The Relationship Between the NF-${\kappa}B$ Activity and Anti-inflammatory Action of Surfactant in the Acute Lung Injury of Rats (백서의 급성폐손상에서 surfactant의 항염증작용과 호중구의 NK-${\kappa}B$ 활성과의 관계)

  • Background : The therapeutic effects of surfactants on acute lung injury derive not only from their recruiting action on collapsed alveoli but also from their anti-inflammatory action in the alveolar sapce. This study evaluated the anti-inflammatory action of a surfactant in an acute lung injury model of rats by neutrophils were recollected from the BAL fluid and the NF-${\kappa}B$ activity of the neutrophilic nuclear protein was evaluated. Methods : Male Sprague-Dawley rats weighing approximately 300 gram were divided into 3 groups, which consisted of 6 rats respectively. In the control group, normal saline(3ml/kg) was instilled into the trachea twice with 30 minute interval. In two other groups, acute lung injury was induced by the intra-tracheal instillation of LPS(5mg/kg). Thirty minutes later, either a surfactant(ST group; 30mg/kg) or normal saline(NT group: 3ml/kg) was instilled via the trachea. Twenty-four hours after the LPS instillation, the BAL fluid was retrieved to measure the WBC count and cytokine(IL-$1{\beta}$ and IL-6) levels. The neutrophils were isolated from the BAL fluid and the nuclear protein was extracted to evaluate the NF-${\kappa}B$ activity using a eletrophoretic mobility shift assay(EMSA). Results : The WBC count of the BAL fluid of the ST group($3,221{\pm}1,914{\times}10^3/{\mu}l$) was higher than that of the control group($356{\pm}275{\times}10^3/{\mu}l$)(p<0.05) and lower than that of the NT group($5,561{\pm}1,757{\times}10^3/{\mu}l$)(p<0.05)). The BAL fluid level of IL-$1{\beta}$ from the NT group($2,064{\pm}1,082pg/ml$) was higher than those of the ST group($360{\pm}234pg/ml$)(p<0.05) and the control group(0pg/ml)p<0.05) and control group($49{\pm}62pg/ml$)(p<0.05). The NF-${\kappa}B$ activity of the neutrophilic nuclear protein in the ST group and NT group was similar. Conclusion : The surfactant, attenuates the alveolar inflammation in the acute lung injury of rats model. However, its anti-inflammatory action does no't appear to be mediated by the inhibition of NF-${\kappa}B$ activity.

Studies on the effect in degree of saturation of fats on serum cholesterol level in the rabbit (토끼의 혈청 콜레스테롤 함량에 미치는 지방의 포화도에 관한 연구)

