Menadione Sodium Bisulfite-Protected Tomato Leaves against Grey Mould via Antifungal Activity and Enhanced Plant Immunity

  • Tomato grey mould has been one of the destructive fungal diseases during tomato production. Ten mM of menadione sodium bisulfite (MSB) was applied to tomato plants for eco-friendly control of the grey mould. MSB-reduced tomato grey mould in the 3rd true leaves was prolonged at least 7 days prior to the fungal inoculation of two inoculum densities (2 × 104 and 2 × 105 conidia/ml) of Botrytis cinerea. Protection efficacy was significantly higher in the leaves inoculated with the lower disease pressure of conidial suspension compared to the higher one. MSB-pretreatment was not effective to arrest oxalic acid-triggered necrosis on tomato leaves. Plant cell death and hydrogen peroxide accumulation were restricted in necrotic lesions of the B. cinereainoculated leaves by the MSB-pretreatment. Decreased conidia number and germ-tube elongation of B. cinerea were found at 10 h, and mycelial growth was also impeded at 24 h on the MSB-pretreated leaves. MSB-mediated disease suppressions were found in cotyledons and different positions (1st to 5th) of true leaves inoculated with the lower conidial suspension, but only 1st to 3rd true leaves showed decreases in lesion sizes by the higher inoculum density. Increasing MSB-pretreatment times more efficiently decreased the lesion size by the higher disease pressure. MSB led to inducible expressions of defence-related genes SlPR1a, SlPR1b, SlPIN2, SlACO1, SlChi3, and SlChi9 in tomato leaves prior to B. cinerea infection. These results suggest that MSB pretreatment can be a promising alternative to chemical fungicides for environment-friendly management of tomato grey mould.

  • Xenopus 수정란의 동물극분리편에 IGF-1(Insulin-like Growth Factor-1)을 고농도로 처리해 주었을 때의 기관유도효과를 실험하였다. Activin A는 동물극 분리배양조직으로부터 다양한 기관을 분화시키며 이러한 효과는 처리 시간과 농도에 의존한다. 본 연구에서는 activin A 뿐만 아니라 IGF-1을 복합처리하여 특정 기관의 분화양상을 관찰하였다. Activin A는 100ng/ml 의 농도로, IGF-1은 0-500 ng/ml의 범위로 조합 처리하였다. 분리편은 정상배가 st. 43에 이를 때까지 배양하였으며, 이때 activin A를 100ng/ml의 고농도로 처리하면 조직이 파괴되는 것이 일반적이다. 그리고 신관의 발생에 대해서는 activin A와 retinoic acid의 복합처리가 매우 효과적인 방법으로 알려져 있으나, IGF-1의 첨가에 의해 신관을 비롯한 다른 조직들이 분화되었다. 또한, 눈의 분화는 activin A 1-100ng/m1와 IGF-1 500ng/ml의 농도범위에서 일어났다. Activin A의 저농도 (1ng/ml)처리에서는 혈구양세포가 분화하고 배양조직은 풍선처럼 부풀게되나 IGF-1의 첨가로 눈이 발생하게 되어 activin A상의 눈의 발생농도범위가 확대되었다.

Basic fibroblast growth factor가 osteopontin 유전자 발현에 미치는 영향 (Effect of basic fibroblast growth factor on osteopontin gene expression)

  • Fibroblast growth factor(FGF)는 세포의 성장과 이동, 분화와 생존과 관련된 여러가지 과정을 조절하는 것으로 알려져 있다. 이들의 prototype인 FGF-1과 FGF-2는 FGF receptors (FGFRs)를 통해서 세포내로 신호를 전달하는데, 두개봉합부의 조기융합을 보이는 craniosynostosis는 FGFRs중, 특히 FGFR-2의 point mutation에 의해서 야기된다. 최근 여러 보고에서 FGF/FGFR 신호전달은 골아세포의 분화에 있어 필수적인 역할을 한다고 하였으며, bFGF soaked bead를 쥐 두개골의 시상봉합부의 mid-mesenchyme과 osteogenic front부위에 적용하였을 때 osteopontin(OPN) 유전자의 발현을 유도한다고 하였다. 이에 본 연구에서는 ST-2 cell line를 이용한 in vitro실험에서 bFGF가 OPN 유전자 발현에 미치는 영향과 그 기전을 Northern blot analysis를 통해서 연구하고자 하였다. 1 ng/ml bFGF의 투여는 OPN, fibronectin 유전자 발현을 증가시켰으며, type I collagen 유전자 발현은 감소시켰으나 alkaline phosphatase 유전자 발현에는 영향을 미치지 않았다. OPN은 그 발현양상이 bFGF의 농도 증가에 따라 증가하는 양상으로 나타났으며, 시간결과에 따른 발현양상도 bFGF 투여 3시간째부터 bFGF를 투여한 군에서 대조군에 비해 높게 나타났으며 이는 24시간까지 시간의 경과에 따라 증가하는 양상을 보였다. 단백질 합성 억제제인 cycloheximide를 처리한 군에서는 OPN의 증가 양상을 보이지 않았는데 이는 bFGF에 의한 OPN유전자 발현이 새로운 전사조절 단백질 합성 등의 여러 단계를 거쳐서 일어남을 의미한다. 결론적으로 bFGF는 새로운 전사조절 단백질의 합성을 통해서 OPN 유전자 발현을 농도 및 시간 의존적으로 증가시킨다.

