Cloning, Expression and Hormonal Regulation of Steroidogenic Acute Regulatory Protein Gene in Buffalo Ovary

  • In mammalian ovary, steroidogenic acute regulatory (StAR) protein mediates the true rate-limiting step of transport of cholesterol from outer to inner mitochondrial membrane. Appropriate expression of StAR gene represents an indispensable component of steroidogenesis and its regulation has been found to be species specific. However, limited information is available regarding StAR gene expression during estrous cycle in buffalo ovary. In the present study, expression, localization and hormonal regulation of StAR mRNA were analyzed by semi-quantitative RT-PCR in buffalo ovary and partial cDNA was cloned. Total RNA was isolated from whole follicles of different sizes, granulosa cells from different size follicles and postovulatory structures like corpus luteum and Corpus albicans. Semi-quantitative RT-PCR analyses showed StAR mRNA expression in the postovulatory structure, corpus luteum. No StAR mRNA was detected in total RNA isolated from whole follicles of different size including the preovulatory follicle (>9 mm in diameter). However, granulosa cells isolated from preovulatory follicles showed the moderate expression of StAR mRNA. To assess the hormonal regulation of StAR mRNA, primary culture of buffalo granulosa cells were treated with FSH (100 ng/ml) alone or along with IGF-I (100 ng/ml) for 12 to 18 h. The abundance of StAR mRNA increased in cells treated with FSH alone or FSH with IGF-I. However, effect of FSH with IGF-I on mRNA expression was found highly significant (p<0.01). In conclusion, differential expression of StAR messages was observed during estrous cycle in buffalo ovary. Also, there was a synergistic action of IGF-I on FSH stimulation of StAR gene.

Effect of the Induced Molting with Low-Protein and Energy Diet on the Postmolt Performance and Egg Quality of Layers (저단백질, 저에너지 사료를 이용한 유도환우가 산란계의 생산성 및 난질에 미치는 영향)

  • This work was carried out to investigate the effect of the induced molting with low-protein and energy diet on the postmolt performance of layers. Eighty White Leghorn layers (61-old-wk) with over 80% egg production were used for 12 weeks in this work. Treatments were non-molt control (CO), fasting treatment for 10 days (FW), molt treatment with low-protein and low-energy molting diet for 3 weeks (LO), and molt treatment with standard molting diet for 3 weeks (ST) as 4 treatments (2 replications/treatment and 10 birds/replication). Body weight (BW) loss was 26.5% of initial BW at 2 week in FW treatment, and was 17.3% and 14.2% in both LO and ST treatments (P<0.05). Layers in molting treatments were fed with commercial layer diet after completion of molting since BW of layers was recovered for 7 weeks. Heart weight ratio was shown as 0.47, 0.43, 0.46 and 0.46% at CO, FW, LO and ST treatments, respectively, and liver weight ration of body weight was shown as 2.56, 1.30, 1.47 and 1.52%, respectively. Thus, those of molting treatments decreased compared to non-molt control (P<0.05). Oviduct weight ratio were shown as 3.95, 1.17, 1.54 and 1.67%, respectively, and similar with the results of liver (P<0.05). Feed intake decreased at LO and ST treatments during molting period and increased from the 5th week compared to control. Egg production decreased at 1 week in molt treatment and stopped at 2 week in FW, 3 week in LO, and 4 week in ST treatments. The birds started to lay egg at 4 week in FW and at 5 week in LO and ST treatments. Egg production was recovered until 50% at 6 week in FW and was recovered as 66.1 and 71.6% at 8 week in LO and ST treatment, respectively. Egg weights were similar among all treatments. Eggshell thicknesses were 0.41, 0.47, 0.46 and 0.46 mm at CO, FW, LO and ST treatment, respectively, but the higher in molt treatments than control (P<0.05). Eggshell breaking strength was 3.83 and 3.81 kg/$cm^2$ in FW and LO treatment, respectively, and high compared to control. However, eggshell breaking strengths were 3.54 and 3.78 kg/$cm^2$ and were not statistically different. Haugh units were 75.6, 81.1, 80.6 and 79.9 in 4 treatments and high in molt treatment. Finally, dietary low-protein and low-energy may induce molt as few effect on performance of layers.