  • The effect in degree of saturation and unsaturation of dietary added oils on the serum cholesterol level in the rabbit was studied for a kperiod of 4 weeks using isocalories and isonitrogenous diets. The subject rabbits were divided into 10 feeding groups such as control-1 (Basal diet only), A group (Basal+sesame oil), B group (Basal+perilla oil), C group (Basal+soybean oil), D group (Basal+rice bran oil), Control-A(Basal+casein), A-1 group (Basal+sesame oil+ casein), B-1 group (Basal+perilla oil+casein), C-1 group (Basal+soybean oil+casein) and D-1 group (Basal+rice bran oil+casein). The results are summarized as follows: 1. Body weight gains per week of the perilla oil fed group were higher than anyother groups during the experimental period. 2. Food efficiency ratios for the group of perilla oil fed were 1.041, 0.781, 0.520 and 0.431 for 1st, 2nd, 3rd and 4th week, respectively. 3. In the group of perilla oil and Casein fed, food efficiency ratios for the experimental period were 0.887, 0.823, 0.489 and 0.437 for 1st, 2nd, 3rd and 4th week, respectively. 4. It is investigated that the food efficiency ratio for perilla oil fed groups was higher than the group of perilla oil and casein fed. 5. Calorie efficiency ratios for perilla oil fed group were 0.018, 0.036, 0.024 and 0.020 for 1st, 2nd, 3rd and 4th week, respectively. Calorie efficiency ratios for perilla oil and casein fed group were 0.028, 0.030, 0.024 and 0.020 for 1st, 2nd, 3rd and 4th week, respectively. 6. Serum cholesterol was 72.8mg% for the group of perilla oil (6gr) and casein(6gr) fed, and liver cholesterol was 460.5mg% for the same group. 7. Serum triglyceride was 130.7mg% for the group of perilla oil (6gr) and casein (6gr) fed. 8. Blood glucose was 40.34mg% for control-l and 96.4mg% for control-A, respectively. Blood glucose was 120.4mg% for group Band 1l0.7mg% for group B-1, respectively. 9. The degree of saturation/unsaturation for perilla oil (SFA/USFA) was 7.8/92.2 and nonessential fatty acid/essential fatty acid(NEFA/EFA) was 26.3/73.7. In this conditions, serum and liver cholesterol was lower than anyother conditions for this experimental period. 10. For the perilla oil fed group, serum cholesterol was 105.5mg% for pleic acid/linoleic acid(18.5/58.5) and 72.8mg% for linoleic acid/linolenic acid(15.2/58.5). In this group, triglyceride was 132.5mg% for oleic acid/linoleic acid and 130.5mg% for linoleic acid/linolenic acid. 11. There are positive correlation between serum cholesterol and saturated fatty acid $({\gamma}=0.78)$, and unessential fatty acid $({\gamma}=0.41)$. There are negative correlation between serum cholesterol and unsaturated fatty acid$({\gamma}=-0.78)$ and essential fatty acid$({\gamma}=0.77)$, respectively. 12. The range of most effective diet for serum cholesterol level lowering was nonessential fatty acid/essential fatty acid(26.3/73.7), saturated fatty acid/unsaturated fatty acid(7.8/92.2) and added oil (6gr)/added casein protein(6gr).

  • To investigate the effects of phosphate($P_2O_5$) fertilization levels on the dry matter(DM) yield, nutrition yield, and $P_2O_5$ efficiency of forages in mixed sward of alfalfa-grasses, a field experiment arranged by randomized complete block design with five treatments(0, 50, 100, 150 and 200kg $P_2O_5$/ha) was conducted at Livestock Experiment Station in Suwon, September, 1990 to Feb~ary, 1993. During two years, average DM yields of forages increased as $P_2O_5$ fertilization level was increased, but no significant difference was found between DM 10,754kg of $P_2O_5$ 150kg/ha and DM 10,845kg of $P_2O_5$ 200kg/ha. Cmde protein and energy productivity of forages increased as $P_2O_5$ fertilization level was increased, and the increasing degree was highest between $P_2O_5$ l00kg and 150kg/ha feriilization. With increasing $P_2O_5$ fertilization, mineral contents tended to increase in P, K and WCa+ Mg equivalent ratio, but not to be regular in the other elements. Efficiency of $P_2O_5$ was highest at $P_2O_5$ 150kg/ha fertilization, which produced DM 12.8kg. net energy lactation 75.4MJ. starch equivalent 6.lkStE, and total digestible nutrients 8.6kg per Ikg $P_2O_5$.

Characterization of Isocitrate Lyase from Micrococcus luteus (Micrococcus luteus에서 정제한 Isocitrate Lyase의 특성)

  • The isocitrate lyase extracted from Micrococcus luteus was purified 38.8 folds with the overall yield of 10.2%, by the ammonium sulfate fractionation, DEAE-cellulose, 1st Sephadex G-200 and 2nd Sephadex G-200 column chromatography. The purified enzyme showed to be a single protein band by polyacrylamide gel electrophoresis. The molecular weight of the purified enzyme was estimated 60,000 by the SDS-polyacry]amide gel electrophoresis. The apparent Michaelis constant, Km value for isocitrate was 0.95 mM. The optimum pH and temperature of the purified enzyme were pH 7.5 and $40^{\circ}C$, respectively. The enzyme was activated by $Mg^{2+}$ and inhibited by $Mn^{2+}$, $Ca^{2+}$, $Cu^{2+}$, $Zn^{2+}$ and $CO^{2+}$. In addition, the activity of isocitrate lyase was increased by glutathione and 2-mercaptocthanol at 5 mM and cysteine at I mM.