가공방법에 따른 치과용 합금의 기계적 특성 및 생물학적 안정성 평가 (Evaluation of Mechanical Characteristic and Biological Stability of Dental Alloys by the Manufacture Method)

  • 치과보철물의 소재는 생물 화학적, 물리적 안정성을 위하여 우수한 생체친화성이 요구된다. 그래서 본 연구에서는 새로운 기술인 레이저 선택용융 가공법으로 제작된 Co-Cr합금(SC시편), Ti합금(ST시편)을 일반적으로 사용되는 주조법으로 제작된 Co-Cr합금(CC시편), Ni-Cr합금(CN시편)과 비교분석하여 기계적 특성과 생물학적 안정성 평가를 실시하였다. 기계적 특성 평가를 위하여 인장시험을 통한 탄성계수를 측정하였다. 그리고 생물학적 안정성 평가를 위하여 유사체액(lactic acid-NaCl solution)을 이용한 용출물질 분석과 용출배지를 이용한 세포독성을 관찰하였다. 그 결과 레이저 선택용융 가공법으로 제작한 Co-Cr 합금이 주조법으로 제작한 Co-Cr합금보다 탄성계수가 높게 나타났다. 그리고 용출물질 분석과 세포독성 평가에서 레이저 선택용융 가공법으로 제작한 Co-Cr합금의 생물학적 안정성이 양호한 것으로 평가되었다. 결과에 의하여 레이저 선택용융 가공법으로 제작한 합금이 치과보철물 제작을 위한 합금으로 적용할 수 있는 것으로 판단된다.

Prednisolone acetate의 실험적 투여로 인한 말의 혈액학적 변화 (Hematologic Changes by Experimental Administration of Prednisolone Acetate in Horse)

  • The study was carried out to investigate the changes of total WBC, neutrophil, lymphocyte, monocite, eosinophil in the horses'blood of 3 groups which randomly assigned 3 heads to each group. In group A(daily therapy), each horse was iniected, intramuscularly, 100 mg of prednisolone acetate/day for 3 days, consecutively. In group B(alternate-day therapy), each horse was injected, 100 mg of prednisolone acetate/day 4 times on alternate days, intramuscularly. In control group, each horse was injected, intramuscularly, the equivalent volumes/day of saline solution fur 3 days, consecutively. The results were summarized as fellows: 1. Total WBC counts in group A rapidly increased until day 4th, which the counts reached the $maximum(13.10{\pm}0.13{\times}10^{3}/{\mu}l$)(p<0.05), and decreased into control group range until day loth. In group B rapidly increased until day 1st, and gradually increased until day 71% which the counts reached the maximum ($12f3{\pm} 0.20$)0<0.05), and decreased into control group range until day 16th. 2. Neutrophil counts in group A rapidly increased until day 4HL which the coun% reached the maxi um($10.85{\pm} 0.08$)0<0.05), and decreased into control group range until day loth. In group B rapidly increased until day 1st and slowly increased until day 7th, which the counts reached the maximum$(9.76{\pm}0.22)0<0.05)$, and decreased into control group range until day 16th. 3. Lymphocyte counts in group A and B rapidly decreased until day 4th, which the counts reached the minimum(2.14{\pm}0.12 and 2.09{\pm}0.20$, respectively) (p<0.05), and increased into control group range until day 10th. Group A and B, however, had no significant effects each other on Iymphocyte counts during the study. 4. Monocite and eosinophil counts in group A and B remained in the range of control group during the study. Eosinophil in group A and B slightly decreased on day 4th, significantly%<0.05), but we could not generally be reliable the result because of severe standard deviations. In conclusion, we fecund that group A had more rapidly increase and decrease than group B on hematologic counts of WBC, but group B had longer period than group A on the hematologic effects of WBC, and found not differences between daily therapy and alternate-day therapy.

