 엽초(葉鞘)와 절간(節間)이 고당형(高糖形)이었다. 조기(早期) IR667과 만기(晩期) 두품종이 출수(出穗)4주후(週後)에 절간(節間) 보다 엽초(葉鞘)에서 고당형(高糖形)이 강화(强化)되었다. 2) 상위(上位)3절위(節位) 엽초(葉鞘) 및 간(稈)이 탄수화물전류(炭水化物轉流)에서 같은 방향(方向)으로 작용(作用)하는 것 같다. 그들간엔 상위(上位)로 갈수록 출수기(出穗期)에는 당경향(糖傾向)을, 4주후(週後)에는 전분경향(澱粉傾向)을 보이며 이러한 현상은 진흥(振興)에서 뚜렷하였다. 3) 작기(作期)가 늦을수록 탄수화물함량(炭水化物含量)(당(糖)+전분(澱粉))이 높고 이 경향(傾向)은 출수(出穗)4주후(週後)와 IR667에서 뚜렷하여 저온(低溫)에 의(依)한 전류조해(轉流阻害)를 나타내는것 같다. 4) 수(穗)로의 전류속도(轉流速度)(일당입중증가(日當粒重增加))는 단기재배(單期栽培)에서 빠르고 진흥(振興)보다 IR667이 빨리 최고속도(最高速度)에 도달(到達)하였다. 5) 작기(作期)가 늦을수록 수량(收量)에 대(對)한 출수전(出穗前) 탄수화물(炭水化物)의 기여율(奇與率)은 커지며 엽초중심형(葉鞘中心形)인 IR667이 언제나 컸다. 6) 당(糖)/전분비(澱粉比)는 주(主)로 품종(品種)의 대사특성(代謝特性)으로, 이차적(二次的)으로 그러나 상당한 정도로 환경요인(環境要因)에 의하여 결정(決定)되는것 같다.