Suppression of Human GD3 Synthase (hST8Sia I) Expression Induced by Retinoic Acid in Human Melanoma SK-MEL-2 Cells (흑색종세포주 SK-MEL-2에서 레티노이드에 의한 GD3합성효소(hST8Sia I)의 발현억제)

  • To elucidate the mechanism underlying the suppressive regulation of hST8Sia I expression in retinoic acid (RA)-induced SK-MEL-2 cells, we characterized the promoter region of the hST8Sia I gene. Functional analysis of the 5‘-flanking region of the hST8Sia I gene by the transient expression method showed that the -1146 to -646 region, which contains putative binding sites for transcription factors c-Ets-1, CREB, AP-1 and NF-kB, functions as the RA-repressive promoter in SK-MEL-2 cells. Site-directed mutagenesis and ChIP analyses indicated that the NF-kB binding site at -731 to -722 is crucial for the RA-induced repression of hST8Sia I in SK-MEL-2 cells. In addition, the transcriptional activity of hST8Sia I suppressed by RA in SK-MEL-2 cells was strongly inhibited by extracellular signal-regulated protein kinase (ERK) inhibitor U0126 and protein kinase C (PKC) inhibitor GO6976, as determined by RT-PCR and luciferase assay of hST8Sia I promoter containing the -1146 to -646 regions. These results suggest that RA markedly modulates transcriptional regulation of hST8Sia I gene expression through the PKC/ERK signal pathway in SK-MEL-2 cells.

Effect of herbal acupuncture with Ginseng Radix Extract at ST36 and GB39 on the growth of mice

  • Objectives: To investigate the effect of Ginseng herbal acupuncture at GB39 and ST36 on growth in mice. Methods: Mice were divided into four groups (n=10); G-ST36 group treated with Ginseng Radix herbal acupuncture at right ST36, G-GB39 group treated with the same at GB39, Control-ST36 group and Control-GB39 group injected with saline at right ST36 and GB39 respectively. The saline injection and herbal acupuncture were performed 3 times a week for 3 weeks. The length( total height, body length and tail length), weight and the protein efficiency ratio were measured as well as IGF-I level in serum Results: The weights of Ginseng herbal acupuncture groups were not significantly increased compared to the control groups but the protein efficiency ratios were significantly higher in Ginseng herbal acupuncture groups compared to the control groups. The increases of the total length, body length and tail length were significantly higher in Ginseng herbal acupuncture groups than the control groups. The serum IGF-I level of G-GB39 was significantly increased compared to control-GB39, not that of G-ST36 compared to control-ST36. Conclusions: Ginseng herbal acupuncture at GB39 and ST36 are both effective to promote growth in terms of height and weight. GB39 may be effective for growth by way of increasing IGF-I, and more closely related to growth in height than ST36.

  • The experiment was canied out to investigate the effects of cutting frequencies (3, 4 and 5 cuts per year) in relation to fertilization of mineral nitrogen on crude protein yield and the contents of crude protein and acid detergent fibre in orchardgms(Dacty1is glomerata L.). Tne results are as follows : 1. The content of crude protein increased according to the increase in cutting frequency and showed the highest in the 1st cut in all treatments. At 4 and 5 cutting frequencies per year, the content of crude protein increased in a large extent according to the increased fertilization of mineral nitrogen. However at 3 cutting frequency, the content of cmde protein appeared higher for no application of mineral nitrogen than for the treatments with fertilization. 2. 'Ihe content of acid detergent fibre(ADF) was considerably reduced at 4 and 5 cutting frequencies and in all treatments, the lowest were recorded in the 1st cut or in the last cut. At 3, 4 and 5 cutting frequencies, the highest ADF were shown in the 2nd cut, 3rd cut and 3rd cut respectively. However, the general trend was that the content of ADF was greatly reduced according to increased level of mineral nitrogen fertilization. 3. The yeild of cmde protein was the highest at 5 cutting frequency(l.2 ton DMIha) and it indicated that the yield of cmde protein increased in accordance to the increase in fertilization of mineral nitrogen and cutting frequency. 4.   • Ganglioside GD1a is specifically formed by the addition of sialic acid to ganglioside GM1a by ST3 ${\beta}$-galactoside ${\alpha}$-2,3-sialyltransferase 2 (ST3GAL2). Above all, GD1a are known to be related with the functional regulation of several growth factor receptors, including activation and dimerization of epidermal growth factor receptor (EGFR) in tumor cells. The activity of EGF and EGFR is known to be a very important factor for meiotic and cytoplasmic maturation during in vitro maturation (IVM) of mammalian oocytes. However, the role of gangliosides GD1a for EGFR-related signaling pathways in porcine oocyte is not yet clearly understood. Here, we investigated that the effect of ST3GAL2 as synthesizing enzyme GD1a for EGFR activation and phosphorylation during meiotic maturation. To investigate the expression of ST3GAL2 according to the EGF treatment (0, 10 and 50 ng/ml), we observed the patterns of ST3GAL2 genes expression by immunofluorescence staining in denuded oocyte (DO) and cumulus cell-oocyte-complex (COC) during IVM process (22 and 44 h), respectively. Expression levels of ST3GAL2 significantly decreased (p<0.01) in an EGF concentration (10 and 50 ng/ml) dependent manner. And fluorescence expression of ST3GAL2 increased (p<0.01) in the matured COCs for 44 h. Under high EGF concentration (50 ng/ml), ST3GAL2 protein levels was decreased (p<0.01), and their shown opposite expression pattern of phosphorylation-EGFR in COCs of 44 h. Phosphorylation of EGFR significantly increased (p<0.01) in matured COCs treated with GD1a for 44 h. In addition, ST3GAL2 protein levels significantly decreased (p<0.01) in GD1a ($10{\mu}M$) treated COCs without reference to EGF pre-treatment. These results suggest that treatment of exogenous ganglioside GD1a may play an important role such as EGF in EGFR-related activation and phosphorylation in porcine oocyte maturation of in vitro.