An Optimization of AAV-82Q-Delivered Rat Model of Huntington's Disease

  • Objective : No optimum genetic rat Huntington model both neuropathological using an adeno-associated virus (AAV-2) vector vector has been reported to date. We investigated whether direct infection of an AAV2 encoding a fragment of mutant huntingtin (AV2-82Q) into the rat striatum was useful for optimizing the Huntington rat model. Methods : We prepared ten unilateral models by injecting AAV2-82Q into the right striatum, as well as ten bilateral models. In each group, five rats were assigned to either the 2×1012 genome copies (GC)/mL of AAV2-82Q (×1, low dose) or 2×1013 GC/mL of AAV2-82Q (×10, high dose) injection model. Ten unilateral and ten bilateral models injected with AAV-empty were also prepared as control groups. We performed cylinder and stepping tests 2, 4, 6, and 8 weeks after injection, tested EM48 positive mutant huntingtin aggregates. Results : The high dose of unilateral and bilateral AAV2-82Q model showed a greater decrease in performance on the stepping and cylinder tests. We also observed more prominent EM48-positive mutant huntingtin aggregates in the medium spiny neurons of the high dose of AAV2-82Q injected group. Conclusion : Based on the results from the present study, high dose of AAV2-82Q is the optimum titer for establishing a Huntington rat model. Delivery of high dose of human AAV2-82Q resulted in the manifestation of Huntington behaviors and optimum expression of the huntingtin protein in vivo.

Effects of forage level and chromium-methionine chelate supplementation on performance, carcass characteristics and blood metabolites in Korean native (Hanwoo) steers

  • A feeding trial was carried out to determine the effects of chromium methionine (Cr-Met) chelate and forage level over two years, $1^{st}$ fattening and $2^{nd}$ fattening period on growth parameters, carcass characteristics and blood metabolites of 46 Korean native (Hanwoo, Bos Taurus, $BW=183{\pm}44kg$) steers. Treatments were: 1) Steers in the low forage (LF) group were fed diets that consisted of 60% concentrate and 40% forage; 2) Steers in the high forage (HF) group were fed diets that consisted of 40% concentrate and 60% forage. Following the $1^{st}$ fattening period, steers ($BW=480{\pm}37.6kg$) were randomly assigned to four treatment groups: LF (40 F plus no Cr-Met supplementation in the $2^{nd}$ fattening period), LFCM (40LF plus added 400 ppb of Cr-Met during the $2^{nd}$ fattening period), HF (60 F plus no added Cr-Met during the $2^{nd}$ fattening period) and HFCM (60 F plus added 400 ppb of Cr-Met in the $2^{nd}$ fattening period). Dry matter intake of the treatment diets did not differ during the raising and $1^{th}$ fattening period (P > 0.05). The ADG in the raising period showed no difference between the 40 F and 60 F groups (P > 0.05). Carcass characteristics including rib-eye area and meat yield index were higher in HF than the other treatment groups (P < 0.05). The HF group tended to show a higher (P = 0.08) marbling score than the LF group whereas the HFCM group showed a higher marbling score than the LFCM group (P < 0.05). HDL was higher and LDL lower in groups fed with Cr-Met than in other groups whereas glucose showed the lowest value in HF group (P < 0.05). Triglyceride (TG), Cholesterol, PUN and total protein (TP) were the same among all treatment groups (P > 0.05). The Insulin concentration in the blood was significantly higher for the HFCM group than for the LF, LFCM and HF groups (P < 0.05). It is concluded that supplementation of chromium-methionine chelate could improve meat quality in beef steers.