냉동보관된 지방세포의 동종이식 (Allogenic Grafting of Cryopreserved Fat Cell)

  • Purpose: The most effective methods of harvesting, preparing, and injecting autologous fat grafts have been inconsistent and conflicting. With its limitation as resorption in fat grafting, handling various techniques affect adipocyte survival, and is crucial to optimizing its long-term survival. To improve graft survival, re-implantation of cryopreserved adipocytes was developed. In addition, adipocytes do not induce immune rejection in response to non-self lymphocytes in a mixed lymphocyte reaction. The purpose of this study is to analyze the changes in cryopreserved adipocytes so as to determine the most efficient long-term storage period, and to analyze the changes in cryopreserved allografted adipocytes so as to determine the efficacy of cryopreserved adipocytes allografting. Methods: Fat tissues were harvested from the inguinal and retroperitoneal fat pad of mice. After the centrifugation of the harvested fat tissues, they were disintegrated with collagenase. The adipocytes were obtained by centrifugation of the disintegrated fat tissues. The adipocytes were treated as follows: (1) They were examined for weight and then frozen at $-20^{\circ}C$(n=25). For four months, each five frozen samples were taken and examined for weight and histologic changes in the 1st week, the 1st month, the 2nd month, the 3rd month, and the 4th month, respectively. (2) The adipocytes were immediately frozen at $-20^{\circ}C$(n=125). For four months, five frozen samples were taken, and allografted in the same time period as above. Finally, for four months, five cryopreserved allografted adipocytes were taken and examined for histologic changes in the same time period as above. Results: (1) Significant weight changes and histologic findings with inflammatory and destructive changes were observed in the cryopreserved adipocytes in three months. (2) Significant fat necrotic changes in the histologic changes with Hematoxylin and eosin stain were observed in the cryopreserved allografted adipocytes since the first week, independent of the freezing period. Conclusion: The study results show that the adipocytes that were cryopreserved for more than three months underwent obvious weight reductions and necrotic changes, and the adipocytes that were allografted without freezing were viable for four months, but the cryopreserved allografted adipocytes had obvious necrotic changes since the first week regardless of the freezing period.

양혈장근건보탕(養血壯筋健步湯)과 전침의 병용치료가 손상된 척수신경 및 운동기능에 미치는 영향 (Effects of Yanghyuljanggeungunbo-tang(Yangxuezhuangjinjianbu-tang) and Electrical Acupuncture on the Spinal Nerve Injury and the Motor Function)

  • Objectives : The purpose of this study was to investigate the effects of Yanghyuljanggeungunbo-tang(Yangxuezhuangjinjianbu-tang, YGKT) and electrical acupuncture treatment in spinal cord injury(SCI)-induced rats. Methods : The subjects were divided into 5 groups ; Normal, Control-no treatment after SCI, Experimental I(Exp. I)-taken with YGKT 500 mg/kg $0.5m{\ell}$ daily after SCI. Experimental II(Exp. II)-taken with electrical acupuncture after SCI and Experimental III(Exp. III)-taken with YGKT 500 mg/kg $0.5m{\ell}$ and electrical acupuncture after SCI. After each operation, the present author observed cytological changes, the motor behavior recovery and nerve regeneration by analysis of the motor behavior tests, EMG, hematological(AST, ALT, WBC), histological and immunological changes. Rats were tested by Motor behavior test at 1st, 3rd, 7th, 14th and 21st day. Results : 1. All the experimental groups were improved compared with control group in the motor behavior tests including Tarlov test, Basso-Beattle-Bresnahan locomotor rating scale, modified inclined plane test, open field test, grid walk test and narrow beam test. Especially Exp. III was significantly improved among other groups. 2. In EMG test, H and M wave were significantly increased in Exp. III. 3. All the experimental groups were significantly decreased compared with control group in AST, ALT and WBC. 4. NGF, BDNF and Trk B of spinal cord gray matter in all the experimental groups were increased compared with control group. Especially, Exp. III was more effective. 5. In histological observations, muscle contraction and denaturation of gastrocnemius muscle of all the experimental groups were inhibited. Especially, those of Exp. III was more effective. On the observations of liver and kidney, cell atrophy and apoptosis of all the experimental groups were decreased compared with control group. Especially, those of Exp. III was more effective. Conclusions : It can be suggested that YGKT and electrical acupuncture may improve motor behavior, EMG, hematological, histological and immunological findings in SCI-induced rats. Especially, combination of these two treatments will be somewhat better in spinal nerve recovery and motor function improvement.