Effects of Nutritional Supplementation of Cereal-Vegetable Diet on the Growth of Rats (I) - Especially on Protein Metabolism - (곡류.야채식이의 영양소 보완이 흰쥐의 성장에 미치는 영향 (I) - 단백질 대사를 중심으로 -)

  • This study was designed to find out the nutritional defect of general Korean diet and the effective way of nutritional supplementation. Seventy weanling Sprague-Dawley male rats weighing 51.8$\pm$0.9g were blocked into ten groups and fed ten different diets ad libitum for eight weeks: Standard groups(st gp) was given 72% sugar-20% casein diet: Cereal-vegetable group(c-v gp) was fed cereal-vegetable diet(c-v diet) composed of rice, barley, soybean, spinach and cabbage: the other eight groups were fed c-v diets supplemented with casein, vitamin $B_{2}$, calcium, vitamin A, vitamin B2 and A, vitamin A and calcium, vitamin $B_{2}$ and calcium, vitamin A and $B_{2}$and calcium, respectively, on the basis of each nutrients content of standard diet. The results were as follows: 1. Food intakes and body weight gains in all the experimental groups were significantly lower than st gp. Among experimental groups, casein gp and vit B2 gp tended to gain more body weights than c-v gp. 2. Through all the experimental period, F.E.R., pp.E.R., and NDPcal% did nod show significant differences among all the experimental groups and st gp. 3. The weights of liver, kidney, and gastrocnemius were significantly lower in all the experimental groups as compared with st gp. But brain and sex organ weights did not show differences among all the groups. 4. Nitrogen contents of total carcass, liver, and gastrocnemius in all the experimental groups tended to be increased as compared with st gp, and among experimental groups, they tended to be increased by casein supplementation and decreased by ca supplementation. 5. Apparent nitrogen digestibility, urinary and fecal nitrogen excretion, the amount of nitrogen retained, and N.P.U. did not show any significant differences among all the groups. 6. Serum total protein concentrations did not show any significant differences among all the groups.

The Clinical Features of Spinal Leptomeningeal Dissemination from Malignant Gliomas