The Assessment of Management Practices on Foodservice , Clinical Nutrition Service in Hospital Foodservice Operations (병원 급식소의 급식 및 임상영양 서비스 업무 실태 조사)

  • The aim of this research was to examine the management practices related to foodservice.clinical nutrition service provided by hospital foodservice operations. A survey of 30 hospital food nutrition service departments was undertaken and detailed information was collected from each, including surveys of 176 dietitians and 30 foodservice managers. Statistical data analysis was completed using the SAS/win 6.11 package for descriptive analysis and t-test. The results of this study can be summarized as follows : Out of 30 hospitals, 73.3%(22) were directly operated and 26.7%(8) were under contract foodservice management. Licensed number of beds were 768.7, with an average length of 11.4 days. The general characteristics of the dietitians were that 49.4% were aged between 25-29, and 60.8% were ordinary dietitians. 76.7% had bachelor's degree, 15.9% with master's degree and 7.4% were college graduates. Most hospital dietitians had internship training and 35.2% took a training course of 6 to 12 months. The average space of kitchen was 452.52 $m^2$, with 133.63$m^2$ for modified diet space and 18.13$m^2$ for nutrition counselling room space. The total number of normal meals was 1,255.47, with 502.93 of modified meals. The average calorie of normal meals was 2,145.04kcal, with 91.9g of protein contained in normal meal. The total food waste was 351.40kg. An average monthly cost for disposing food waste was 745,171.67 won. 83.3% of the kitchens were on the 1st basement and only 66.7%(20) of 30 hospitals had its own nutrition counselling room. 80% used dishwashers and 66.7% had an exclusive elevator for delivering meals. 80.0% of hospitals used centralized delivery service. 90% had established an area for hygiene division, 70% used dry zone within the dishwashers for sterilization, 66.7% provided spoon and chopstick, and 100% used dish cover. As means of food waste treatment, most hospitals(56.7%) used animal feed by contracters, followed by means of collection by contracters(30%) and in-house high speed fermentation machines(13.3%). It was found that only 33.3% hospitals regularly checked temperatures of the meals given to patients. Total productivity index was 3.72(meals/hour) in average, with an average productivity index for normal diet of 5.41. Average productivity index for modified diet was 4.62. Productivity indices for patient meals and clinical nutrition were 5.01(meals/hour) and 1.12(cases/hour) respectively and hospitals under self-operated foodservice management received higher points on clinical nutrition productivity index(P<0.01) than hospitals under contract foodservice management.

  • The Fibroblast growth factors(FGFs) plays an important role in the control of osteogenesis during skeletal development. Especially, FGF-2 is a potent mesodermal inducer during embryogenesis and FGF receptors (FGFRs) messages are strongly expressed in developing bones. In this study, we investigated the effect of bFGF on osteopontin(OPN) gene expression in ST-2 cells and tried to elucidate the mechanism of its stimulatory effects. The obtain results were as follows; The treatment of bFGF(1ng/ml) upregulates OPN, fibronectin mRNA levels and downregulates type I collagen mRNA levels. But, there was no remarkable difference in alkaline phosphatase mRNA levels between two groups. The OPN gene expression increased in a dose-dependent manner up to 10ng/ml and OPN gene began to occur at around 3h with continuous increase up to 24h then decreased to basal level at 48h. 30 minutues pretreatment with cycloheximide (500ng/ml), a protein synthesis inhibitor, prior to addition bFGF resulted in blocking bFGF induced OPN expression. These results suggest that bFGF increased the level of OPN mRNA in a dose and time-dependent manner via the synthesis of certain transcriptional regulatory proteins.