Upregulation of miR-23b Enhances the Autologous Therapeutic Potential for Degenerative Arthritis by Targeting PRKACB in Synovial Fluid-Derived Mesenchymal Stem Cells from Patients

  • The use of synovial fluid-derived mesenchymal stem cells (SFMSCs) obtained from patients with degenerative arthropathy may serve as an alternative therapeutic strategy in osteoarthritis (OA) and rheumatoid arthritis (RA). For treatment of OA and RA patients, autologous transplantation of differentiated MSCs has several beneficial effects for cartilage regeneration including immunomodulatory activity. In this study, we induced chondrogenic differentiation of SFMSCs by inhibiting protein kinase A (PKA) with a small molecule and microRNA (miRNA). Chondrogenic differentiation was confirmed by PCR and immunocytochemistry using probes specific for aggrecan, the major cartilaginous proteoglycan gene. Absorbance of alcian blue stain to detect chondrogenic differentiation was increased in H-89 and/or miRNA-23b-transfected cells. Furthermore, expression of matrix metalloproteinase (MMP)-9 and MMP-2 was decreased in treated1 cells. Therefore, differentiation of SFMSCs into chondrocytes through inhibition of PKA signaling may be a therapeutic option for OA or RA patients.

  • 늑연골세포는 초기 증식능력이 관절연골세포보다 우수하며, 또한 세포 채취에 용이하다는 장점이 있어, 관절연골의 대안으로 사용된다. FDA의 승인을 받은 합성고분자인 폴리락산-글리콜산 공중합체(PLGA)는 조직공학적 생체재료로 사용되고 있으나, 세포 부착률이 낮고, 염증반응을 야기시킨다고 보고되고 있다. 본 연구에서는 세포의 증식에 영향을 주며 염증반응이 감소된다고 보고된 탈미네랄골분(DBP)을 이용하여, 함량별 DBP/PLGA를 제조한 뒤, 세포를 파종하여 연구를 실시하였다. 세포의 부착 및 증식률을 측정하기 위하여 세포 파종한 후 MTT와 SEM 분석을 수행하였으며, DBP가 세포외 기질 형성에 미치는 영향을 확인하고자 글리코스아미노글라이칸(sGAG)과 콜라겐 함량을 측정하였다. 생체 내 외 환경에서 세포의 부착과 증식에 미치는 영향을 관찰하기 위해 연구를 진행하여, PLGA 지지체보다 DBP/PLGA 지지체가 세포의 성장과 증식에 영향을 주는 것을 확인하였으며, DBP의 함량이 증가할수록 효과가 좋은 것으로 나타났다.

Ganoderma Lucidum Pharmacopuncture for Teating Ethanol-induced Chronic Gastric Ulcers in Rats

  • Objectives: The stomach is a sensitive digestive organ that is susceptible to exogenous pathogens from the diet. In response to such pathogens, the stomach induces oxidative stress, which might be related to the development of both gastric organic disorders such as gastritis, gastric ulcers, and gastric cancer, and functional disorders such as functional dyspepsia. This study was accomplished to investigate the effect of Ganoderma lucidum pharmacopuncture (GLP) on chronic gastric ulcers in rats. Methods: The rats were divided into 4 groups of 8 animals each: the normal, the control, the normal saline (NP) and the GLP groups. In this study, the modified ethanol gastritis model was used. The rats were administrated 56% ethanol orally every other day. The dose of ethanol was 8 g/kg body weight. The normal group received the same amount of normal saline instead of ethanol. The NP and the GLP groups were treated with injection of saline and GLP respectively. The control group received no treatment. Two local acupoints CV12 (中脘) and ST36 (足三里) were used. All laboratory rats underwent treatment for 15 days. On last day, the rats were sacrificed and their stomachs were immediately excised. Results: Ulcers of the gastric mucosa appeared as elongated bands of hemorrhagic lesions parallel to the long axis of the stomach. In the NP and GLP groups, the injuries to the gastric mucosal injuries were not as severe as they were in the control group. Wound healings of the chronic gastric ulcers was promoted by using GLP and significant alterations of the indices in the gastric mucosa were observed. Such protection was demonstrated by gross appearance, histology and immunehistochemistry staining for Bcl-2-associated X (BAX), B-cell lymphoma 2 (Bcl-2) and Transforming growth factor-beta 1 (TGF-${\beta}1$). Conclusion: These results suggest that GLP at CV12 and ST36 can provide significant protection to the gastric mucosa against an ethanol induced chronic gastric ulcer.