  • Objective : The incidence of leptomeningeal dissemination from malignant glioma is rare, so the clinical features of this are not well documented yet We attempted to determine the clinical features of leptomeningeal dissemination from malignant gliomas. Methods : We retrospectively analyzed 11 cases of leptomeningeal dissemination of malignant glioma, who were treated at our institution between 2006 and 2009. We investigated the clinical features of these patients by considering the following factors : tumor locations, the events of ventricular opening during surgery and the cerebrospinal fluid (CSF) profiles, including the cytology. Results : The group was composed of 9 males and 2 females. The histological diagnosis of their initial intracranial tumors were 4 primary glioblastoma, 3 anaplastic astrocytoma, 1 anaplastic oligoastrocytoma, 2 ganglioglioma and 1 pleomorphic xanthoastrocyotma with anaplastic features. The mean age of the patients at the time of the initial presentation was $42.8{\pm}10.3$ years. The mean time between surgery and the diagnosis of spinal dissemination was $12.3{\pm}7.9$ (3-28) months. The mean overall survival after dissemination was $2.7{\pm}1.3$ months. All our patients revealed a history of surgical opening of the ventricles. Elevated protein in the CSF was reported for eight patients who had their CSF profiles checked. Conclusion : We propose that in the malignant gliomas, the surgical opening of ventricles can cause the spinal leptomeningeal dissemination and the elevated protein content of CSF may be a candidate marker of leptomeningeal dissemination.

Clinical Significance of CLDN18.2 Expression in Metastatic Diffuse-Type Gastric Cancer

  • Purpose: Isoform 2 of tight junction protein claudin-18 (CLDN18.2) is a potential target for gastric cancer treatment. A treatment targeting CLDN18.2 has shown promising results in gastric cancer. We investigated the clinical significance of CLDN18.2 and other cell-adherens junction molecules (Rho GTPase-activating protein [RhoGAP] and E-cadherin) in metastatic diffuse-type gastric cancer (mDGC). Materials and Methods: We evaluated CLDN18.2, RhoGAP, and E-cadherin expression using two-plex immunofluorescence and quantitative data analysis of H-scores of 77 consecutive mDGC patients who received first-line platinum-based chemotherapy between March 2015 and February 2017. Results: CLDN18.2 and E-cadherin expression was significantly lower in patients with peritoneal metastasis (PM) than those without PM at the time of diagnosis (P=0.010 and 0.013, respectively), whereas it was significantly higher in patients who never developed PM from diagnosis to death than in those who did (P=0.001 and 0.003, respectively). Meanwhile, CLDN18.2 and E-cadherin expression levels were significantly higher in patients with bone metastasis than in those without bone metastasis (P=0.010 and 0.001, respectively). Moreover, we identified a positive correlation between the expression of CLDN18.2 and E-cadherin (P<0.001), RhoGAP and CLDN18.2 (P=0.004), and RhoGAP and E-cadherin (P=0.001). Conversely, CLDN18.2, RhoGAP, and E-cadherin expression was not associated with chemotherapy response and survival. Conclusions: CLDN18.2 expression was reduced in patients with PM but significantly intact in those with bone metastasis. Furthermore, CLDN18.2 expression was positively correlated with other adherens junction molecules, which is clinically associated with mDGC and PM pathogenesis.

Bone Healing in Ovariectomized-rabbit Calvarial Defect with Tricalcium Phosphate Coated with Recombinant Human Bone Morphogenetic Protein-2 Genetically Engineered in Escherichia coli

  • Purpose: This study compares the bone formation ability of tricalcium phosphate (TCP) with and without recombinant human bone morphogenetic protein-2 (rhBMP-2) and assesses TCP as a carrier of rhBMP-2. Methods: Bilateral round defects (diameter: 8.0 mm) were formed in the cranium of eight New Zealand white rabbits. The defects were grafted with TCP only (control group) or with rhBMP-2-coated TCP (experimental group). The animals were sacrificed at 1st week, 2nd week, 4th week, and 8th week postoperatively; two rabbits sacrificed each time. The skulls were harvested and subjected to radiographic and histological examination. Results: Radiologic evaluation showed faster bone remodeling in the experimental group than in the control group. Histologic evaluation (H&E, Masson's trichrome stain) showed rapid bone formation, remodeling and calcification in the 1st and 2nd week in the experimental group. Immunohistochemical evaluation showed higher expression rate of osteoprotegerin, receptor activator of nuclear factor ${\kappa}B$ ligand, and receptor activator of nuclear factor ${\kappa}B$ in the experimental group at the 1st and 2nd week than in the control group. Conclusion: rhBMP-2 coated TCP resulted in rapid bone formation, remodeling, and calcification due to rhBMP-2's osteogenic effect. TCP performed properly as a carrier for rhBMP-2. Thus, the use of an rhBMP-2 coating on TCP had a synergic effect on bone healing and, especially, bone remodeling and maturation.