  • We have completed a study to measure the contents of glucose, BUN, creatinine. LDH, and T-protein with respect to a fatigued condition in the bloods of rats which a constant swimming is loaded and to measure the maximun swimming time of mice The test has been carried out as a part of the basic study on the efficacy of B. E. P. (Biological Energy Projector) for emitting a light energy having a specific wavelength out of far-infrared rays. As a result. We have reached the following conclusions: 1. At testing of mice's maximun swimming time, all of B.E.P.(2. 4. 8. 24hrs) treated group have been increased in comparison with the control group, but only 24hrs-B.E.P. treated group significantly increased during 4 weeks. 2. The contents of glucose, BUN. creatinine, LDH, and T-protein measured immediately after the swimming of mice have been distinctly changed but not been significantly changed at their increase and decrease in comparison with the control group. 3. At 3rd day out of the swimming loading, the contents of glucose in the blood serum of the white rat have been distinctly increased in comparison with the control group. And 24hrs-B.E.P treated group surpassed 8hrs-B.E.P. treated group. 4. At 1st and 3rd day, the contents of creatinine in the blood serum of the white rat have been distinctly increased at B.E.P. (8, 24hrs) treated groups in comparison with the control group and have been recovered to the condition of the normal group. 5. After three days, the contents of BUN in the blood serum of the white rat have been significantly decreased in B.E.P.(8, 24hrs) treated groups at 3rd day in comparison with the control group and have been recovered to the condition of the normal group. 6. The contents of LDH in the blood serum of the white rat have been decreased in B.E.P.(8, 24hrs) treated groups at 3rd day in comparison with the control group, in particular 24hrs-B.E.P. treated group has been decreased distinctly than the normal group. 7. The contents of T-protein in the blood serum of the white rat have been distinctly increased in B.E.P. (8, 24hrs) treated groups at 3rd day in comparison with the control and normal group. As the above results, it has been proved that the execise of mice and the fatigue metabolism of rats were influenced by the light energy emitted the B.E.P., and it has been also proved that the external stimulation could be used as a preferable stimulative factor for the biological metabolism. If the clinical training and study are positively achieved, the B.E.P. would be used as curative means and preventive measures for helping human body.

Effects of Systematic Variation Application of Fe, Mn, Cu, and Zn on the Contents of N-compounds(Crude/Pure Protein) in Orchardgrass and White Clover (Fe, Mn, Cu 및 Zn의 Systematic Variation 시비가 Orchardgrass 및 White Clover의 질소화합물(조/순단백질) 함량에 미치는 영향)

  • This pot experiment was conducted to investigate the effects of systematic variation application of Fe, Mn, Cu, and Zn on forage performance of orchardgrass and white clover. The treatments of systematic variation were 0/100, 25/75, 50/50. 75/25, and $100/0\%$ in the Fe/Cu(trial-1). Mn/Zn(trial-2), and Fe+Cu/Mn+Zn(trial-3), respectively The treatments of Fe/Mn/Cu/Zn(trial-4) were composed of $70\%$ for main element and $10\%$ for other 3 elements, respectively. 1. The contents of N-compounds in forages tended to be in inverse proportion to the yields. In the Mn/Zn trial, the 0/l00 to white clover resulted in the relatively high contents of soluble N-compounds and low ratio of pure protein/soluble N-compounds in company with a severe yield decrease. 2. In the Fe+Cu/Mn+Zn trial, the 0/100 and 100/0 resulted in the somewhat high contents of N-compounds in white clover. It was likely to be caused by the concentration effect derived from yield decrease. In addition, the 100/0 resulted in the relatively high content of soluble N-compounds and low ratio of pure protein/soluble N-compounds. The protein synthesis in white clover was likely to be negatively influenced by the 100/0. 3. In the Fe/Mn/Cu/Zn trial, white clover showed the low contents of crude and pure protein at the 1st cut. It was likely to be caused by the unbalanced mutual ratios derived from the high application levels of each single element. 4. In white clover at the 5th cut, the 0/100 of Mn/Zn and 100/0 of Fe+Cu/Mn+Zn resulted in the relatively high content of K. It was likely to be caused by the concentration effect derived from yield decrease.
